Suppressive Effects of Platycodon grandiflorum on the Progress of Carbon Tetrachloride-Induced Hepatic Fibrosis

  • The suppressive effects of Platycodi Radix (Changkil: CK), the root of Platycodon grandiflorum A. DC (Campanulaceae), on the progress of acute carbon tetrachloride $(CCl_4)$-induced hepatic fibrosis were investigated in the rat. CK significantly suppressed $(CCl_4)$-induced hepatic necrosis and inflammation, as determined by the serum enzymatic activities of alanine and aspartate aminotransferase and serum tumor necrosis factor-${\alpha}$ levels, in dose-dependent manners. In addition, the increased hepatic fibrosis after acute $(CCl_4)$ treatment was suppressed by the administration of CK. CK also significantly prevented the elevation of hepatic ${\alpha}$ 1(I) procollagen (type I collagen) mRNA and ${\alpha}$ -smooth muscle actin (${\alpha}$ -SMA) expressions in the liver of $(CCl_4)$-intoxicated rats and also suppressed the induction of ${\alpha}$ -SMA and type I collagen in cultured hepatic stellate cells, in dose-dependent manners. These results suggest that the suppressive effects of CK against the progress of acute $(CCl_4)$-induced hepatic fibrosis possibly involve mechanisms related to its ability to block both hepatic inflammation and the activation of hepatic stellate cells.

Effect of Solanum lyratum Extract on the Hepatotoxicity of Carbon Tetrachloride in Rats (백모등 엑스의 간기능개선 효과)

  • The studies were attempted to investigate the therapeutic effect of Solanum lyratum extract(SLE) on the liver damage induced by carbon tetrachloride in rats. Male rats (Sprangue-Dawley) were treated with subchronic concentration of $CCl_4$, which was 1 mg/kg of $CCl_4$ via per oral administration daily for three days. The level of serum cholesterol, transaminase, bilirubin and BSP were measured. Treatment of SLE significantly decreased the increment in serum transaminase activities, total cholesterol and enlargement of liver but serum bilirubin was not reduced. The protective effect was observed when SLE was given daily for 4 weeks after $CCl_4$ treated for 3 days. SLE administration to rats prevented the development of fatty change, necrosis, and fibrosis induced by $CCl_4$. The results suggest that SLE has appreciable therapeutic effect on $CCl_4$ induced hepatotoxicity.

The Effect of Electro-acupuncture at ST36 on Carbon Tetrachloride-induced Chronic Hepatic Damage in Rats (족삼리 전침자극이 Carbon Tetrachloride 장기투여로 유발된 흰쥐의 만성 간손상에 미치는 영향)

  • Objectives : The purpose of this study is to investigate the effect of electro-acupuncture(EA) at ST36 on $CCl_4{^-}$ induced chronic hepatic damage in rats. Methods : The authors performed several experimental items, including measurements of liver index, hematological analysis for WBC, lymphocytes, neutrophils, monocytes, hemoglobin, and biochemical assays for ALT, AST, ALP in serum, and histological analysis of liver tissue. Results : Liver index(liver weight/body weight), the levels of WBC, Neutrophils and Monocytes in blood, the levels of ALT, AST and ALP in serum which were increased by $CCl_4{^-}$ intoxication were reduced significantly by EA at ST36. The levels of lymphocyte and hemoglobin in blood which were decreased by $CCl_4{^-}$ intoxication were significantly increased by EA at ST36. Conclusions : EA at ST36 has a hepatoprotective effect on chronic hepatic damage in $CCl_4{^-}$ intoxicated rats.

Effect of Phenobarbital Pretreatment on the Hepatotoxicity of Carbon Tetrachloride in Rat (Phenobarbital 전처치가 사염화탄소 급성중독 흰쥐 간세포의 구조에 미치는 영향)

  • The purpose of this study was to evaluate the influence of phenobarbital(PB) on hepatotoxic effect of carbon tetrachloride($CCl_4$) which induces centrilobular necrosis in liver. Rats were injected intraperitoneally $CCl_4$ dissolved in olive oil by a dose of 0.4mg/kg. For change related to PB pretreatment, rats were injected $CCl_4$ 0.4mg/kg after PB pretreatment. The liver samples were taken in 6, 12, 24, 48, 72, and 120 hours after $CCl_4$ and/or FB injection. Extracted liver tissue was examined with light and electron microscopes. The results were sumarized as follows : 1. Light microscopic findings : In $CCl_4$ group, centrilobular necrosis developed from 6 hours after injection, was the most severe in 48 hours, and recovered after 72 hours. In addition to necrosis, fatty change and pale cell change were accompanied. In PB-$CCl_4$ group, necrosis occurred from 6 hours after $CCl_4$ injection and continued to 72 hours, and the degree of necrosis was more severe than that of $CCl_4$ group and pale cell change was decreased. 2. Electron microscopic findings: In $CCl_4$ group, the early principal change was clumping and vesicular dilatation of endoplasmic reticulum. In PB-$CCl_4$ group, the degenerative change of endoplasmic reticulum was aggrevated and the mitochondria also revealed severe degenerative change. According to the results, it was revealed that $CCl_4$ hepatotoxicity primarily began with the damage of endoplasimic reticulum, then damage of other cell organelles and cell necrosis followed, and these cytotoxic effects were aggrevated by PB pretreatment.

Thermal Degradation Characteristics of Carbon Tetrachloride in Excess Hydrogen Atmosphere (과잉수소 반응조건하에서 사염화탄소의 고온 분해반응 특성 연구)

  • pure compound chloromethanes; methyl chloride, methylene chloride, chloroform and The carbon tetrachloride were used as a model of chlorocarbon system with Cl/H ratio to investigate thermal stability and hydrodechlorination process of carbon tetrachloride under excess hydrogen atmosphere. The parent thermal stability on basis of temperature required for 99% destruction at 1 second no was evaluated as $875^{\circ}C$ for $CH_3Cl$, $780^{\circ}C$ for $CH_2Cl_2$, $675^{\circ}C$ for $CHCl_3$ and $635^{\circ}C$ for $CCl_4$. Chloroform was thermally less stable than $CCl_4$ at fairly low temperatures $(<570^{\circ}C).$ The lion of $CCl_4$ became more sensitive to increasing temperature, and $CCl_4$ was degraded CHCl3 at above $570^{\circ}C.$ The number and quantity of chlorinated products decreases with increasing temperature for the Product distribution of $CCl_4$ decomposition reaction system. Formation of non-chlorinated hydrocarbons such as $CH_4$, $C_2H_4$ and C_2H_6$ increased as the temperature rise and particularly small amount of methyl chloride was observed above $850^{\circ}C$ in $CC1_4$/$H_2$ reaction system. The less chlorinated products are more stable, with methyl chloride the most stable chlorocarbon in this reaction system.

Protective Effects of Extract of Puerariae radix on Hepatic injury Induced by Carbon Tetrachloride In Rat. (갈근 추출물이 사염화탄소로 유발된 흰쥐의 간 손상에 대한 보호효과)

  • This study was done to investigate the protective effects of ethanol extract puerariae radix(Pr) on carbon tetrachloride($CCl_4$) intoxicated rats. Male sprague Dawley rats(200~210g)was used. experimental groups were divided into normal group, $CCl_4$-control group, and ethanol extract $CCl_4$-treated group. $CCl_4$-treated groups were injected with $CCl_4$ 0.6mg/kg.b.w(i.p). The activities of Alanine aminotransferase(ALT), Aspartate aminotransferase(AST), Alkaline phosphatase(ALP), Glutamyltranspeptidase(${gamma}$-GT), Lactate dehydrogenase(LDH) in extract pretrated group was significantly decreased(p<0.05) compared to the $CCl_4$-control group. The contents of triglyceride, cholesterol and lipid peroxide were significantly decreased(p<0.05). whereas content of HDL-choresterol was significantly increased.(p<0.05). In addition, activities of hepatic catalase(CAT), glutathione peroxidase(GSH-Px) in the extract pretreated rats were significantly decreased(p<0.05) compared to the $CCl_4$-control group. but the content of glutathione(GSH) was significantly increased(p<0.05). These results suggest that extract of puerariae radix(Pr) has hepatoprotective effect in the $CCl_4$-intoxicated rats.

Solubilization by $\beta$-Cyclodextrin: A Fluorescence Quenching Study

  • Solubility of carbon tetrachloride ($CCl_4$) in water increases appreciably in presence of $\beta$-cyclodextrin ($\beta$CD). $CCl_4$ is a very good quencher of 1-naphthol (1ROH) fluorescence. By studying the quenching of fluorescence of 1ROH included in $\beta$CD cavity, it was found that there is an increase in the availability of $CCl_4$ around $\beta$CD in the aqueous medium. This could help to rationalize the enhanced solubility of $CCl_4$.

Protective Effects of Extracts of Hovenia dulcis Thunb on Hepatotoxicity in Carbon Tetrachloride Intoxicated Rats (지구자나무 추출물이 사염화탄소로 유발된 흰쥐의 간손상에 미치는 영향)

  • This study was performed to investigate the protective effects of Hovenia dulcis Thunb on hepatotoxicity in carbon tetrachloride-intoxicated rats. Male Sprague-Dawley rats (220~240 g) were used as experimental groups, which were divided into 7 groups; Control group, $CCl_4$-treated group, hexane fraction pretreated and $CCl_4$-treated group, chloroform fraction pretreated and $CCl_4$-treated group, ethylacetate fraction pretreated and $CCl_4$-treated group, butanol fraction pretreated and $CCl_4$-treated group, $H_2O$ fraction pretreated and $CCl_4$-treated group. After 6 days, the activities of aminotransferase, contents of cholesterol, TG and hepatic lipid peroxide content in chloroform fraction pretreated and $CCl_4$-treated group were significantly decreased (p<0.05) compared to the only $CCl_4$-treated group. The content of glutathione and activities of GST in chloroform fraction pretreated and $CCl_4$-treated group were also significantly increased (p<0.05) compared to the only $CCl_4$-treated group. In addition, activities of SOD, catalase and GSH-Px in chloroform fraction pretreated and $CCl_4$-treated group were significantly decreased (p<0.05) compared to the only $CCl_4$-treated group. These results indicated that the chloroform fraction of Hovenia dulcis Thunb methanol extract showed hepatoprotective effect in carbon tetrachloride-intoxicated rats.

Effect of a Water Soluble Dimethyl Dimethoxy Biphenylate Derivative on the Carbon Tetrachloride Induced Hepatotoxicity in Rats (수용성 Dimethyl Dimethoxy Biphenylate 유도체의 간염 치료 효과)

  • A water-soluble DDB derivative (Bis{2-(methylamino)ethyl}-4,4-dimethoxy-5,5',6,6'-dimethylenedioxy-biphenyl-2,2'-dicarboxylate, DDB-S) was synthesized and its therapeutic effects on the liver damage induced by carbon tetrachloride in rats were evaluated. Oral administration of DDB-S reduced the aspartate aminotransferase(AST) and alanine aminotransferase(ALT) activities and increased total protein and albumin contents in the serum of the carbon tetrachloride intoxicated rat. Therapeutic effects of DDB-S by intravenous injection was also investigated using carbon tetrachloride intoxicated rats. Histological studies showed that IV injection of DDB-S had improved the typical necrosis around centrilobular area in liver tissue due to the carbon tetrachloride intoxication and also prevented the elevation of liver weigh/body weight ratio. IV administration of DDB-S to $CCl_4-treated$ rats significantly decreased AST & ALT activities and also prevented the decrease of aniline hydroxylation activity of the liver. These results indicate that i.v. administration of DDB-S is very effective in recovering the liver function in $CCl_4-treated$ rats.

Inhibition Effects of Zanthoxylum schinifolium and Its Active Principle on Lipid Peroxidation and Liver Damage in Carbon Tetrachloride-treated Mice (산초 및 그 활성성분이 사염화탄소를 부여한 Mouse에 있어서 지질과산화 및 간손상 억제에 미치는 영향)

  • The present study was undertaken to investigate the inhibition effects of sancho(Zanthoxylum schinifolium) on lipid peroxidation and liver damage in carbon tetrachloride tetracloride-treated mice. Mice aged 3 weeks old were fed diets containing either dry sancho powder, methanol extract or hyperoside isolated from sancho at the various levels for 2 weeks(sancho, methanol extract from sancho) or 3 days (hyperoside) before $CCl_{4}$ treatments. Seed coats of sancho added to diet with the levels from 1.25 to 5.0% significantly reduced(p<0.001) thiobarbituric acid reactive substances(TBARS) formation in liver and serum of mice treated with carbon tetrachloride. $CCl_{4}$ treatments significantly increased (p<0.001) in liver TBARS comparing with those of the untreated control, while methanol extract from the seed coat of sancho added to diet with the levels from 0.5 to 1.0% significantly reduced (p<0.01) to 20mg per kg of body weight showed significant reduction(p<0.01, p<0.001) of liver TBARS in mice treated with $CCl_{4}$. $CCl_{4}$ treatments significantly increased(p<0.05) in serum TBARS comparing with those the levels from 0.5 to 1.0% significantly reduced(p<0.01) serum TBARS in mice treated with $CCl_{4}$, and administration of hyperoside with the levels from 10 to 20mg per kg of body weight showed significant reduction(p<0.01, p<0.001) of serum TBARS in mice treated with $CCl_{4}$. $CCl_{4}$ significantly increased(p<0.001) in serum AST activities comparing with those of the untreated control, while either addition(0.5, 1.0%) of methanol extract from sancho or administration(10, 20mg/kg) of hyperoside significantly reduced(p<0.001, p<0.05) serum AST activities in $CCl_{4}$-treated mice. $CCl_{4}$ treatments showed significant increases(p<0.001) in serum ALT activities comparing with those of the untreated control, while either addition(0.5, 1.0%) of methanol extract from sancho or administration (10, 20mg/kg) of hyperoside showed moderate decreases(p<0.01, p<0.001) serum ALT activities in $CCl_{4}$-treated mice.
