Effect of Bulblets Size Oriented from Tissue Culture on Growth and Bulb Enlargement of Lilium Oriental Hybrids Grown in Highlands

  • This study was conducted to investigate the effect of bulblet size on growth and bulb enlargement of Lilium Oriental hybrids, such as 'Sorbonne', 'Siberia', and 'Casa Blanca', grown in areas of high altitude (780 m), in 2009. The bulblets were oriented from tissue culture and planted to produce virus-free bulblets. All the three cultivars showed high vitality, with more than 93% producing shoots, even from small bulblets with a bulb circumference (BC) of less than 6 cm. Bulblets with BC 9-10 cm showed an increased height and number of leaves rapidly to induce the phase conversation from vegetative growth to reproductive growth. The bulblets of BC 9-10 cm in 'Sorbonne', 'Siberia', and 'Casa Blanca' had 2.0, 2.9, and 2.8 flower bud sets per plant, respectively. However, the flowers from those bulblets were not adequate as standard bulbs to produce cut-flowers for the domestic market. The larger bulblets showed more prosperous characteristics in growth and development of Lilium Oriental hybrids. Small bulblets had a high occurrence of viruses and leaf blight symptoms during cultivation, indicating the aggravated disease symptoms in the previously infected bulbs. 'Sorbonne' cultivars showed a high rate of enlargement of bulbs, and small bulblets under BC 6 cm produced more than 23% of the standard bulbs of BC 14-16 cm, which could produce cut-flowers for export. However, 'Casa Blanca' bulblets with BC 6-9 cm produced low bulbs of BC 14-16 cm at 21% level. 'Casa Blanca' cultivars had more bulb roots, longer root lengths, and higher weights than 'Sorbonne' and 'Siberia' cultivars. 'Casa Blanca' cultivars with BC over 9-10 cm grew rapidly and produced a high quantity of bulbs. In these results, 'Sorbonne' bulblets of BC 9-10, 'Siberia' bulblets of BC 10-12 cm, and 'Casa Blanca' bulblets of BC 10-12 cm produced 89%, 86%, and 93% of the standard bulbs of BC 14-16 cm, respectively. It is recommended that bulblets larger than the above sizes be used to produce cut-flowers for the export market. Experiment results suggest that production of bulbs larger than BC 18 cm requires bulblets that are larger than BC 12-14 cm of the three cultivars for the highest quality cut-flowers.

  • Amaryllis ($Hippeastrum{\times}johnsonii$) is commonly propagated by three methods: seeds, offset bulblets, and twin scaling. Since the number and size of bulblets produced by these methods are low, we conducted an experiment to evaluate different bulb cutting methods for propagation. For this purpose, bulbs (circumference of 30 - 35 cm) were sectioned or notched into 8, 12, or 16 segments or twin-scaled into 48, 72, or 96 segments for bulblet formation. Our results show that the largest number of bulblets was produced by the twin scaling method, but they take longer to grow to a final, commercial size. Sectioning and notching resulted in larger, but fewer bulblets than twin-scaling. Compared to notching, sectioning provided more space for the bulblets to grow, and is therefore the recommended method. While increasing the number of sections cut from a single bulb resulted in a larger number of bulblets, the diameter of the bulblets decreased. Therefore, sectioning the bulb into 8 segments was the best method for producing an acceptable number of vigorous bulblets.

  • This experiment was conducted to clarify the effects of physiological properties in culture media on the proliferation and the acclimation of bulblets of several kinds of lilies cultured in vitro. Proliferation rate of bulblets from bulblet of Oriental hybrid ‘Marco Polo’, ‘Casa Blanca’ and Asiatic hybrid ‘Jolanda’ was higher in solid MS media compared with bulblets from scale Acclimation rate of regenerated bulblets was high in solid media which was pre-cultured in vitro, compared with liquid media. Sprouting rate of bulblets from bulblet was higher than bulblets from scale.

  • This research was conducted to find the most effective and economical artificial propagation method of Lycoris squamigera among chipping, half-chipping, twin-scaling, coring, scooping and notching, in order to investigate the number of bulblets obtained from one bulb and the number of bulbs treated by a man in a day for artificial propagation. We got 42.2 bulblets from one bulb by twin-scaling, 23.2 bulblets by half-chipping, 18.2 bulblets by chipping, 14.3 bulblets by notching, 1 bulblet by coring and also 1 bulblet by scooping. Although we got the most bulblets by twin-scaling, it took the most time to treat the bulbs for artificial propagation (200 bulbs per day) and the bulblets formed were very small. But 400 bulbs were treated in a day by chipping and the bulblets formed were comparatively large. In addition, machines could be used for chipping, so chipping was considered to be the most effective and economical artificial propagation method.

  • Regeneration and growth of bulblets from bulblet-derived bulb-scale segment of Lilium longiflorum (cv Georgia) were investigated. Bulblets were initiated on bulb scales taken from bulblets on MS medium containing 0.05 mg/L 2,4D with 3% sucrose or 0.02 mg/L 2,4D with 9% sucrose. Benzyladenine promoted the differentiation of bulblets but inhibited the growth of differentiating bulblets. The growth of bulblet was promoted by supplying 1/2 strength 1/2 NH$_4$NO$_3$ concentration in MS medium containing 12% sucrose.

  • Shoot clusters were induced from bulb scales of Lilium longiflorum 'Geogia', and proliferated on medium containing 0.5 mg/L BA and 0.5 mg/L IAA. Thereafter, these shoot clusters were cultured in 5 L air-lift bioreactors to form and grow normal bulblets. Number of bulblets increased on medium with 30 g/L sucrose, but growth of bulblets was effective on medium with 60 g/L sucrose. The number of bulblets from shoot clusters had no differences, though bulblet growth was very effective on medium with between full and double strength of MS salts. The inoculation of 100 g shoot clusters as a cultural material was suitable for formation and growth of bulblets in 5 L bioreactors. Air-lift type was more effective for the formation and growth of bulblets than that in ebb and flood one, and 200∼300 mL$.$min$^{-1}$ injection of air was suitable in growth of bulblets.

Effect of In Vitro Culture conditions on Ex Vitro Sprouting of Bulblets of Oriental Lilium Hybrid cv. Casa Blanca (기내 배양환경이 오리엔탈 백합 '카사블랑카' 의 기외 발아에 미치는 영향)

  • This study was carried out with Lilium oriental hybrid cv. Casa Blanca to observe the effect of in vitro culture conditions on ex vitro sprouting of bulblets. Low temperature (15$^{\circ}C$) inhibited the growth of in vitro bulblets while high temperature ($25^{\circ}C$) enhanced the growth. Bulblets cultured at 15$^{\circ}C$ did not show dormacy while those cultured at 2$0^{\circ}C$ ,$25^{\circ}C$ had a longer dormancy period. High sucrose concentration (9%) induced longer dormancy. Dormancy period was also prolonged in bulblets cultured in vitro at high temperature ($25^{\circ}C$). Dormancy period was more affected by in vitro culture temperature rather than sucrose concentration. Physiological dormancy was released more rapidly when bulblets were cultured at $25^{\circ}C$ for 6 weeks and further transferred at 15$^{\circ}C$ and cultured for another 12 weeks. Treatment of ABA induced the dormancy in Lilium bulblets but when bulblets were subjected to chilling treatment (4$^{\circ}C$ for 8 weeks) nearly 100% sprouting were observed. The medium containing 1.0 mg/L BA or 1.0 mg/L fluridone was also effective to produce non-dormant bulblets.

Effects of Growth Regulators and Sucrose Concentrations on the Bulblet Formation through In Vitro Culture of Scale Segment in Nerine bowdenii (네리네(Nerine bowdenii)의 기내 인편배양시 자구형성에 미치는 생장조절제와 Sucrose 농도의 영향)

  • The twin-scale segments of nerine (Nerine bowdenii) were cultured to investigate the influence of NAA, BA and sucrose concentrations on in vitro bulblet formation. The formation of bulblets from twin-scale segments showed a good response both the percentage of bulblet formation and the number of bulblets per explant on MS medium supplemented with 1mg/L NAA and 2 mg/L BA. Formation of bulblet showed the highest efficiency on medium containing 30g/L, and the formation of bulblets was strongly inhibited on medium containing over 90g/L. When the twin-scale segments formed bulblets were subcultured three times to the same medium by 60 day subculture interval, the number of bulblets per explant was 6.5, 7.3 and 8.2 in order of first, second and third. The bulblets over 3mm in diameter were hypertrophied and rooted after transferring to the hormone-free MS medium. The plantlets over 50mm in height were successfully acclimatized in the soil mixed with the same volume of vermiculite and perlite, and the survival rate was over 95%.

Effect of Inorganic Salts in MS Medium, Sucrose, and Activated Charcoal on Bulblet Formation from in Vitro Bulbscales in Lilium Oriental Hybrid 'Casa Blanca' (MS 배지 무기물, 당 및 활성탄의 농도가 Lilium Oriental Hybrid 'Casa Blanca'의 기내인편으로부터 자구형성에 미치는 영향)

  • The effects of MS salt strength, sucrose, and cultural conditions on bulblet formation and growth were investigated to optimize the conditions for micropropagating bulblets from in vitro bulb scales of Lilium Oriental Hybrid 'Casa Blanca'. There was no difference on bulblet formation in the range of 1/2~2 $\times$ strength of MS salt, but it was inhibited remarkably in 3 $\times$ strength of MS salt. The growth of regenerated bulblets was most stimulated on MS basal medium. Favorable bulblet formation and its growth from bulb scales were achieved when grown on the media with 1 : 2 or 1 : 3 in the ratio of NH$_4$^+ : NO$_3$^- , as well as on MS basal medium (NH$_4$^+ : NO$_3$^- = about l : 2). Therefore, MS basal medium was very suitable for bulblet formation and growth from bulb scales. Bulblet formation was inhibited but its growth was stimulated with increase sucrose concentration in the medium. The growth of regenerated bulblets was very effective on the media with 9~12% sucrose. Addition of activated charcoal (AC) to the medium inhibited bulblet formation from bulb scales, but enhanced the growth of regenerated bulblets. Especially, the medium containing 1 g/L AC was most effective on the growth of bulblets. No difference was found on bulblet formation and growth from bulb scales under light and dark conditions. In vitro micropropagation of L. Oriental Hybrid 'Casa Blanca' was supposed very reasonable to enhance the growth of the bulblets after forming of bulblets from in vitro bulb scales, and then, subculture the bulb scales from the grown bulblets on MS medium with 9% sucrose and 1 g/L AC.

  • The experiment were carried out to establish the optimal condition of bulblet size, low temperature treatment and time of incubation in acclimatization on stem emergence of Lilium oriental hybrids. The large size bulblet was better than middle and small size bulblets in percentage of stem emergence and weight of bulblets. The highest percentage of stem emergence was 95% in large size bulblet 'Acapulco' but small size bulblet of 'Casablanca' was no stem emergence. Low temperature treatment(5℃)for breaking dormancy was needed at least more than 9weeks. Weight of bulblets and percentage of stem emergence was not good in 5℃ treatment for 6 weeks.
