Screening of Genetic Variations in Korean Native Duck using Next-Generation Resequencing Data

  • Korean native ducks (KNDs) continue to have a high preference from consumers due to their excellent meat quality and taste characteristics. However, due to low productivity and fixed plumage color phenotype, it could not secure a large share in the domestic market compared to imported species. In order to improve the market share of KNDs, the genetic characteristics of the breed should be identified and used for improvement and selection. Therefore, this study was conducted to identify the genetic information of colored and white KNDs using next-generation resequencing data and screening for differences between the two groups. As a result of the analysis, the genetic variants that showed significant differences between the colored and white KND groups were mainly identified as mutations related to tyrosine activity. The variants were located in the genes that affect melanin synthesis and regulation, such as EGFR, PDGFRA, and DDR2, and these were reported as the candidate genes related to plumage pigmentation in poultry. Therefore, the results of this study are expected to be useful as a basis for understanding and utilizing the genetic characteristics of KNDs for genetic improvement and selection of white broiler KNDs.

무릎관절 골관절염이 있는 개에서 침술 적용 효과에 대한 역학적 보행분석 1례 (Kinetic Gait Analysis of a Dog with Knee Osteoarthiritis Treated with Acupuncture)

  • Force plate analysis was used to describe the effect of acupuncture on right knee osteoarthritis (OA) in a seven-year-old hound breed dog weighing 27 kg. The acupuncture treatment was performed twice a week for 3 weeks. Ground reaction forces (GRFs) and subjective clinical scores in both hind limbs were obtained prior to and 72 hours after each treatment. Right vertical peak force was increased as the dog was acupunctured. Although their sensitivity was lower than that of the GRF data, the subjective clinical scores reflected an improvement. In addition, vertical peak force of normal left hind limb was also increased during this trial. These results indicate that gait analysis may be valuable objective method in future evaluation of acupuncture for OA of the stifle joint.

Potential influence of κ-casein and β-lactoglobulin genes in genetic association studies of milk quality traits

  • Objective: From a review of published information on genetic association studies, a meta-analysis was conducted to determine the influence of the genes ${\kappa}-casein$ (CSN3) and ${\beta}-lactoglobulin$ (LGB) on milk yield traits in Holstein, Jersey, Brown Swiss, and Fleckvieh. Methods: The GLIMMIX procedure was used to analyze milk production and percentage of protein and fat in milk. Models included the main effects and all their possible two-way interactions; not estimable effects and non-significant (p>0.05) two-way interactions were dropped from the models. The three traits analyzed used Poisson distribution and a log link function and were determined with the Interactive Data Analysis of SAS software. Least square means and multiple mean comparisons were obtained and performed for significant main effects and their interactions (p<0.0255). Results: Interaction of breed by gene showed that Holstein and Fleckvieh were the breeds on which CSN3 ($6.01%{\pm}0.19%$ and $5.98%{\pm}0.22%$), and LGB ($6.02%{\pm}0.19%$ and $5.70%{\pm}0.22%$) have the greatest influence. Interaction of breed by genotype nested in the analyzed gene indicated that Holstein and Jersey showed greater influence of the CSN3 AA genotype, $6.04%{\pm}0.22%$ and $5.59%{\pm}0.31%$ than the other genotypes, while LGB AA genotype had the largest influence on the traits analyzed, $6.05%{\pm}0.20%$ and $5.60%{\pm}0.19%$, respectively. Furthermore, interaction of type of statistical model by genotype nested in the analyzed gene indicated that CSN3 and LGB genes had similar behavior, maintaining a difference of more than 7% across analyzed genotypes. These results could indicate that both Holstein and Jersey have had lower substitution allele effect in selection programs that include CSN3 and LGB genes than Brown Swiss and Fleckvieh. Conclusion: Breed determined which genotypes had the greatest association with analyzed traits. The mixed model based in Bayesian or Ridge Regression was the best alternative to analyze CSN3 and LGB gene effects on milk yield and protein and fat percentages.

다변량 모체효과 모형을 이용한 돼지 경제형질의 분산성분 추정 (Estimation of Variance Component on Swine Economic Traits using Multivariate Maternal Animal Model)

  • 본 연구에서는 2000년부터 2008년 사이에 출생한 Duroc종, Landrace종 및 Yorkshire종 종돈의 총 31,455두의 농장검정 자료를 이용하여 다형질 동물 모형과 다변량 모체효과 모형에 의한 돼지 경제형질의 유전적 분산성분을 추정하였다. 품종별 조사된 각 형질에 있어서 다변량 모체효과 모형을 이용하여 추정된 상가적 효과에 의한 유전력이 모체효과에 의한 유전력보다 모두 높게 나타났으며, 다형질 동물 모형에서 상가적 효과만 고려했을 때의 유전력이 다변량 모체효과 모형에서 추정된 상가적 효과의 의한 유전력보다 대부분 높게 추정되었다. 다변량 모체효과 모형을 이용하여 추정된 모체효과에 의한 육종가와 표현형가와의 상관관계에 있어서 대부분 낮은 정(+)의 상관이거나 강한 부(-)의 상관관계를 보여 품종별 각 형질에 있어서 모체효과를 적절히 고려하여 육종가와 표현형의 상관관계를 높인다면 보다 나은 개량을 도모 할 수 있을 것으로 사료된다.

습지총량제 도입에 따른 문제점과 개선방안에 대한 연구 (Studies on Problems and Improvement of Introducing No Wetland Loss)

  • Wetlands considered to be an important natural resources because they provide biodiversity and habitat for species to breed and survive. In this regard, many countries in the world provide wetlands and have policies and laws to protect them. In Korea we hosted RAMSAR COP-10 at Changwon in 2008 representing Korea's endeavor to wetland protection. But, in the process of development Wetlands are easy target to be lost due its easy access and the laws and regulations to protect them are relatively weak. Thus, this research focused on the introduction of No Wetland Loss(NWL) and we can achieve wetland protection in the economy market ways. Thus, NWL should consider 1) a clear definition of NWL and harmony and consensus of introduction of NWL, 2) considering most wetlands be private we need a financial support for securing wetlands, 3) inventories for wetlands in Korea, draw a line of demarcation, technic to evaluate wetlands, 4) wetland restoration considering function of ecosystem not total amount of wetlands. Wetland protection should be a part of Basic Law of Water Management which in its progress and we need further studies on wetland protection because of watershed management, deserted agricultural paddies, etc.

Identification of the SNP (Single Necleotide Polymorphism) of the Stearoyl-CoA Desaturase (SCD) Associated with Unsaturated Fatty Acid in Hanwoo (Korean Cattle)

  • Fatty acid composition of beef intramuscular tissue is an important trait because high proportions of mono-unsaturated fatty acid are related to favorable beef flavor. In this study, we investigated the effects of genetic factors, such as stearoyl-CoA desaturase (SCD), on beef carcass traits, including fatty acid composition, in the Hanwoo. Analysis of fatty acids in Hanwoo was performed using a breed raised in Gyeonbuk province (n = 395). Compared to the homozygote, the GA, CT, and CT genotypes of exon 5 in the SCD polymorphism showed a higher content of oleic acid (p<0.05) and higher contents of mono-unsaturated fatty acid (p<0.05) and marbling scores (p<0.05) in intramuscular fat. Results of haplotype analysis showed a significant presence of unsaturated fatty acids and marbling score in the $ht1^*ht2$ and $ht2^*ht2$ groups (p<0.05). Furthermore, haplotype effects more powerful than a single gene were also observed. These ht1 and ht2 types also showed a significant difference in unsaturated fatty acids and marbling score, affecting beef flavor in the Hanwoo groups. Therefore, it can be inferred that the ht1 and ht2 types might be valuable new markers for use in improvement of Hanwoo.

생물방제균 Bfacillus subtilis YB-70의 외부 Urease 유전자 도입과 길항력 증강

  • To genetically breed powerful multifunctional antagonistic bacteria, the urease gene of alkalophilic Bacillus pasteurii was transferred into Bacillus subtilis YB-70 which had been selected as a powerful biocontrol agent against root-rotting fungus Fusarium solani. Urease gene was inserted into the HindIII site of pGB215-110 and designated pGU266. The plasmid pGU266 containing urease gene was introduced into the B. subtilis YB-70 by alkali cation transformation system and the urease gene was very stably expressed in the transformant of B. subtilis YB-70(pGU266). The optimal conditions for the transfomation were also evaluated. From the in vitro antibiosis tests against F. solani, the antifungal activity of B. subtilis YB-70 containing urease gene was much efficient than that of the non-transformed strain. Genetic improvement of B. subtilis YB-70 by transfer of urease gene for the efficient control seemed to be responsible for enhanced plant growth and biocontrol efficacy by combining its astibiotic action and ammonia producing ability.

African Indigenous Cattle: Unique Genetic Resources in a Rapidly Changing World

  • At least 150 indigenous African cattle breeds have been named, but the majority of African cattle populations remain largely uncharacterized. As cattle breeds and populations in Africa adapted to various local environmental conditions, they acquired unique features. We know now that the history of African cattle was particularly complex and while several of its episodes remain debated, there is no doubt that African cattle population evolved dramatically over time. Today, we find a mosaic of genetically diverse population from the purest Bos taurus to the nearly pure Bos indicus. African cattle are now found all across the continent, with the exception of the Sahara and the river Congo basin. They are found on the rift valley highlands as well as below sea level in the Afar depression. These unique livestock genetic resources are in danger to disappear rapidly following uncontrolled crossbreeding and breed replacements with exotic breeds. Breeding improvement programs of African indigenous livestock remain too few while paradoxically the demand of livestock products is continually increasing. Many African indigenous breeds are endangered now, and their unique adaptive traits may be lost forever. This paper reviews the unique known characteristics of indigenous African cattle populations while describing the opportunities, the necessity and urgency to understand and utilize these resources to respond to the needs of the people of the continent and to the benefit of African farmers.

Biodiversity and Conservation of Indian Sheep Genetic Resources - An Overview -

  • Indian subcontinent is a rich source of diverse ovine germplasm, and only very few countries have such a large number of breeds with wide genetic diversity. This vast ovine biodiversity in India is being eroded rapidly and more than 50% of sheep breeds are currently under threat. It is noteworthy that the characterization of Indian sheep breeds was last done in the first half of the century since then no recent estimates are available and surveys in majority of the regions/breeds are far from complete. Starting in 1985 National Bureau of Animal Genetic Resources (NBAGR), Karnal, India, therefore, initiated activities aimed at determining the status and compiling information on indigenous farm animals including sheep. This report represents attempts made to date on the basis of field/literature surveys and additional activities on molecular characterization to ascertain their status including distribution, population changes, breed diversities and risk status. The need, mode and mechanisms of conservation are also described. Involvement of several agencies for evaluation, improvement, conservation programmes and recommendations made for effective characterization and conservation of sheep biodiversity are highlighted. This publication would promote action particularly at national level to improve the information base on domestic Indian breeds of sheep and provide input into national domestic sheep diversity conservation policy decisions.

Application of genomics into rice breeding

  • By the progress of genome sequencing, infrastructures for marker-assisted breeding (MAB) of rice came to be established. Fine mapping and gene isolation have been conducted using the breeding materials derived from natural variations and artificial mutants. Such genetic analysis by the genome-wide dense markers provided us the knowledge about the many genes controlling important traits. We identified several genes or quantitative trait loci (QTL) for heading date, blast resistance, eating quality, high-temperature stress tolerance, and so on. NILs of each gene controlling heading date contribute to elongate the rice harvest period. Determination of precise gene location of blast resistance gene pi21, allowed us to overcome linkage drag, co-introduction of undesirable eating quality. We could also breed the first practical rice cultivar in Japan with a brown planthopper resistance gene bph11 in the genetic back-ground of an elite cultivar. Discovery of major and minor QTLs for good eating quality allowed us to fine-tune of eating quality according to the rice planting area or usage of rice grain. Many rice cultivars have bred efficiently by MAB for several traits, or by marker-assisted backcross breeding through chromosome segment substitution lines (CSSLs) using genetically diverse accessions. We are also systematically supporting the crop breeding of other sectors by MAB or by providing resources such as CSSLs. It is possible to pyramid many genes for important traits by using MAB, but is still difficult to improve the yielding ability. We are performing a Genomic Selection (GS) for improvement of rice biomass and grain yield. We are also trying to apply the genome editing technology for high yield rice breeding.
