Numerical Analysis on the Characteristics of Supersonic Steam Jet Impingement Load (초음속 증기제트의 충돌하중 특성에 대한 수치해석 연구)

  • Structures, systems and components of nuclear power plants should be able to maintain safety even in the event of design-basis accidents such as high-energy line breaks. The high-pressure steam jet ejected from the broken pipe may cause damage to the adjacent structures. The ANSI/ANS 58.2 code has been adopted as a technical standard for evaluating the jet impingement load. Recently, the U.S. NRC pointed out the non-conservativeness of the ANSI/ANS 58.2, because it does not take into account the blast wave effect, dynamic behavior of the jet, and oversimplifies the shape and load characteristics of the supersonic steam jet. Therefore, it is necessary to improve the evaluation method for the high-energy line break accident. In order to evaluate the behavior of supersonic steam jet, an appropriate numerical analysis technique considering compressible flow effect is needed. In this study, numerical analysis methodology for evaluating supersonic jet impingement load was developed and verified. In addition, the conservativeness of the ANSI/ANS 58.2 model was investigated using the numerical analysis methodology. It is estimated that the ANSI jet model does not sufficiently reflect the physical behavior of under-expanded supersonic steam jet and evaluates the jet impingement load lower than CFD analysis result at certain positions.

Effect of Surface Treatments of Stainless Steels on Oxidation Behavior Under Operating Condition of IT SOFC Interconnect (IT SOFC 인터커넥터 구동 조건에서의 스테인레스 소재의 산화거동에 미치는 표면전처리의 영향)

  • Solid oxide fuel cells (SOFCs) have many attractive features for widespread applications in generation systems. Recently, stainless steels have attractive materials for metallic bipolar plate because metallic bipolar plates have many benefits compared to others such as graphite and composite bipolar plates. SOFC operates on high temperature of about $800{\sim}1000^{\circ}C$ than other fuel cell systems. Thus, many studies have attempted to reduced the operation temperature of SOFC to about $600{\sim}800^{\circ}C$, which is the intermediate temperature (IT) of SOFC. Low cost and high-temperature corrosion resistance are very important for the practical applications of SOFC in various industries. In this study, two specimens, 304 and 430 stainless steels with and without different pre-surface treatments on the surface were investigated. And, specimens were exposed at high temperature in the box furnace under oxidation atmosphere of $800^{\circ}C$. Oxidation behavior have been investigated with the materials exposed at different times (100 hrs and 400 hrs) by SEM, EDS and XRD. By increasing exposure time, the amount of metal oxide increased in the order like; STS304 < STS430 and As-received < As-polished < Sand-blast specimens.

An in-silico approach to design potential siRNAs against the ORF57 of Kaposi's sarcoma-associated herpesvirus

  • Kaposi's sarcoma-associated herpesvirus (KSHV) is one of the few human oncogenic viruses, which causes a variety of malignancies, including Kaposi's sarcoma, multicentric Castleman disease, and primary effusion lymphoma, particularly in human immunodeficiency virus patients. The currently available treatment options cannot always prevent the invasion and dissemination of this virus. In recent times, siRNA-based therapeutics are gaining prominence over conventional medications as siRNA can be designed to target almost any gene of interest. The ORF57 is a crucial regulatory protein for lytic gene expression of KSHV. Disruption of this gene translation will inevitably inhibit the replication of the virus in the host cell. Therefore, the ORF57 of KSHV could be a potential target for designing siRNA-based therapeutics. Considering both sequence preferences and target site accessibility, several online tools (i-SCORE Designer, Sfold web server) had been utilized to predict the siRNA guide strand against the ORF57. Subsequently, off-target filtration (BLAST), conservancy test (fuzznuc), and thermodynamics analysis (RNAcofold, RNAalifold, and RNA Structure web server) were also performed to select the most suitable siRNA sequences. Finally, two siRNAs were identified that passed all of the filtration phases and fulfilled the thermodynamic criteria. We hope that the siRNAs predicted in this study would be helpful for the development of new effective therapeutics against KSHV.

Plant Virome Analysis by the Deep Sequencing of Small RNAs of Fritillaria thunbergii var. chekiangensis and the Rapid Identification of Viruses

  • Thunberg fritillary (Fritillaria thunbergii), a perennial used in traditional Chinese herbal medicine, is a members of the family Liliaceae. The degeneration of germplasm is a severe problem in the production of Fritillaria thunbergii var. chekiangensis. However, no information about viral infections of F. thunbergii var. chekiangensis has been reported. In this study, we sequenced the small RNAs of F. thunbergii var. chekiangensis from leaves and bulbs, and viruses were identified using a phylogenetic analysis and BLAST search for sequence. In addition, multiplex reverse transcriptase-polymerase chain reaction (RT-PCR) was used to rapidly detect viruses in this variety. Our study first reported that five viruses infected F. thunbergii var. chekiangensis. Among them, fritillary virus Y (FVY), lily mottle virus (LMoV), Thunberg fritillary mosaic virus (TFMV), and hop yellow virus (HYV) had been reported in F. thunbergii, while apple stem grooving virus was first reported in the genus Fritillaria. A multiplex RT-PCR method was developed to rapidly test the four viruses FVY, LMoV, TFMV, and HYV in F. thunbergii var. chekiangensis. Our results provide a better understanding of the infection of F. thunbergii var. chekiangensis by viruses and a basic reference for the better design of suitable control measures.

Flow-Chart for Influence Estimation of Underwater Blasting (수중발파의 영향평가를 위한 Flow-Chart)

  • In this study, ground vibration values and damping coefficient produced by underwater blasting were measured and analyzed. Equations of vibration, $V=K(SD)^{-0.536}$, were presented from quantitative experiment results. The K Values are classified with 1.507, 2.005 and 2.939 respectively at 50%, 90% and 95% reliability. Also, hydrospace noise in aquafarm and noise in atmosphere as well as ground vibrations were measured, and maximum values of these results were 86.8dB(A), 147.8dB(A), 0.244cm/s, respectively. Equations of hydrospace noise, $SL=293.2SD^{-0.164}$, was presented from quantitative experiment results. Also, the flow-chart for influence estimation and underwater blast design was presented from these results. The results of the study may be applied for the evaluation of the influence on aquafarm as a basic data before having main underwater blasting at construction sites.

Field Demonstration of Pre-Fracturing for Controlling Noise and Vibration (선행이완발파의 진동 및 소음 제어 효과 검증을 위한 발파 시험 시공)

  • Pre-fracturing is the blasting method to weaken the rock mass prior to the main excavation. This study aims to evaluate the effectiveness of pre-fracturing by using half the explosive charge typically employed in conventional blasting designs. Field tests conducted at a quarry in Gapyeong showed that noise levels were reduced by 2.7 dB due to the decreased amount of explosive per blast hole, and vibration levels were controlled to the precision vibration control blasting standard. Rock weakening was confirmed through induced cracks observed on the surface and core samples, and it was noted that the weakening effect of the blasting decreased as the burden increased. The vibrations from conventional blasting were found to be lower than those from pre-fracturing. This was attributed more to the geological conditions, such as joints, rather than the blasting design factors like explosive amount, burden, and the number of free face.

Intestine Bacterial Microbiota of Asian Hornet (Vespa Velutina Nigrithorax) and Honey Bee (등검은말벌과 꿀벌의 장내 세균 군집 비교)

  • BACKGROUND: The Asian hornet (Vespa velutina nigrithorax), a wasp species, has attacked honey bee populations and affected the beekeeping industry in Korea over the past 15 years. However, little research has been done with this invasive species. In this study, we investigated the intestine bacterial microbiota of Asian hornets and honey bees to design an attractive trap for Asian hornets. METHODS AND RESULTS: Genomic DNAs isolated from the intestine microorganisms of Asian hornets and honey bees were utilized to amplify bacterial 16S rDNA for the comparative sequence analysis. The next generation sequencing analysis identified that the orders Flavobacteriales as the most abundant intestinal microorganisms in Asian hornets, showing a clear difference compared to honey bees in which Aeromonadales are dominant. We also report five newly identified 16S rDNA sequences of Asian hornet intestinal bacteria. According to the sequence blast search, these five bacteria belong to the genera Thalassomonas, Caedobacter, Vampirovibrio, Alkaliphilus and Calothrix. CONCLUSION: While Asian hornets and honey bees show similar intestine bacterial diversity, the relative ratio of bacterial populations is different. providing useful information to design pest control agents specifically targeting Asian hornets.

A Fundamental Study about the Applicability of Mud Flat as a Concrete Admixture and Filler (갯벌의 콘크리트용 혼화재 및 채움재로서의 활용가능성에 대한 기초적 연구)

  • Recently, review on viability of various industrial by product and natural materials as raw material for concrete has been actively done in aspect of environment-friendly issue and depletion of natural resource. This study conducted fundamental study on the possibility of utilizing mud flat as admixture and filling material for concrete. First, chemical analysis on the viability of mud flat as admixture was done and the researchers compared it with the substance of fly ash and blast furnace slag. According to the result, substance content was proven to be inadequate. In addition, as the replacement rate of mud flat increased, compressive strength and tensile strength decreased. According to the estimated result of chemical substance analysis, possibility of utilizing mud flat as admixture was low. According to the result of experiment done as filling material, 10% ~ 30% replacement rate of mud flat manifested more than 8 Mpa of compressive strength of block which may be utilized for secondary product. However, additional experiment such as making block is required afterward. According to the result of flow experiment, as the replacement rate of mud flat increased, flow value decreased, and through chloride content analysis test, it was proven that mud flat is inappropriate to be applied as steel beam using structure since it has high content of sodium. It may be utilized as products that does not use steel beam such as internal brick.

  • Abstract The characteristics of bed rock in the study area was classified by means of the crack coefficient estimated from the seismic velocities of in-situ and intact rocks. Various statistical methods were investigated in order to minimize the possible errors in estimating the predictive equation of blasting vibration and to enhance the determination coefficient $R^2$, for more reliable estimation. The determination coefficient showed the highest in the analysis for those groups using weighting function with the number of samples. The analysis for the weighting function employed with standard coefficient and variance also enhanced the determination coefficients significantly compared to the others, but the reliability was slightly lower than results obtained former method. Therefore the most reliable predictive equation of blasting vibration was found to be obtained from a regression analysis of the mean vibration level using the weighting of same distance groups within 15m with the same explosive charge weight per delay. The coefficients, K and n 317.4 and -1.66, respectively, when using the square root scaling, and 209.9 and -1.66, respectively, when using the cube root scaling. The analysis also showed that the square root scaling may be used in the distance less than 31m form the blast source, and the cube root scaling in the distance more than 31m for safe design.

Evaluation of Applicability of CMD-SOIL Recycled Resources as Ground Improvement Material for Deep Mixing Method (심층혼합공법용 지반개량재로서 순환자원을 재활용한 CMD-SOIL의 적용성 평가)

  • As port development in soft ground is actively promoted for international logistics and transportation, the Deep Mixing Method (DMM) is continuously applied to form an improved column body directly in the ground by mixing cement with soil to secure the stability of the structure. However, in the case of cement, there is a problem of emitting a lot of greenhouse gases during the production process, so the development and use of new alternative materials are socially required to achieve the national goal of carbon neutrality. Accordingly, in this study, CMD-SOIL, developed to induce a hardening reaction similar to cement by recycling recycled resources, was used as a ground improvement material for the DMM. In addition, it was attempted to determine the possibility of replacing cement by conducting on-site test construction and evaluating applicability. As a result of the study, the compressive strength of CMD-SOIL compared to the design reference strength was 1.46 to 2.64 times higher in the field mixing test and 1.2 to 5.03 times higher than in the confirmed boring. In addition, the ratio (λ) of the compressive strength in the field to the design reference strength was 0.63 to 1.14, which was similar to the previous research results. Therefore, in the case of CMD-SOIL, it is possible to express the compressive strength necessary to secure stability, and there is no difference in applicability compared to existing materials such as ordinary portland cement and blast furnace slag cement, so it was analyzed that it could be used as a ground improvement material for the DMM.
