Ultrastructural Changes in the Mouse Liver Cell Treated with Ginseng Extract for Damaged Liver by Carbon Tetrachloride (사염화탄소(四鹽化炭素)에 의한 간상해시(肝傷害時) 인삼(人蔘)이 간세포(肝細胞)의 미세구조(微細構造)에 미치는 영향(影響))

  • Election microscopic investigation were conducted on the mature mouse (B.W. about 25g) liver which was treated with Ginseng water extract during three, six and nine days respectivelly after carbon tetracholride injection into abdominal cavity. The results obtained were as follows: The liver cells of control group treated with carbon tetrachloride alone were restored slowly. The liver cells of experimental group treated with Ginseng water extract after carbon tetrachloride injection, however, were shown the appearance of well-developed ${\gamma}-ER$ and Golgi complex, marked aggregation of glycogen particles, and a number of large lipid droplets which are attached markedly with glycogen particles as compared to the control group. As these findings, it could be suggested that Ginseng gives an activation to restore the damaged liver cells.

Protective Effects of Extracts of Diospyrus kaki Folium against Hepatotoxicity in Carbon Tetrachloride Intoxicated Rats (감잎 추출물이 사염화탄소로 유발된 흰쥐의 간손상에 미치는 효과)

  • This study was done to investigate the protective effects of Diospyrus kaki Folium on carbon tetrachloride intoxicated rats. Male Sprague-Dawley rats (200~230g) was used. Experimental groups were divided into normal group and $CCl_{4}$-treated group and then $CCl_{4}$-treated groups were divided into 7 groups: only $CCl_{4}$-treated group, methanol extract pretreated group, butanol fraction pretreated group and $H_{2}O$ fraction pretreated group, $CCl_{4}$ treated groups were injected with $CCl_{4}$ 0.6 mg/kg (ip). The activities of ananine aminotransferasein hexane, chloroform and $H_{2}O$ fraction pretreated groups were significantly decreased (p<0.05) compared to the only $CCl_{4}$-treated group and also activity of aspartate aminotransferase and hepatic malondialdehyde content in $H_{2}O$ fraction pretreated group were significantly decreased (p<0.05) compared to the only $CCl_{4}$-treated group. The activity of alkaline phosphatase in chloroform and $H_{2}O$ fraction pretreated groups were decreased compared to the only $CCl_{4}$-treated group but the difference was not statistically significant. The activity of ${\gamma}$-glutamyltranspeptidase in hexane, chloroform and ethylacetate fraction pretreated groups were significantly decreased (p<0.05) compared to the only $CCl_{4}$-treated group. The contents of cholesterol and triglyceride were not influenced in any fraction pretreated groups, but HDL-cholesterol content in hexane and chloroform fraction pretreated groups were significantly increased (p<0.05) compared to the only $CCl_{4}$-treated group. Therefore, these results demonstrate a possible hepatoprotective role of $H_{2}O$ fraction of Diospyrus kaki Folium methanol extract in carbon tetrachloride intoxicated rats.

The Pharmacokinetics of Nimodipine After Oral Administration in Rabbits with Hepatic Failure

  • The pharmacokinetics of nimodipine, following a single 16 mg/kg oral dose, was investigated in rabbits with hepatic failure induced by 0.5 mL/kg (mild), 1.0 mL/kg (moderate) and 2.0 mL/kg (severe) of carbon tetrachloride $(CCl_{4}$ : olive oil = 20 : 80, v/v). The plasma concentrations of nimodipine were determined by a high performance liquid chromatographic assay. The levels of sGOT and sGPT in rabbits with mild $(86.2{\pm}29.0\;and\;98.5{\pm}33.1\;unit/dL)$, moderate $(168.1{\pm}61.2\;and\;196.2{\pm}66.0\;unit/dL)$ and severe $(292.7{\pm}82.2\;and\;314.2{\pm}99.8\;unit/dL)$ hepatic failure were significantly increased compared to the control $(38.0{\pm}10.1\;and\;32.4{\pm}10.2\;unit/dL)$. The area under the plasma concentration-time curve (AUC) of nimodipine was significantly increased in mild $(131.7{\pm}28.1%)$, moderate $(168.8{\pm}32.8%)$ and severe $(204.6{\pm}58.3%)$ carbon tetrachloride-induced hepatic failure rabbits compared to the control (100%) rabbits. The volume of distribution $(V_{d})$ and the total body clearance $(CL_{t})$ of nimodipine were significantly decreased in all hepatic failure groups. The elimination rate constant $(K_{el})$ of nimodipine was significantly decreased in moderate and severe carbon tetrachloride-induced hepatic failure rabbits. There was a correlation between sGOT (y= 1.01x+241, r=0.993) or sGPT (y=0.92x +243, r=0.997) value and the AUC of nimodipine in the rabbits with hepatic failure. These findings suggest that the hepatic metabolism of nimodipine was inhibited by carbon tetrachloride-induced hepatic failure rabbits, resulting in the decrese in $V_{d}$ and $CL_{t}$ of nimodipine in the rabbits with mild, moderate and severe hepatic failure.

The Therapeutic Effects of Korean Red Ginseng on Carbon Tetrachloride- and Galactosamine-induced Hepatotoxicity in Rats (홍삼의 사염화탄소 및 갈락토사민 유발 간독성에 대한 치료효과)

  • In this study, we investgated the effect of Red Ginseng (KRG) on liver damage induced by carbon tetrachloride (CTC) and galactosamine (GalN) in rats using indicator enzymes such as serum alanine/aspartate aminotransferases, sorbital dehydrogenase, lactate dehydrogenase, and ${\gamma}$-glutamyltransferase. Treatment of KRG restored these enzyme activities to near normal level compared to CTC or GalN treatment alone. Treatment of KRG also enhanced hepatic microsomal enzyme system, malondialdehyde formation, and depletion of reduced glutathione content, which were reduced by CTC or GalN. We also found that the decreased activities of glutathione S-transferase and glutathine reductase but not ${\gamma}$-glutamycysteine synthetase after KRG treatment restored to normal level. These results indicate that KRG has potent therapeutic activity against CTC- and GalN-induced hepatotoxicity in rat.

Effects of Pretreated Korean Red Ginseng on Carbon Tetrachloride and Galactosamine-induced Hepatotoxicity in Rats (홍삼의 전처리에 의한 사염화탄소 및 갈락토사민 유발 간독성에 대한 보호효과)

  • In present study, we examined whether or not the pretreatment of Korean Red Ginseng (KRG) could protect hepatotoxicity induced by carbon tetrachloride (CCl$_4$) and D-galatosamine (GalN). For this study, we not only tested activity of various plasma enzymes (AST, ALT, SDH, LDH), which are used as indicators of liver disease, but also checked the change of liver components such as lipid, glutathione and cytochromes content, and several liver enzyme activity. Pretreatment of KRG for two weeks significantly reduced the elevated plasma enzyme activities induced by CCl$_4$ and GalN. Pretreatment of KRG also restored the hepatic enzymes, malonedialdehyde formation, and depletion of reduced glutathione content induced by CCl$_4$ and GalN to near normal level. However, ${\gamma}$-glutamylcysteine synthetase activity was lot affected by KRG. These results suggest that KRG shows the hepatoprotective effect by reducing lipid peroxidation, by reducing the activity of free radical generating enzymes, and by preserving the hepatic glutathione.

Hepatoprotective Effects of the Water Extract of Protaetia brevitarsis Larva Against Carbon Tetrachloride-Induced Liver Injury in Rats (사염화탄소에 의해 유도된 흰쥐의 간장해에 미치는 지잠의 보호효과)

  • Hepatoprotective effects of the water extract of Protaetia brevitarsis larva (PB) was investigated in carbon tetrachloride ($CCI_{4}$) treated male Sprague-Dawley rats. PB administration protected rats against ALT, AST and LDH elevations induced by $CCI_{4}$, as well as the severity of liver damage. PB recovered the decrease in serum level of high-density lipoprotein-cholesterol and the increase in serum level of low-density lipoprotein-cholesterol induced by $CCI_{4}$. In histopathological observation, massive fatty change and necrosis in the centrilobular area, degenerative change including pyknosis of nucleus and swelling of parenchymal cell induced by $CCI_{4}$ were clearly protected by PB. These histopathological findings paralleled with the serum biochemical results. The present results demonstrated that the water extract of PB may have the hepatoprotective effect against $CCI_{4}$-induced liver damage in vivo.

Influence of Thyroxine on the Hepatotoxicity of Carbon Tetrachloride ($CC1_4$의 간장독작용(肝臟毒作用)에 미치는 Thyroxine의 영향(影響))

  • Calvert et al. formulated the hypothesis that carbon tetrachloride ($CCl_4$) acted on the central nervous system to produce and intensify sympathetic discharge which resulted in anoxic necrosis of the liver. Recknagel suggested that the essential feature of $CCl_4$ hepatotoxicity depended on the cleavage of it to $CCl_3$(free radical) and the peroxidative decomposition of cytoplasmic membrane structural lipids. And there are many reports which show the increase of adrenergic activity in hyperthyroidism. In this paper, the influence of thyroxine on the hepatotexicity of carbon tetrachloride was investigated in mice. The results obtained were summarized as follows; 1) Hepatic total lipid and lipid peroxide contents were slightly decreased by L-sodium thyroxine injection(4mg/kg/day for 4days or 6days), but hepatic glycogen content was significantly decreased. 2) Hepatic total lipid and lipid peroxide contents and serum lactic dehydrogenase activity were significantly increased by $CCl_4$ (4 ml/kg single dose or triple dose: 4ml/kg/day for 3days), but hepatic glycogen content was significantly decreased. 3) The increase of hepatic total lipid and lipid peroxide contents and serum lactic dehydrogenase activity induced by $CCl_4$ were significantly inhitited by the pretreatment of thyroxine. 4) The decrease of hepatic glycogen induced by $CCl_4$ was not affected by the pretreatment of thyroxine.

  • Object : This study was carried out to examine the effect of Hwangryunhaedoktang on the liver and kidney of rats induced by carbon tetrachloride and ethanol. Methods : The histopathological changes were examined after preadministration of 100 and 200mg/100g Hwangryunhaedoktang. Results : The perivenular ballooning degeneration and infiltration of basophilic inflammatory cells were decreased in the experimental group when compared with the control group. No abnormalities were observed in the kidneys of both groups. But, interstitial nephritis was observed only in the experimental group. Conclusion : This study suggests that Hwangryunhaedoktang may be of benefit to the prevention of hepatotoxicity induced by carbon tetrachloride and ethanol, and it is more effective in high dosages.
