MREIT of Postmortem Swine Legs using Carbon-hydrogel Electrodes

  • Magnetic resonance electrical impedance tomography(MREIT) has been suggested to produce cross-sectional conductivity images of an electrically conducting object such as the human body. In most previous studies, recessed electrodes have been used to inject imaging currents into the object. An MRI scanner was used to capture induced magnetic flux density data inside the object and a conductivity image reconstruction algorithm was applied to the data. This paper reports the performance of a thin and flexible carbon-hydrogel electrode that replaces the bulky and rigid recessed electrode in previous studies. The new carbon-hydrogel electrode produces a negligible amount of artifacts in MR and conductivity images and significantly simplifies the experimental procedure. We can fabricate the electrode in different shapes and sizes. Adding a layer of conductive adhesive, we can easily attach the electrode on an irregular surface with an excellent contact. Using a pair of carbon-hydrogel electrodes with a large contact area, we may inject an imaging current with increased amplitude primarily due to a reduced average current density underneath the electrodes. Before we apply the new electrode to a human subject, we evaluated its performance by conducting MREIT imaging experiments of five swine legs. Reconstructed conductivity images of the swine legs show a good contrast among different muscles and bones. We suggest a future study of human experiments using the carbon-hydrogel electrode following the guideline proposed in this paper.

Simple Electrochemical Immunosensor for the Determination of Rabbit IgG Using Osmium Redox Polymer Films

  • An amperometric immunosensor for the determination of rabbit IgG is proposed. The immunoassay utilizes a screen-printed carbon electrode on which osmium redox polymer is electrodeposited. This immunoassay detects 0.1 ng/ml of rabbit IgG, which is ${\sim}10^2$ fold higher than the most sensitive enzyme amplified amperometric immunoassay. The assay utilizes a screen-printed carbon electrode which was pre-coated by a co-electrodeposited film of an electron conducting redox hydrogel and a rabbit IgG. The rabbit IgG in the electron conducting film conjugates captures, when present, the anti-rabbit IgG. The captured anti-rabbit-IgG is labeled with horseradish peroxidase (HRP) which catalyzes the two-electron reduction of $H_2O_2$ to water. Because the redox hydrogel electrically connects HRP reaction centers to the electrode, completion of the sandwich converts the film from non-electrocatalytic to electro-catalytic for the reduction of $H_2O_2$ to $H_2O$ when the electrode is poised at 200 mV vs. Ag/AgCl.

  • In electrochemical glucose sensing, the enhancement of the sensitivity and the response time is essential in developing stable and reliable sensors, especially for continuous glucose monitoring. We developed a method to increase the sensitivity and to shorten the response time for the sensing upon the appropriate addition of single wall carbon nanotube onto the osmium polymer-based hydrogel electrode. Also, the background stabilization is dramatically enhanced.

Electrochemical Properties of Activated Carbon Capacitor Adopting a Proton-conducting Hydrogel Polymer Electrolyte (수소이온전도성 고분자 겔전해질을 적용한 활성탄소계 전기이중층 캐패시터의 전기화학적 특성)

  • An electric double-layer capacitor (ELDC) of activated carbon electrode is prepared using a proton-conducting hydrogel polymer electrolyte, which is composed of poly(vinyl alcohol), silicotungstic acid, $H_3PO_4$, and deionized water. A solid film by evaporating the hydrogel polymer electrolyte is also prepared for comparison. The hydrogel polymer electrolyte also acts as a separator with the thickness of about $80{\mu}m$ and the room-temperature ionic conductivity of $10^{-2}S\;cm^{-1}$. The EDLC containing the symmetric electrodes of activated carbon shows the specific capacitance of $58F\;g^{-1}$ at $100mV\;s^{-1}$ with a good cycle life, implying that the hydrogel polymer electrolyte is very promising for use in EDLCs.

  • Magnetic Resonance Electrical Impedance Tomography (MREIT) is a new bio-imaging modality providing cross-sectional conductivity images from measurements of internal magnetic flux densities produced by externally injected currents. Recent MREIT studies demonstrated successful conductivity image reconstructions of postmortem and in vivo canine brain. However, the whole head imaging was not achieved due to technical issues related with electrodes and noise in measured magnetic flux density data. In this study, we used a new carbon-hydrogel electrode with a large contact area and injected 30 mA imaging current through a canine head. Using a 3T MREIT system, we performed a postmortem canine experiment and produced high-resolution conductivity images of the entire canine head. Collecting magnetic flux density data inside the head subject to multiple injection currents, we reconstructed cross-sectional conductivity images using the harmonic $B_z$ algorithm. The conductivity images of the canine head show a good contrast not only inside the brain region including white and gray matter but also outside the brain region including the skull, temporalis muscle, mandible, lingualis proprius muscle, and masseter muscle.

Amperometric Detection of DNA by Electroreducation of O2 in an Enzyme-Amplified Two-Component Assay

  • The two-component type enzyme amplified amperometric DNA assay is described to use an ambient $O_2$ of the substrate of the DNA labeling enzyme. Although the assay detects DNA only at > 0.5M concentration, a concentration $\~10^6$ fold higher than the sandwich-type enzyme amplified amperometric DNA assay, it can be run with an always available substrate. The assay utilizes screen-printed carbon electrodes (SPEs) which were pre-coated by a co-electrodeposited film of an electron conducting redox hydrogel and a 37-base long single-stranded DNA sequence. The DNA in the electron conducting film hybridizes and captures, when present, the 37-base long detection-DNA, which is labeled with bilirubin oxidase (BOD), an enzyme catalyzing the four-electron reduction of $O_2$ to water. Because the redox hydrogel electrically connects the BOD reaction centers to the electrode, completion of the sandwich converts the film from non-electrocatalytic to electrocatalytic for the reduction of $O_2$ to water when the electrode is poised at 200 mV vs. Ag/hgCl. The advantage or the assay over the earlier reported sandwich type enzyme amplified amperometric DNA assay, in which the amplifying enzyme was horseradish peroxidase, is that it utilizes ambient $O_2$ instead of the less stable and naturally unavailable $H_2O_2$.

  • A hydrogel electrolyte consisting of 6 M KOH aqueous solution, potassium polyacrylate (PAAK, 3 wt.%), and a hydrophilic silica OX50 (1 wt.%) was prepared to use as an electrolyte medium coated on a Scimat separator of activated carbon supercapacitor. The silica particle distributed homogeneously on surface pores of the separator to increase ionic conductivity and electrochemical stability of the hydrogel electrolyte. The silica addition also involved superior specific capacitance even at higher scan rates due to decrease in interfacial resistance between hydrogel electrolyte and activated carbon electrode.

  • A hydrogel electrolyte consisting of potassium poly(acrylate) (PAAK) (3 wt%) in 6 M KOH aqueous solution is coated on poly(acrylonitrile) nonwoven separator to examine high-rate characteristics of activated carbon supercapacitor adopting the separator. The hydrogel is homogeneously coated on the surface pores of the nonwoven separator. The electrolyte uptake of the PAAK hydrogel maintains for 24 days higher than 230% and the coated separator shows slightly lower ionic conductivity ($2.9{\times}10^{-2}Scm^{-1}$) than that ($3.6{\times}10^{-2}Scm^{-1}$) of using 6 M KOH only. The activated carbon supercapacitor adopting the coated separator shows a specific capacitance higher than $27Fg^{-1}$ at $1000mVs^{-1}$ and a retention ratio higher than 97% after the 1000th cycle. This is due to strong interfacial contact of coated hydrogel electrolyte between the activated carbon electrode and the nonwoven separator.

  • A field effect transistor(FET) type dissolved carbon dioxide($pCO_{2}$) sensor with a double layer structure of hydrogel membrane and $CO_{2}$ gas permeable membrane was fabricated by utilizing a $H^{+}$ ion selective field effect transistor(pH-ISFET) with Ag/AgCl reference electrode as a base chip. Formation of hydrogel membrane with photo-crosslinkable PVA-SbQ or PVP-PVAc/photosensitizer system was not suitable with the photolithographic process. Furthermore, hydrogel membrane on pH-ISFET base chip could be fabricated by photolithographic method with the aid of N,N,N',N'-tetramethyl othylenediarnine(TED) as $O_{2}$ quencher without using polyester film as a $O_{2}$ blanket during UV irradiation process. Photosensitive urethane acrylate type oligomer was used as gas permeable membrane on top of hydrogel layer. The FET type $pCO_{2}$ sensor fabricated by photolithographic method showed good linearity (linear calibration curve) in the range of $10^{-3}{\sim}10^{0}\;mol/{\ell}$ of dissolved $CO_{2}$ in aqueous solution with high sensitivity.
