Anti-diabetic effect of purple corn extract on C57BL/KsJ db/db mice

  • BACKGROUND/OBJECTIVES: Recently, anthocyanins have been reported to have various biological activities. Furthermore, anthocyanin-rich purple corn extract (PCE) ameliorated insulin resistance and reduced diabetes-associated mesanginal fibrosis and inflammation, suggesting that it may have benefits for the prevention of diabetes and diabetes complications. In this study, we determined the anthocyanins and non-anthocyanin component of PCE by HPLC-ESI-MS and investigated its anti-diabetic activity and mechanisms using C57BL/KsJ db/db mice. MATERIALS/METHODS: The db/db mice were divided into four groups: diabetic control group (DC), 10 or 50 mg/kg PCE (PCE 10 or PCE 50), or 10 mg/kg pinitol (pinitol 10) and treated with drugs once per day for 8 weeks. During the experiment, body weight and blood glucose levels were measured every week. At the end of treatment, we measured several diabetic parameters. RESULTS: Compared to the DC group, Fasting blood glucose levels were 68% lower in PCE 50 group and 51% lower in the pinitol 10 group. Furthermore, the PCE 50 group showed 2-fold increased C-peptide and adiponectin levels and 20% decreased HbA1c levels, than in the DC group. In pancreatic islets morphology, the PCE- or pinitol-treated mice showed significant prevention of pancreatic ${\beta}$-cell damage and higher insulin content. Microarray analyses results indicating that gene and protein expressions associated with glycolysis and fatty acid metabolism in liver and fat tissues. In addition, purple corn extract increased the phosphorylation of AMP-activated protein kinase (AMPK) and decreased phosphoenolpyruvate carboxykinase (PEPCK), glucose 6-phosphatase (G6pase) genes in liver, and also increased glucose transporter 4 (GLUT4) expressions in skeletal muscle. CONCLUSIONS: Our results suggested that PCE exerted anti-diabetic effects through protection of pancreatic ${\beta}$-cells, increase of insulin secretion and AMPK activation in the liver of C57BL/KsJ db/db mice.

Child health promotion program in South Korea in collaboration with US National Aeronautics and Space Administration: Improvement in dietary and nutrition knowledge of young children

  • BACKGROUND/OBJECTIVES: Childhood obesity has become a global epidemic. Development of effective and sustainable programs to promote healthy behaviors from a young age is important. This study developed and tested an intervention program designed to promote healthy eating and physical activity among young children in South Korea by adaptation of the US National Aeronautics and Space Administration (NASA) Mission X (MX) Program. SUBJECTS/METHODS: The intervention program consisted of 4 weeks of fitness and 2 weeks of nutrition education. A sample of 104 subjects completed pre- and post- surveys on the Children's Nutrition Acknowledgement Test (NAT). Parents were asked for their children's characteristics and two 24-hour dietary records, the Nutrition Quotient (NQ) at baseline and a 6-week follow-up. Child weight status was assessed using Korean body mass index (BMI) percentiles. RESULTS: At baseline, 16.4% (boy: 15.4%; girl: 19.2%) of subjects were overweight or obese (based on $BMI{\geq}85%tile$). Fat consumption significantly decreased in normal BMI children ($48.6{\pm}16.8g$ at baseline to $41.9{\pm}18.1g$ after intervention, P < 0.05); total NQ score significantly increased from 66.4 to 67.9 (P < 0.05); total NAT score significantly improved in normal BMI children (74.3 at baseline to 81.9 after the program), children being underweight (from 71.0 to 77.0), and overweight children (77.1 at baseline vs. 88.2 after intervention, P < 0.001). CONCLUSIONS: The 6-week South Korean NASA MX project is feasible and shows favorable changes in eating behaviors and nutritional knowledge among young children.

Effect of 4 Weeks' Walking Exercise with Blood Flow Restriction on Insulin Resistance, Adipokines and Gut Hormones in Middle Aged Obese Women (혈류를 제한한 4주간의 걷기운동이 중년 비만여성의 인슐린저항성과 아디포카인, Gut hormone에 미치는 영향)

  • This study was conducted to investigate the effect of walking exercise with blood flow restriction (BFR) on insulin resistance, adipokines and gut hormones in middle aged obese women. Eleven obese women (BMI > 25kg/m2; body fat > 30%) wore pneumatic pressure belts at both femurs and performed walking exercise consisting of walking for 2 min and resting for 1min twice per day, 3 days/wk for 4 weeks. Insulin significantly increased after exercise (p<0.05), while glucose increased slightly, but not significantly. Additionally, HOMA-IR decreased significantly after exercise (p<0.05), while adiponectin and visfatin increased, but not significantly. Finally, ghrelin (p<0.05) and GLP-1 (p<0.05) increased significantly after exercise. These results suggest that walking exercise with blood flow restriction for 4 weeks improves insulin resistance, adipokines, and gut hormones in obese middle-aged women. Therefore, high intensity walking exercise with blood flow restriction for short periods of time has more positive effects on prevention and treatment of obesity-related metabolic diseases.

A Study on the Cestodes in yheyagya chalcogramma (해산어류 명태(Theragra chalcogramma)에 기생하는 조충류 조사)

  • This study was performed to investigate the intestinal parasites (especially cestoda) in the guts of Theragra chalcogramma in Korea. The cestode larvae collected were morphologically observed and measured, and at the same time, microscopical study was done with the section slides of the cestode larvae. They were identified on the basis of the descriptions by Schmidt and Yamaguti. The results obtained in this study are as follows: 1. The cestode larvae were identified as Khawia sinesis, Eulacistorhynchus chiloscyllius, Tentacularia coryphaenga, Pssudonybelinia odontasantha and Nybelinia Zingualis. These are new species reported in Korea. 2. The characteristics of Khawia sinesis are its scolex lacking loculi and broad, fat and fimbriate shape. Its neck is not separated from the body but a little constricted. 3. Eulacistorhynchus chiloscyzzius is characterized by fairly long and acraspedote scolex. The bulbs are long; retractor muscles are attached to bases of the bulbs. The two bothridia are oval or round in shape and lacking posterior notch. The tentacles are long and poeciloacanthus; double chainette present. 4. Tentacularia coryphaenea is characterized by its long craspedote and subcylindrical scolex. The bothridia are separated, without free borders and spinous. The tentacles are short, slender, armed with solid hooks in spirals, similar except the base of tentacle. The tentacle sheaths are not twisted. The bulbs are ellipsoidal. 5. Pseudonybelinia odontacantha is characterized by its craspedote scolex. The tentacles are inserted near anterior margin of bothridia and armed with hooks in spirals ascending left to right. The hooks are similar, throughout tentacle, with feeble curve and provided with a tooth-like protuberance on the ventral side. The bulbs are three times longer than its width. The posterior margins of bothridia have a pair of eversible ciliated pits; or fossettes. 6. Nybelinia lingualis is characterized by its short and craspedote scolex. The bothridia are separated, with free boarders. The tentacles are cylindrical, armed with solid and similar hooks in quincunxes.

A Study on the Efficiency Evaluation of Ultrasound Therapy Using Varicose Vein Simulated Tissue Phantom and Tissue Equivalent Phantom (하지정맥류 모사 생체조직 팬텀과 조직등가 팬텀을 이용한 초음파 치료효과 평가에 관한 연구)

  • Because of the expectation of the non-invasive treatment effect, Various studies on the treatment of varicose veins using focused ultrasound are reported. In this study, the bio-tissue phantom and tissue equivalent phantom that can be applied to estimation of ultrasonic varicose veins treatment effect. Each phantom was evaluated for its usefulness by evaluating the acoustic characteristics and the shrinkage rate according to the ultrasonic irradiation. A multi-layer structure phantom with three layers of skin, fat, and muscle was constructed considering the structure of the tissue where the varicose veins occurred. The materials constituting each layer were made to have characteristics similar to human body. In addition, the multi-layered phantoms with blood vessel mimic tube, with bovine blood vessel, and with animal tissue were fabricated. The degree of shrinkage of blood vessel mimic material and vascular tissue according to ultrasonic irradiation was evaluated using B-mode image. As the results of this study, it was thought that the proposed phantom could be used effectively in the evaluation of ultrasonic varicose veins treatment. In addition, it is thought that these phantoms could be applied to the development of varicose vein treatment device using the focused ultrasound and the verification of the therapeutic effect.

A Study on the Analysis of Risk Factors and Correlations of Coronary Artery Disease of the Examinee taking Coronary Computed Tomography Angiography in a Comprehensive Health Improvement Center (종합검진 수검자의 관상동맥 전산화단층 혈관조영술 검사에서 관상동맥질환의 위험요인과 관련성 분석)

  • The purpose of this study is to evaluate the usefulness of coronary computed tomography angiography of coronary artery disease for the purpose fo health screening according to gender and age. In addition the association between hematological factors (Glucose, total cholesterol, visceral fat, body mass index, triglycerides, high density lipoprotein (HDL), low density lipoprotein (LDL)) and coronary artery disease is investigated. A retrospective analysis of coronary computed tomography angiography with 299 subjects showed that the detection rate of coronary artery disease was higher in men over 50 years of age and it was statistically significant. In particular, the prevalence rate of men (37.9%) was about 2 times higher than that of women (17.0%). Glucose and HDL as hematological factors associated with coronary artery disease were statistically significant. The prevention and management of coronary artery disease seems to require the control of glucose and high density lipoprotein (HDL). Although it is not statistically significant with other hematological factors, the need for management of coronary artery disease was identified. the coronary computed tomography angiography of coronary artery has higher radiation doses than other CT scans. Therefore, for the purpose of screening, coronary computed tomography angiography should be considered in consideration of the sex and age of the examinee, and detection of coronary artery disease through other non-invasive tests should be prioritized over coronary computed tomography angiography.

A Study on Dietary Mineral Intakes, Urinary Mineral Excretions, and Bone Mineral Density in Korean Postmenopausal Women (폐경 후 여성의 무기질 섭취량과 소변 중 배설량 및 골밀도에 관한 연구)

  • The purpose of this study was to determine the urinary Ca, P, Mg, Zn, Cu, and Mn levels and bone mineral density (BMD) in sixty-two postmenopausal women. The study was conducted through anthropometric checkup, 24-hour recall, 24-hour urine and bone mineral density using DEXA. Average age, height, weight and body fat of the subjects were respectively 65.39 years, 150.19 cm, 58.03 kg and 37.22%. The average spine and femoral neck BMD of subjects were -2.19, -3.13. The mean intakes of Ca, P and Mg were 524.7 mg, 993.10 mg, and 254.6 mg and those of Zn, Cu and Mn were 8.6 mg, 1.5 mg, and 3.5 mg. The average 24-hour urinary excretion of Ca (UCa), P (UP) and Mg (UMg) were 161.07 mg, 673.68 mg, and 99.87 mg. The average 24-hour urinary excretion of Zn (UZn), Cu (UCu) and Mn (UMn) were 366.50 ${\mu}g$, 22.57 ${\mu}g$, and 1.55 ${\mu}g$. Ca intake showed significantly positive correlations with urinary UCa (p < 0.05), UMg (p < 0.01) and spine BMD (p < 0.05). P intake showed significantly positive correlations with UCa (p < 0.05), UMg (p < 0.05) and UZn (p < 0.05). Mg intake showed significantly positive correlations with UZn (p < 0.05) and Mn intake showed significantly positive correlations with UCa (p < 0.05). Multiple regression analysis indicates that Ca intake and UMg is the most important factor to increase spine BMD. On the other hand, UCa is the most important factor to decrease spine BMD. Higher femoral neck BMD was related to UP, while lower femoral neck BMD was related to UCa. In conclusion, Dietary intake of Ca showed positive effect of spine BMD, while excessive P intake showed negative effect on BMD due to increases in UCa, UMg and UZn. Further studies are required to investigate the relationship between bone metabolism and mineral excretion.

Effects of Fish Oil Supplementation on Growth Performance, Fatty Acid Composition of Longissimus Muscle and Carcass Characteristics in Hanwoo Steers (Fish Oil의 첨가가 한우 거세우의 육성성적, 배최장근의 지방산 조성 및 도체특성에 미치는 영향)

  • Twenty Hanwoo steers (average body weight=440.8±32.4kg) were used to investigate the effects of fish oil supplementation on growth performance, fatty acid composition of longissimus muscle and carcass characteristics. The experiment was done with two treatment groups; FO-0, without fish oil and FO-3, supplemented with 3% fish oil. Total gain and average daily gain (ADG) of steers were similar between two groups. Fish oil supplementation had no effects on contents of protein, ether extract and ash in longissimus muscle. Contents of isoleucine and glycine in longissimus muscle were decreased by fish oil supplementation (p<0.05), but content of cystein was increased by fish oil supplementation (p<0.05). Fish oil supplementation decreased contents of myristic acid and eicosenoic acid in longissimus muscle (p<0.05), but increased contents of oleic acid and arachidonic acid in longissimus muscle (p<0.05). Contents of eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) in longissimus muscle were increased by fish oil supplementation p<0.05). Carcass weight, back fat thickness, rib-eye area, yield index and yield grade were similar between two groups. Meat color was improved by fish oil supplementation (p<0.05), Ratio of quality grade ‘1 or over’ increased by fish oil supplementation. Therefore, the present study indicating that fish oil supplementation had positive effects on content of oleic acids in relation to flavor of beef, contents of EPA and DHA in relation to human health and ratio of quality grade ‘1 or over’.

Changes of Thyroid Hormone Levels in Plasma of Rats during Cold Exposure (저온환경 적응에 있어서의 갑상선 Hormone의 변동)

  • The importance of thyroid hormones for the survival of rats in the cold is along-established fact. Hypothyroid animals are unable to survive in a cold environment. It was also reported that acute exposure of rats, guinea pigs and rabbits to cold produced an increased secretion of TSH and thereby thyroid hormone secretion within 10 to 30 min, but this increase of thyroid activity disappeared quite rapidly during warming. However, in human study no significant difference was found in the concentration of $T_4$, TSH and cortisol between summer and winter. But plasma $T_3$ concentration was increased significantly in winter in 56 adult men. On the other hand, it has been also known that catecholamines are important in the maintenance of body temperature of rat exposured to cold. Abundant evidences suggest that the sympathetic nervous system is involved in the activation of nonshivering thermogenesis and that thyroid hormone metabolism and secretion are influenced by catecholamines and consequently by the activity of the sympatheticadrenal system. Many of the metabolic effects of catecholamines are associated with an increase in the level of cAMP mediated through activation of adenylate cyclase which converts ATP to cAMP. Other studies have shown that thyroid hormones affect the amount of adenylate cyclase present in the adipose tissue. On the other hand. it was also reported that a particulate cAMP phosphodiesterase activity in fat cells was modulated by the action of thyroid hormones. The objective of the present study was to determine the interaction between thyroid activity and cyclic nucleotides during acute exposure to cold. Albino rats weighing around 200 g were used as the experimental animal. The room temperature group was kept at $25^{\circ}C$ and the cold-exposured group was kept at $4^{\circ}C$ for 1 week or 2 weeks. Each group was subdivided into three subgroups; control, KI, and MTU group. At the end of experiment the animals were etherized and blood was taken from abdominal aorta for $T_4,\;T_3$ and cyclic nucleotides. The determinations of $T_3,\;T_4$ and cyclic nucleotides were carried out with a radioimmunoassay(RIA) method. The results were summerized as followings. 1) A significant increase of thyroid weight was observed in rats exposured to cold for 2 weeks. Furthermore, in rats administered MTU while to exposure to cold the thyroid weight was also increased significantly. 2) After 2 weeks $T_3$ concentration in the plasma of cold-exposured rats was significantly increased in KI group and MTU group as well as in control group. On the contrary, after 2 weeks of cold exposure $T_4$ level was decreased in control group. 3) In the case of cyclic nucleotides, plasma cAMP was increased in the control group after 1 or 2 weeks of cold exposure. However, cAMP level in plasma was rather significantly decreased in KI group and MTU group as well as in control group.

Some Factors Affecting Bone Mineral Status of Premenopausal Women

  • This study was conducted to investigate dietary and other factors affecting bone mineral density (BMD) in Korean premenopausal women. Seventy-eight premenopausal women who visited health promotion center for health examinations volunteered to participate in this study and they were divided into two groups according to the bone status by T-score : normal or osteopenic group and osteoporotic group. The demographic and general characteristics, and dietary intake were surveyed using the questionnaire. BMDs of the lumbar spine and femoral neck of subjects were measured by dual energy X-ray absorptiometry. Serum samples were measured for Lipid concentration, and calcium, phosphorus, alkaline phosphatase as bone formation indicators. Urine was analysed for creatinine as bone resolution indicators. The results are as follows:The mean BMDs of the lumbar spine and femoral neck were 1.21 0.02g/cm$^2$ and 0.97 0.04g/cm$^2$, respectively and the BMD levels of osteoporotic group were significantly lower than that of normal or osteopenic group (p<0.001, respectively). Height in osteoporotic group was significantly lower than that of normal or osteopenic group (p<0.01), and bodyweight did not show any significant difference but had a lower tendency. Mean daily intake of energy was 1720 52ka1. When nutrient intake was compared with recommended dietary allowances (RDA) of subjects, calcium, Fe, vitamin A and riboflavin intakes showed means lower than RDA. The nutrient intake did not show any significant difference between normal or osteopenic group and osteoporotic group except intakes of protein fat and niacin. Serum and urine levels did not show any significant differences between normal or osteopenic group and osteoporotic group and all were within normal range, however, serum alkaline phosphatase level of osteoporotic group was significantly higher than that of normal or osteopenic group (p<0.001). Height showed positive correlations with lumbar spine bone mineral density (LBMD, r=0.332, p<0.01), no correlation was found with femoral neck bone mineral density (NBMD). Age, age at menarche, bodyweight, body mass index (BMI) and obesity showed no correlation with BMD. The BMD of the lumbar spine was significantly and positively related to the intake of niacin and vitamin C (r=0.236, p<0.05; r=0.274, p<0.05). Serum levels of calcium and phosphorus showed a negative correlation with LBMD (r=-0.698, p=0.0001; r=-0.503, p=0.0001, respectively). The results suggested that the BMDs of the lumbar spine was positively related to the intake of niacin and vitamin C in premenopausal women. Therefore, this study confirmed that one of the most effective way to minimize bone loss would be higher intake of niacin and vitamin C rich foods and habitual physical activity may have a beneficial effect on BMD in premenopausal period.
