The Effect of Extract Powder from Fresh and Black Garlic on Main Components in Serum and Organs of Streptozotocin-Induced Diabetic Rats (생마늘 및 흑마늘 추출분말이 Streptozotocin 유발 당뇨 흰쥐의 혈청 및 장기 내 주요성분에 미치는 영향)

  • In this study, we examined the biological activity and synergistic effects of an extract powder of 1% and 3%, each fresh (FGP) and black garlic (BGP) in streptozotocin-induced diabetic rats. Blood glucose content was significantly lower in FGP and BGP groups than control group. Glycosylated Hb was significantly higher in streptozotocin induced diabetic control group than normal group, but significantly lower in FGP and BGP groups. Total cholesterol content of the FGP and BGP groups were lower than control group, but not shown the significant difference between garlic fed groups. HDL-cholesterol concentrations of the FGP and BGP fed groups were significantly higher than control group, except of 1% BGP group. LDL and VLDL-cholesterol contents were significantly lower in 3% FGP group, and the same tendency atherogenic index and cardiac risk factor. GOT, GPT, and ${\gamma}$-GTP activity of serum were lower in FGP and BGP fed groups than control group. Glycogen contents in liver significantly higher than control group, and has not significantly difference between normal group. TBARS content was no significantly difference in the liver and serum, but in the kidney, 3% FPG and BGP fed groups were significantly lower than other experimental group. DPPH radical scavenging activity of liver has not significant difference among experimental groups, but activity was higher garlic extract powder fed groups in serum and kidney. These results indicate that dietary supplements of fresh and black garlic extract powder was contributed to lower of blood glucose, loss prevention of glycogen in liver and improve of lipid metabolism.

  • Two consecutive studies were conducted to evaluate the effects of dietary supplementation with fermented garlic powder (FGP) or fermented garlic fluid (FGF) on growth performance, immune responses, and disease resistance of olive flounder Paralichthys olivaceus. In experiment I, olive flounder (BW: 65 g) were fed four experimental diets formulated to contain 0%, 0.5%, 1%, and 1.5% FGP (designated as FGP-0, FGP-0.5, FGP-1, and FGP-1.5, respectively). After the 10-weeks feeding trial, feed intake was significantly lower in fish fed the FGP-0.5 and FGP-1.0 diets, as compared to those fed the control diet. Fish fed the FGP-0 and FGP-0.5 diets showed significantly lower survival, as compared to the other treatments. Dietary supplementation with FGP resulted in higher non-specific immune responses than the FGP-0 group. Plasma cholesterol and triglyceride levels decreased as dietary FGP level increased. In experiment II, olive flounder (BW: 65 g) were fed four experimental diets for 10 weeks. The diets were prepared with a commercial expanded pellet to have 0%, 0.25%, 0.5%, and 1% FGF (designated as FGF-0, FGF-0.25, FGF-0.5, and FGF-1, respectively) by adsorption. At the end of the second feeding trial, feed intake was significantly lower in fish fed the FGF-0 diet, as compared to other treatments. Fish fed the FGF-0.25 and FGF-0.5 diets exhibited significantly lower cholesterol levels, as compared to other treatments. Lysozyme activity significantly increased with increases in dietary FGF. Cumulative mortality in a challenge test with Streptococcus iniae was significantly lower in the fish groups fed FGF-supplemented diets than in fish fed the control diet. The results of this study indicated that dietary supplementation with FGP or FGF can enhance the non-specific immune responses and disease resistance of olive flounder against S. iniae.

  • We report non-specific immune responses and disease resistance against Vibrio anguillarum, Streptococcus iniae and Edwardsiella tarda by dietary supplementation of fermented garlic powder (FGP) in olive flounder for the first time. Four isonitrogenous (45% crude protein) and isocaloric (17.1 MJ/kg) diets were formulated to have 0%, 0.5%, 1% and 2% of the FGP (G-0, G-0.5, G-1 and G-2). The experimental diets were fed to juvenile olive flounder averaging 23.4 g in triplicate groups (90 fish/group) in a flow-through system. After a five-week feeding trial, healthy fish with similar sizes from each tank were selected and injected with 1 ml of three bacteria each to evaluate disease resistance of the fish. During the 5-week feeding trial, the weight gain, specific growth rate, feed conversion ratio, protein efficiency ratio, and survival of the fish were not significantly affected by the experimental diets. However, feed intake was significantly lower (P<0.05) in the fish fed the G-2 diet compared with the control group. Hemoglobin, myeloperoxidase activity, cholesterol and HDL-cholesterol were not different between the dietary groups. However, hematocrit, nitroblue tetrazolium (NBT) activity, and lysozyme activity were increased (P<0.05) with an increment of dietary FGP. Plasma triglyceride of the fish fed the G-0.5 diet was significantly lower than that of fish fed the control diet. The cumulative mortality was lower in the fish fed diets containing FGP compared with the control group in the challenge test except for the bacteria Edwardsiella tarda. The results in this study indicate that dietary supplementation of FGP can enhance the non-specific immune responses and disease resistance of olive flounder against V. anguillarum and S. iniae.

Effect of Supplementation of Acanthopanax senticosus on Growth Performance, Blood Biochemical Profiles and Expression of Pro-Inflammatory Cytokines in Broiler Chicks (육계에서 가시오갈피 급여에 따른 생산성, 혈액 생화학적 성상 및 면역 사이토카인 발현에 미치는 영향)

  • This study was performed to examine the effects of dietary Acanthopanax senticosus (AS) on growth performance, immune organ weights, blood biochemical parameters and the expression of pro-inflammatory cytokines in broiler chicks. A total of 120 4-day-old birds were given a basal diet (CON) or a basal diet supplemented with 0.5% (AS1) or 1.0% (AS2) AS powder until the birds were 35 days of age. There was no difference in body weight, total gain, feed intake or immune organ weights among the treatment groups. However, the feed conversion ratio in the AS2 group was lower (p<0.05) than that in the CON group. Serum biochemical components, including AST (aspartate aminotransferase), ALT (alanine aminotransferase), albumin and total protein, were not affected by the dietary treatments, whereas glucose and triglyceride levels increased (p<0.05) in the AS2 group compared with the CON group. The AS1 group exhibited decreased mRNA expression (p<0.05) of IFN-${\gamma}$ in white blood cells and iNOS in the liver compared with the CON group. The other pro-inflammatory cytokines were unaffected by dietary AS supplementation, although there was a trend towards decreased expression of these genes, including those encoding Il-$1{\beta}$, IL-6 and TNF-${\alpha}$. In conclusion, dietary supplementation with 0.5% AS decreased the expression of several pro-inflammatory cytokines without affecting growth performance, suggesting that this supplement might be applicable as an immunoregulatory feed additive in broiler chicks.

Influence of Houttuynia cordata Thnub Powder Mixtures on Plasma Lipid Composition in Pigs (어성초 분말 혼합 식이가 돼지의 혈장 지질성분에 미치는 영향)

  • This study was designed to investigate the influence of Eosungcho (Houttuynia cordata Thunb) mixture on the plasma lipids of pigs. Experimental groups were divided into 3 groups: control group(0%), 5% and 10% Eosungcho feeding groups. Plasma concentration of total cholesterol, atherosclerotic index, free cholesterol and cholesteryl ester were significantly lower in the Eosungcho powder 5% and 10% feeding groups than control group. Plasma contents of HDL-cholesterol, cholesteryl ester ratio, triglyceride and phospholipid were significantly different between 10% Eosungcho feeding group and control group. Plasma concentration of LDL-cholesterol was significantly love in 10% Eosungcho feeding group($76.5{\pm}5.4mg/dL$) than in control group($86.5{\pm}5.6mg/dL$). Plasma concentration of VLDL and chylomicron were showed similar tendency to LDL-cholesterol. Also, blood sugar, GOT and GPT concentration were significantly lower in 10% Eosungcho feeding group than in control group. In control group, the ratio of saturated fatty acid to unsaturated fatty acid was 58.1 : 51.9, but in 5% and 10% Eosungcho feeding groups, those were 42.1 : 57.9 and 41.0 : 59.0, respectively. The component of main fatty acid was different between control group and Eosungcho feeding groups. In Eosungcho feeding groups, main unsaturated fatty acids were oleic acid(18:1) and linoleic acid(18:2). From the results, addition of Eosungcho powder might be effective in improving the plasma lipid components in pigs.

TISSUE CHANGE AFTER EMBEDDING GELATIN MATRIX IMPLANT(FFIBREL®) IN SUBCUTANEOUS TISSUE OF RATS;HISTOLOGIC, IMMUNOHISTOCHEMICAL AND SCANNING ELECTRON MICROSCOPIC STUDY (백서의 피하조직에 Gelatin Matrix Implant (Fibrel®) 매식시 조직변화에 관한 연구)

  • GMI (Fibrel${(R)}$) is one of the dermal filling substances which have been successfully used for the treatment of depressed cutaneous scar and wrinkles. It's major components are; Gelatin powder, which provides a framework for the clot to form and remains stable under the scar, and ${\varepsilon}$-aminocaproic acid, which inhibits the production of fibrinolysin, and Plasma, which provides the necessary ingredients for collagen synthesis. GMI has advantages of low immunogenicity and increased longevity. It has been known to induce fibroblast activity and promote new collagen synthesis. We used 34 Sprague-Dawley rats which were bred under the same condition and duration. 18 of experimental animals were undergone cardiac puncture, and their blood were collected, centrifugated, and stored in freezer. Out of 16 animals, control group were injected with 2ml plasma into the subcutaneous tissue of Lt. scapular, while experimental group were implanted of 2 ml GMI into the Rt. same area. Experimental animals were sacrificed at the 3rd day, 5th day, 1st week and 2nd week respectively after implantation of GMI. To observe the histopathologic change of GMI and surrounding tissue reaction of GMI, we had examined with H&E staining, immunohistochemical staining with vimentin, ${\alpha}$-SMA, S-100 under LM and SEM. The obtained results were as follows ; 1. In LM study, the inflammatory cell infiltrations and granulation tissue formation were observed, and muscle tissues were well attached with adipose tissues in the control group. In the experimental group, inflammatory cell infiltrations had been observed by the 2nd week and irregular adipiose tissues and well differentiated mesenchymal tissues were examined. 2. In immunohistochemical study, the experimental group of ${\alpha}$-SMA study, there were a prominent positive response on endothelial development of granulation tissues and mesenchymal tissues compare with the control group. In vimentin study, positive response on mescenchymal fibroblast continued to 2nd week, but negative in the control group. In S-100 study, both groups were positively responded on irregular adipose tissues. 3. In SEM study, collagen fibers were embedded by the plasma by the 5th day in the control group, and in the 3rd day experiment GMI were resorved but communited with collagen fiber till the 1st week. Collagen fibers were infilt-rated into GMI at the 2nd week and the infilltrated GMI were conglomerated with the mature adipose cells and the collagen fibers. From the above results, GMI implantation in the subcutaneous tissue of Sprague-Dawley rat, the mild infiltration of inflammatory cells were showed till 2nd week and the granulation tissues were observed. GMI were nearly resorbed till 2nd week, but well attached with adipose tissue and collagen fibers. The endothelium and fibroblasts were actively proliferated. Adipose tissues and mesenchymal tissue cells were observed. As already expressed, GMI showed resorptive change in course of time without any early immune reaction, and seemed to induce fibroblast activity and promote new collagen synthesis.
