Previous experiment was performed under abiotic condition. Steady-state abiotic experiments in the sand-tank reactor with air flowing through the reactor headspace demonstrated that oxygen supply through the water table interface into the saturated zone was enhanced when an LNAPL (dodecane) pool was present at the water table. Biotic condition was considered in this study. Biotic experiments performed after inoculating the reactor with Pseudomonas putida mt-2, which does not grow on dodecane, indicated that the enhanced oxygen supply in the presence of the LNAPL pool also enhanced biodegradation of a solute (glucose) plume passing beneath the LNAPL pool at steady-state.
Background: Ambrosia trifida is a highly invasive annual plant, but effective control methods have not been proposed. Among various eradication methods, cutting is a simple measure to control invasive plants, and sowing seeds of native plants may effectively increase biotic resistance to invasion. In this study, we conducted a field experiment with two treatments: cutting and sowing seeds of six native or naturalized plants. Results: We found a significantly lower A. trifida abundance after cutting than in the control (77% decrease). Sowing seeds of native species did not provide any additional benefit for the control of A. trifida, but increased the importance values and diversity of other native vegetation. The abundance of A. trifida was negatively correlated with that of other plant taxa based on plant cover, biomass, and density. However, biotic resistance of sown plants was not effective to control invasion because A. trifida was so competitive. Conclusions: We concluded that cutting is an effective measure to control Ambrosia trifida while sowing seeds of native plants can increase native plant diversity.

Recently, there has been increased attention to the development of new plant cultivars with enhanced resistance to biotic and abiotic stress. To develop ginseng cultivars with such traits, systematic breeding programs and comprehensive field studies are prerequisites. In this study, we applied a pure-line selection method to identify a ginseng cultivar with enhanced stress resistance. Phenotypic and agronomic characteristics, seed yield, and physiological responses to biotic and abiotic stresses were investigated according to the guidelines of the International Union for the Protection of New Varieties of Plants (UPOV). In the newly developed 'Kowon' cultivar, the time of emergence, flowering, and berry maturity were intermediate between those of the controls, 'Yunpoong' and 'Chunpoong'. The stem length of 'Kowon' was intermediate, whereas the root length was shorter and the main root diameter was greater than those of 'Chunpoong'. In local adaptability tests conducted in three regions, the yield of 'Kowon' was $666kg{\cdot}10a^{-1}$; 27% and 4% higher than that of 'Chunpoong' and 'Yunpoong'. Diseases such as Alternaria blight, Phytophthora blight, mulberry mealybug, and nematode infestation did not occur in 'Kowon'; and it also exhibited moderate resistance to damping-off and anthracnose. In these cases, yellow spots occurred on aerial parts and the rusty skin of the root, and it exhibited moderate resistance at high temperatures. Our study demonstrates that 'Kowon', which has a high root weight and enhanced biotic/abiotic stress resistance, is a superior cultivar that could increase farmers' income.

The mitogen-activated protein kinase (MAPK) signaling cascade is critical for regulating plant defense systems against various kinds of pathogen and environmental stresses. One component of this cascade, the MAP kinase kinases (MAPKK), has not yet been shown to be induced in plants following biotic attacks, such as those by insects and fungi. We describe here a gene coding for a blast (Magnaporthe grisea)- and insect (Nilaparvata lugens)-responsive putative MAPK kinase, OmMKK1 (Oryza minuta MAPKK 1), which was identified in a library of O. minuta expressed sequence tags (ESTs). Two copies of OmMKK1 are present in the O. minuta genome. They encode a predicted protein with molecular mass 39 kDa and pI of 6.2. Transcript patterns following imbibition of plant hormones such as methyl jasmonic acid (MeJA), ethephone, salicylic acid (SA) and abscisic acid (ABA), as well as exposure to methyl viologen (MV), revealed that the expression of OmMKK1 is related to defense response signaling pathways. A comparative analysis of OmMKK1 and its O. sativa ortholog OsMKK1 showed that both were induced by stress-related hormones and biotic stresses, but that the kinetics of their responses differed despite their high amino acid sequence identity (96%).

It has been known that endophyte (Neotyphodium coenophialum) is beneficial to tall fescue (Schedonorus phoenix Scop.) because the mutualistic endophyte is able to confers tolerance against abiotic and biotic stresses to tall fescue. However, this fungal endophyte produces toxic alkaloid resulting in negative effects on animal performance. Recently, Non-toxic endophyte have been developed and inserted into tall fescue to avoid detrimental effect on animal but remaining positive influence on tall fescue. In order to keep this beneficial impact, it is essential to have endophyte infected tall fescue through vertical transmission from maternal plants to seeds. Little research has been carried out on endophyte transmission. To get basic information related to endophyte transmission, experiment was conducted to examine the effect of plant genotype on endophyte transmission under water stresses. Overall endophyte concentration in seeds was higher than that in panicles and endophyte concentration in seeds and panicles relied on plant. This study revealed that drought is not a critical component to control the endophyte transmission from maternal plants to seeds. Plant genotype is an important factor controlling the endophyte transmission from plant to seed.
Overwintering plants should survive the various biotic and abiotic stresses that occur during winter. Previous studies indicated that active oxygen species are involved in freezing, dehydration, anoxia and pathogen infections. As the importance of the events that occur in the intercellular compartment became apparent in disease resistance, we examined the nature of intercellular antioxidant enzymes in order to access their possible involvement in the winter hardiness of barley. The levels of intercellular peroxidase, catalase, and SOD activities on the unit protein basis were 394, 18, and 9% of those of cellular activities, respectively. Major intercellular peroxidase isoforms consisted of four neutrals and four basic forms; whereas major cellular isoforms were two basic forms. Out of the two major catalase isoforms a higher molecular weight form was predominantly abundant in both cellular and intercellular compartments. Among the five major cellular SOD isoforms, three were also present in the intercellular compartment. The presence of substantial amounts of intercellular antioxidant enzymes in overwintering barley leaves may suggest the involvement of these enzymes in the tolerance mechanism to the various stresses that occur during winter.
Tall fescue (Schedonorus phoenix Scop.) is resistant to abiotic and biotic stresses through a symbiotic relationship with Neotyphodium coenophialum. However, this endophyte has been considered detrimental since it produces toxic alkaloids to animals. It is vital to understand mutuality between these two to maximize positive impact of the endophyte on agri-ecosystem. Little research has been conducted on endophyte transmission mechanism in planta. To provide basic information related to endophyte transmission, an experiment was conducted to examine the effect of endophyte genotype and water stress on endophyte transmission by imposing soil moisture deficits at different stages of panicle development. There was water stress effect on endophyte frequency but not on concentration, whereas endophyte genotype significantly influenced endophyte concentration in pseudostem of tall fescue at boot stage. Reproductive tillers showed greater endophyte frequency and concentration. Endophyte frequency in florets or seeds depended on position within panicle. There was no drought effect on endophyte concentration, but showed the effect of endophyte genotype on endophyte concentration in florets and seeds. Overall endophyte concentration in seeds was higher. From this study, we may conclude that although water stress reduced endophyte frequency in vegetative tiller, water stress does not have effect on endophyte transmission, suggesting that drought is not an important factor controlling the endophyte transmission from plant to seed. Endophyte genotype and seed position in a panicle affected endophyte transmission, indicating that these two factors are involved in endophyte transmission and may determine seed transmission of endophyte in tall fescue.
We evaluated the field application feasibility that biologically derived substances (Naphthoquinone derivate: NQ 2-0) can be used for the eco-friendly mitigation of natural harmful cyanobacterial blooms in freshwater. We conducted a 30 ton scale mesocosm experiment to investigate the effects of NQ 2-0 on biotic and abiotic factors in water collected from Gi-heung reservoir. In the mesocosm experiments, the abundance of Microcystis sp. was continuously increased in the control. However, the Microcystis sp. cell density was sharply decreased on the $10^{th}$ day. In the treatment, NQ 2-0 showed the strong and selective algicidal activity toward the target cyanobacteria (Microcystis sp.). Accordingly, the algicidal activity of NQ 2-0 compound increased gradually until $10^{th}$, $15^{th}$ days and algal biomass was decreased to 99.4 and 100 %, respectively. NQ 2-0 compound was not only selective algicidal activity but also the growth of other phytoplankton and increased the Shannon-Wiener diversity index of phytoplankton. In the mesocosm experiments, the dynamics of biotic (bacteria, heterotrophic nanoflagellate, ciliates, zooplankton) and abiotic (water temperature, dissolved oxygen, pH, conductivity, nutrients) factors remained unaffected. These results suggest that NQ 2-0 could be a selective and ecologically safe algicide to mitigate harmful cyanobacterial blooms. In addition, it is believed that NQ 2-0 will play a major role in forming a healthy aquatic ecosystem by facilitating habitat and food supply of aquatic organisms.
Pinus densiflora stands are common secondary forest communities on infertile soils in Korea. The stands are results of long severe past biotic pressure such as cutting, burning and grazing. These could be regarded as biotic climax in Korea. Because of their prevalent occurrence, relatively simple species and age composition, and their domestic economic importance, study of their distributional patterns may give some basic knowledge for better utilization of land resources in Korea. To detect distributional patterns and interspecific associations ten pine stands, each of which was homogenious with respect to topography and physiognomy, were subjectively selected from pine stands in Kyunggi Province near Seoul in 1969 and were made object of this study. Four contiguous systematic samples of count for trees, shrubs and seedlings from belt transects were collected from homogeneous areas within ten natural pine stands. The belt transect was 64m or 128m in length, and 1m, 2m or 4m in width. Basic units within the transect ranged from 64 to 256. The data from the contiguous transects were analysed in terms of multiple split-plot experiment. Departure from randomness of stem distribution, i.e., pattern, was tested in terms of variance mean ratio. For the detection of association between species, correlation coefficient was calculated for different block sizes. The values of ${\gamma}$ were tested by the usual t-test. Fine trees within one of the stands showed significant regular distribution through out the blocks. Within other eight stands pines were randomly distributed at basic unit with 4$\times$4m, 2$\times$2m, 2$\times$1m and 1$\times$1m. One significantly clumped distribution at basic unit 2$\times$2m, however, was observed from one of the pine stands. These randomly distributed groups were themselves significantly regularly distributed throughout the blocks for four pine stands. For the other four pine stands, in addition to the random distribution at the basie unit(the primary random group), randomly distributed groups with 32m dimension(the secondary random groups) were also observed. Both the primary and the secondary random groups were significantly regularly distributed at the rest of blocks. Pine seedlings were not distributed randomly thoughout the blocks. Within three of the ten stands they were contagiously distributed. Important shrub species underneath pines such as Querus serrata, Q. acutissima, Leapedeza intermedia, Rhododendron Yedoense var. poukhanenae, Juniperus utilis, Rhododendron mucronulatum var. ciliatum shnwed consistently similar distributional pattern with the pine at each stand. The shrub species pairs; Rhododendron Yedoense var. poukhanenae/Quercus serrata, Rhododendron mucronulatum var. ciliatum/Lespedeza intermedia were significantly negatively associated from 1m to 4m dimensional block sizes but became significantly positively associated from 8m sized blocks on. On the other hand the shrub species pairs; Lespedeza intermedia/Robinia Pseudoacacia, and Lespedeza bicolor var, japonica/Lespedeza intermedia were also significnalty negatively associated from 1m to 8m sized blocks but became significantly positively associated from 16m sized blocks on. The associational pattern between Rhododendron mucronul tun var. poukhanenae and Lespedeza intermedia was not consistent throughout the stands. In some stands negative associations were observed throughout the blocks except NS 32. From these observatons micro-edaphic variation within the pine stands seems not to be great enough to cause distributinal difference of pine trees within the ten pine stands. Among each species and pine seedings, however, the edaphic variation within the pine stands may be great enought to cause distributional variation.
Effects of various concentrations of elicitors for enhancement in valepotriates and valerenic acids production in adventitious root culture of Valeriana fauriei were investigated. Although the growth of adventitious roots was suppresed by addition of biotic or abiotic elicitors, the production of valepotriates and valerenic acids was enhanced. Addition of $100\;{\mu}M$ methyl jasmonate or 1 $g/{\ell}$ yeast extract to the root culture of V. fauriei resulted in the optimal production of valepotriates ($12.56\;{\pm}\;0.78\;mg/{\ell}$) and valerenic acids ($10.63\;{\pm}\;1.1\;mg/{\ell}$), respectively.
