Comparison Packing Materials for Proper MAP of Mixed Sprout Vegetables (혼합 싹채소의 MAP를 위한 포장재 비교)

  • A comparison was made of the effect of different packaging materials on storability of mixed sprout vegetables (alfalfa, broccoli, cabbage, radish, and red radish) stored at $8^{\circ}C$. They were packaged by several kinds of films that were 50 fm thickness low-density polyethylene film (PE 50), 50 fm thickness polypropylene film (PP 50), $50{\mu}m$ thickness ceramic film (CE 50), $25{\mu}m$ thickness ceramic film (CE 25), $10\sim13{\mu}m$ thickness polyethylene film (wrap), and polyethylene terephthalate box with ventilation hole (box). The fresh weight of mixed sprouts packaged with different materials decreased below to 2% except box packages that decreased around 7% compared with initial weight. The atmospheres that were developed inside the different materials during storage differed significantly. The carbon dioxide and oxygen concentration of packages were kept properly with 5% in CE 25, but the PE 50 and CE 50 treatments showed higher carbon dioxide and lower oxygen concentration. So the off-flavor of mixed sprouts was more severe in the PE 50 and CE 50 treatment. The ethylene concentration of packages showed lowest in box treatment, followed by PP 50, wrap and CE 25 treatment. The overall quality of mixed sprouts was lower in PE 50 and CE 50 than CE 25 treatment after 10 days storage, supposed to be resulted from highest ethylene concentration and lowest oxygen concentration. Conclusionally, the CE 25 film that showed below 1% fresh weight loss, 5% carbon dioxide, 5% oxygen and below 4 ppm ethylene concentration in package may be a proper packaging material for mixed sprout vegetables.

Isolation and Identification of Antagonistic Bacterium Active against Sclerotinia sclerotioum Causing Sclerotinia Rot on Crisphead Lettuce (결구상추 균핵병균(Sclerotinia sclerotioum)에 대한 길항세균의 분리 및 동정)

  • The fungus genus Sclerotinia contains a number of important plant pathogens. Vegetable growers in our country are probably most familiar with Sclerotinia sclerotiorum, the causes of sclerotinia rot on crisphead lettuce. S. sclerotiorum has a wide host range which can include lettuce as well as crops such as broccoli, cabbage, carrots, celery, beans, peppers, potatoes, stocks, and tomato. Some fungicides, including benomyl, are effective in some crops, but not all. So, we isolated a antagonistic bacteria that are active on sclerotinia rot caused by S. sclerotiorum and that can be used to control it. About 702 strains had been isolated from soil around plant roots in the field. Ten strains showed strong antifungal activity against S. sclerotiorum. In pot test for antagonistic activity, A-7 strain showed high control value against the pathogen when compared with others. The strain was, therefore, selected as a biocontrol candidate against sclerotinia rot and its biochemical properties and 16S rDNA sequence was analyzed. The A-7 strain was highly related to Bacillus subtilis and B. amyloliquefaciens. To confirm precise identification, we had performed gyr A gene sequences analysis. Its sequence had 96% similarity with B. amyloliquefaciens. Consequently, the isolate was identified as B. amyloliquefaciens A-7.

Establishment of Analytical Method for Pymetrozine Residues in Crops Using Liquid-Liquid Extraction(LLE) (액-액 분배법을 활용한 작물 중 pymetrozine의 잔류분석법 확립)

  • Polar pesticides like pymetrozine (log $P_{ow}$: -0.18) are known to be difficult to analyze. The analytical method of pymetrozine using hydromatrix included in the official method of KFDA was uncommon and provided ambiguous evidence to confirm both the identity and the quantity. Therefore, precise single residue analytical method was developed in representative crops for using liquid-liquid extraction (LLE). The pymetrozine residue was extracted with methanol from 11 representative crops which comprised apple, blueberry, broccoli, cabbage, cherry, crown daisy, hulled rice, Korean cabbage, potato, rice and watermelon. The extract was purified serially by liquid-liquid extraction (LLE) and silica solid phase extraction (SPE). For rice and hulled rice samples, n-hexane partition was additionally adopted to remove nonpolar interferences, mainly lipids. The residue levels were analyzed by HPLC with DAD, using $C_8$ column. LOQ (limit of quantitation) of pymetroizinie was 1 ng (S/N > 10) and MQL (method quantitation limit) was 0.01 mg/kg. Mean recoveries from 11 crop samples fortified at three levels (MQL, 10 ${\times}$ MQL and 50 ${\times}$ MQL) in triplicate were in the range of 83.1~98.5% with coefficients of variation (CV) of less than 10%, regardless of sample type, which satisfies the criteria of KFDA. The method established in this study could be applied to most of crops as an official and general method for analysis of pymetrozine residue.

Effect of Plant Extracts with Superoxide Dismutase-like Activity on Survival of Fruit Flies under Oxidative Stress (Superoxide Dismutase유사활성을 지닌 식물체가 Oxidative Stress를 받고 있는 초파리의 수명에 미치는 영향)

  • Biological effect of aqueous extracts of 12 plants which showed superoxide dismutase (SOD)-like activity in vitro was evaluated using Drosophila melanogaster. Survival percentage of the flies was a criterion of effect when the flies were exported to paraquat, which generated superoxide anion radical in vivo. When flies were co-administered with paraquat and aqueous extracts of garlic, lettuce, kiwi, and nameko, they showed no defensive effect against of oxygen toxicity. If flies were exposed to 60 mM paraquat after adaptation to feed containing plant extracts with SOD-like activity for 10 days, however, survival percentage of flies fed with phytochemicals was $35{\sim}63%$ while that percentage of flies fed without phytochemical was only 11%. This result indicated that adaptation of flies to plant extracts with SOD-like activity could prevent the flies from oxidative injury. On the other hand, lettuce, kiwi, nameko, onion, persimmon, fern brake and cauliflower showed a reparative effect on an oxidative stress. Dropwort, shiitake, agaric mushroom and broccoli did not show such an effect.

Bacterial Biocontrol of Sprouts through Ethanol and Organic Acids (새싹채소 오염세균의 에탄올과 유기산에 의한 제어)

  • Although bacterial outbreaks from ready-to-eat foods such as sprouts have increased, the information on microbial biocontrol by means of sanitizers is limited. Twenty sprouts of red cabbage, baby radish, alfalfa, and broccoli each were collected from the wholesale markets in Seoul. Ethanol and organic acids including acetic acid, citric acid, and lactic acid were used to control the amount of mesophilic bacteria and Bacillus cereus. Sanitizer mixtures of ethanol and organic acids showed a remarkable reduction of viable counts and, in particular, the sanitizer mixture comprised of 20% ethanol and 1% lactic acid seemed to be best by a reduction of 7~8 log CFU/g on the sprouts after a 10 minute exposure. At the same time, the sanitizer comprised of 20% ethanol and 1% lactic acid showed the same reduction for Escherichia coli, Salmonella typhimurium, B. cereus, Staphylococcus aureus, and Enterococcus faecalis, with more effect toward the Gram-negative bacteria. Sensory evaluation by texture, browning, off-flavour, and overall acceptability seemed better just after treatment when compared to one day after treatment with the sanitizer. Therefore, the sanitizers of ethanol-organic acid might be an effective means to control the bacterial contamination of sprouts in palce of sodium hypochloric acid.

Effects of Crop Rotations on Potato Yield, Soil Chemical and Microbiological Properties in Organic Farming System (윤작이 감자 수량, 토양 화학성 및 미생물 활성에 미치는 영향)

  • The objective of this study was to determine crop rotation effects on potato yield, soil chemical and microbiological properties from a short-term field experiment from 2010 to 2011 in Jeju Island, Korea. Potato cropping systems included continuous and rotation sequences of soybean (Glycine max(L.) Merr.), barley (Hordeum vulgare var. hexastichon), rapeseed (Brassica napus L.) and broccoli (Brassica oleracea var. italica). Crop rotations increased the yields of potato from 31% to 52% compared with continuous potato. Marketable yield of potato was highest under soybean plus rapeseed rotation by $20.97MT\;ha^{-1}$ and lowest under continuous cropping by $11.95MT\;ha^{-1}$. The incidence and severity of scab disease was significantly lower in tubers from crop rotation with soybean plus barley. Differences in marketable tuber yields among rotations were associated with potato scab disease. Especially, incidence and severity of potato scab were strongly correlated with soil pH, exchangeable calcium, and bacteria population of the soil. Crop rotations significantly increased soil pH, available phosphate, exchangeable K and Ca, especially in crop rotations with soybean plus barley or rapeseed. Soil microbial biomass C of crop rotations with soybean plus barley or rapeseed, was also significantly higher compared with monoculture. In conclusion, crop rotation may decrease the incidence of soil-born pathogen by increasing soil chemical properties and soil microbial biomass. Overall, potato crop productivity was generally maintained in rotations that contained soybean plus barley or rapeseed but declined under continuous cropping system.

Sulforaphane-Induced Apoptosis was Regulated by p53 and Caspase-3 Dependent Pathway in Human Chondrosarcoma, HTB-94 (Sulforaphane에 의한 p53 및 caspase-3 의존 신호전달계를 통한 인간 연골암 세포주 HTB-94에서의 세포사멸 기전 연구)

  • Sulforaphane (SFN) is an isothiocyanate, isolated from glucoraphanin in broccoli and other cruciferaous vegetables. Recent studies have revealed that SFN induces anti-proliferation and apoptosis by cell cycle arrest in various cancer cells. In this study, we investigated the effect of SFN induced apoptosis in chondrosarcoma HTB-94 cells. SFN caused suppression of proliferation and apoptosis in a dose-dependent manner as determined by cell phenotype, MTT assay and FACS analysis in HTB-94 cells. Treatment of SFN led to caspase-3 activation and p53 accumulation as determined by Western blot analysis. Also, SFN significantly induced DNA fragmentation and nuclear degradation though activation of caspase-3, as detected by DNA electrophoresis and immunostaining, respectively. Our results indicate that SFN-induced apoptosis was regulated by p53 and caspase-3 dependent pathways. Furthermore, SFN may act as a potent anti-proliferation agent, and as a promising candidate for molecular-targeting chemotherapy against human chondrosarcoma cells.

Factors Affecting Efficiency of Anther Culture and Phenotypic Characteristics of Anther -derived Progeny in Broccoli (녹색꽃양배추 (Brassica oleracea L. var. italica)의 약배양 효율증진과 약유래 계통의 특성)

  • This study was carried out to clarify several factors affecting embryogenesis from anther culture of nine cultivars in Brassica oleracea L. var. italica and to investigate the characteristics of plants derived from anther culture. Androgenesis from anther culture was elevated on the B5 medium supplemented with 0.1mg/L NAA, 0.1mg/L 2.4-D and 10% sucrose. Embryo production in liquid medium was five-fold higher than solid medium. High temperature treatment at 35$^{\circ}C$ for one day before transfer to culture room maintained at $25^{\circ}C$ had effective to induce embryogenesis of cultured anthers but extended treatment at 35$^{\circ}C$ decreased significantly the percent of embryogenesis. Frequency of embryogenesis from cultured anthers exhibited significant difference from 2.8% in 'Green Valiant' to 21% in 'Haisi' as affected by genotypes. Percent of spontaneously dihaploid among regenerated plants from anther culture was ranged from 62 to 74% as affected by the genotypes. Characteristic in relation to plant height, number of leaves and branches, and size of head from anther-derived plants showed differential variation in 'Rokguray' and 'Haisi'. Among these charaters obtained from two cultivars, five lines were selected for early maturity, long plant height and large head. Selected lines were used as breeding meterials for F$_1$ hybrid.

Anti-proliferative Effects of the Isothiocyanate Sulforaphane on the Growth of Human Cervical Carcinoma HeLa Cells (Sulforaphane에 의한 HeLa 인체자궁경부함세포의 증식 억제 기전 연구)

  • Sulforaphane, an isothiocyanate derived from hydrolysis of glucoraphanin in broccoli and other cruciferous vegetables, was shown to induce phase II detoxification enzymes and inhibit chemically induced mammary tumors in rodents. Recently, sulforaphane is known to induce cell cycle arrest and apoptosis in human cancer cells, however its molecular mechanisms are poorly understood. In the present study, we demonstrated that sulforaphane acted to inhibit proliferation and induce morphological changes of human cervical carcinoma HeLa cells. Treatment of HeLa cells with $10{\mu}M\;or\;15{\mu}M$ sulforaphane resulted in significant G2/M cell cycle arrest as determined by flow cytometry. Moreover, $20{\mu}M$ sulforaphane significantly induced the population of sub-G1 cells (9.83 fold of control). This anti-proliferative effect of sulforaphane was accompanied by a marked inhibition of cyclin A and cyclin-dependent kinase (Cdk)4 protein and concomitant induction of Cdc2, Cdk inhibitor p16 and p21. However, sulforaphane did not affect the levels of cyelooxygenases and telomere-regulatory gene products. Although further studies are needed, the present work suggests that sulforaphane may be a potential chemoprevetive/ chemotherapeutic agent for the treatment of human cancer cells.

Changes in Preference for Grape Seed Oil Dressing according to Mirepoix au Maigre Content Level (Mirepoix Au Maigre 함량 수준에 따른 포도씨유 드레싱의 수용도 변화)

  • The purpose of this study was to develop an excellent salad dressing using grape seed oil, which contains high levels of unsaturated fatty acids. First, a vinaigrette was made with grape seed oil and vinegar and divided into five portions. Then, five types of mirepoix au maigre ($M_1,M_2,M_3,M_4,M_5$) were made with apple, onion, carrot, garlic, tomato puree, and tomato ketchup by mixing in a main material vessel. The samples were then allowed to ripen for three days. The control group $M_1$ was named $GD_1$, and the experimental groups $M_2,\;M_3,\;M_4,\;and\;M_5$ were named $GD_2,\;GD_3,\;GD_4,\;and\;GD_5$. respectively. To measure receptiveness, a sensory test was conducted using a 7 point category scale. The test concentrated on flavor, taste, color, agreeability, aftertaste, and overall acceptability: the panel consisted of 15 subjects According to the results, $GD_4$ had the best flavor, color and agreeability. The samples with the most preferred taste and aftertaste were $GD_4\;and\;GD_5$. Between $GD_4\;and\;GD_5$, there were no noticeable differences in taste, aftertaste, or overall acceptability, but differences in color and agreeability did exist. Thus, $GD_4$ proved to be the most favorable product. To determine the best complementing vegetables for $GD_4$ yam($VE_1$), broccoli sprouts($VE_2$), mustard leaves($VE_3$), beet leaves($VE_4$), cucumber($VE_5$), and lettuce(VE6) were selected as samples, and a sensory test was conducted. Each vegetable has its own peculiarities. According to the results, $VE_2$ had the nicest vegetable fragrance recording the highest mean value. The vegetable with the most preferred taste, aftertaste, and agreeability were $VE_2\;and\;VE_6$. $VE_1\;and\;VE_6$ had the nicest color and contained the brightest color as well. The highest overall acceptability was awarded to $VE_2\;and\;VE_6$ and there was no remarkable difference between thorn at a level of 0.05. In conclusion, $VE_2\;and\;VE_6$ were the best complementing vegetables with grape seed oil dressing. However, to commercialize such a dressing, many follow-up studies must be conducted.
