BACKGROUND: Many strawberry growers are utilizing daylength extension by using incandescent bulb or fluorescent lamp to break dormancy of strawberry induced by low temperature and short day conditions. Conventional incandescent bulb and fluorescent lamp consume a lot of electricity and have short longevity. Red light known for most efficient wavelength for daylength extension light of short-day plant and long-day plant. This study was conducted to verify the effects of red light to enhance growth and to increase production of strawberry (Fragaria ${\times}$ ananassa Duch. cvs. "Seolhyang") METHODS AND RESULTS: Three red light (660nm) of 0.70, 0.87, and $1.05{\mu}mol/m^2/s$ (PAR) and conventional incandescent bulb of 40 Lux were treated respectively under the pot experiment. All treatment irradiated from 18:00 to 24:00 for 6 hours. Red light treatment tend to increase leaf stem number, flower stem number, weight of flower stem, crown weight, root weight, and leaf area of strawberry then incandescent bulb treatment. In field experiment, red light of $0.7{\mu}mol/m^2/s$ (PAR) and conventional incandescent bulb of 40 Lux were irradiated respectively. Field experiment showed that the leaf number, leaf weight, and crown weight of strawberry increased than those of incandescent bulb control with red LED of $0.7{\mu}mol/m^2/s$ (PAR). Red LED treatment increased the fruit number over 15g than incandescent bulb. Furthermore, red LED treatment decreased fruit number below 15g of strawberry than incandescent bulb treatment. Therefore, We believed that red LED treatment increased marketable fruit number by increment of weight of each fruit. Consequently, marketable fruit number, fruit weight, and fruit production of strawberry were increased than those of incandescent bulb by 5 %, 2.9 %, and 8.5 % respectively, but not showed significantly differences. CONCLUSION: These results presumably due to directly enhanced photosynthesis of strawberry leaves and activated action of Pfr phytochrome form by red light. In conclusion, red LED of 660nm could be used for daylength extension light source to enhance production of strawberry.
Amaryllis ($Hippeastrum{\times}johnsonii$) is commonly propagated by three methods: seeds, offset bulblets, and twin scaling. Since the number and size of bulblets produced by these methods are low, we conducted an experiment to evaluate different bulb cutting methods for propagation. For this purpose, bulbs (circumference of 30 - 35 cm) were sectioned or notched into 8, 12, or 16 segments or twin-scaled into 48, 72, or 96 segments for bulblet formation. Our results show that the largest number of bulblets was produced by the twin scaling method, but they take longer to grow to a final, commercial size. Sectioning and notching resulted in larger, but fewer bulblets than twin-scaling. Compared to notching, sectioning provided more space for the bulblets to grow, and is therefore the recommended method. While increasing the number of sections cut from a single bulb resulted in a larger number of bulblets, the diameter of the bulblets decreased. Therefore, sectioning the bulb into 8 segments was the best method for producing an acceptable number of vigorous bulblets.
This study was conducted to establish a regeneration system of plantlets through callus culture induced from bulb scales of Lillium‘Gelria’. Friable callus was induced very easily from bulb scales, and grew vigorously on medium lacking growth regulators. In media with 0.5∼ 1.0 mg/L kinetin and 0.1 ∼ 1.0 mg/L NAA, 100% of explants produced callus. Proliferation of callus was actively occurred on media containing 0.1 ∼ 1.0 mg/L kinetin and 0.1 ∼ 1.0 mg/L NAA. Callus proliferation and regeneration of bulblets from callus were occurred simultaneously. Light condition was more effective for the callus proliferation and solid medium was better than liquid medium. Althrough callus was proliferated vigorously on media containing 0.1 ∼ 1.0 mg/L BA and NAA, the frequncy of plantlet regeneration was better on medium without growth regulators, then on medium with 0.1 mg/L BA and NAA.
In this study, we developed a species-specific PCR assay for rapid and accurate detection of three Xanthomonas species, X. axonopodis pv. poinsettiicola (XAP), X. hyacinthi (XH) and X. campestris pv. zantedeschiae (XCZ), based on their draft genome sequences. XAP, XH and XCZ genomes consist of single chromosomes that contain 5,221, 4,395 and 7,986 protein coding genes, respectively. Species-specific primers were designed from variable regions of the draft genome sequence data and assessed by a PCR-based detection method. These primers were also tested for specificity against 17 allied Xanthomonas species as well as against the host DNA and the microbial community of the host surface. Three primer sets were found to be very specific and no amplification product was obtained with the host DNA and the microbial community of the host surface. In addition, a detection limit of $1pg/{\mu}l$ per PCR reaction was detected when these primer sets were used to amplify corresponding bacterial DNAs. Therefore, these primer sets and the developed species-specific PCR assay represent a valuable, sensitive, and rapid diagnostic tool that can be used to detect three specific pathogens at early stages of infection and may help control diseases.
The regeneration and growth of bulblets from the bulb scale segments of Hyacinthus orientalis L. cv. Pink Pearl were more efficient in IBA than IAA at the same concentrations (1.0 and 3.0 mg/l). The normal (base-down) orientation of explants was more effective for bulblet regeneration and root growth than the inverted (base-up) orientation. The growth of bulblets and roots was increased higher in the perlite than the agar medium. These results suggested that the alternate culture system, first cultured in the agar medium for bulblet regeneration, and then in the perlite medium for bulblet growth, may be more useful for efficient in vitro culture of hyacinth (H. orientalils) cv. Pink Pearl.
This study was performed to investigate the role of plant growth substances on the bulbing of cold type of garlic (Allium sativum L. cv. Uiseongmaneul) during long and short day conditions. The change in endogenous plant hormones such as abscisic acid (ABA), jasmonic acid (JA) and sugar contents in leaf blade and sheath was examined during the growth stage from bulb differentiation (starting at April 16) to bulbing (April 24 to May 18) in cold type of garlic. In the long day condition, ABA contents were higher than short day condition and ABA contents of leaf sheath were higher than leaf blade. ABA contents of growth stage in garlic were not changed during growth stage from April 16 to May 2, however it rapidly increased during bulbing (May 2 to May 18). On the other hand, endogenous JA contents in short day condition did not change in long day condition, it increased from April 16 to May 2. JA contents in the leaf sheath (33.85-62.04 $ng{\cdot}g^{-1}$ DW) were higher than leaf blade (15.39-30.04 $ng{\cdot}g^{-1}$ DW). These results showed that garlic bulb differentiation and bulbing was induced by JA in leaf sheath. In long day condition, total sugar contents in the leaf blade were increased from bulb differentiation (April 16) to bulbing (May 4) and it was decreased during bulbing (May 4 to May 18) while the total sugar content in leaf sheath were gradually increased from bulb differentiation (April 16) to bulbing (May 18). In conclusion, our results showed that there is a significant correlation between the bulb development of garlic and hormonal content in the leaf sheath.
Most orchids are propagated by tissue culture. To survey the viral infection of tissue cultured Orchids, total RNA was extracted from in vitro Cymbridium and Phalaenopsis spp. collected from companies producing tissue-cultured orchids, and RT-PCR analysis was conducted with primer pairs specific to Cymbidium mosaic virus (CymMV) and Odontoglossum ring spot virus(ORSV), which are infecting wide range of orchid genera. The bulb size of Cymbidium infected with CymMV and ORSV was compared with healthy one at 10 months after planting in vitro orchids in the glasshouse. The CymMV or ORSV infection in 97 Cymbidium and 55 Phalaenopsis plants was 84.5 and 89.1 %, respectively. Mixed infection was found in 52.6 and 47.3% of Cymbidium and Phalaenopsis tested, whereas virus-free orchids were 15.5 and 10.9%, respectively. The CymMV and ORSV reduced the bulb size by 2.7-50% depending on the cultivars of Cymbidium. The both viruses caused yellowing, mottle and mosaic with or without necrosis in 4 Cymbidium cultivars.
The garlic cultivation area is moved by change of grown environment due to global warming. It is important to predict changes in cultivation area, quantity and quality of each crop. This study was carried out to estimate the yield and response of garlic growth by the rapid temperature changes in the greenhouse with thermostat control system. Seedlings of Namdo garlic were planted on September 27, 2012 and harvested on May 30, 2013. The used treatments for a rapid temperature change in March-April-May were T0 (control): $6.0-10.4-17.2^{\circ}C$, T1: $6.0-5.4(-5)-17.2^{\circ}C$, T2: $6.0-10.4-22.2(+5)^{\circ}C$ and T3: $6.0-5.4(-5)-22.2(+5)^{\circ}C$. Total dried weight per plant of garlic significantly increased by 5.0% for T2, but T1 and T3 decreased by 12.5 and 4.6%, respectively, compared to T0. Total yields of bulb within the temperature change as T2 and T0 increased significantly (p<0.05), as compared to T1. Decreasing temperature significantly (p<0.05) reduced plant height, SPAD reading, crude protein and fiber contents etc., as compared to T0 and T2. ABA contents gradually increased with time but IAA content rapidly decreased. Conclusively, growth and yield of garlic were more affected by decreased temperature than increased temperature at bulb development stage.
Lily symptomless virus (LSV), Lily mottle virus (LMoV), and Cucumber mosaic virus (CMV) are the most prevalent viruses infecting lilies in Korea. Leaf and bulb samples showing characteristic symptoms of virus infection were collected in 2012, and 80 field samples were analyzed by reverse transcription polymerase chain reaction (RT-PCR). The infection frequencies were 79% for LMoV, 5% for LSV, and 3% for CMV. The LMoV coat protein gene was amplified and cloned into the pET21d(+) expression vector to develop serological diagnostic tools to detect LMoV. The resulting carboxy-terminal His-tagged coat proteins were expressed in Escherichia coli strain BL21 (DE3) by induction with IPTG. The recombinant proteins were purified using Ni-NTA agarose beads and used as an antigen to produce polyclonal antibodies in laying hens. The resulting egg yolk immunoglobulin (IgY) specifically recognized LMoV from infected plant tissues in immunoblotting assays and had comparable sensitivity to that of a mammalian antibody. In addition, method of immunocapture RT-PCR using this IgY was developed for sensitive, efficient, and rapid detection of LMoV. Based on these results, large-scale bulb tests and detection of LMoV in epidemiological studies can be performed routinely using this IgY. This is the first report of production of a polyclonal IgY against a plant virus and its use for diagnosis.
These experiments were carried out to examine efficacy of soil solarization for control of pink root disease by means of mulching with transparent polyethylene sheets in the hot season. The effects of soil solarization on incidence of pink-root disease caused by Pyrenochaeta terrestris and on onion growth and on populations of soil fungi were investigated. Solarization was dramatically effective in reducing pink root incidence in onion seedling and harvested onion bulb. A 30-day and 40-day solarization treatment significantly improved seedling survival and increased yield of 'Changnyeong-deago' onion while decreasing incidence of pink root. Populations of soil fungi from fields planted to onion were assayed on selective media. Solarization treatment was effective in reducing populations of P. terrestris, Pythium spp., and Rhizoctonia sp. in soil. Increase of yield of onion bulbs was associated with control of soil-borne pathogenic fungi. Soil solarization had beneficial effects on yield, bulb diameter, or incidence of pink root.
