Physicochemical and Sensory Characteristics of Beef-bone Broths Prepared under Atmospheric Pressure and Overpressure (상압과 가압조건에서 제조한 소뼈 용출액의 이화학 및 관능 특성)

  • This study was conducted to compare the physicochemical and sensory characteristics of beef-bone broths prepared under atmospheric pressure (BBAP) and overpressure (BBOP). Beef-bone was boiled in water (bone/water=1:2, w/w) for 1, 2, 4, 6, and 12 h under atmospheric pressure and overpressure ($121^{\circ}C$, 1.25 atm). The BBOP broth samples were found to contain significantly higher amounts of solid, crude protein, crude ash, collagen, and chondroitin sulfate than the BBAP broth samples for a given boiling time (p<0.05). In addition, the Ca and Mg contents in the BBAP samples were higher than those in the BBOP samples, whereas the P, Na, and K contents were higher in BBOP than BBAP. The L value of BBAP was also significantly higher than that of BBOP (p<0.05). Further, as the boiling time increased, turbidity increased in the BBAP samples. In the quantitative descriptive analysis, the BBOP samples exhibited stronger brownness, transparency, meaty off-odor, meaty off-flavor, and sulfuric odor than the BBAP samples.

A Study on the Bioavailability of Dietary Calcium Sources (칼슘급원식품의 체내이용성 연구)

  • This study was conducted to investigate the effect of various types of calcium sources on calcium metabolism. Sprague-Dawley male rate weighing approximately 82g were divided into 6 groups and fed experimental diets containing about 0.2% calcium for 4 weeks. Perilla leaves, dried sea mustard, mulberry leaves, loach, skim milk powder, and CaCO3 were used as calcium sources for this study. Food intake of experimental groups showed no significant difference from that of control group, but food efficiency ratio were higher in group fed loach as a calcium source. Apparent calcium absorption from perilla leaves, and skim milk powder groups as good as that in CaCO group. Femur length showed no significant difference among exjerimental groups with different calcium sources. The breaking force of bone was higher in loach and dried sea mustard groups. Weight, ash weight, and calcium content of the femur were higher in the loach diet group than in the others. Thus, calcium from not only skim milk powder but also perilla leaves, dried sea mustard, mulberry leaves, and loach appears readily available and all of these can be recommended as calcium sources.

  • By extracting the sample of Ulmus davidiana Planch(Ulmaceae), which was known to have the protection of damaged organ and the anti-inflammation action, it was experimented whether it is available for the application of treatment of osteoporosis. In the previous experiment, the extracts from Ulmus davidiana Planch(Ulmaceae) were confirmed to inhibit Cathepsin K through treating the cell of long bone, which contains osteoclast. Through this, it is suggested that Ulmus davidiana Planch(Ulmaceae) can play a role of prodrug as an inhibitor of absorbing bone ash in the treatment of osteoporosis. In the present experiment, a research in vitro Ulmus davidiana Planch(Ulmaceae) on the growth and sensibilization of osteoblast in a state that induced osteosis by using the cell tissue of MC3T3-El pre-osteoblastic was conducted. As a result, it could be confirmed that Ulmus davidiana Planch(Ulmaceae) has the strengthening function by enhancing the dosage and the activity of ALP depending on the time. The dosage was observed at the minimum of $50{\mu}g/m{\ell}$ and the maximum of $150{\mu}g/m{\ell}$. The enhancement in bone morphogenetic protein-2 at $100{\mu}g/m{\ell}$ UD could be observed, and it also increased the concentration of ALP mRNA within the cell of MC3T3-El. At $60{\mu}g/m{\ell}$ UD which indicated a little increase in Type I collagen mRNA for a long time of culture. However, it was shown to sharply inhibit the expression of gene in the culture between 15-20 days. These results suggest that Ulmus davidiana Planch(Ulmaceae) has an influence upon bone metabolism through thje sensibilization of osteoblast. Therefore, it could be known that utilized Ulmus davidiana Planch(Ulmaceae) can be positively applied for the general disease of bone metabolism through future studies.

Effects of antler, red ginseng, safflower, ipriflavone and estrogen on body weight, bone mineral density, organ weight and histological change in ovariectomized rats (녹용, 홍삼, 홍화, ipriflavone 및 estrogen이 난소적출 rat의 체중, 골밀도, 장기중량 및 조직학적 변화에 관한 연구)

  • This study was peformed to elucidate the effects of antler, red ginseng, safflower seed, ipriflavone and estrogen on ovariectomized rats. The rats were fed with Ca and P deficient diet for five weeks to induce osteoporosis. After this period, these animals were fed with normal for and treated every other day with antler(600mg/kg, PO), red ginseng(200mg/kg, PO), safflower(200mg/kg, PO), ipriflavone(80mg/kg, PO) and estrogene(400$\mu\textrm{g}$/kg, IM) for S weeks. During the treatment, the rats were examined for body, organ, femur and femur ash weights. And electron-microscopical examinations were also performed on femora. The results are summarized as follows : 1. The body weights of the antler, red ginseng, safflower, ipriflavone and estrogen-treated groups after five weeks of treatment, were 303.0$\pm$3.0g, 273.3$\pm$4.4g, 288.3$\pm$5.1g, 300.5$\pm$5.2g, 320.5$\pm$4.2g, respectively. They, except the red ginseng-treated group, were significantly higher than 278.4$\pm$2.7g of normal control group(P<0.05). And the body weight of non-treatment group was 29it.2$\pm$5.5g which was higher than that of control group, too. 2. The weights of left femur after five weeks of treatment were showed 0.83$\pm$0.2g by antler, 0.89$\pm$0.2g by red ginseng, 0.87$\pm$0.2g by safflower 0.83$\pm$0.1g by ipriflavone and 0.78$\pm$0.1g by estrogen-treated group. They were a little lower than 0.97$\pm$0.1g of normal control group. Non-treatment group showed 0.83$\pm$0.1g which was lower than that of control group but not significant. 3. The weights of ashed left femora after five weeks of treatment were showed 0.36$\pm$0.01g by antler, 0.40$\pm$0.01g by red ginseng, 0.41$\pm$0.01g by safflower, 0.36$\pm$0.01g by ipriflavone and 0.35$\pm$0.01g by estrogen-treated group. Those results were significantly lower than 0.43$\pm$0.01g of normal control group. And non-treatment group showed 0.29$\pm$0.01g of femoral ash which was significantly lower than that showed by control group(p<0.05). 4. The femoral Ca contents of the antler, red ginseng, safflower, ipriflavone and estrogen-treated rats were 19.6$\pm$0.7%, 17.8$\pm$1.1%, 18.8$\pm$0.9%, 18.0$\pm$1.1%, 15.3$\pm$1.1%, respectively, after five weeks of treatment. The femoral Ca contents of antler and safflower-treated groups were higher than that of control group which showed 18.2$\pm$0.9% of Ca The non-treatment group, showing 15.3$\pm$0.7%, had significantly lower content of Ca than normal group(p<0.05). 5. The weights of liver, spleen, and kidney in the osteoporosis-induced rats during the treatment with antler, red ginseng, safflower, jpriflavone and estrogen showed not significant changes. 6. The connections among the trabeculae of control group were maintained well, showing no bone loss. However, the connections among the trabeculae of non-treatment group were thinner than those of control group and were almost disconnected and the lacunae were found to be broadened. Antler and safflower-treated group showed only slight changes. The recovery rates were prominent in antler, safflower, estrogen-treated groups but they were insignificant in red ginseng and ipriflavone-treated groups.

Evaluation of Relative Bioavailability of 25-Hydroxycholecalciferol to Cholecalciferol for Broiler Chickens

  • This study was conducted to evaluate the relative bioavailability (RBV) of 25-hydroxycholecalciferol (25-OH-$D_3$) to cholecalciferol (vitamin $D_3$) in 1- to 21-d-old broiler chickens fed with calcium (Ca)- and phosphorus (P)-deficient diets. On the day of hatch, 450 female Ross 308 broiler chickens were assigned to nine treatments, with five replicates of ten birds each. The basal diet contained 0.50% Ca and 0.25% non-phytate phosphorus (NPP) and was not supplemented with vitamin D. Vitamin $D_3$ was fed at 0, 2.5, 5.0, 10.0, and $20.0{\mu}g/kg$, and 25-OH-$D_3$ was fed at 1.25, 2.5, 5.0, and $10.0{\mu}g/kg$. The RBV of 25-OH-$D_3$ was determined using vitamin $D_3$ as the standard source by the slope ratio method. Vitamin $D_3$ and 25-OH-$D_3$ intake was used as the independent variable for regression analysis. The linear relationships between the level of vitamin $D_3$ or 25-OH-$D_3$ and body weight gain (BWG) and the weight, length, ash weight, and the percentage of ash, Ca, and P in femur, tibia, and metatarsus of broiler chickens were observed. Using BWG as the criterion, the RBV value of 25-OH-$D_3$ to vitamin $D_3$ was 1.85. Using the mineralization of the femur, tibia, and metatarsus as criteria, the RBV of 25-OH-$D_3$ to vitamin $D_3$ ranged from 1.82 to 2.45, 1.86 to 2.52, and 1.65 to 2.05, respectively. These data indicate that 25-OH-$D_3$ is approximately 2.03 times as active as vitamin $D_3$ in promoting growth performance and bone mineralization in broiler chicken diets.

Dietary Calcium and Non-phytin Phosphorus Interaction on Growth, Bone Mineralization and Mineral Retention in Broiler Starter Chicks

  • An experiment was conducted to study the requirement of calcium (Ca) and non-phytin phosphorus (NPP) in commercial broilers during starter phase. Seven hundred and twenty day-old Vencob male broiler chicks were randomly distributed into 144 stainless steel battery brooders, 5 birds in each. Four levels each of Ca (6, 7, 8, and 9 g/kg) and NPP (3, 3.5, 4, and 4.5 g/kg diet) were fed in a factorial design in a corn-soya basal diet. Levels of dicalcium phosphate and oyster shell grit were adjusted to obtain the desired levels of Ca and NPP. Each diet was fed ad libidum to chicks in 9 battery brooders from one d to 21 d of age. Body weight gain and feed intake were depressed (p<0.01) by increasing the dietary Ca level (8 and 9 g/kg) at lower levels of NPP (3 and 3.5 g/kg diet). The growth depression observed at lower NPP level was alleviated by reducing the Ca content to 6 g/kg diet. The tibia ash content and tibia breaking strength increased with increase in both Ca (>6 g/kg) and NPP (>3 g/kg) levels. The leg abnormality score decreased (p<0.01) with increase in NPP content in the diet at all levels of Ca tested. The serum Ca and inorganic P levels were increased with increase in the level of the respective mineral in the diet, but the serum concentration of Ca and P were inversely related to the level of NPP and Ca, respectively /kg diet. In general, the excretion of macro minerals (Ca, and P), and micro minerals {zinc (Zn), manganese (Mn), iron (Fe), and copper (Cu)} was significantly lower at lower levels of Ca and NPP tested (6 and 3 g/kg diet, respectively). The mineral excretion increased with increase in dietary Ca and NPP levels, more conspicuously at the disproportionate ratio of these minerals (>2:1, Ca and NPP). Similarly, the retention of Zn, Mn, and Fe in liver was significantly higher (p<0.01) at lower levels of Ca and NPP tested. Results from this study indicate that the commercial broilers do not require more than 3 g NPP and 6g Ca/kg diet during starter phase (up to 21 d of age) for optimum weight gain, feed efficiency and utilization of Ca, P, Zn, Mn, Fe and Cu. However, the requirements of these minerals for optimum bone mineralization were higher than the levels suggested above.

Requirement of Non-phytate Phosphorus in Synthetic Broiler Breeder Diet

  • An experiment was conducted to study the laying performance, shell quality, bone mineralization, hatchability of eggs and performance of progeny (weight at day one and 14 d of age, P content in day old chick, leg score and survivability of chicks) of synthetic broiler breeders fed different levels of non-phytate phosphorus (NPP). Six levels of NPP (1.2, 1.8, 2.4, 3.0, 3.6 and 4.2 g/kg diet) at a constant calcium (Ca) level (30 g/kg) in a maize-soya-deoiled rice bran based diet were tested. Levels of dicalcium phosphate, shell grit and deoiled rice bran were adjusted to achieve the desired levels of NPP and Ca. Each level of NPP was fed with a weighed quantity of feed (160 g/b/d) to 40 female broiler breeders from 25 to 40 weeks of age housed in individual cages. Each bird was considered as a replicate. Egg production, feed/egg mass, body weight, egg weight, shell weight, shell thickness, egg specific gravity, serum Ca content and tibia breaking strength were not influenced (p>0.05) by the variation in dietary NPP levels tested. Increasing the dietary levels of NPP did not influence the hatchability of eggs, phosphorus (P) contents both in egg yolk and day old chick, chick body weight at day one and 14 d of age, leg score and survivability of chicks up to 14 d of age. Maximum response ($p{\leq}0.01$) in shell breaking strength, tibia ash and serum inorganic P contents were observed at NPP levels of 2.09, 2.25 and 3.50 g per kg diet, respectively. The retention of Ca increased, while the P retention decreased ($p{\leq}0.01$) with increasing dietary levels of NPP. Though maximum responses in shell breaking strength, bone ash and serum inorganic P were observed at NPP higher than 1.2 g/kg diet, the broiler breeder performance in terms of egg production, shell quality, hatchability of eggs and progeny performance and their survivability was not influenced by dietary NPP concentrations. It is concluded that synthetic broiler breeders maintained in cages do not require more than 1.2 g NPP/kg diet with a daily intake of 192 mg NPP/b/d during 24 to 40 weeks of age.

Optimization of Olive Flounder Paralichthys olivaceus Size as a Raw Material for Sikhae and Quality Characteristics of Sikhae with Suitable Olive Flounder Paralichthys olivaceus Weight (식해 소재로서 넙치(Paralichthys olivaceus) 크기의 최적화 및 이를 활용한 식해의 품질 특성)

  • This study was conducted to optimize the size of olive flounder Paralichthys olivaceus (OF) as a material of sikhae and to investigate the quality characteristics. The results on the protease activity of OF meat, protein and ash contents of the bone, and yields and hardness of fish bone during fermentation time suggest that the suitable fish weight for sikhae was less than 250 g. The proximate compositions of the OF sikhae fermented under optimum condition (fermentation for 9 days at 15℃), were 73.0% moisture, 12.0% crude protein, 1.3% crude fat and 2.4% ash. The salinity, titration acidity and amino acid nitrogen contents per 100 g sikhae were 1.7 g, 2.46 g, and 311.3 mg, respectively. The lactic acid bacteria concentration in the sikhae were 8.84 log CFU/g, which were higher than those (5.78-6.62 log CFU/g) of 5 kind of commercial flounder sikhae. The functional properties, such as ACE inhibitory activity (69.0%), antioxidative activity (69.3%), α-glucosidase inhibitory activity (22.7%), xanthine oxidase inhibitory activity (88.2%), and nitrite scavenging activity (96.4%) of the sikhae were superior to those of 5 kind of commercial flounder sikhae.

Influence of UV Irradiation Interval on Mineral Content in Metatarsus of Broiler Chicks (자외선 조사간격이 브로일러 병아리의 중족골 광물질 함량에 미치는 영향)

  • A study was conducted to investigate the concentrations of Ca, P and ash in metatarsal bone of broiler chicks exposed to UV light in different Interval. Day-old Hubbard broiler chicks (199=10 control+3 irradiation interval $\times$ 9 elapsed time $\times$ 7 replicate) were fed vitamin D3 deficient diet for 3 wk in a windowless subdued-light room and exposed to 297 nm UVB light by 0.068 mJ/$\textrm{cm}^2$ three times In 0, 12 or 24 h interval. The metatarsal bones were taken at 0, 6, 12, 18, 24, 48, 96, 144 or 240 h after last irradiation, separated from adhering tissue, ether extracted, dried and ashed. The Ca concentration was measured by atomic absorption spectrophotometry and P by ammonium metavanadate colorimetry. When the birds were continuously exposed to UVB light for 30 min without interval, the Ca content in metatarsus increased gradually according to the time after irradiation and reached the highest value 16.75% at 240 h after exposure. The P content also increased gradually until 144 h, where it was 9.75%. The ash content in metatarsus increased continuously until 240 h, the final time in this research, where 42.75% was shown. As 10 min three times irradiation in 12 h interval was applied to the chicks, the metatarsal Ca presented a small peak(13.31%) at 12 h after irradiation and a large peak(16.91%) at 144 h. P content showed a small peak(7.18%) at 12 h and a large level(8.34%) at 240 h. Ash content increased continuously until 240 h, where it was 46.53%. The small peaks in Ca and P concentration were thought to be derived from preirradiation at 12 and 24 h before final irradiation for 10 min. When 24 h interval system was treated, the peak value of Ca content(24.18%) occurred earlier(96 h) than those in 0 and 12 h interval systems. P content also showed the maximum value at 96 h(7.29%). Ash content presented an increasing trend until 240 h, where 45.75% was appeared. In respecting the method of UVB irradiation, the peak value of Ca content in metatarsus appeared earlier in 24 h interval system than in other systems. Meanwhile the ash contents in metatarsus of birds exposed to UVB light in 12 and 24 h interval procedures were higher than those in 0 h interval one. Therefore, it was concluded that a daily 10 min irradiation of UVB light would be desirable for increasing the Ca and ash content in metatarsus of brolier chicks.

Efficacy of New 6-Phytase from Buttiauxella spp. on Growth Performance and Nutrient Retention in Broiler Chickens Fed Corn Soybean Meal-based Diets

  • A total of 420 day-old male Ross chicks were weighed at d 1 of life and assigned to test diets to assess the efficacy of a new Buttiauxella spp. phytase expressed in Trichoderma reesei. Diets were: positive control (PC) adequate in nutrients and negative control (NC) diet (40% and 17% less available phosphorous (P) and calcium (Ca), respectively) supplemented with 6 levels of phytase 0, 250, 500, 750, 1,000, and 2,000 phytase units (FTU)/kg of diet. All diets had titanium dioxide as digestibility marker and each diet was allocated to ten cages (6 birds/cage). Diets were fed for 3 wk to measure growth performance, apparent retention (AR) on d 17 to 21 and bone ash and ileal digestibility (AID) on d 22. Growth performance and nutrient utilization was lower (p<0.05) for NC vs PC birds. Phytase response in NC birds was linear (p<0.05) with 2,000 FTU showing the greatest improvement on body weight gain (20%), feed conversion (7.4%), tibia ash (18%), AR of Ca (38%), AR of P (51%) and apparent metabolizable energy corrected for nitrogen (5.1%) relative to NC. Furthermore, phytase at ${\geq}750FTU$ resulted in AID of total AA commensurate to that of PC fed birds and at ${\geq}1,000FTU$ improved (p<0.05) AR of P, dry matter, and N beyond that of the lower doses of phytase and PC diet. In conclusion, the result from this study showed that in addition to increased P and Ca utilization, the new Buttiauxella phytase enhanced growth performance and utilization of other nutrients in broiler chickens in a dose-dependent manner.
