Iron succinyl casein encapsulated alginate beads for the treatment of iron deficiency anemia

  • Iron deficiency is the most common nutritional problem worldwide. Oral iron supplementation programs have failed because of noncompliance and gastrointestinal toxicity. The purpose of this study was to explore the possibility of alginate gel bead as an oral controlled release system of iron supplements and increase the stability of iron succinyl casein (ISC). Alginate beads containing ISC were prepared by the gelation of sodium alginate with calcium cations. The release profiles of ISC were investigated according to the concentration of polymer, the drug/sodium alginate ratio, the concentration and type of cation, curing time and pH of calcium chloride solution. Ion-Selectrodes based on Calcium Ferrite (칼슘 페라이트 이온-선택성 막전극)

  • The ion-selectrodes based on calcium ferrite were prepared by mixing calcium oxide with ferric oxide on molar ratio of 0.6:1.4, 0.8:1.2, 1.0:1.0, respectively. The specimens were sintered at $1400^{\circ}C$ for 2hrs in $O_2$ and for 20min in $H_2$ atmosphere. The selectrode sintered in hydrogen atmosphere showed better responded potential and wider range of responded concentration than selectrode sintered in oxygen atmosphere. The ion-selectrodes base on calcium ferrite(0.6:1.4 molar ratio.) showed the highest potential to bivalent cations such as $Ca^{2+}$, $Ba^{2+}$, $Mg^{2+}$, $Zn^{2+}$ and halogen anions such as $I^-$, $Br^-$ in the range of $10^{-1}{\sim}10^{-7}M$. It showed good agreement with theoretical nernstian values.

Effect of Calcium Intake and Health Status on Blood Lipids in Middle - aged Women (장년기 여성에서 칼슘영양상태와 건강상태가 혈중 지질성분에 미치는 영향)

  • The purpose of this study was to confirm that calcium intake and healthy status play a role in reducing the risk of cardiovascular disease, to assure that calcium intake and regular exercise are important in reducing serum lipid levels in middle-aged women. Daily nutrient intakes were analyzed by convenient method. Anthropometric measurement such as body weight and height and blood pressure were measured. The subjects divided into two groups : women in group I under 50 years of age($\leq$49 yr group) and those in group II 50 years up($\geq$50 yr group). Average ages of group I and II were 43.87 and 53.46 years. The nutrients intake of subjects were higher than Korean recommended dietary allowances(KRDA) except calorie, iron, calcium, vitamin B1. They showed significant difference each other in vitamin B2(p<0.05), calcium(p<0.01) intake. Mean daily calcium intake was 540.88mg in $\leq$49 yr group and 519.50mg in $\geq$50 yr group. The levels of serum cholesterol, triglyceride, blood pressure, Atherogenic Index(AI), LDL/HDL and Cardiac Index(CI) were increased with age. The triglyceride and total cholesterol concentrations and LDL/HDL in $\geq$50 yr group were significantly higher than those of $\leq$49 yr group. There was a highly significantly positive correlation between age and triglyceride, total cholesterol and Atherogenic Index(AI). Triglyceride, total cholesterol, LDL-cholesterol concentration and Atherogenic Index(AI) were positively correlated with body weight and WHR(waist-hip ratio). Also ther was a significantly negative correlation between exercise and Cardiac Index(CI). Also a significantly negative correlation was found between Ca intake and LDL-cholesterol. Exercise and Cardiac Index(CI) also had a significantly negative relation. The results suggest that increased habitual physical activity and calcium intake should be recommended as a way of decreasing blood lipids and blood pressure in middle-aged women.

  • Composite films of zinc calcium alginate were prepared by a film maker from 7 wt% sodium alginate solution and then they solidified into 3, 5 wt% content $ZnCl_2$, $CaCl_2$ solution followed by washing and drying at room temperature. The characteristics were measured with several methods (antimicrobial activity, water solubility, swelling ratio and viscosity, SEM, EDS) and the film properties were investigated. Composite films of zinc calcium alginate showed an increase in the water resistance by increasing $ZnCl_2$ and $CaCl_2$ content and the antimicrobial test showed that the calcium alginate as well as zinc alginate films result in excellent antimicrobial activity in the two strains, Staphylococcus and Escherichia coli. The results show the possible improvement of the physical properties of composite films.

  • Alteration of intracellular calcium ion Concentration by adding of either calcium ionophore A23187 or EGTA in culture medium at 24 hr after cell plating resulted in remarkable changes in the progression of differentiation of chick embryo myoblast. When separated myoblast proteins using two-dimensional gel electrophoresis, synthesis patterns of several proteins changed upon the addition of either A23187 or EGTA. Treatment of A23187 and calciumactivated neutral protease at 24 hr after initial plating caused an increase in the rate of fusion compared to control culture. However, EGTA inhibited the myoblast fusion to a marked degree. A23187 treated at 24hr also increased the activity of protein kinase C during the fusionprogressed period. It seems that intracellular calcium ion plays an important role in the myoblast differentiation in vitro together with the protein kinase C and calcium-activated neutral protease.

  • The synthesis and crystallization of amorphous calcium carbonate($CaCO_3$.$nH_2 O$) obtained from gas-liquid reaction between aqueous solution of calcium hydroxide and carbon dioxide at 15~$50^{\circ}C$ are investigated by electrical conductometry, XRD and TEM. The results are as follows: The initial reaction products prior to the formation of precipitated calcium carbonate is amorphous calcium carbonate. The electrical conductivity values in the slurry are decreased during the formation of amorphous calcium carbonate which covers particle surface of calcium hydroxide and retard the dissolution of calcium hydroxide into the solution. that amorphous calcium carbonate is unstable in the aqueous solution and crystallizes finally to calcite by the through-solution reaction. While amorphous calcium carbonate crystallizes into chain-like calcite, the conductivity values are recovered rapidly and the apparent viscosity of slurry containing higher concentration of calcium hydroxide increase. At below pH 9.5, chain-like calcite separates into individual particles to form precipitated calcium carbonate. The formation and synthetic temperature range of amorphous calcium carbonate is most suitable a primary decreasing step(a-step) at $15^{\circ}C$ in the electrical conductometry.

Preparation of Alginate Microspheres Using Membrane Emulsification Method (막유화법에 의한 알지네이트 Microsphere의 제조)

  • We prepared monodispersed calcium alginate microspheres by controlling various conditions of emulsification procedure using a lab-scale batch type membrane emulsification system equipped with SPG (Shirasu porous glass) tubular membranes. We determined the effects of process parameters of membrane emulsification (ratio of dispersed phase to continuous phase, alginate concentration, emulsifier concentration, type and concentration of stabilizer, transmembrane pressure, concentration of crosslinking agent, stirring speed and membrane pore size) on the mean size and size distribution of alginate microspheres. The increase of the ratio of dispersed phase to continuous phase, transmembrane pressure and alginate concentration led to the increase in the mean size of alginate microspheres. On the contrary, the increase in emulsifier concentration, stirring speed of the continuous phase and concentration of the crosslinking agent caused the reduction of the mean size of microspheres. Through controlling these parameters, monodisperse alginate microspheres with about $6{\mu}{\textrm{m}}$ of the mean size and 1.1 of the size distribution value were finally prepared in case of the using SPC membrane with the pore size of $2.9{\mu}{\textrm{m}}$.

Action of Serotonin on Sodium-Potassium Activated ATPase in Rabbit Red Cell Membrane (토끼 적혈구막의 NaK ATPase의 활성도에 대한 serotonin의 작용)

  • The action of serotonin on the sodium plus potassium activated ATPase activity in the rabbit red cell membrane has been investigated. The experiments were also designed to determine the mechanism of action of serotonin on the ATPase activity. The following results were obtained. 1) The NaK ATPase activity of rabbit red cell ghosts is stimulated by low concentration of serotonin but inhibited by higher concentration, and the concentration of serotonin for maximal activity is about 2mM. The pH optimum for the serotonin sensitive component is 8.0. 2) The activating effect of serotonin on the ATPase, with a given concentration of sodium in the medium, is increased by raising the potassium concentration but the ratio of activity is decreased. 3) The activating effect of serotonin on the ATPase, with a given concentration of potassium in the medium, is increased by raising the sodium concentration but the ratio of activity is decreased. 4) The ATPase activity is increased by small amounts of calcium but inhibited by larger amounts and the ratio of activity by serotonin is decreased by small amounts of calcium but increased by larger amounts. 5) The action of serotonin on the ATPase activity was not related to the amino group of lysine, the hydroxyl group of threonine, the carboxyl group of aspartic acid, or the imidazole group of histidine. 6) The action of serotonin on the ATPase activity is due to sulfhydryl group of the enzyme of NaK ATPase.

Preparation of Alginate Microspheres by Rotating Membrane Emulsification (회전 막유화에 의한 알지네이트 미소 구체의 제조)

  • When preparing calcium alginate microspheres using rotating membrane emulsification that rotates SPG (Shirasu porous glass) tubular membrane in the continuous phase, the optimal conditions of rotating membrane emulsification process parameters for producing monodisperse microspheres were determined. We determined the effects of process parameters of rotating membrane emulsification (the rotating speed of membrane module, the transmembrane pressure, the ratio of dispersed phase to continuous phase, the alginate concentration, the emulsifier concentration, the stabilizer concentration, the crosslinking agent concentration, and the membrane pore size) on the mean size and size distribution of alginate microspheres. As a result, the size of the microspheres decreased as the rotating speed of membrane module, the emulsifier concentration, and the crosslinking agent concentration increased among the process parameters of rotating membrane emulsification. On the contrary, as the ratio of dispersed phase to continuous phase, the transmembrane pressure, and the alginate concentration increased, the size of the microspheres increased. In the rotating membrane emulsification using an SPG membrane with a pore size of 3.2 ㎛, it was possible to finally prepare monodisperse alginate microspheres with a particle size of 4.5 ㎛ through the control of process parameters.
