Effects of IAA, IBA, and Media on Growth and Saikosaponin Biosynthesis in Bupleurum falcatum Hairy Root Culture (시호 모상근 배양에서 생장과 시호 사포닌 생합성에 미치는 배지와 IAA 및 IBA의 영향)

  • We examined the effects of various basal media and exogenous auxins on the growth and saikosaponin production in hairy root cultures of Bupleurum falcatum. Of the various media tested, 2RCM medium was superior for growth and saikosaponin content, in which saikosaponin a, c, and d was produced, however MS medium was resulted in extremely low level in saikosaponin production. By the treatment of 0.5 mg/L IBA the growth rate was increased by about 60% in 2RCM medium, whereas both IAA and IBA showed a potent inhibition effect in saikosaponin biosynthesis by the treatment of 0.01 mg/L~5mg/L.

Effect of Siho (Bupleurum falcatum L.) Extracts on Liver Function in $CCl_4$ Intoxicated rats (시호추출물이 간장기능 회복에 미치는 영향)

  • Effect of Siho (Bupleurum falcatum L.) extracts on liver function were investigated in CCl4 intoxicated rats. Plasma $\alpha$-fetoprotein and total protein concentration showed a tendency to decrease in Siho extracts groups. However plasma albumin concentration showed no significantly different in all treatment groups. Activities of plasma asparate aminotransferase and alanine aminotransferase in Siho extracts groups showed a lower values than control group. Alkan phosphatase and lactate dehydrogenase activities showed a tendency to decrease in Siho extracts groups. However ${\gamma}$-glutamyl transferase activity showed no significantly different in all treatment groups. Concentration of plasma triglyceride and total cholesterol showed no significantly different in all treatment groups.

  • This study was conducted to determine the optimum planting dates and density of one year old Bupleurum falcatum L. to improve its quality. Furthermore, the effect of cultural methods and plant growth regulators$(GA_3,\;IAA,\;Kinetin)$ on the quality of B. falcatum were also investigated. In this study, Jeongseon cultivar collected in Korea and Mishima cultivar introduced from Japan were used. Some of the results obtained are as follows. Jeongseon cultivar showed less stem branches and shoot weight compared to Mishima. However, Jeongseon cultivar showed tall plant height, high root fresh and dry weight, and high levels of SSa and TSS, but low SSc content than that of Mishima. Although fresh and dried root weight of both cultivars were not affected by plant growth regulators treatment. compared with the untreated one, increasement of TSS content were made by $GA_3$ 100 ppm, IAA 50 ppm or 10 ppm and kinetin 10 ppm treatment in Jeongseon cultivar, and by $GA_3$ 10 ppm and IAA 100 ppm or 10ppm treatment in Mishima cultivar applied on June 1.

Effects of Irrigation and Sowing Time on Growth and Yield of Bupleurum falcatum L. (시호(柴胡) 생육(生育)과 수량(收量)에 대한 파종기(播種期) 및 초기관수(初期灌水) 효과(效果))

  • When Bupleurum falcatum field was irrigated three times with 20mm at intervals of 10days after sowing, establishment was inproved (64%) and root yield increased to 58kg/10a. And in early growing stage, 30mm irrigation six times at intervals of 10days increased the number of harvested plants per square meter and resulted in yield increase by 26% over yield from natural plot. 20mm irrigation in sowing time at intervals of 10days and 30mm irrigation in early growing stage at intervals of 10days kept adequate soil moisture content (soil moisture tension: $0.39{\sim}0.49bar)$ and resulted in better establishment, growth and yield of Buleurum falcatum.

Characteristics of Flowering and Pollination in Bupleurum falcatum (시호의 개화 및 수분양식)

  • This study was carried out to understand the flowering and pollination characteristics in Bupleurum falcatum. Estimated total node numbers of primary branches were 106.6, and the total numbers of florets were theoretically 9,622 until secondary branches flower. In the sizes of floral organs according to the branch order, all of ray, pedicel, petal, pistil, stamen, ovary surface, and sepals have a tendancy to be smaller as this order: main stem> 1st branch> 2nd branch. As the branch development proceeds, the size of florets was getting smaller. Each flowering periods, especially stamen emergence, in different branch positons and in intra-branch were clearly divided. In case of florets, umbellets, and compound umbels enveloped in the gauze and paper, they showed 0% fertilization rate. However, the plants which were entirely enveloped in gauze led to the partial fertilization. Consequently, it was admitted that B. falcatum was a partly allogamous plant, fertilized in the way of entomophily. In respect of pol­linatability according to the steps of floral organs maturation, this plant was able to bear fruit after 8 days (Pistil maturation). It was at maturity stages, especially 11 through 13 days, that the maximum fer­tilization rate appeared.

Medicinal Components in Bupleurum Species (시호의 약리성분 특성)

  • This review deals briefly with the various medicinal components(mainly saikosaponins), their biological activities and the variation of their contents by different cultivation environment and plant parts in Bupleurum species. Bupleuri radix, a crude drug, is the root of Bupleurum falcatum L. (Korea, Japan), B. chinense(China), and their related species (Umbelliferae). There are over 120 species in Bupleurum genus throughout world, mainly Asian area, and over 5 species in Korea, investigated up to now. These plants contain many physiological active compounds and the principal components are saikosaponins. Major activities of this crude drug and saikosaponins are the anti-inflammatory and antihepatotoxic activities. Saikosaponins and their derivatives in Bupleurum spp. have been chemically studied, isolated and identified over 70 compounds in over 50 species. Other components, physiologically active ones, also have been investigated, which are the groups of lignan, flavonoid, essential oil, polyacetylene, polysaccharide, etc. Saikosaponins belong to the group of triterpenoid saponin chemotaxonomically and occur the accumulation and turnover in plant tissues through secondary metabolism, mevalonic acid pathway. The contents and kinds of saikosaponins and other components in Bupleurum spp. plants are various due to different species and growing environments, as the plant growth characters and yield are various. Most of medicinal plants as well as Bupleurum species are very useful as agricultural products and traditional medicines, and also are very valuable as genetic resources and natural products. So we need to collect, evaluate, preserve, and utilize various medicinal plants, and also to under-stand secondary metabolism and improve the breeding and cultivation techniques for the safe production of crude drugs with high quality and yielding.

  • This experiment was conducted to obtain the basic information for the increase ofgermination percent by the seed diameter, specific gravity, different grain filling period and stratification in Bupleurum falcatum seeds. The seed size of B.falcatum ranged from 2.0 to 3.9mm in diameter, The large seed showed the higher germinability, The optimum specific gravity of salt selection for seed screening was 1.05 resulting in screening out seeds with high germinability over 70%.The seeds needed grain filling period for 60 days or longer after flowering to keep high germinability The optimum period of stratification treatment to promote seed germination was about 80 days.

Effects of Bupleurum falcatum Extract on the Survival of Cancered ICR Mouse and the Growth of Cancer Cells such as J774A.1 Cells and L1210 Cells (시호추출물의 ICR 발암생쥐의 생존율 및 J774A.1 세포와 L1210 세포의 증식에 미치는 영향)

  • The current investigation was carried out to find out the anticancer activity of the methanol extract from Buplerum falcatum against cancered ICR mouse and cancer cell lines such as J774A.1 and L1210 cells. Extract of Buplerum falcatum displayed the considerable augmentation(134%) of the survival of ICR mouse bearing Sarcoma 180 cancer. In addition, the cytotoxic effects of methanol extract of Buplerum falcatum against J774A.1 cells and L1210 cells were found to show $IC_{50}$ values of $57.3\;{\mu}g/ml$ and $54.6\;{\mu}g/ml$, respectively. In contrast to such cytotoxicity against cancer cells, the extract exerted only meagre toxicity against normal lymphocytes. The increased generation of $O_2^-$ and the considerably increased activities of super-oxide dismutase(SOD) and glutathione peroxidase(GPx) of both J774A.1 cells and L1210 cells in the presence of Buplerum falcatum extract implied that the observed cytotoxicities may have resulted from the detrimental effect of reactive oxygen species(ROS) evoked by Buplerum falcatum extract on the cancer cells.
