Korean red ginseng, the processed root of Panax ginseng Meyer, has been frequently used for various therapeutic purposes in oriental medicine. The present study investigated the possible effect of Korean red ginseng extract (RGE) for the treatment of liver fibrosis in mice injected with carbon tetrachloride ($CCl_4$) for 4 wk. Liver injuries were assessed by blood biochemistry and histopathology in mice treated with $CCl_4$ alone or $CCl_4$+ RGE (30, 100, and 300 mg/kg). Concomitant treatment with RGE and $CCl_4$ (three times/wk for 4 wk) effectively inhibited liver fibrosis as evidenced by decreases in plasma alanine and aspartate aminotransferases, as well as by the percentages of degenerative regions, numbers of degenerative hepatocytes, and collagen accumulation in hepatic parenchyma. Treatment with $CCl_4$ for 4 wk increased mRNA levels of transforming growth factor ${\beta}1$ and plasminogen activator inhibitor 1 in fibrogenic liver, whereas RGE (30, 100, and 300 mg/kg) significantly blocked the induction of fibrogenic genes by $CCl_4$. Similarly, RGE also prevented transforming growth factor ${\beta}1$-mediated induction of fibrogenic genes in human hepatic stellate cell lines. More importantly, RGE markedly reduced the number of ${\alpha}$-smooth muscle actin-positive cells in liver tissue. This study implies that RGE efficaciously protects against the liver fibrosis induced by chronic $CCl_4$ treatment, and may therefore have potential to treat liver disease.

The present study was carried out to investigate the pretreatment of effects of water extract obtained from Eucommia ulmoides leaf (ELE) on hepatotoxicity of carbon tetrachloride ($CCl_4$) -treated rats. Thirty two healthy male Sprague-Dawley rats were divided into four groups, i.e. normal group (N), $CCl_4$-treated group (T), ELE-treated group (E), and ELE-treated group after injection of $CCl_4$(TE). The administration of $CCl_4$increased the activities of aspartate aminotransferase, alanine aminotransferase and lactate dehydrogenase in serum, but their activities were significantly decreased by the addition of ELE. Alkaline phosphatase activity in serum was significantly decreased in $CCl_4$-treated group, while the activity was not decreased by the extracts. Higher levels of HDL cholesterol was found in E and TE group which showed lower levels of total and LDL-cholesterol. Histopathologic findings by light microscopic examination showed fatty change, ballooning degeneration, acidophilic degeneration, spotty necrosis and zonal necrosis in $CCl_4$treated liver tissue. But the degree of liver damage was not identified in liver tissue of the TE group. These results indicated that Eucommia ulmoides leaf water extracts led migtigation of liver demage induced with $CCl_4$.
The purpose of this study was to evaluate the influence of high does carbon tetrachloride($CCl_4$) on the hepatotoxic effect of dimethylnitrosamine(DMN) which induces acute hemorrhagic necrosis in liver. Rats were injected intraperitoneally DMN dissolved in physiologic saline by a dose of 40mg/kg. For changes related to $CCl_4$ pretreatment, rats were injected intraperitoneally $CCl_4$ dissolved in olive oil by a dose of 0.4mg/kg, and then injected DMN. The livers were extracted from the rats 3, 6, 12, 24, 48, 72, and 120 hours after $CCl_4$ and/or DMN injection. Liver tissues were examined with light and electron microscopes. The results were summarized as follows ; Light microscopic findings : Severe centrilobular hemorrhagic necrosis developed from 12 hours after injection of DMN and continued to 120 hours. On injection of DMN after $CCl_4$ pretreatment, Massive necrosis occurred early. But active regenerative changes were produced in 24 hours. In 120 hours, the liver recovered in almost normal appearance. The degree of necrosis in pretreated group was similar to that in DMN injection only, and the time of recovery was faster in preteated group. Electron microscopic findings ; The early change was mainly disorganization of RER in DMN injection, and clumping and vesicular dilatation of ER in injection of $CCl_4$. In pretreatment group the early change was similar in appearance with $CCl_4$ group, but severer in degree. According to the results, it was revealed that acute toxic effect of DMN was recovered more rapidly in pretreatment group. Thus it was suggested that $CCl_4$ had protective effect in DMN hepatotoxicity.
This study aimed to evaluate the protective effect of cordycepin-increased Cordyceps militaris strain on carbon tetrachloride ($CCl_4$)-induced hepatotoxicity and oxidative stress in rats. Male Sprague-Dawley rats were randomly divided into five groups (n=6) based on six dietary categories: normal (N), $CCl_4$ control (C), $CCl_4$ plus Paecilomyces japonica (CPJ) (3%, w/w), $CCl_4$ plus C. militaris (CCM) (3%, w/w), and $CCl_4$ plus cordycepin-increased C. militaris ($CCM{\alpha}$) (3%, w/w). The activities of the liver marker enzymes ALT, AST, and LDH and the levels of lipid peroxidation were increased in the $CCl_4$-treated groups, but these parameters were significantly decreased in the $CCM{\alpha}$ group. The TBARS content in the liver homogenate, microsome, and mitochondrial fractions of the C group was significantly elevated compared with the N group. However, in the $CCl_4$-treated groups, $CCM{\alpha}$ group was significantly lowered in the TBARS levels of hepatic homogenate and microsomal fractions. The C group showed a significant decrease in the levels of plasma and hepatic glutathione, whereas they were significantly increased in the $CCM{\alpha}$ group. Accordingly, cordycepin-increased C. militaris may be an ideal animal model for studying hepatoprotective effects.
Object : This study was carried out to examine the effect of Hwangryunhaedoktang on the liver and kidney of rats induced by carbon tetrachloride and ethanol. Methods : The histopathological changes were examined after preadministration of 100 and 200mg/100g Hwangryunhaedoktang. Results : The perivenular ballooning degeneration and infiltration of basophilic inflammatory cells were decreased in the experimental group when compared with the control group. No abnormalities were observed in the kidneys of both groups. But, interstitial nephritis was observed only in the experimental group. Conclusion : This study suggests that Hwangryunhaedoktang may be of benefit to the prevention of hepatotoxicity induced by carbon tetrachloride and ethanol, and it is more effective in high dosages.
In the present study, Thespesia populnea (Malvaceae) bark was extracted with methanol and water. The extracts were vacuum dried to yield the respective methanol (MET) and aqueous extract (AET). The extracts were evaluated for hepatoprotective activity against carbon tetrachloride $(CCl_{4})$ induced liver damage at 2 dose levels (250 and 500 mg/kg). The biochemical parameters observed in serum were total bilirubin, alkaline phosphatase (ALP), serum glutamate oxaloacetate transaminase (SGOT), serum glutamate pyruvate transaminase (SGPT) levels and total protein. Aspartate transaminase (AST), alanine transaminase (ALT) and total protein levels in liver were also evaluated. Histopathological study on the liver tissue was also performed. The extracts exhibited dose dependent reduction in total bilirubin, ALP SGOT, SGPT, AST, ALT and increase in total protein (serum and liver) levels. The extracts also exhibited only mild hepatocytic damage compared to the $CCl_{4}$ Treated group. MET was found to exhibit higher hepatoprotection than AET.

The protective effect of Bifidobacterium breve K-110, K-111 and B. infantis K-525 on carbon tetrachloride induced hepatotoxicity in rats was investigated. Among them, B. infantis K-525 had the most potent hepatoprotective activity. It reduced serum aspartate aminotransferase and alanine aminotransferase levels to 51% and 80% of the $CCl_{4}-treated$ groups, respectively. In rat liver homogenate intoxificated with $CCl_{4}$, B. infantis K-525 inhibited in vitro as well as in vivo lipid peroxidation more than the other Bifidobacteria.
Hepatoprotective activity of methanol extract of Angelica tenuissima Nakai on the $CCl_4$-induced hepatotoxicity was investigated. To elucidate the hepatoprotective activity and free radical scavenging effect, we examined alanine aminotransferase (ALT), aspartate aminotransferase (AST), albumin, total protein, cholesterol, malondialdehyde (MDA) levels in serum and activities of superoxide dismutase (SOD), catalase (CAT) in hepatic tissue as compared with those of carbon tetrachloride-induced rats. The action mechanism also has been estimated by quantative analysis of cytochrome P450 (CYP), NADPH-CYP reductase for phase I metabolism and glutathion (GSH), glutathion S-transferase (GST) level for phase II metabolsim. Treatment of Angelica tenuissima methanol extract significantly lowered the levels of alanine aminotransferase and aspartate aminotransferase. In addition, the levels of cholesterol, triglyceride, MDA, CAT were decreased, and SOD was activated. This result indicates that the hepatoprotective effect of Angelica tenuissima methanol extract on the CCl4-induced hepatotoxicity would be originated from reduction of the NADPH-CYP reductase, GSH and the enhancement of the activities of GST, CYP.
This study was to investigate the preventive effects in anti-oxidation of Ulva lactuca extract fractions (ULEF) against $CCl_4$ toxification in liver total homogenate and mitochondrial fraction of ULEF-pretreated and carbon tetrachloride $(CCl_4)-posttreated$ rats. ULEF was intraperitoneally administered into rats at dose of 1 ml/kg for14 days. On the day 15, 3.3 ml/kg of $CCl_4$ dissolved in olive oil (1:1) was injected 12 hr before anesthetization. Activities of superoxide dismutase(SOD) in mitochondrial fraction were measured and catalase (CAT), glutathione peroxidase (GPx), malondialdehyde (MDA) in liver total homogenate. SOD, CAI and GPx were higher in the ULEF-pretreated and $CCl_4-posttreated$ group than those in the $CCl_4-posttreated$ group, and the pretreatment of ULEF decreased MDA. To investigate effects of Jujube methanol extract on the carbon tetrachloride-induced liver damage in rats, experimental animals were divided into 4 groups; control group(CON), Jujube methanol extracttreated group(JME), $CCl_4$- treated groups(CCl), and Jujube methanol extract and $CCl_4$-treated group (JMC). Each group was sacrificed after 2 or 4week feeding and determined the activities of serum transaminase(GOT, GPT) and hepatic xanthine oxidase, superoxide dismutase(SOD), catalase and glutathione peroxidase(GSH-Px), and hepatic contents of thiobarbituric acid-reactants(TBARS) and glutathione in liver. The activities of sGOT and sGPT, and the hepatic content of TBARS after $CCl_4$-treatment were markedly increased, compared to CON, but those levels were significantly decreased by the pretreatment of Jujube methanol extract, especially in sGOT after 2 and 4 week and TBARS after 4week respectively. Xanthine oxidase activity was increased by $CCl_4$- treatment as compared to CON, but it was also inhibited by the pretreatment of Jujube methanol extract for 2 and 4 week. The activities of SOD, catalase and GSH-Px were elevated by $CCl_4$-treatment, compared to CON, but those elevated activities were showed significant decreasing effect by pretreatment of Jujube methanol extract after 2 and 4week as compared to CON, however, hepatic catalase activity was not affected significantly. These results suggest that Jujube methanol extract is believed to be a possible protective effect for the carbon tetrachloride-induced hepatotoxicity in rats.
