A Unique Gene Expression Signature of 5-fluorouracil

  • To understand the response of cancer cells to anticancer drugs at the gene expression level, we examined the gene expression changes in response to five anticancer drugs, 5-fluorouracil, cytarabine, cisplatin, paclitaxel, and cytochalasin D in NCI-H460 human lung cancer cells. Of the five drugs, 5-fluorouracil had the most distinctive gene expression signature. By clustering genes whose expression changed significantly, we identified three clusters with unique gene expression patterns. The first cluster reflected the up-regulation of gene expression by cisplatin, and included genes involved in cell death and DNA repair. The second cluster pointed to a general reduction of gene expression by most of the anticancer drugs tested. A number of genes in this cluster are involved in signal transduction that is important for communication between cells and reception of extracellular signals. The last cluster represented reduced gene expression in response to 5-fluorouracil, the genes involved being implicated in DNA metabolism, the cell cycle, and RNA processing. Since the gene expression signature of 5-fluorouracil was unique, we investigated it in more detail. Significance analysis of microarray data (SAM) identified 808 genes whose expression was significantly altered by 5-fluorouracil. Among the up-regulated genes, those affecting apoptosis were the most noteworthy. The down-regulated genes were mainly associated with transcription-and translation-related processes which are known targets of 5-fluorouracil. These results suggest that the gene expression signature of an anticancer drug is closely related to its physiological action and the response of caner cells.

  • In this work, a variety of polymer based jelly phantoms suitable for the hyperthermia operations to human organs was synthesized in order to confirm the possibility of auxiliary cancer therapy. Specifically, using an appropriate material composition including polyethylene, Jelly phantoms for brain was prepared and characterized their electrical properties suitable for the monitoring the effect of electromagnetic wave from code division multiple access (CDMA) and personal communication service (PCS) on the human body. In the future, after injection of ferromagnetic nanoparticle into the jelly phantoms, new approach to propose the cancer therapy can be anticipated by monitoring the degree of temperature rise in human body using the photograph of Infrared camera.

  • Microarray classification typically possesses two striking attributes: (1) classifier design and error estimation are based on remarkably small samples and (2) cross-validation error estimation is employed in the majority of the papers. A Microarray data of ovarian cancer consists of the expressions of thens of thousands of genes, and there is no systematic procedure to analyze this information instantaneously. In this paper, gene markers are selected by ranking genes according to statistics, popular classification rules - linear discriminant analysis, k-nearest-neighbor and decision trees - has been performed comparing classification accuracy of data selecting gene markers and not selecting gene markers. The Result that apply linear classification analysis at Microarray data set including marker gene that are selected using ANOVA method represent the highest classification accuracy of 97.78% and the lowest prediction error estimate.

  • Next-generation sequencing (NGS) has enabled whole genome and transcriptome single nucleotide variant (SNV) discovery in cancer and method of the most fundamental being determining an individual's genotype from multiple aligned short read sequences at a position. Bayesian algorithm estimate parameter using posterior genotype probabilities and other method, EM algorithm, estimate parameter using maximum likelihood estimate method in observed data. Here, we propose a novel genotype-calling system and compare and analyze the effect of sample size(S = 50, 100 and 500) on posterior estimate of sequencing error rate, somatic mutation status and genotype probability. The result is that estimate applying Bayesian algorithm even for 50 of small sample size approached real parameter than estimate applying EM algorithm in small sample more accurately.

  • Nowadays, a lot of related data obtained from these research could be given a new present meaning to accomplish the original purpose of the whole research as a human genome project. In such a thread, construction of gene expression analysis system and a basis rank analysis system is being watched newly. Recently, being identified fact that particular sub-class of tumor be related with particular chromosome, microarray started to be used in diagnosis field by doing cancer classification and predication based on gene expression information. In this thesis, we used cDNA microarrays of 3840 genes obtained from neuronal differentiation experiment of cortical stem cells on white mouse with cancer, created system that can extract informative gene list through normalization separately and proposed combination method for selecting more significant genes. And possibility of proposed system and method is verified through experiment. That result is that PC-ED combination represent 98.74% accurate and 0.04% MSE, which show that it improve classification performance than case to experiment after generating gene list using single similarity scale.

  • A fuzzy classifier which needs various analyses of features using genetic algorithms is proposed. The fuzzy classifier has a simple structure, which contains a classification part based on fuzzy logic theory and a rule generation part using genetic algorithms. The rule generation part determines optimal fuzzy membership functions and inclusion or exclusion of each feature in fuzzy classification rules. We analyzed recognition rate of a specific object, then added finer features repetitively, if necessary, to the object which has large misclassification rate. And we introduce repetitive analyses method for the minimum size of string and population, and for the improvement of recognition rates. This classifier is applied to two examples of the recognition of iris data and the recognition of Thyroid Gland cancer cells. The fuzzy classifier proposed in this paper has recognition rates of 98.67% for iris data and 98.25% for Thyroid Gland cancer cells.

Deep Learning based Skin Lesion Segmentation Using Transformer Block and Edge Decoder (트랜스포머 블록과 윤곽선 디코더를 활용한 딥러닝 기반의 피부 병변 분할 방법)

  • Specialists diagnose skin cancer using a dermatoscopy to detect skin cancer as early as possible, but it is difficult to determine accurate skin lesions because skin lesions have various shapes. Recently, the skin lesion segmentation method using deep learning, which has shown high performance, has a problem in segmenting skin lesions because the boundary between healthy skin and skin lesions is not clear. To solve these issues, the proposed method constructs a transformer block to effectively segment the skin lesion, and constructs an edge decoder for each layer of the network to segment the skin lesion in detail. Experiment results have shown that the proposed method achieves a performance improvement of 0.041 ~ 0.071 for Dic Coefficient and 0.062 ~ 0.112 for Jaccard Index, compared with the previous method.

The Bethedsa System 2001 Workshop Report (The Bethesda System 2001의 최신지견)

  • The Bethesda System (TBS) was first developed in 1988 for the need to enhance the communication of the cytopathologic findings to the referring physician in unambiguous diagnostic terms. The terminology used in this reporting system should reflect current understanding of the pathogenesis of cervical/vaginal disease so the framework of the reporting system should be flexible enough to accommodate advances in medicine including virology, molecular biology, and pathology. Three years after the Introduction of TBS, the second Bethesda workshop was held to set or amend diagnostic criteria for each categories of TBS. TBS 1991 is now widely used. The third Bethesda workshop, The Bethesda System 2001 Workshop, was held in National Cancer institute Bethesda, Maryland from April 30 to May 2, 2001. Again, the goals of this workshop were to promote effective communication and to clarify in reporting cervical cytopathology results to clinicians and to provide with the information to make appropriate decisions about diagnosis and treatment. Nine forum groups were made and there were Web-based bulletin board discussions between October, 2000 and the first week of April, 2001. On the basis of bulletin board comments and discussions, the forum moderators recommended revised terminologies in the Workshop. Hot discussions were followed after the presentation by forum moderators during the workshop. Terminologies confusing clinicians and providing no additional informations regarding patient management were deleted in the workshop to clarify the cervicovaginal cytology results. Any informations related to the patient management were encouraged to add. So 'Satisfactory for evaluation but limited by...' of 'Specimen Adequacy' catergory was deleted. Terminology of 'Unsatisfactory' was further specified as 'Specimen rejected' and 'Specimen processed and examined, but unsatisfactory'. Terminologies of 'Benign Cellular Change' and 'Within Normal Limits' were combined and terminology was changed to 'Negative for intraepithelial lesion or malignancy'. In General categorization, category 'Other' was newly inserted and the presence of 'Endometrial cells' in women over 40 years old can be checked. Although the category 'Benign Cellular Change' was deleted, the organisms or reactive changes of this category can be listed in the descriptive diagnoses. Terminologies of ASCUS and AGUS were changed to atypical squamous cell and atypical glandular cell, respectively. Diagnostic term of 'Adenocarcinoma in situ', which is highly reproducible with reliable diagnostic criteria, was newly Inserted. The category of hormonal evaluation was deleted. Criteria for liquid-based specimen were discussed. Reporting by computer-assisted cytology was discussed and terminology for automated review was newly inserted. This is not the final edition of Bethesda 2001. The final document can be prepared before the ASCCP meeting in which Consensus Guidelines for the Management on Cytology Abnormalities and Cervical Precursors will develop in September 2001.

Effect of Ikiyangeumhaedoc-tang on the tumor and metastasis (익기양음해독탕의 항암 및 항전이효과에 관한 연구)

  • Objective : Ikiyangeumhaedoc-tang(IYHT) has an effect of nourishing Yin(陰) and Jin(津), and has been used to cancer patient effectively. In order to prove the anticancer's and antimetastic effect of IYHT experimentally, studies were done. Methods : We evaluated the cytotoxic activity on HT-1080 cells as well as inhibitory effect on activity of DNA topoisomerase Ⅰ, cell adhesion, cell invasion and proliferation of HUVEC cells induced by bFGF and measured the expression of mRNA(uPA, MMP2, TIMP2), p-ERK protein, recovery effect of gap junctional intercellular communication by $H_{2}O_2$ and survival time of ICR mice bearing sacoma-180. Results : IYHT showed the inhibitory effect on DNA topoisomerase Ⅰ in the concentration of $100{\mu}g/ml,\;500{\mu}g/ml$ and the dosage-dependent inhibitory effect on the adhesion of HT-1080. The concentration of 1mg/ml of IYHT inhibited 15% of adhesion compared with control. IYHT decreased the expression of uPA, but not in MMP2, TIMP2 by RT-PCR and inhibited the expression of p-ERK effectively in the concentration of more than $500{\mu}g/ml.$ IYHT recovered the inhibited gap junctional intercellular communication by $H_{2}O_2$ to the level of 60% of normal control in the concentration of $400{\mu}g/ml$ but, did not extended the mean survival time of sarcoma 180-bearing mouse. Conclusions : It was concluded that IYHT could be applied usefully for prevention and treatment of human cancer, And also experimental study for the evaluation of molecular biological study and antimetastatic research would be recommended in the near future.

The Prayer Experiences of Patients with End-Stage Cancer (말기암환자의 기도 경험)

  • Purpose: To explore the meaning of prayer in Korean patients with end-stage cancer who profess Christianity or Buddhism, given the significant differences between these religions. Methods: The Colaizzi (1978) analysis method was employed. In-depth interview were performed with 13 participants (seven Christians and six Buddhists) who were admitted to a University-affiliated hospital in Korea. Results: The six categories emerged: 1) communication with God, 2) mind discipline, 3) spiritual growth, 4) mysterious experiences, 5) perception of death and after-life, and 6) various forms of prayers. Conclusion: The participants' prayer experiences were described in a religious context. Christians believed that prayer is communication with God while Buddhists regarded it as disciplining of minds. Despite some differences between the religious groups, a general meaning of prayer was a desperate desire to solve their health issues by relying on God or someone who is more powerful than themselves. They also experienced personal and spiritual growth through prayer. This study explains the phenomenon of prayer experiences and shows that prayer is an important coping mechanism.
