불등가사리 분획물의 암세포 성장 억제 효과 (Growth Inhibition Effects of Cancer Cell Lines by Gloiopeltis furcata Fractions in Vitro)

  • 본 실험은 홍조해조류의 하나인 불등가사리를 추출 후 각 용매별로 분획하여 암세포 성장억제 효과와 QR유도활성 효과 등 생리활성을 연구하였다. 불등가사리를 이용하여 4종의 인체 암세포주HepG2, HeLa, MCF-7및 HT-29와 정상 간세포에 대한 암세포 증식억제 실험을 한 결과 사용한 4종의 암세포주에서 모두 시료첨가 농도에 의존적으로 증식 저지 효과가 나타났고, 특히 불등가사리의 methanol 분획층인 GFMM에서는 매우 낮은 농도의 시료첨가에도 불구하고 괄목할 만한 높은 암세포 성장 억제효과를 나타내었으며, 정상 간세포에서는 모든 분획층에서 약한 세포증식 억제율을 나타내어 정상 세포에 미치는 독성이 낮음을 확인하였다. 특히 본 시료는 여성암의 대표적인 유방암과 자궁경부암의 암세포 성장저지효과가 탁월하였다. 한편, 사용한 4가지 암세포주 중 유일하게 QR을 가지고 있는 HepG2를 이용한 암 예방지표인 QR 효소 유도 활성 여부를 측정한 결과 분획물 첨가농도를 10, 20, 30 및 40 $\mu$g/mL로 첨가하였을 때 GFMM의 첨가농도 $20\mu g/mL$에서 대조군에 비해 약 2배 이상의 높은 QR 유도효과를 나타내어 최종농도 $40\mu g/mL$에서는 2.6의 암 예방 QR 유도효과를 나타내었다.

Synergistic Induction of Apoptosis by the Combination of an Axl Inhibitor and Auranofin in Human Breast Cancer Cells

  • Axl receptor tyrosine kinase has been implicated in cancer progression, invasion, and metastasis in various cancer types. Axl overexpression has been observed in many cancers, and selective inhibitors of Axl, including R428, may be promising therapeutic agents for several human cancers, such as breast, lung, and pancreatic cancers. Here, we examined the cell growth inhibition mediated by R428 and auranofin individually as well as in combination in the human breast cancer cell lines MCF-7 and MDA-MB-231 to identify new advanced combination treatments for human breast cancer. Our data showed that combination therapy with R428 and auranofin markedly inhibited cancer cell proliferation. Isobologram analyses of these cells indicated a clear synergism between R428 and auranofin with a combination index value of 0.73. The combination treatment promoted apoptosis as indicated by caspase 3 activation and poly (ADP-ribose) polymerase cleavage. Cancer cell migration was also significantly inhibited by this combination treatment. Moreover, we found that combination therapy significantly increased the expression level of Bax, a mitochondrial proapoptotic factor, but decreased that of the X-linked inhibitor of apoptosis protein. Furthermore, the suppression of cell viability and induction of Bax expression by the combination treatment were recovered by treatment with N-acetylcysteine. In conclusion, our data demonstrated that combined treatment with R428 and auranofin synergistically induced apoptosis in human breast cancer cells and may thus serve as a novel and valuable approach for cancer therapy.

  • 3',4'-Disenecioylkhellactone is one of khellactone-type coumarins isolated from the roots of Peucedanum japonicum Thunb. However, its pharmacological effects are still little understood. In the present study, we investigated the inhibitory effect of 3',4'-disenecioylkhellactone on growth of gastric cancer cells. 3',4'-Disenecioylkhellactone strongly suppressed cell proliferation and induced caspase-mediated apoptosis in AGS human gastric cancer cells. Analysis of phospho-antibody arrays revealed 3',4'-disenecioylkhellactone effectively suppressed signal transducer and activator of transcription 3 (STAT3) tyrosine phosphorylation. 3',4'-Disenecioylkhellactone decreased STAT3 translocation to the nucleus and expression of STAT3 target genes. In addition, we examined the level of STAT3 activation in several gastric cancer cells and found that the inhibition of STAT3 phosphorylation by 3',4'-disenecioylkhellactone was associated with gastric cancer cell proliferation. Taken together, this study provides evidence for the first time that 3',4'-disenecioylkhellactone may be a potential therapeutic agent for the prevention or treatment of gastric cancer.

Growth Inhibition and Apoptosis Induction of Gastric Cancer Cells by Copper (II) Glycinate Complex

  • The in vitro cytotoxic effects of newly synthesized copper (II) glycinate complex were investigated in two gastric cancer cell lines of SNU484 and SNU638 cells. The complex inhibited the growth and decreased the viability of both gastric cancer cells in a dose-dependent manner. Gastric cancer tells treated with the complex exhibited the features of apoptosis, as demonstrated by fragmentation of chromosomal DNA, activation of caspase-3-like enzyme, and cleavage of poly[ADP-ribose] polymerase (PARP). With the treatment of copper (II) glycinate complex, the active form of caspase-3 was observed in SNU484 cells, but not in SNU638 cells, indicating that an alternative pathway of apoptosis might have been triggered in SNU638 cells. In conclusion, copper (II) glycinate complex induces apoptosis of SNU484 and SNU638 gastric cancer cells, and it is suggested that novel copper (II) glycinate complex is highly active against human gastric cancer cells.

Natural Products for Cancer-Targeted Therapy: Citrus Flavonoids as Potent Chemopreventive Agents

  • Targeted therapy has been a very promising strategy of drug development research. Many molecular mechanims of diseases have been known to be regulated by abundance of proteins, such as receptors and hormones. Chemoprevention for treatment and prevention of diseases are continuously developed. Pre-clinical and clinical studies in chemoprevention field yielded many valuable data in preventing the onset of disease and suppressing the progress of their growth, making chemoprevention a challenging and a very rational strategy in future researches. Natural products being rich of flavonoids are those fruits belong to the genus citrus. Ethanolic extract of Citrus reticulata and Citrus aurantiifolia peels showed anticarcinogenic, antiproliferative, co-chemotherapeutic and estrogenic effects. Several examples of citrus flavonoids that are potential as chemotherapeutic agents are tangeretin, nobiletin, hesperetin, hesperidin, naringenin, and naringin. Those flavonoids have been shown to possess inhibition activity on certain cancer cells' growth through various mechanisms. Moreover, citrus flavonoids also perform promising effect in combination with several chemotherapeutic agents against the growth of cancer cells. Some mechanisms involved in those activities are through cell cycle modulation, antiangiogenic effect, and apoptosis induction.Previous studies showed that tangeretin suppressed the growth of T47D breast cancer cells by inhibiting ERK phosphorylation. While in combination with tamoxifen, doxorubicin, and 5-FU, respectively, it was proven to be synergist on several cancer cells. Hesperidin and naringenin increased cytotoxicitity of doxorubicin on MCF-7 cells and HeLa cells. Besides, citrus flavonoids also performed estrogenic effect in vivo. One example is hesperidin having the ability to decrease the concentration of serum and hepatic lipid and reduce osteoporosis of ovariectomized rats. Those studies showed the great potential of citrus fruits as natural product to be developed as not only the source of co-chemotherapeutic agents, but also phyto-estrogens. Therefore, further study needs to be conducted to explore the potential of citrus fruits in overcoming cancer.

Antitumor Effects of Water Extracts of Panax notoginseng on NCI-H460 Tumor Regression Model

  • Objective: This study aimed to investigate the antitumor effects of water extracts of Panax notoginseng (WEPN) in NCI-H460 human lung cancer cell xenografted nude mice. Materials and Methods: We cultured NCI-H460 cell lines and xenografted them to nude mice. The mice were divided into 3 groups; positive control group, NCI-H460+150 mg/kg WEPN-treated group, and NCI-H460+300 mg/kg WEPN-treated group. They had been raised and treated in 28 days. We checked their body weight and tumor weight and volumes twice a week and their absolute organ weight and microhistological observation at the final day. We also calculated their tumor inhibition rate (I.R.), mean survival time and percent increase in life span (% ILS). Results: Body weight of WEPN (300 mg/kg) treated mice tended to slightly greater increase than those of the positive control group, but had no significance. Tumor volume (measurement with a caliper) of WEPN-treated mice tended to be lower than that of the positive control group. Inhibition rate (I.R.) of the WEPN group decreased more than the positive control group, but had no significance. Results of tumor weights and volume (plethysmography) had no significance. Mean survival time and percent increase in life span (% ILS) in the WEPN 300 mg/kg treatment group were higher than those of any other group (p<0.05). In absolute organ weights, the WEPN (150-300 mg/kg) treatment group decreased liver weights (p<0.05). Liver tissue of mice treated with WEPN (300 mg/kg) did not show any specific lesions. Conclusion: We suggest that WEPN may have potential as a growth inhibitor of solid tumors induced by NCI-H460 without any side effects. However, this study has limitations in proving anti-tumor effects of WEPN, so further studies to overcome those limitations will be needed.

상황을 이용한 한의학적 보건기능 개선제에 의한 인체폐암세포의 증식억제에 관한 연구 (Down-regulation of COX-2 and hTERT Expression by Healthful Decoction Utilizing Phellinus Linteus in Human Lung Carcinoma Cells)

  • The objective of the present study was to investigate the effects of aqueous extract from the healthful decoction utilizing Phellinus linteus (HDPL) on the growth of human lung carcinoma A549 cells. HDPL treatment declined the cell viability of A549 cells in a concentration-dependent manner and the anti-proliferative effects by HDPL treatment were associated with morphological changes such as membrane shrinking and cell rounding up. HDPL treatment did not affect the distribution of the cell cycle. Western blot analysis and RT-PCT data revealed that the levels of tumor suppressor p53 and cyclin-dependent kinase inhibitor p21WAF1/CIP1 in HDPL-treated A549 cells were remained unchanged. However, HDPL treatment inhibited the expression of cyclooxygenase-2 (COX-2) mRNA and protein in a concentration-dependent fashion. Additionally, the expression of human telomerase reverse transcriptase (hTERT), a main determinant of the telomerase enzymatic activity, was progressively down-regulated by HDPL treatment. Taken together, these findings suggest that HDPL-induced inhibition of human lung cancer cell proliferation is associated with the inhibition of several major growth regulatory gene products, such as COX-2 and hTERT, and HDPL may have therapeutic potential in human lung cancer.

miR-372 Regulates Cell Cycle and Apoptosis of AGS Human Gastric Cancer Cell Line through Direct Regulation of LATS2

  • Previously, we have reported tissue- and stage-specific expression of miR-372 in human embryonic stem cells and so far, not many reports speculate the function of this microRNA (miRNA). In this study, we screened various human cancer cell lines including gastric cancer cell lines and found first time that miR-372 is expressed only in AGS human gastric adenocarcinoma cell line. Inhibition of miR-372 using antisense miR-372 oligonucleotide (AS-miR-372) suppressed proliferation, arrested the cell cycle at G2/M phase, and increased apoptosis of AGS cells. Furthermore, AS-miR-372 treatment increased expression of LATS2, while over-expression of miR-372 decreased luciferase reporter activity driven by the 3' untranslated region (3' UTR) of LATS2 mRNA. Over-expression of LATS2 induced changes in AGS cells similar to those in AGS cells treated with AS-miR-372. Taken together, these findings demonstrate an oncogenic role for miR-372 in controlling cell growth, cell cycle, and apoptosis through down-regulation of a tumor suppressor gene, LATS2.

Inhibition of Cervical Cancer Cell Growth by Gene Silencing of HPV16 E6 Induced by Short-interfering RNA

  • The Human Papilloma Virus (HPV) infection has been strongly associated with pathogenesis of uterine cervix carcinoma. HPV type 16, a causative agent of uterine cervix carcinoma, encodes the E6 and E7 oncogenes, expression of which is pivotal for malignant transformation and maintenance of malignant phenotypes. To develop a gene therapy for HPV-related carcinoma, We investigated the effect of E6 short-interfering RNA (E6 siRNA) on the expression of this oncogene and on the growth of HPV 16-related uterine cervix carcinoma cells. SiHa cells, a uterine cervix carcinoma cell line, which contain a single copy of HPV 16 integrated in the chromosome and express the E6 and E7 oncogenes. Before 24 hr of transfection, cells were seeded and transfected with control plasmid or E6 siRNA-expressing plasmid. The mRNA was analysed by reverse transcriptase polymerase chain reaction (RT-PCR). The cell growth rate was investigated by MTT method. The E6 mRNA level in SiHa cells was decreased in HPV 16 E6 siRNA-expression vector transfected cells and a decrease in the growth of these cells was also observed. From these results. it is evident that E6 siRNA played a role in suppression of growth of SiHa cells and has a fair chance as a candidate for gene specific therapy for HPV related uterine cervix carcinoma.

siRNA-mediated Silencing of Notch-1 Enhances Docetaxel Induced Mitotic Arrest and Apoptosis in PCa Cells

  • Purpose: Notch is an important signaling pathway that regulates cell fate, stem cell maintenance and the initiation of differentiation in many tissues. It has been reported that activation of Notch-1 contributes to tumorigenesis. However, whether Notch signaling might have a role in chemoresistance of prostate cancer is unclear. This study aimed to investigate the effects of Notch-1 silencing on the sensitivity of prostate cancer cells to docetaxel treatment. Methods: siRNA against Notch-1 was transfected into PC-3 prostate cancer cells. Proliferation, apoptosis and cell cycle distribution were examined in the presence or absence of docetaxel by MTT and flow cytometry. Expression of $p21^{waf1/cip1}$ and Akt as well as activation of Akt in PC-3 cells were detected by Western blot and Real-time PCR. Results: Silencing of Notch-1 promoted docetaxel induced cell growth inhibition, apoptosis and cell cycle arrest in PC-3 cells. In addition, these effects were associated with increased $p21^{waf1/cip1}$ expression and decreased Akt expression and activation in PC-3 cells. Conclusion: Notch-1 promotes chemoresistance of prostate cancer and could be a potential therapeutic target.
