New Retention Mechanism of Mononucleotides with Buffer Concentrations in Ion-suppressing RP-HPLC

  • HPLC separation of ionic samples tends to be more complicated and difficult to understand than that of non-ionic compounds. On the other hand, band spacing is much more easily manipulated for ionic than for neutral samples. Ion-suppression RP-HPLC method was used with organic modifier and aqueous buffer solution. In this work, five mononucleotides of cytidine-5-monophosphate (5-CMP) disodium salt, uridine-5-monophosphate disodium salt (5-UMP), guanosine-5-monophosphate disodium salt (5-GMP), inosine-5-monophosphate disodium salt (5-IMP), and adenosine-5-monophosphate disodium salt (5-AMP) were examined. Acetic acid and sodium phosphate were used as buffers, and methanol as an organic modifier. A new relationship between the retention factor and the buffer concentration at a fixed modifier content (5% of methanol) could be expressed by following: K = (k(sub)-1 + k(sub)0 (k(sub)B/k(sub)S)/(1 + (k(sub)B/k(sub)S) C(sub)B(sup)a), where C(sub)B was the concentration of buffer. Using this relationship, the calculated values closely matched the experimental data. The derived relationship showed that as the buffer concentration increased, the retention factor approached a certain value, and this was buffer dependent.

Inhibitory Effect of Scopoletin on U46619-induced Platelet Aggregation through Regulation of Ca2+ Mobilization

  • Platelet aggregation is essential for hemostatic process in case of blood vessels damages. However, excessive platelet aggregation can cause cardiovascular disorders including atherosclerosis, thrombosis and myocardial infarction. Scopoletin is usually found in the roots of genus Scopolia or Artemisia, and is known to have anticoagulant and anti-malarial effects. This study investigated the effect of scopoletin on human platelet aggregation induced by U46619, an analogue of thromboxane $A_2(TXA_2)$. Scopoletin had anti-platelet effects by down-regulating $TXA_2$ and intracellular $Ca^{2+}$ mobilization ($[Ca^{2+}]_i$), the aggregation-inducing molecules generated in activated platelets. On the other hand, scopoletin increased the levels of cyclic adenosine monophosphate (cAMP) and cyclic guanosine monophosphate (cGMP), which are known to be intracellular $Ca^{2+}$ antagonists. This resulted in inhibition of fibrinogen binding to ${\alpha}IIb/{\beta}_3$ in U46619-induced human platelet aggregation. In addition, scopoletin inhibited the release of adenosine trisphosphate (ATP) in dose-dependent manner. This result means that the aggregation amplification activity through the granule secretion in platelets was suppressed by scopoletin. Therefore, we demonstrated that scopoletin has a potent antiplatelet effect and is highly likely to prevent platelet-derived vascular disease.

Chemical Characteristics of Commercial Korean Soy Sauce Produced by Alkali Treatment Following Acidic Hydrolysis

  • Alkali treatment following acidic hydrolysis (ATAH) is a commonly used practice for reducing the levels of monochloropropanediol in commercial Korean soy sauce. This study investigated the chemical compounds produced in commercial Korean soy sauce made by ATAB. The levels of amino-N, total acidity, pH, salinity and Brix of the soy sauce were 6.66%, 2.52%, 19.81%, 4.57 and 35.01, respectively. The major fatty acids were C18:2n-6, C16:0, Cl8:ln-9 and C18:3n-6. The concentration of polyunsaturated fatty acids were especially high, with of C18:2n-6 (49.8%) being the highest followed by C18:3n-6 (3.8%) and C18:3n-3 (2.4%) in that order. Among the non-volatile organic acids, the concentration of levulinic acid (1,206.28 mg/100 g) was the highest, while the taste value of citric acid was the highest. Among the ATP related compounds, IMP concentration (31.19 mg/100 g) was highest followed by AMP, hypoxanthine and GMP in that order. The concentrations of free and total amino acids in soy sauce were 6,136.94 mg/100 g and 8,702.76 mg/100 g, respectively. On the other hand, the taste value of glutamic acid, a major amino acid flavor determinant in soy sauce, was highest of all the amino acids, which is desirable since most free amino acids such as methionine, histidine and phenylalanine have a bitter taste that detracts from the flavor of soy sauce.

GS 283의 평활근 억제 작용기전 (Mechanisms Underlying the Inhibitory Effect of GS 283 in Various Smooth Muscles)

  • Tetrahydroisoquinoline유도체인 GS 283의 약리학적 특성을 흰쥐 흉부대동맥, 기니픽 결장띠 및 토끼 장간막 동맥 및 흰쥐 뇌를 사용하여 조사하였다. 혈관 평활근에서 GS283은 고 $K^+$에 의한 수축을 농도 의존적으로 억제하여 $Ca^{2+}$ 길항작용을 보였다. 또한 ${alpha}_1$- 수용체 자극에 의한 수축도 억제하였다. GS 283의 혈관이완 작용은 propranolol영향을 받지 않으므로 ${\beta}$-수용체 자극작용에 의한 것이 아니었다. 세포내 칼슘이온과 근장력 변화를 동시에 측정하였을 때 GS 283의 억제효과는 조직내 형광의 증가를 수반했다. 이 증가는 fura 2형광에 의한것이 아니라 내인성 pyridine nucleotide에 의한 것이며 이는 GS 283이 미토콘드리아 기능을 억제하는 효과가 있음을 시사했다. 흰쥐 뇌의 cAMP와 cGMP 의존성 phosphodiesterase에 대한 GS 283의 $K_i$,값은 2.5와 6.7mM이었다. 이상의 결과에서 GS 283의 약리 작용은 $Ca^{2+}$ 길항작용, ${\alpha}_1$- 수용체억제 작용 및 cyclic nucleotide 의존성 phosphodiesterase 억제 등 다양한 작용이 있으며 평활근 수축 억제에 대한 GS283 작용에는 $Ca^{2+}$ 길항이 가장 중요한 요인이 될 것으로 생각된다.

비지의 발효과정중 발효미생물 및 성분변화 (Microbiological Studies and Biochemical Changes in Fermenting Soybean Curd Residue during Fermentation)

  • 비지의 발효조건을 확립하여 비지의 발효에 관여하는 주요 미생물 두 균주를 분리, 동정하였으며 또한 발효과정중 유리 아미노산의 함량, 핵산관련물질의 함량, 환원당 및 소당류의 함량 변화를 조사하였다. 비지의 발효조건은 $55^{\circ}C$에서 48시간 발효시킨 비지가 재래식으로 발효시킨 비지와 가장 유사하였다. 발효과정중 pH는 서서히 증가하였으며 비지의 발효에 관여하는 주요 미생물군은 Bacillus subtilis와 Bacillus lichemiformis로 동정되었다. 비지의 발효과정중 수분 함량은 48시간 발효 후에는 80.8%에서 58.4%로 감소하였으나 다른 일반성분은 거의 변화가 없었다. 총 유리 아미노산 함량은 발효과정을 통하여 급격히 증가하였다. 또한 발효된 비지에 있어서 유리 아미노산의 종류 및 양도 증가하였는 데 Phe>Lys>His>Tyr>Glu>Asp 의 순서로 증가하였다. 핵산관련물질의 변화에 있어서 처음 36시간 동안은 거의 변화가 없다가 48시간 발효 후에는 1/6배로 감소하였으며 5'-IMP와 5'-GMP 혼합구는 처음 36시간 동안은 서서히 증가하다가 48시간 발효 후에는 약 3.5배로 증가하였다. 한편 환원당 함량은 비지의 발효과정중 계속 증가하였으며 stachyose 및 raffinose 혼합구는 미량이기는 하나 점차 감소하였다. 이로부터 비지의 발효과정중 5'-IMP와 5'-GMP의 증가는 유리 아미노산 및 환원당의 증가와 함께 발효된 비지의 맛성분에 영향을 주는 중요한 요인이라 생각되며 발호된 비지를 메주제조시 혼합하여 콩의 대체 원료로 이용하는 것에 대한 연구가 필요하다고 생각된다.

  • Carp, Cypinus carpio, has been esteemed as one of the most tasty fresh water fishes in Korea. But little study on its food has been reported. The composition of fatty acids, amino acids, nucleotides and their related compounds as chemical component of carp muscle were analyzed by gas-liauid chromatography, amino acid autoanalyzer and high speed liauid chromatography. The fatty acids of the carp muscle lipid consisted of the large amount of $C_{18}\;:\;2,$ $C_{18}\;:\;1,$ $C_{18}\;:\;3,$ $C_{16}\;:\;0,$ $C_{16}\;:\;1,$ acids, small amount of $C_{16}\;:\;2,$ $C_{18}\;:\;0,$ $C_{18}\;:\;4,$ $C_{14}\;:\;0,$ $C_{14}\;:\;1,$ $C_{12}\;:\;0,$ acids, and $C_{13}\;:\;1,$ $C_{13}\;:\;0,$ $C_{20}\;:\;1,$ $C_{17}\;:\;0,$ $C_{15}\;:\;1,$ $C_{17}\;:\;1,$ $C_{22}\;:\;1$ acids were smaller. In fresh carp, the results showed that inosine $17.25{\mu}mole/g$, hypoxanthine $10.06{\mu}mole/g$ were dominant and the content of GMP, IMP, AMP, UMP, CMP were 1.56, 1.36, 0.92, 0.49, $0.34\:{\mu}mole/g$ on dry base, respectively. Histidine and lysine were dominant amino acids in carp extract, having 49.4% (214.6mg%), 33.9% (147.1mg%) of total free amino acid contents, respectively, but the content of glycine, serine, alanine and glutamic acid were low, and arginine, aspartic acid, threonine, proline, valine, methionine, isoleucine, phenylalanine were detected in trace amount. The content of amino acid composition showed that glutamic acid, aspartic acid, lysine, leucine, alanine, valine and arginine were the most abundant amino acids, while such amino acids as glycine, isoleucine, phenylalanine, threonine, histidine, methionine, serine and tyrosine were low, and proline was detected in trace amount.

(-)-Epigallocatechin Gallate Inhibits the Pacemaker Activity of Interstitial Cells of Cajal of Mouse Small Intestine

  • The effects of (-)-epigallocatechin gallate (EGCG) on pacemaker activities of cultured interstitial cells of Cajal (ICC) from murine small intestine were investigated using whole-cell patch-clamp technique at $30^{\circ}C$ and $Ca^{2+}$ image analysis. ICC generated spontaneous pacemaker currents at a holding potential of -70 mV. The treatment of ICC with EGCG resulted in a dose-dependent decrease in the frequency and amplitude of pacemaker currents. SQ-22536, an adenylate cyclase inhibitor, and ODQ, a guanylate cyclase inhibitor, did not inhibit the effects of EGCG. EGCG-induced effects on pacemaker currents were not inhibited by glibenclamide, an ATP-sensitive $K^+$ channel blocker and TEA, a $Ca^{2+}$-activated $K^+$ channel blocker. Also, we found that EGCG inhibited the spontaneous $[Ca^{2+}]_i$ oscillations in cultured ICC. In conclusion, EGCG inhibited the pacemaker activity of ICC and reduced $[Ca^{2+}]_i$ oscillations by cAMP-, cGMP-, ATP-sensitive $K^+$ channel-independent manner.

Effects of Schisandra chinensis fruit extract and gomisin A on the contractility of penile corpus cavernosum smooth muscle: a potential mechanism through the nitric oxide - cyclic guanosine monophosphate pathway

  • BACKGROUND/OBJECTIVES: This study evaluated the effects and molecular mechanisms of the Schisandra chinensis fruit extract (SC) and its major compound gomisin A (GA), on the contractility of rabbit penile corpus cavernosum smooth muscle (PCCSM). MATERIALS/METHODS: PCCSM was exposed to SC or GA after appropriate pretreatment with nitric oxide synthase (NOS) blocker, guanylate cyclase blocker, adenylyl cyclase blocker or protein kinase A blocker. Subsequently, we evaluated the cyclic nucleotide in the perfusate by radioimmunoassay, protein expression level of neuronal NOS (nNOS) and endothelial NOS (eNOS) by western blot, and the interaction of SC or GA with udenafil and rolipram. RESULTS: Both SC and GA induce PCCSM relaxations in a concentration-dependent manner. Pretreatment with NOS blocker, guanylate cyclase blocker, adenylyl cyclase blocker or protein kinase A blocker result in significantly decreased relaxation. SC and GA also induce the levels of cyclic nucleotide in the perfusate in a concentration-dependent manner. Perfusion with GA also showed significantly higher levels of eNOS protein. Furthermore, the udenafil and rolipram induced relaxations of PCCSM were enhanced after exposure to SC and GA. Our results indicate that SC and GA induce the relaxation of PCCSM via the nitric oxide (NO)-cGMP and cAMP signaling pathways. CONCLUSIONS: The SC and GA are potential alternative treatments for men who want to consume natural products to ameliorate erectile function, or who do not respond to the commercially available medicines.

소복축어탕(小腹逐瘀湯)이 흰쥐 흉부대동맥의 혈관운동성에 미치는 영향 (Effects of Water Extract of Sobokchuko-tang on Rat Blood Vessels)

  • This study was carried out to examine the relaxational response to the water extract of Sobokchuko-tang (SCT) in rat thoracic aorta. Segments of thoracic aorta obtained from rats immediately after delivery were mounted in organ baths superfused on a polygraph. It was found that the thoracic aorta segments responded to the SCT with dose-dependent vasorelaxation. At $10^{-7}M$ phenylephrine-induced contraction, the contractile response of thoracic aortic rings were inhibited by 99.2%. 75.3%. 42.4% and 35.8% after addition of the respective concentration (1O. 30. 100. $300{\mu}g$/ml) of SCT. This vasorelaxation of rat thoracic aorta was endothelium-dependent. The action mechanisms of vasorelaxation are thought to be concerned with the role of NO. cAMP and cyclooxygenase. but not with cGMP.

서방출형 이산화염소 가스 젤팩 첨가가 닭가슴육 저장 중 지질산패 및 정미성분의 변화 (Changes in Lipid Oxidation and Taste Compounds of Chicken Breast Meat by Slow-released ClO2 Gas Gel-pack during Storage)

  • To extend the shelf-life of chicken breast meat, samples were treated with gel-packs containing slow-released $ClO_2$ gas at 7-15 ppm for 8 days at $4^{\circ}C$. And the changes in lipid oxidation and taste compounds of the samples were investigated. TBARS value of the chicken breast was slightly increased during storage. TBARS value of gas treatments was similar to the control during storage. There were 14 fatty acids in the chicken breast. And there was no change in the fatty acid composition during storage, and there was no significant difference between the control and gas treatments. The content of free amino acids was gradually increased during storage. The content of free amino acids were not significantly different between the control and gas treatments during storage. The content of GMP in the control and gas treatments were decreased during storage. However, gas treatments showed slightly higher content than that of control. AMP was not significantly different between the control and gas treatments. IMP gradually decreased during storage and the content of inosine and hypoxanthine was increased. IMP, inosine and hypoxanthine contents of gas treatment were similar to control, but the control tended to change more rapidly than those of gas treatments.
