Biokinetics of Protein Degrading Clostridium cadaveris and Clostridium sporogenes in Batch and Continuous Mode of Operations

  • A quantitative real-time polymerase chain reaction (QPCR) was applied to estimate biokinetic coefficients of Clostridium cadaveris and Clostridium sporogenes, which utilize protein as carbon source. Experimental data on changes in peptone concentration and 16S rRNA gene copy numbers of C. cadaveris and C. sporogenes were fitted to model. The fourth-order Runge-Kutta approximation with non-linear least squares analysis was employed to solve the ordinary differential equations to estimate biokinetic coefficients. The maximum specific growth rate (μmax), half-saturation concentration (Ks), growth yield (Y), and decay coefficient (Kd) of C. cadaveris and C.sporogenes were 0.73 ± 0.05 and 1.35 ± 0.32 h-1, 6.07 ± 1.52 and 5.67 ± 1.53 g/l, 2.25 ± 0.75 × 1010 and 7.92 ± 3.71 × 109 copies/g, 0.002 ± 0.003 and 0.002 ± 0.001 h-1, respectively. The theoretical specific growth rate of C. sporogenes always exceeded that of C. cadaveris at peptone concentration higher than 3.62 g/l. When the influent peptone concentration was 5.0 g/l, the concentration of C.cadaveris gradually decreased to the steady value of 2.9 × 1010 copies/ml at 4 h Hydraulic retention time (HRT), which indicates a 67.1% reduction of the initial population, but the wash out occurred at HRTs of 1.9 and 3.2 h. The 16S rRNA gene copy numbers of C. sporogenes gradually decreased to steady values ranging from 1.1 × 1010 to 2.9 × 1010 copies/ml. C. sporogenes species was predicted to wash out at an HRT of 1.6 h.

  • This study was carried out to evaluate the economic value of Tyvek planting technique in Korean citrus. The Tyvek planting technique was developed by national institute of horticultural and herbal science in Korea rural development administration. This technology could be used to improve efficiency and practicality of cultivation of citrus. The research results show that the technological value of Tyvek planting technique was evaluated 42,163(discount rate 6%)-55,018(discount rate 8%) million won. IRR is greater than discount rate(6-10%). NPV is greater than zero. B/C ratio IRR is greater than 20. The economic validity of Tyvek planting technique is identified by the results of technology evaluation.

Association of SNP Markers on Chromosomes 3 and 9 with Body Weight in Jeju Horses (제주마에서 3번 및 9번 염색체상의 단일염기변이와 생체중과의 관련성 연구)

  • This study was conducted to investigate the association of single nucleotide polymorphism (SNP) markers on equine chromosomes (ECA) 3 and 9 with body weight in Jeju horses. We used DNA samples and body weight data of 320 horses provided by the Livestock Promotion Agency, Jeju Special Self-Governing Province, and the Korean Racing Association, respectively. We genotyped all the experimental animals using nine SNP markers located on ECA 3 (BIEC2-808466, BIEC2-808543, BIEC2-808967, and BIEC2-809370) and ECA 9 (BIEC2-1105370, BIEC2-1105372, BIEC2-1105377, BIEC21105505, and BIEC2-1105840). These markers were selected due to their effects on body conformation traits in horses. The joint effect of the genotypes of the two SNP markers (BIEC2-808467 and BIEC2-1105377) regarding body weight were also evaluated. The estimated breeding value (EBV) of body weight was obtained as the dependent variable for association analyses using a linear mixed model. Significant associations were detected between SNP markers (BIEC2-808543, BIEC2-808967, BIEC2-809370, BIEC2-1105370, BIEC2-1105372, and BIEC2-1105377) and the body weight EBV. In addition, the joint genotype effect of the BIEC2-808467 and BIEC2-1105377 on the body weight EBV was significant. These results indicate that the SNP markers, which showed their significant effects on body conformation, can be used as genetic markers to improve the efficiency of the selective breeding program for the body weight traits in Jeju horses.

Alternative Sweetener of Sucrose by using Threshold Value and Effects of Salt Addition on the Sweetness (역치를 활용한 설탕 대체 감미료의 적용 및 감미도에 대한 식염의 효과)

  • In this study the expression of the sweetness by using threshold value and effects of salt addition on the sweetness was carried out in model system. Threshold value was 0.4% in sucrose solution and was 0.75-0.80% in the mixture of sweeteners such as low glutinous starch syrup and reduced starch syrup with sucrose solution. The equation between mixture of sweeteners and relative sweetness ($0-20^{\circ}Brix$) on sucrose was induced. The sweetness of red bean paste prepared with low glutinous starch syrup and reduced starch syrup were estimated as 32.78-34.32 by using threshold value, and those lower than 55 of red bean paste prepared with sucrose alone. The synergistic effect of sweetness was observed when 0.05% salt in sugar solution (2.0%) and 0.2% salt in red bean paste were added, respectively. It showed that the addition of salt could be increase the degree of sweetness and decrease the quantity of sweeteners.

Application of Gamma Ray Irradiation to the Microbiological Safety of Dried Seafood Products (건조 수산물의 미생물학적 안전성 확보를 위한 감마선 조사 기술의 이용)

  • This study evaluated the effects of gamma ray irradiation on the safety of dried seafood products. Dried salted squid, Engraulis japonica, Hizikia fusiformis, Mytilus coruscus, and Porphyra tenera were gamma-irradiated at doses of 0, 1, 3, and 5 kGy. The total bacterial populations were then enumerated on total plate count agar, and bacteria isolated from the samples were identified by 16S rDNA sequencing. In addition, the isolated strains were inoculated in the products to determine $D_{10}$ values. The total bacterial populations in the dried seafood products ranged from 3.40 to 6.59 log CFU/g, and those of yeasts and molds ranged from 2.21 to 4.56 log CFU/g. The sequence analysis identified Staphylococcus sp. as the most common species in the dried seafood products, except for dried P. tenera. The $D_{10}$ values of the contaminating bacteria were lower than 0.7 kGy, except for Deinococcus sp., which had a value of 1.39 kGy. This study demonstrated that gamma irradiation could be used to improve the safety of dried seafood products.

Antibacterial Activity of Pharbitin, Isolated from the Seeds of Pharbitis nil, against Various Plant Pathogenic Bacteria

  • This study aimed to isolate and characterize antibacterial metabolites from Pharbitis nil seeds and investigate their antibacterial activity against various plant pathogenic bacteria. The methanol extract of P. nil seeds showed the strongest activity against Xanthomonas arboricola pv. pruni (Xap) with a minimum inhibition concentration (MIC) value of $250{\mu}g/ml$. Among the three solvent layers obtained from the methanol extract of P. nil seeds, only the butanol layer displayed the activity with an MIC value of $125{\mu}g/ml$ against Xap. An antibacterial fraction was obtained from P. nil seeds by repeated column chromatography and identified as pharbitin, a crude resin glycoside, by instrumental analysis. The antibacterial activity of pharbitin was tested in vitro against 14 phytopathogenic bacteria, and it was found to inhibit Ralstonia solanacearum and four Xanthomonas species. The minimum inhibitory concentration values against the five bacteria were $125-500{\mu}g/ml$ for the n-butanol layer and $31.25-125{\mu}g/ml$ for pharbitin. In a detached peach leaf assay, it effectively suppressed the development of bacterial leaf spot, with a control value of 87.5% at $500{\mu}g/ml$. In addition, pharbitin strongly reduced the development of bacterial wilt on tomato seedlings by 97.4% at $250{\mu}g/ml$, 7 days after inoculation. These findings suggest that the crude extract of P. nil seeds can be used as an alternative biopesticide for the control of plant diseases caused by R. solanacearum and Xanthomonas spp. This is the first report on the antibacterial activity of pharbitin against phytopathogenic bacteria.

Supercritical $CO_2$ Extraction of Sesame Oil with High Content of Tocopherol (초임계 이산화탄소를 이용한 토코페롤 고함유 참기름 추출)

  • The characteristics of sesame oil containing one of natural antioxidant, ' $\gamma$-tocopherol', were studied with the supercritical $CO_2$ extraction. Although $\gamma$-tocopherol has a lower vitamin E value in biological systems than $\alpha$-tocopherol, it is a more potent antioxidant with in oils. For the research of various factors influence to the $\gamma$-tocopherol contents increment, we have checked roasting time and temperature, as well as pressure, temperature and flow rate of supercritical fluid. As a result, we found that the $\gamma$-tocopherol content was maintained constant under the condition of roasting temperature over $200^{\circ}C$. With the longer roasting time, $\gamma$-tocopherol content was increased. Except 250 bar, the $\gamma$-tocopherol content was maintained constant under the condition of the various pressure of supercritical fluid. But $\gamma$-tocopherol content was increased with lower flow rate of supercritical fluid from 1 $m{\ell}$/L to 3 $m{\ell}$/L. When the extraction performance with the supercritical fluid was compared to the conventional compressed extraction, $\gamma$-tocopherol content was increased up to 1.6 times.

An Evaluation of the Anti-thrombosis and Antioxidant Activities of Different Parts of Dystaenia takeshimana (섬바디 부위별 추출물의 항혈전 및 항산화 활성)

  • Dystaenia takesimana is an endemic plant found only in Korea, especially on Ulleung Island. The leaves and roots of D. takesimana have been used as food, forage, and oriental medicine. Anti-bacterial, anti-inflammation, antioxidant, and α-glucosidase inhibition biological activities have been reported in the plant's root extract. However, studies concerning the anti-thrombosis activities of D. takesimana are still in the rudimentary stage. In this study, the extracts of the leaf (DT-L), stem (DT-S), and root (DT-R) of D. takesimana were prepared using 70% ethanol, and their anti-thrombosis activities were evaluated. DT-L extracts (0.25 mg/ml) showed strong inhibitions against platelet aggregation, comparable to aspirin, with strong radical scavenging activities. Furthermore, the DT-L extract did not show any RBC hemolysis up to 1 m/ml. The ant-coagulation and antioxidant activities of the DT-S extract were ignorable. While the DT-R extract showed inhibitions against thrombin and blood coagulation factors, it also showed strong platelet aggregation. This is a first report of the anti-thrombosis activities of D. takesimana, and our results suggest that DT-L could be developed as a valuable bioresource for high value-added products.

Comprehensive Characterization of Mutant Pichia stipitis Co-Fermenting Cellobiose and Xylose through Genomic and Transcriptomic Analyses

  • The development of a yeast strain capable of fermenting mixed sugars efficiently is crucial for producing biofuels and value-added materials from cellulosic biomass. Previously, a mutant Pichia stipitis YN14 strain capable of co-fermenting xylose and cellobiose was developed through evolutionary engineering of the wild-type P. stipitis CBS6054 strain, which was incapable of co-fermenting xylose and cellobiose. In this study, through genomic and transcriptomic analyses, we sought to investigate the reasons for the improved sugar metabolic performance of the mutant YN14 strain in comparison with the parental CBS6054 strain. Unfortunately, comparative whole-genome sequencing (WGS) showed no mutation in any of the genes involved in the cellobiose metabolism between the two strains. However, comparative RNA sequencing (RNA-seq) revealed that the YN14 strain had 101.2 times and 5.9 times higher expression levels of HXT2.3 and BGL2 genes involved in cellobiose metabolism, and 6.9 times and 75.9 times lower expression levels of COX17 and SOD2.2 genes involved in respiration, respectively, compared with the CBS6054 strain. This may explain how the YN14 strain enhanced cellobiose metabolic performance and shifted the direction of cellobiose metabolic flux from respiration to fermentation in the presence of cellobiose compared with the CBS6054 strain.
