본 연구에서는 인간 간암세포주 HepG2세포에서 $H_2O_2$로 인해 유도된 산화적 스트레스에 대한 방사선 돌연변이 블랙베리 ${\gamma}$-B201 추출물의 세포보호 효능에 대하여 알아보고자 하였다. ${\gamma}$-B201 추출물 처리 시 $H_2O_2$로 처리된 HepG2 세포에서 ROS 생성이 억제되었으며, 세포 생존율을 증가시키고 LDH의 방출을 억제함을 확인하였다. Comet assay를 통해 DNA 손상 정도를 분석한 결과, $H_2O_2$로 인해 유도된 산화적 스트레스에 의한 DNA 손상은 ${\gamma}$-B201 추출물의 처리에 의해 감소하였다. 또한, HepG2 세포에 $H_2O_2$ 처리에 의해 저하된 항산화 효소인 SOD와 CAT의 활성을 ${\gamma}$-B201 추출물에 의해 증가시킴으로써 $H_2O_2$로 인해 유도된 산화적 스트레스로부터 HepG2 간세포를 보호하는 것으로 판단되며, ${\gamma}$-B201 추출물이 간 손상 보호 및 간 기능 개선 효과를 갖는 기능성 소재로서 활용될 수 있는 것으로 사료된다.
요즘은 "손 안의 컴퓨터"라 불리는 smartphone이 전 세계적으로 열풍이다. 휴대전화가 보급되어 일반사람들도 이동하면서 전화가 가능하게 된 지 얼마 되지도 않았다. Blackberry, apple, HTC, nokia, samsung, LG 등 수많은 제조업체들이 이미 smartphone 시장에 뛰어든 지 오래이며, 이들 간의 경쟁은 갈수록 치열해지고 있다. 이 논문은 이러한 우리들의 생각을 반영하여 만들게 된 프로그램에 대한 것이고 따라서, "smartphone OS 중 가장 개발자를 위한다"는 평을 받고 있는 Android 기반의 P2P 프로그램에 대한 것이다. P2P는 많은 사람들이 파일공유를 목적으로 사용하는 프로그램으로, 이미 컴퓨터에서는 많이 사용되고 있다. 이는 트래커 서버 운영을 기반으로 한다. Android smartphone에서도 이러한 수요는 앞으로 급증할 것으로 보여서, 이 프로그램의 개발은 유저의 요구를 반영할 수 있는 의미있는 작업이라 하겠다.
본 연구에 사용한 식물체는 Rubus fruticosus L. cv. BB21은 Rubus fruticosus L. ${\times}$ R. parvifolius L. 교잡 블랙베리 V3품종에서 방사선을 처리하여 돌연변이로 선발 육종된 V9개체 중 다시 방사선을 처리하여 선발육종 된 품종이다. 이러한 우량계통의 신속한 증식을 이루기 위하여 조직 배양을 통한 효과적인 방법을 구명하고자 배지 및 생장조절물질 등의 처리에 따른 안정적인 생산을 위해 본 연구가 수행되었다. 땅속에서 채집된 근맹아 절편체를 0.1, 0.5, 1.0, 2.0 mg/L BAP가 첨가된 MS와 1/2MS 배지에 치상하여 6주간 배양한 후, 다경의 형성률을 조사하였다. 형성된 다경으로부터 산업적으로 이용하기 위한 안정된 생산을 위하여, 신초의 잎을 제거한 후 절간배양을 수행하여 다수의 소식물체를 증식하고자 여러종류의 배지 즉, AD (Anderson 1980), B5 (Gamborg 1968), MS (Murashige and Skoog 1962), SH (Schenk and Hildebrandt 1972), QL (Quoirin and Lepoivre 1977), WPM (Lloyd and McCown 1981)배지와 상기의 각각의 동일배지의 무기염류의 수준을 절반으로 한 1/2배지에 배양하였다. 초기의 생육(30일간 배양 시)에 있어서 생장은 배지 간에 차이를 나타내어 1/2AD, 1/2QL 배지에서 가장 양호한 초장을 나타내었으며, 절간수도 약 3.8, 4.4개를 나타내었으며, 뿌리, 생중량 다경유도 모두 양호하였다. 90일간 배양한 경우에서는 MS배지가 초장이 가장 양호하였으며 AD, SH 순으로 양호한 결과를 나타내었다. 식물체의 토양 순화는 신초와 뿌리가 잘 발달된 정상적인 식물체를 모래, 토양, 그리고 버뮤큘라이트가 혼합된 토양(1:1:1, v/v)에 각각 직접 이식하여 실시한 결과, 식물체의 발근율과 생존률(약98%)을 나타내었다. 이상의 결과를 통하여 블랙베리 묘목의 대량생산이 가능하여 농가에 원활한 공급이 가능할 것으로 예상된다.
본 연구에서는 국내에서 재배되거나 야생의 나무딸기류 20 종류를 수집하여 무게, 수분함량, 과즙의 당도, pH, 산도, pH를 측정하고, 과즙의 당 조성 및 유기산 조성 분석하여 각각의 특성을 검토하였다. 실험에 사용한 나무딸기류 과일의 무게는 $0.9{\sim}7.8g$이었고, 과일의 수분함량은 $82.0{\sim}90.3%$였다. 나무딸기류 과즙의 당도는 $6.9{\sim}16.9%$(Brix)로 다양하였다. HPLC로 분석한 나무딸기류 과즙의 당은 glucose(총당 함량의 $40.7{\sim}84.0%$(w/w))와 fructose($6.5{\sim}57.5%$)가 주를 이루고 있었고, 일부 과일에는 sucrose와 xylose도 미량 존재하였다. 과즙의 pH는 $2.8{\sim}3.9$였고, 적정산도는 $0.6{\sim}2.9%$(w/v, citric acid)였다. 나무딸기류 과즙의 유기산을 HPLC로 분석한 결과, citric acid(총산 함량의 $14.6{\sim}87.5%$(w/w))와 malic acid ($9.6{\sim}87.1%$)가 주를 이루고 있었고, 일부 과일에는 tartaric acid, succinic acid, oxalic acid 등이 함유되어 있었다.

More effective treatments are needed for non-alcoholic fatty liver disease (NAFLD). We hypothesized that water extracts of blackberry fruits (BF) and leaves (BL) and their combinations (BFL) reduce fat deposition in HepG2 cells and modulate shor-tchain fatty acids (SCFA) and fecal bacteria in vitro. HepG2 cells were treated with BF, BL, BFL1:2, and BFL1:3 for 1 h, and 0.5 mM palmitate was added to the cells. Moreover, low ($30{\mu}g/mL$) and high doses ($90{\mu}g/mL$) of BL and BF were applied to fecal bacteria in vitro, and SCFA was measured by GC. BL, BF, BFL1:2, and BFL1:3 reduced triglyceride deposition in the cells in a dose-dependent manner, and BFL1:2 and BFL1:3 had a stronger effect than BF. The content of malondialdehyde, an index of oxidative stress, was also reduced in BL, BF, and BFL1:2 with increasing superoxide dismutase and glutathione peroxidase activities. The mRNA expression of acetyl CoA carboxylase, fatty acid synthase, and sterol regulatory element-binding protein-1c was reduced in BL, BF, BFL1:2, and BFL1:3 compared to the control, and BFL1:2 had the strongest effect. By contrast, the carnitine palmitolytransferase-1expression, a regulator of fatty acid oxidation, increased mostly in BFL1:2 and BFL1:3. Tumor necrosis factor-${\alpha}$ and interleukin-$1{\beta}$ expression was reduced in BL compared to that in BF and BFL1:2 in HepG2 cells. Interestingly, BL increased propionate production, and BF increased butyrate and propionate production and increased total SCFA content in fecal incubation. BF increased the contents of Bifidobacteriales and Lactobacillales and decreased those of Clostridiales, whereas BL elevated the contents of Bacteroidales and decreased those of Enterobacteriales. In conclusion, BFL1:2 and BFL1:3 may be potential therapeutic candidates for NAFLD.
Factors affecting regeneration of different Rubus varieties (blackberry, raspberry and their hybrid) were examined and a reliable regeneration system was established. Media for stock plant maintenance were tested; different explants and media were investigated to find the best circumstances for the regeneration. The effect of the commonly used antibiotics was studied to determine the most suitable one for selection of the transformants. We found that both MS and LS media supplemented by $20\;gL^{-1}$ sucrose are suitable for the stock plant maintenance. The optimal hormone content for the stock plants is $0.125\;mgL^{-1}$ 6-benzylaminopurine (BAP) with $0.01\;mgL^{-1}$ indole-3- butyric acid (IBA). The highest regeneration rate was observed on medium containing MS salts with B5 vitamins complemented with glucose, sucrose, maltose, $10\;gL^{-1}$ each, supplemented with benzylaminopurine riboside (BAR) ($2\;mgL^{-1}$) and indole-3-acetic acid (IAA) ($0.1\;mgL^{-1}$). The regenerated shoots appeared directly from the cut edges, without callus phase. Hygromycin and geneticin proved to be good selection agents for the Rubus explants, but due to their severe effect on the tissues we propose to use marker-free constructions for the transformation.
Colored compounds adsorption from the textile dyeing effluents on activated carbons produced from various indigenous vegetable sources by zinc chloride activation is studied. The most important parameters in chemical activation were found to be the chemical ratio of $ZnCl_2$ to feed (3:1), carbonization temperature (460-470 $^{\circ}C$) and time of activation (75 min). The absorbance at 511 nm (red effluent) and 615 nm (blue effluent) are used for estimation of color. It is established that at optimum temperature ($50^{\circ}C$), time of contact (30-40 min) and adsorbent loading (2 g/L), activated carbons developed from rain tree (Samanea saman) saw dust and blackberry (Randia formosa) tree saw dust showed great capability to remove color materials from the effluents. It is observed that adsorption of reactive dyes by all types of activated carbons is more than that of disperse dyes. It is explained that because of its acidic nature the activated carbon can adsorb better reactive dye particles containing large number of nitrogen sites and $-SO_3Na$ group in their structure. The use of activated carbons from the indigenous biomass would be economical, because saw dusts are readily available waste worldwide.
The bramble cultivated widely in South Korea, which is known as Bokbunja-ddal-gi, is regarded as having originated from Korean native Rubus coreanus. This study was carried out to obtain basic phylogenetic information on Korean cultivated bramble (KCB) by comparing the internal transcribed spacer (ITS) regions with those of R. coreanus, blackberry (R. lanciniatus), black (R. occidentalis) and red (R. idaeus) raspberry. Sequences of the ITS 1 suggest that some KCB accessions share a significant similarity with both R. occidentalis and R. coreanus in the ITS 1 region. The ITS 2 sequences of the three KCB accessions clustered more closely to those of two R. occidentalis accessions than to those of R. coreanus. These results suggest that there exist variations in the sequences of ITS among KCB accessions and KCB accessions are more closely related to black raspberry than R. coreanus in the ITS regions.
Korean cultivated bramble, which is known as Bokbunja-ddal-gi is regarded to be originated from Korea native Rubus coreanus. However, little scientific evidence and significant morphological differences between Korean cultivated bramble(KCB) and R. coreanus throw doubt on the ancestry of KCB. This study was carried out to obtain phylogenetic information on KCB by comparing its nuclear genomic background with those of R. coreanus, black(R. occidentalis) and red(R. idaeus) raspberry, blackberry(R. lanciniatus) and R. crataegifolius. A total of 99 random amplified polymorphic DNA(RAPD) markers were generated and used for phylogenetic analysis of 76 Rubus accessions. Accessions of each species were grouped into each distinct subclade by the RAPD markers at a similarity coefficient of about 0.59. The KCB subclade formed a clade with R. occidentalis and R. crataegifolius subclades at a similarity coefficient of 0.47. The R. coreanus subclade formed a clade with R. idaeus, R. lanciniatus and R. crataegifolius subclades at a similar similarity coefficient. Only one KCB accession from Hoengsung was included in R. coreanus subclade. The accession shows leaf and flower characteristics different from the rest of the KCB accessions. The phylogenetic relationship inferred from the RAPD markers suggests that the nuclear genomic background of KCB accessions which show morphological similarity to black raspberry is more closely related to black raspberry than to R. coreanus. This brings about the need for close scientific evaluations on the ancestry of KCB at both morphological and molecular levels.
피부의 주름은 노화과정의 결과로 유도되어지는데, 비효소적 glycation 반응은 주름생성의 원인 중에 하나로 단백질의 비가역적 가교결합에 의해 생성된 최종당화산물(AGEs)이라고 불리는 갈색화합물을 생성한다. 비교적 반감기가 긴 collagen은 쉽게 glycation이 일어나고 collagen의 가교결합(cross-linking)이 일어나면서 여러 단계의 반응을 거쳐 최종적으로 생성된 AGEs가 피부에 축적되어 피부의 탄력감소 및 주름을 일으킨다고 알려져 있다. 피부 내에 glycation된 collagen의 축척은 피부의 탄력성을 유지하는 것을 방해한다. 수종의 식용작물로부터 항주름소재를 개발하기 위하여 메탄올 및 열수추출물을 조제한 후 AGEs 형성을 억제하는 활성을 검색하였다. 먼저 서양향신료 중에서는 월계수잎, 계피, 정향, 오레가노, 로즈마리, 사보리, 팔각과 한약재 중에서는 복분자, 달개비, 삼지구엽초, 가자와 해당화의 메탄올 및 열수추출물에서 최종당화산물 생성 억제활성이 높았다. 그러나 국내 식물 중에서는 단지 참취 및 녹차 추출물의 메탄올과 열수추출물만이 높은 활성을 보였으며 해조류 중에서는 활성이 높은 시료는 없었다.
