Effect of sperm penetration of oocytes after in vitro fertilization (IVF) with cauda epididymal spermatozoa in Hanwoo bull after feeding of timothy hay (티모시 건초 급여 한우 씨수소 정소상체 정자의 수정 효과)

  • In this study, we examined effect of sperm penetration of oocytes after in vitro fertilization (IVF) with cauda epididymal spermatozoa in Hanwoo bull after feeding of timothy hay. One testicle with epididymides was castrated from one Hanwoo bull (14 months of age) and spermatozoa recovered from cauda epididymis and cryopreserved. As control, frozen Hanwoo semen was used. Matured cumulus oocyte complexes were co-incubated with frozen-thawed cauda epididymal spermatozoa for 12 or 18 hours. After IVF, presumptive zygotes were cultured in modified synthetic oviductal fluid. In experiment 1, we examined sperm penetration rate at 12 hours of IVF with epididymal sperm. Total penetration rate among cauda epididymis and control was similar(mean${\pm}$standard error, cauda epididymis and control vs. $49.7{\pm}11.3$ and $54.4{\pm}12.8%$). In experiment 2, cleavage and blastocyst developmental rate were evaluated at day 2 and day 8 after IVF for 18 hours. Cleavage rate among cauda epididymis and control was similar(cauda epididymis and control vs. $81.2{\pm}3.4$ and $82.7{\pm}2.5%$). However, blastocyst developmental rate of cauda epididymis group was significantly higher than that of control group(cauda epididymis and control vs. $24.4{\pm}1.6$ and $12.2{\pm}2.8%$, p<0.05). In conclusion, cauda epididymal spermatozoa in Hanwoo bull has high embryo developmental competence and can be used as an alternative to ejaculated frozen sperm in vitro.

Effect of knife castration on leukocyte cytokine expression and indicators of stress, pain, and inflammation in Korean cattle bull calves

  • Objective: This study investigated the effects of surgical castration on behavior, physiological and inflammatory indicators, and leukocyte cytokine mRNA levels in Korean cattle bull calves. Methods: Nineteen Korean cattle bull calves (average body weight, 254.5 kg; average age, 8.2 months) were divided into two treatment groups: control (n = 9) and castration (n = 10). Surgical castration was performed using Newberry knives and a Henderson castrating tool. Blood was obtained just before castration (0 h) and at 0.5 h, 6 h, 1 d, 3 d, 7 d, and 14 d after castration. Plasma cortisol (PC), saliva cortisol (SC), plasma substance P, and plasma haptoglobin concentrations, and the leucocyte mRNA levels of the interleukin-1-alpha (IL1A), interleukin-1-beta (IL1B), interleukin-1 receptor antagonist (IL1RN), and interleukin-6 (IL6) genes were analyzed. Results: Castration decreased (p<0.01) the average daily gain and gain/feed ratio. Castration reduced the time spent eating (p<0.001) and the eating frequency (p<0.01) and increased (p<0.001) the lying frequency. Castration temporarily increased (p<0.05) circulating PC and SC concentrations at 0.5 h after castration. Castration temporarily increased (p<0.05) plasma substance P concentrations at 1 d after castration. Castration increased (p<0.05) plasma haptoglobin concentrations at 1 and 3 d after castration. Castration increased (p<0.05) leukocyte mRNA levels of the IL1A, IL1B, IL1RN, and IL6 genes at 6 h after castration. Conclusion: Castration temporarily induced stress and expression of leucocyte inflammatory cytokine genes in Korean cattle bull calves.

Brodie's Abscess in a Pit Bull Terrier Dog (핏불테리어 견에서 발생한 브로디씨 농양)

  • A 3-year-old Pit bull terrier dog had left hindlimb lameness for 3-weeks. On plain radiographs, geographic osteolysis with well-defined edges and marginal sclerosis was identified in medial aspect of left proximal tibialmetaphysis. Surgical debridement and curettage was performed. Coagulase negative Staphylococcus spp. grew on a culture specimen obtained during surgery, and Brodie's abscess was diagnosed.

A Study on the physicochemical Properties and Palatability of Hanwoo Beef Loin in $1^+$ Grade (한우 $1^+$ 등급육의 이화학적 특성과 기호성에 관한 연구)

  • The physicochemical properties and palatability of Hanwoo(cow, steer, and bull) beef loin(1+ carcass grade) were studied. pH(5.56~5.69), moisture content(67.6~69.0%), crude protein content(17.8~18.7%), and crude fat content(11.9~13.8%) of Hanwoo beef loin did not show significant difference between the types of Hanwoo. L(44.5~46.9), a(19.9~20.6), and b(10.3~11.6) color parameters, water holding capacity (69.4~70.5%), volatile basic nitrogen content(9.86~11.28mg%), thiobarbituric acid value(0.148~0.158 mg malonaldehyde/kg), and cooking loss(26.3~27.9%) of beef loin did not show significant difference between the types of Hanwoo either. Hardness, myofibrillar fragmentation index(MFI), and chewiness of beef loin were 6.1~6.9 dyne/cm2, 50.3~54.9, and 39.5~ 52.0g, respectively. No differences were found in hardness and MFI between the types of Hanwoo, while chewiness of cow was much lower than that of steer and bull(p<0.05). Cow was the best in palatability.

EFFECT OF HOT SEASON ON LIVE SPERMATOZOA WITH INTACT ACROSOME IN HOLSTEIN BULLS

  • Semen characteristics were examined to find the deterioration of the percentage of live spermatozoa with intact acrosome during hot season using 5 Holstein bulls located in Shirnizu-cho Hokkaido Japan. Spermatozoal viability and acrosomal status were observed simultaneously with triple-stain technique for each spermatozoon. Spermatozoa were divided in four categories (live spermatozoa with intact acrosome, live spermatozoa without intact acrosome, dead spermatozoa with intact acrosome and dead spermatozoa without intact acrosome). Bull and collection month had significant effects on semen characteristics (p < 0.01). The percentage of live spermatozoa with intact acrosome and the percentage of live spermatozoa had the lowest least squares mean by collection month in August (72.7% and 76.7%). These two characteristics indicated the obvious deterioration during hot season. But the fluctuation of these two characteristics were not parallel and the differences between the two characteristics were largest during July to September. The present results indicate the necessity for the simultaneous determination of viability and acrosomal status of each Holstein bull's spermatozoa in order to keep fertility above an acceptable minimum level during hot season.

  • Through a floral study and resource investigation of Korean mushrooms during the year 1993, five species of Agaricales, Pseudoclitocybe cyathiformis (Bull.: Fr.) Sing.; Pholiota brunnescens A.H. Smith & K. Hesler; Coprinus angulatus Peck; Rhodophyllus bisporus Hongo; Suillus viscidipes Hongo and one form of Gasteromycetes, Lysurus mokusin (L.: Pers.) Fr.f. sinensis (Lloyd) Kobayashi, were found and described new to Korean flora. Pseudoclitocybe is described as an unrecorded genus to Korea. The color names cited are from Kornerup & Wanscher's Methuen Handbook of Colour (1984). All the specimens are deposited in the RDAGB's and ASIK's herbarium.

Asymmetric Information Spillovers between Trading Volume and Price Changes in Malaysian Futures Market

  • This study aims to examine the dynamics of price changes and trading volume of Kuala Lumpur Options and Financial Futures Exchange (KLOFFE) from 2000 to 2008. With augmented analysis, our results support two hypotheses. First, under information spillover, our findings support noise traders' hypothesis as the time span for variance of past trading volume to cause variance of current return is found to be asymmetric under bull and bear markets. Second, looking at the dynamic relation between volume and volatility of price changes, our findings support Liquidity-Driven Trade hypothesis as past trading volume and subsequent volatility of return exhibit positive correlation. In terms of investors' behavior in response to the news, we find that investors are more risk taking in bull market and more risk reverse in bear market. Our study suggests that investors should adjust their strategy in the futures market in a dynamic manner as the time span of new information arrival is not consistent. Also, uninformed investors with information asymmetry should expect noninformational trading from informed investors to establish their desired positions for better liquid position.

Atrial Flutter with Poor Ventricular Response in a Bull Terrier Dog (불테리어종 개에서 발생한 미약한 심실반응이 있는 심방조동)

  • A six-year-old intact female Bull Terrier dog (weighing 17 kg) was referred to the Veterinary Teaching Hospital with exercise intolerance and occasional syncope as the primary complaint. Diagnostic studies revealed a rare case of atrial flutter with complete heart block and aberrant ventricular activation from the ectopic focus, possibly due to idiopathic dilated cardiomyopathy. Although the dog was responded with cardiac medication for right-sided heart failure and anti-arrhythmic agent (dofetilide), the dog died suddenly of cardiac arrest after 11 months of treatment.
