Ephrin signaling is involved in various morphogenetic events, such as axon guidance, hindbrain segmentation, and angiogenesis. We conducted a yeast two-hybrid screen using the intracellular domain (ICD) of EphrinB1 to gain biochemical insight into the function of the EphrinB1 ICD. We identified the transcriptional co-repressor xTLE1/Groucho as an EphrinB1 interacting protein. Whole-mount in situ hybridization of Xenopus embryos confirmed the co-localization of EphrinB1 and a Xenopus counterpart to TLE1, xTLE4, during various stages of development. The EphrinB1/xTLE4 interaction was confirmed by co-immunoprecipitation experiments. Further characterization of the interaction revealed that the carboxy-terminal PDZ binding motif of EphrinB1 and the SP domain of xTLE4 are required for binding. Additionally, phosphorylation of EphrinB1 by a constitutively activated fibroblast growth factor receptor resulted in loss of the interaction, suggesting that the interaction is modulated by tyrosine phosphorylation of the EphrinB1 ICD.
Monoclonal antibody (mAb) Tg786 against Toxoplasma gondii has been found to detect a 42-kDa rhoptry protein (ROP6) which showed protease activity and host cell binding characteristics after secretion. Using the mAb, a colony containing a 3'-UTR was probed in a T. gondii cDNA expression library. A full length cDNA sequence of the rhoptry protein was completed after 5'-RACE, which consisted of 1,908 bp with a 1,443 bp ORF. The deduced amino acid sequence of ROP6 consisted of a polypeptide of 480 amino acids without significant homology to any other known proteins. This sequence contains an amino terminal stop transfer sequence downstream of a short neutral sequence, hydrophilic middle sequence, and hydrophobic carboxy terminus. It is suggested that the ROP6 is inserted into the rhoptry membrane with both N- and C-termini.
Src-family kinases are critically involved in the control of cytoskeleton organization and in the generation of integrin-dependent signaling responses, inducing tyrosine phosphorylation of many signaling and cytoskeletal proteins. Activity of the Src family of tyrosine kinases is tightly controlled by inhibitory phosphorylation of a carboxy-terminal tyrosine residue, inducing an inactive conformation through binding with its SH2 domain. Dephosphorylation of C-ter tyrosine, as well as its deletion of substitution with phenylalanine in oncogenic Src kinases, leads to autophosphorylation at a tyrosine in the activation loop, thereby leading to enhanced Src activity. Beside this phophorylation/dephosphorylation circuitry, cysteine oxidation has been recently reported as a further mechanism of enzyme activation. Mounting evidence describes Src activation via its redox regulation as a key outcome in several circumstances, including growth factor and cytokines signaling, integrin-mediated cell adhesion and motility, membrane receptor cross-talk as well in cell transformation and tumor progression. Among the plethora of data involving Src kinase in physiological and pathophysiological processes, this review will give emphasis to the redox component of the regulation of this master kinase.
The maize lipoxgyenase-1 is a non-traditional dual positional specific enzyme and the reaction proceeds via enzyme-initiated catalysis. Bioinformatic analysis indicated that the maize lipoxygenase-1 is structurally more similar to soybean LOX1 than pea LOXN2 in that it has an additional external loop (residues 318-351) in the carboxy-terminal catalytic domain. We analyzed the dependence of product distribution on concentration of linoleic acid and monitored the formation of hydroperoxyoctadecadienoic acid as a function of enzyme concentration. Product distribution was strongly influenced by substrate concentration, such that kinetically-controlled regioisomers were enriched and thermodynamically-controlled regioisomers were depleted at high substrate concentration. Kinetic studies indicated that the formation of hydroperoxyoctadecadienoic acid saturated rapidly in an enzyme concentration-dependent manner, which implied that reactivation by reoxidation of inactive Fe(II) failed to occur. Our results support the previously proposed enzyme-initiated catalytic mechanism of the maize lipoxgyenase-1 and reveals that a substrate molecule serves as a hydrogen atom donor in its enzyme-initiated catalysis.
발광 박테리아인 Photobacterium species의 lux 오페론에서 발견된 riboflavin 생합성에 관여하는 유전자들(ribI,II,III,IV)의 기능을 조사하였다. 대장균에서 이들 유전자가 포함된 재조합 플라스미드를 발현시켰을 때 상당량의riboflavin이 합성되는 것을 확인하였으며, 또한 이들 유전자들(ribI,II,III,IV)의 기능을 riboflavin에 대하여 종속 영양체인 대장균 돌연변이주(BSV 11,18)를 이용한 유전학적인 방법과 생화학적 방법으로 분석한 결과, 이들은 각각 riboflavin synthase, 3,4-dihydroxy-2-butanone 4-phosphate (DHBP) synthase, lumazine synthase, GTP cyclohydrolase II활성도를 갖는 단백질을 코드하는 것으로 밝혀졌다. 이는Photobacterium species의 riboflavin 유전자 체계가 riboflavin 생합성에 관여하는 모든 5개의 유전자들이 한 오페론에 존재하는 Bacillus subtilis와 주요 riboflavin 유전자들이 분리되어 있는 대장균과는 다른, 중간적인 형태를 갖는다는 것을 나타낸다.
키네신-1은 모터 도메인이 있는 두 개의 중쇄(KHC 또는 KIF5)와 모터 도메인이 없는 두 개의 경쇄(KLC)로 구성된 이형사량체 단백질이다. KIF5에는 KIF5A, KIF5B 및 KIF5C의 세 가지 subtype이 있으며, 카르복실(C)-말단 영역을 제외하고는 아미노산 상동성이 높다. KIF5A는 세포 내에서 화물을 운반하며, KIF5A의 C-말단 영역에 결합하는 매개 단백질은 키네신-1과 화물 사이를 연결한다. 키네신-1의 세포내 수송을 조절하는 단백질은 아직 충분히 확인되지 않았다. 본 연구는 리소좀의 세포 내 수송에 관여하는 ADP-ribosylation GTPase-activating protein 1 (ArfGAP1)과 KIF5A와의 결합을 확인하였다. KIF5A는 ArfGAP1의 C-말단 영역에 결합하고, ArfGAP1은 KIF5A의 C-말단 영역에 결합하지만 KIF5B, KIF5C, 키네신 경쇄 1 (KLC1) 또는 KIF3A와는 결합하지 않았다. ArfGAP 도메인을 가진 다른 동질형인 SMAP1과는 결합하지 않았다. 세포에서 KIF5A는 ArfGAP1과 같은 위치에서 발현하며, KIF5A, KIF5B 및 KLC1와 같이 면역 침전하였다. 그러나, KIF3B와는 같이 면역 침전하지 않았다. 이러한 결과들은 키네신-1은 세포내 화물 수송에서 ArfGAP1에 의해 조절될 수 있음을 시사한다.

MBL 단백질은 세균, 바이러스, 곰팡이, 기생충 등의 탄수화합물 구조에 결합하여 식균 작용을 돕거나 보체경로를 활성화 시킨다. MBL 단백질은 C-말단이 탄수화물을 인식하는 도메인이며, 연결 목 부위와 콜라겐 부위로 구성되어 있다. 혈청 내의 MBL 농도가 낮으면 높은 빈도로 면역결핍현상이 관찰된다고 알려져 있다. MBL 단백질의 기능과 유전에 대해 많은 연구가 되어져 왔으나 아직 MBL 단백질 복합체 등에 대한 연구는 많이 이루어져 있지 않다. 따라서 MBL 연구에 필수적인 MBL cDNA 제조와 재조합 단백질의 합성, 그리고 재조합 단백질을 항원으로 사용하여 polyclonal antibody를 생산한 연구 결과를 보고하는 바이다. 본 연구결과로 획득한 MBL cDNA, 재조합 단백질과 anti-MBL 항체는 앞으로의 MBL 연구에 절대적으로 필요한 도구가 될 것으로 생각된다.
