Extraction of seven major compounds from Agastache rugosa (Fisch. & C.A.Mey.) Kuntze: optimization study using response surface methodology

  • The purpose of this study is to demonstrate the potential enhancement of the flavonoid contents from Agastache rugosa, which can be obtained as raw materials for functional products in the food medicine industry by identifying important factors for efficient preparation to save costs and time in terms of economic factors. For this reason, response surface methodology using Box-Behnken design was used to optimize the extraction conditions for the maximum yield of seven major compounds from A. rugosa. The optimum conditions were obtained with an ethanol concentration of 60.0%, a temperature of 50 ℃, and an extraction time of 33.6 min, meaning that the regression analysis fits the experimental data well. Under these conditions, the seven major compounds 1-7 had observed values of 2.169, 2.135, 0.697, 2.485, 0.105, 1.247, and 0.551%, respectively. These results show that the observed values are in good agreement with the predicted values in the regression model. This process for optimization study exhibited a basic protocol for obtaining stable ingredients from A. rugosa that are appropriate for the development of effective functional products.

Experimental and statistical investigation of torque coefficient in optimized surface piercing propeller

  • The interaction of the blade of surface-piercing propellers (SPPs) with the water/air surface is a physical phenomenon that is difficult to model mathematically, so that such propellers are usually designed using empirical approaches. In this paper, a newly developed mechanism for measuring the torque of SPPs in an open water circuit is presented. The mechanism includes a single-component load cell and a deformable torque sensor to detect the forces exerted on the propeller. Deformations in the sensor elements lead to changes in the strain gauge resistance, which are converted into voltage using a Wheatstone bridge. The amplified signal is then recorded by a 16-channel data recording system. The mechanism is calibrated using a 6-DoF calibration system and a Box-Behnken design, achieving 99% accuracy through multivariate regression and ANOVA. Finally, the results of performance tests on a 4-blade propeller were presented in the form of changes in the torque coefficient as a function of feed rate. The results show that the new mechanism is 8% more accurate than conventional empirical methods.

Laser micro-drilling of CNT reinforced polymer nanocomposite: A parametric study using RSM and APSO

  • The present experimental investigation focuses on finding optimal parametric data-set of laser micro-drilling operation with minimum taper and Heat-affected zone during laser micro-drilling of Carbon Nanotube/Epoxy-based composite materials. Experiments have been conducted as per Box-Behnken design (BBD) techniques considering cutting speed, lamp current, pulse frequency and air pressure as input process parameters. Then, the relationship between control parameters and output responses is developed using second-order nonlinear regression models. The analysis of variance test has also been performed to check the adequacy of the developed mathematical model. Using the Response Surface Methodology (RSM) and an Accelerated particle swarm optimization (APSO) technique, optimum process parameters are evaluated and compared. Moreover, confirmation tests are conducted with the optimal parameter settings obtained from RSM and APSO and improvement in performance parameter is noticed in each case. The optimal process parameter setting obtained from predictive RSM based APSO techniques are speed=150 (m/s), current=22 (amp), pulse frequency (3 kHz), Air pressure (1 kg/cm2) for Taper and speed=150 (m/s), current=22 (amp), pulse frequency (3 kHz), air pressure (3 kg/cm2) for HAZ. From the confirmatory experimental result, it is observed that the APSO metaheuristic algorithm performs efficiently for optimizing the responses during laser micro-drilling process of nanocomposites both in individual and multi-objective optimization.

Comparison of response surface methods for the optimization of an upflow anaerobic sludge blanket for the treatment of slaughterhouse wastewater

  • This study was aimed at using the Central Composite Design (CCD) and Box-Behnken Design (BBD) to compare the efficiency and to elucidate the main interacting parameters in the upflow anaerobic sludge blanket (UASB) reactor, namely: Organic Loading Rate (OLR), Hydraulic Retention Times (HRT) and pH at a constant temperature of 35℃. Optimum HRT (15 h), OLR (3.5 kg.m-3.d-1) and pH (7) resulted in biogas production of 5,800 mL/d and COD removal of 80.8%. BBD produced a higher desirability efficiency of 94% as compared to the CCD which was 92%. The regression quadratic models developed with high R2 values of 0.961 and 0.978 for both CCD and BBD, respectively, demonstrated that the interaction models could be used to pilot the design space. BBD model developed was more reliable with a higher prediction of biogas production (5,955.4 ± 225.3 mL/d) and COD removal (81.5 ± 1.014%), much close to the experimental results at a 95% confidence level. CCD model predictions was greater in terms of COD removal (82.6 ± 1.06% > 80.8%) and biogas production (4,636.31 mL/d ± 439.81 < 5,800 mL/d) which was less than the experimental results. Therefore, RSM can be adapted for optimizing various wastewater treatment processes.

양돈용 사료 첨가제 개발을 위하여 구기자 부산물로부터 메탄올수용액을 이용한 총 폴리페놀 추출조건 최적화 (Optimization of Aqueous Methanol Extraction Condition of Total Polyphenol from Spent $Lycium$ $chinense$ Miller to Develop Feed Additives for Pig)

  • This study was conducted to develop a functional feed additive for pig with spent $Lycium$ $chinense$ Mill fruit. We investigated the optimum conditions for the extraction of polyphenol from spent $Lycium$ $chinense$ Mill using methanol. Methanol concentration as a solvent for extraction, extraction time and the volume of solvent per a gram of solid (ground spent Lyceum chinense Mill) were selected as parameters. Three levels of parameters were configured according to Box Behnken experiment design, a fractional factorial design, and total 15 trials were employed. Total polyphenol concentration from each trial was used as response from experiment system and effects of parameters on total polyphenol extraction efficiency were determined using response surface model. As a result, all terms in analysis of variance, regression ($p$ = 0.001), linear ($p$ = 0.002), square ($p$ = 0.017) and interaction ($p$ = 0.047) was significant and adjusted determination coefficient ($R^2$) was 94.7%. Total polyphenol extraction efficiency was elevated along increased methanol content and decreased solvent to solid ratio. However extraction time did not affect the efficiency. This study provides a primary information for the optimum extraction conditions to maximize total polyphenol recovery from spent Lycium chinens Mill fruit and this result could be applied to re-use of argo-industrial by-products and to develop of functional feed additives in organic farming.

Optimization of the whole extract of Zarawand Mudaharaj (Aristolochia rotunda L.) root by Response Surface Methodology (RSM)

  • The chemical constitution of a drug has been accepted as an important basis for pharmacological action in Unani medicine. Various dosage forms have been developed on this concept, such as decoctions (Joshanda), infusions (Khesanda), extract (Rub / Usara), and syrup. Zarawand Mudaharaj (ZM.) / Aristolochia rotunda L. root was subjected to extraction process using Soxhlet's apparatus by using Response Surface Methodology (RSM) to design the number of random runs of the extracts with variation in the factors of temperature, the concentration of ethanol in water, time for extraction, for optimizing and maximizing the yield concentration. The data obtained, was analyzed with regression equation and ANOVA two-way summary to interpret the interaction of the factors for yield maximization. Minitab version 18 was used to design and analyze data. Validation of the optimum conditions for maximum yield of the whole extract of ZM. Root was carried out by re-run of the extract using the optimized conditions. The maximum yield percentage thus obtained using RSM was 20.87% whereas using these optimum conditions 21.35 % yield was obtained thereby validating the method. The association between the response functions and the process variables was identified by a three-factor recorded Box-Behnken design. In the present study RSM is used because itis a cheap and affordable method to optimize maximum yield percentage which may be reliably used by researchers. The study set in the surface conditions for ZM. root extraction by the Soxhlet apparatus for maximizing the yield percentage.

UV 공정을 이용한 N-Nitrosodimethylamine (NDMA) 광분해 및 부산물 생성에 관한 연구: 박스-벤켄법 실험계획법을 이용한 통계학적 분해특성평가 및 반응모델 수립 (A study on the Degradation and By-products Formation of NDMA by the Photolysis with UV: Setup of Reaction Models and Assessment of Decomposition Characteristics by the Statistical Design of Experiment (DOE) based on the Box-Behnken Technique)

  • 본 연구는 광분해 산화공정으로 난분해성 물질인 N-Nitrosodimethylamine (NDMA)인 제거 및 부산물 생성 특성을 파악하기 위한 3개의 독립변수 (자외선 강도($X_1:\;1.5{\sim}4.5\;mW/cm^2$, 초기 NDMA 농도($X_2:\;100{\sim}300\;uM$), pH(X3:3~9))와 4개의 종속변수(NDMA 제거율($Y_1$), dimethylamine (DMA) 생성농도($Y_2$), dimethylformamide (DMF) 생성농도($Y_3$) 및 $NO_2$-N 생성농도($Y_4$))로 구성된 박스-벤켄 설계를 이용한 실험계획을 적용시켜 예측 모델과 광분해 산화 최적조건을 수립하였다. 실험결과 2시간 광분해 후 NDMA는 거의 완전히 제거되었으며 DMA, DMF와 $NO_2$-N은 NDMA 광분해와 동시에 부산물로 생성되었다. 광분해 최적의 조건을 얻기 위해 정준분석을 수행하여 최적 점 (반응값, 독립변수 조건)과 예측반응모델을 수립한 결과, 다음과 같은 결과를 얻었다 ($Y_1=117+21X_1-0.3X_2-17.2X_3+{2.43X_1}^2+{0.001X_2}^2+{3.2X_3}^2-0.08X_1X_2-1.6X_1X_3-0.05X_2X_3$ ($R^2$ = 96%, Adjusted $R^2$ = 88%)와 99.3% ($X_1:\;4.5\;mW/cm^2$, $X_2:\;190\;uM$, $X_3:\;3.2$), $Y_2=-101+18.5X_1+0.4X_2+21X_3-{3.3X_1}^2-{0.01X_2}^2-{1.5X_3}^2-0.01X_1X_2-0.07X_1X_3-0.01X_2X_3$ ($R^2$= 99.4%, 수정 $R^2$ = 95.7%)와 35.2 uM ($X_1:\;3\;mW/cm^2$, $X_2:\;220\;uM$, $X_3:\;6.3$), $Y_3=-6.2+0.2X_1+0.02X_2+2X_3-{0.26X_1}^2-{0.01X_2}^2-{0.2X_3}^2-0.004X_1X_2+0.1X_1X_3-0.02X_2X_3$ ($R^2$= 98%, 수정 $R^2$ = 94.4%)와 3.7 uM ($X_1:\;4.5\;mW/cm^2$, $X_2:\;290\;uM$, $X_3:\;6.2$), $Y_4=-25+12.2X_1+0.15X_2+7.8X_3+{1.1X_1}^2+{0.001X_2}^2-{0.34X_3}^2+0.01X_1X_2+0.08X_1X_3-3.4X_2X_3$ ($R^2$= 98.5%, 수정 $R^2$ = 95.7%)와 74.5 uM ($X_1:\;4.5\;mW/cm^2$, $X_2:\;220\;uM$, $X_3:\;3.1$). 반응표면분석법 중 하나인 박스-벤켄법은 UV 광분해에 의한 NDMA 분해 및 부산물 생성에 대한 통계학적 및 수학적인 결과 및 최적의 운전조건을 제시하였다. 예측모델의 검정을 통하여 박스-벤켄법은 매우 높은 신뢰성을 보였다.

Application of Factorial Experimental Designs for Optimization of Cyclosporin A Production by Tolypocladium inflatum in Submerged Culture

  • A sequential optimization strategy based on statistical experimental designs was employed to enhance the production of cyclosporin A (CyA) by Tolypocladium inflatum DSMZ 915 in a submerged culture. A 2-level Plackett-Burman design was used to screen the bioprocess parameters significantly influencing CyA production. Among the 11 variables tested, sucrose, ammonium sulfate, and soluble starch were selected, owing to their significant positive effect on CyA production. A response surface methodology (RSM) involving a 3-level Box-Behnken design was adopted to acquire the best process conditions. Thus, a polynomial model was created to correlate the relationship between the three variables and the CyA yield, and the optimal combination of the major media constituents for cyclosporin A production, evaluated using the nonlinear optimization algorithm of EXCEL-Solver, was as follows (g/l): sucrose, 20; starch, 20; and ammonium sulfate, 10. The predicted optimum CyA yield was 113 mg/l, which was 2-fold the amount obtained with the basal medium. Experimental verification of the predicted model resulted in a CyA yield of 110 mg/l, representing 97% of the theoretically calculated yield.

Optimization of Tannase Production by Aspergillus niger in Solid-State Packed-Bed Bioreactor

  • Tannin acyl hydrolase, also known as tannase, is an enzyme with important applications in the food, feed, pharmaceutical, and chemical industries. However, despite a growing interest in the catalytic properties of tannase, its practical use is very limited owing to high production costs. Several studies have already demonstrated the advantages of solid-state fermentation (SSF) for the production of fungal tannase, yet the optimal conditions for enzyme production strongly depend on the microbial strain utilized. Therefore, the aim of this study was to improve the tannase production by a locally isolated A. niger strain in an SSF system. The SSF was carried out in packed-bed bioreactors using polyurethane foam as an inert support impregnated with defined culture media. The process parameters influencing the enzyme production were identified using a Plackett-Burman design, where the substrate concentration, initial pH, and incubation temperature were determined as the most significant. These parameters were then further optimized using a Box-Behnken design. The maximum tannase production was obtained with a high tannic acid concentration (50 g/l), relatively low incubation temperature ($30^{\circ}C$), and unique low initial pH (4.0). The statistical strategy aided in increasing the enzyme activity nearly 1.97-fold, from 4,030 to 7,955 U/l. Consequently, these findings can lead to the development of a fermentation system that is able to produce large amounts of tannase in economical, compact, and scalable reactors.

A Dilute-and-Shoot LC-MS/MS Method for Screening of 43 Cardiovascular Drugs in Human Urine

  • A simple, specific, and economical LC-MS/MS method was investigated for the screening of 43 prescribed antihypertensive and related drugs in human urine. The urine samples were simply prepared by diluting and mixing with internal standard before directly introduced to the LC-MS/MS system, which is fast, straightforward, and cost-effective. Fractional factorial, Box-Behnken, and I-optimal design were applied to screen and optimize the mass spectrometric and chromatographic factors. The analysis was carried out on a triple quadrupole mass spectrometer system utilizing multiple reaction monitoring with positive and negative electrospray ionization method. Chromatographic separation was performed on a Thermo Scientific Accucore RP-MS column (50 × 3.0 mm ID., 2.6 ㎛) using two separate gradient elution programs established with the same mobile phases. Chromatographic separation was performed within 12 min. The optimal method was validated based on FDA guideline. The results indicated that the assay was specific, reproducible, and sensitive with the limit of detection from 0.1 to 50.0 ㎍/L. The method was linear for all analytes with coefficient of determination ranging from 0.9870 to 0.9981. The intra-assay precision was from 1.44 to 19.87% and the inter-assay precision was between 2.69 and 18.54% with the recovery rate ranges from 84.54 to 119.78% for all drugs measured. All analytes in urine samples were stable for 24 h at 25℃, and for 2 weeks at -60℃. The developed method improves on currently existing methods by including larger number of cardiovascular medications and better sensitivity of 12 analytes.
