저온 플라즈마 처리된 투과증발막의 부탄올 분리/농축 연구 (Butanol Separation/Concentration by Plasma Treated Pervaporation Membrane)

  • 생물 발효 공정에서 생산되는 부탄올 수용액으로부터 부탄올을 농축시키기 위한 투과증발 공정의 flux와 선택도를 향상시키기 위하여 저온 plasma 처리 공정으로 막을 제조하였다. 플라즈마 처리 조건인 공급 power, 반응 시간, 단량체 공급 속도 등에 따른 영향을 조사하여 (W/FM)t의 최적값이 4.0389$\times$10(sup)9 J.min/kg임을 확인하였으며, 이에 따른 최적의 막 제조 공정을 확립하였다. 여러 가지 유기 화합물로서 저온 플라즈마 처리된 막에 대하여 부탄올 분리 성능을 조사하였으며, 플라즈마 처리된 막에 대하여 물과 부탄올의 상대적 sorption 비와 접촉각을 측정하여 표면 분석을 하고자 하였으며, 그 상관 관계를 조사하였다. 접촉각과 상대적 sorption 비가 증가함에 따라 부탄올의 선택도는 0.186에서 3.525로 향상되었고, 부탄올 질량 flux는 0.042에서 0.567 kg/$m^2$.hr로 향상됨을 볼 수 있었는데, 이는 막의 소수서이 증가함에 따라 막과 부탄올과의 친화력이 증가하였기 때문인 것으로 판단된다. 또한 사용된 유기 화합물과 물 또는 부탄올과의 친화력 정도를 알아보기 위해 heat of mixing을 측정하여 분리 성능과 비교하였으나 뚜렷한 경향성을 얻지 못하였다. 이것은 플라즈마 처리 시 유기 화합물이 분해되어 새로운 화합물을 형성하게 되어 유기 화합물 본래의 특성을 잃어버리기 때문인 것으로 생각된다.

  • The effect of positive inotropic agents on the contractile properties of myocardial muscle were studied in the cat papillary muscle preparation. For the purpose, the effects of ouabain $(1{\times}10^{-6}g/ml)$, norepinephrine (0.05r/m1) and Aconiti tuber butanol fraction (AF(5), $1{\times}10^{-4}$, $5{\times}10^{-4}$, $1{\times}10^{-3}$, $2{\times}10^{-3}g/ml$) on the contractile dynamics of the papillary muscle preparation isolated from right ventricle of cat were observed in terms of the characteristics of isometric twitch and the lengh-tension relation, the force-velocity relation and the load-extension relation of the series elastic component of contractile model of A.V. Hill. All the studied inotropic drugs similary increased the rate and the intensity of the developed isometric tension, while shortened the time from onset of contraction to peak tension and the total duration of contraction. In the afterloaded simultaneous isotonic and isometric contraction, they also similary increased the maximal velocity of shortening accompanied with the increasing the maximum developed force. In the load-extension relation all the drugs, however, had no appreciable influence on the properties of the series elastic component. Increasing the concentration, Aconiti tuber butanol fraction produced more pronounced effect on all the studied parameters of isometric and isotonic contraction of cat papillary muscle preparation. From the aspect of contractile dynamics, it seemed that the positive inotropic effect of ouabain, norepinephrine and Aconiti tuber butanol fraction are similary achieved through an influence on the behavior of the contractile component only.

Photo-protective and Anti-melanogenic Effect from Phenolic Compound of Olive Leaf (Olea europaea L. var. Kalamata) Extracts on the Immortalized Human Keratinocytes and B16F1 Melanoma Cells

  • Ethylacetate and butanol fractions of leaf extracts (OLE) showed the higher contents of total phenolic compounds than hexane and water fractions. Oleuropein contents were $4.21{\pm}0.57,\;3.92{\pm}0.43,\;0.32{\pm}0.03,\;5.76{\pm}0.32$, and $32.47{\pm}0.25mg$/100g for ethanol extract, and hexane, chloroform, ethyl acetate, and butanol fraction, respectively. Treatment of ultraviolet-B (UVB) irradiated cells with 3 OLEs prepared by using ethylacetate and butanol at concentrations 0.001, 0.005, and 0.01% respectively showed significant recovery of cell viabilities. Treatment of dexametason 1 mM reduced tumor necrotic factor (TNF)-${\alpha}$ secretion by about 40%. UVB irradiated immortalized human keratinocytes (HaCaT) cells were treated with 3 different OLEs at the same concentrations. Ethylacetate fraction showed the strongest inhibition activity with respect of reduction of the elevated (TNF)-${\alpha}$. Cytotoxicity of OLEs on the B16-F1 cells was evaluated through thiazolyl blue tetrazolium bromide (MTT) assay. Ethylacetate fraction has no cytotoxicity in the range of 0.005-0.01%. A slight cytotoxicity was observed at the concentration of 0.1% butanol fraction of OLE that caused 10% decrease in cell viability.

  • Clostridium.aetobutylicum Bl8에 의한 부탄올 발효에서 농도, extra nutrient, pH대한 부탄올 생성량을 알아보았다. 실험결과 extra nutrient를 주입했을 때 부탄올의 농도가 주입하지 않았던 배양에서보다 약 17%정도 증가하는 결과를 보였고, pH를 control 했을 때에는 glucose 60g/L 회분 실험을 기준으로 부탄올 생산량이 약 41%정도가 증가하는 것으로 나타났다.

Antimicrobial Effectiveness of Pine Needle Extract on Foodborne Illness Bacteria

  • Fresh pine needles were collected and extracted with 95% methanol and the extract was concentrated to determine its antimicrobial activity. The methanol extract had a considerable inhibitory effect on the tested bacteria, such as Esherichia coli 0157;H7, Listeria monocytogenes, Salmonella typhimurium, and Staphylococcus aureus. The methanol extract of pine needles was further fractionated to chloroform, ethylacetate, butanal, and water fractions. Among these four fractions, the butanol and water fractions, which showed a relatively strong inhibitory effect on all of the tested bacteria, were purified and the minimum ingibitory concentration (MIC) was determined for each microorganism. The MIC raged between 25 mg/ml and 45 mg/ml depending on the microorganism. the purified active fractions were applied to sterilized milk as a model food system to define the antimicrobial effectiveness and it was found that the antimicrobial activities in the water fractions were stronger than those in the butanol fractions.

여러 첨가물의 용매가 Bacillus stearothermophilus가 생산하는 Cyclodextrin Glucanotransferase의 열안정성에 미치는 영향 (Effect of Various Additives and Solvents on Thermostability of Cyclodextrin Glucanotransferase from Bacillus stearothermophilus)

  • Ethylene glycol, glycerol, sorbitol 그리고 sucrose가 Bacillus stearothermophilus가 생산하는 cyclodextrin glucanotransferase(CGYase)의 열안정성에 미치는 영향을 조사하였다. Glycerol, sorbitol 그리고 sucrose가 CGTase의 열안정성에 효과가 있었다. 이 효과는 첨가물의 농도와 밀접한 관계가 있었다. n-Butanol, 1,4-dioxane그리고 n-octane과 같은 유기 용매에서 CGTase의 열안정성을 조사하였다. 1,4-dioxane 과 n-octane은 CGTase의 열안정성을 증가시켰다. 특히, n-octane에서 CGTase를 $75^{\circ}C$에서 90분간 정치시킨 후에도 원래 효소활성의 81를 유지하였다.

  • The esterification reaction of previously obtained l,4-sorbitan with acrylic acid using Novozym 435 was carried out in t-butanol as solvent. Immobilized lipase Novozym 435 showed high enzymatic activity at $50^{\circ}C$ in t-butanol and optimum contents of Novozym 435 added in the esterification reaction was 3%(w/v). The maximum conversion rate was 55.8% when initial concentration was 50g/L and conversion rate of this reaction was 63.5% when the molar ratio of l,4-sorbitan to acrylic acid was 1:3.

  • Ten yeast strains affecting doenjang flavor were isolated from soybean fermented foods (traditional meju and doenjang), among which Zygosaccharomyces sp. Y-2-5, showing excellent growth, glucose consumption, pH, and flavor production, was selected. Higher alcohols produced by Zygosaccharomyces sp. Y-2-5 related to flavor were 2-propanol, 1-propanol, 2-methyl-1-propanol, 1-butanol, and 3.3-dimethyl-2-butanol. Optimal culture conditions for Zygosaccharomyces sp. Y-2-5 were 10% (w/v) NaCl, pH 4.0, 3.0% (w/v) glucose concentration, and inoculation time day 0 or 15 doenjang fermentation.

  • To use onion(Allium cepa L.) residue as raw materials of food product, yields of onion-juice according to various extracting methods and optimal extracting condition of quercetin and its related glycosides were carried out. Onion juices gained by the methods of pressing, rotary crushing, freeze pressing and enzyme treatment. The yield by the method of enzyme treatment was higher than others. The yields of juice from fresh onion and heat-treated onion(8$0^{\circ}C$/10min) by pressing were 66% and 83%, respectively. Ethanol extraction of onion was efficient at 75$^{\circ}C$ and acetic acid extraction was proper at 3% concentration for 3 hours at 11$0^{\circ}C$. The onion extract was fractionated in the order of hexane, chloroform, ethylacetate and butanol to test flavonoids. The highest abundant flavonoids were found in ethylacetate and butanol fraction.

동백(Camellia japonica L.)유박 추출물의 항균활성 (Antimicrobial Activity of Defatted Camellia(Camellia japonica L.) Seeds Extract)

  • To developed natural food preservatives, ethanol and water extracts were prepared from the defatted camellia(Camellia japonica L.) seeds. Antimicrobial activities were examined against 14 microorganisms which were food borne pathogens and/or food poisoning microorganisms and food-related bacteria and yeasts. Ethanol extract showed antimicrobial activities against several microorganisms tested, but not on lactic acid bactria. Especially, minimum inhibitory concentrations(MIC) for yeasts were as low as 1mg/ml. Water extract exhibited antimicrobial activities for the yeasts tested, but almost bacteria were not observed. The ethanol extract was fractionated in the order of hexane, chloroform, ethylaetate and butanol fractions to test antimicrobial activity. The strongest activity for the tested yeasts were found in the butanol fractions, but bacteria were chloroform fraction. Hansenula anomala treated with ethanol were observed by scanning electron micrograph, and the results exhibited morphological changes, including the irregularly contracted cell surface and expanded ellipsoidal shape.
