Iron precipitating organisms play a significant role in the formation of ferric hydroxide precipitate, which acts as strong adsorbent for toxic metal. In this respect four different iron precipitating cultures were isolated from Hutti gold mine surface winze water sample on citrate agar medium. The best isolate was screened out for metal removal study on the basis of fast visual iron precipitation. The selected isolate was identified as Enterobacter sp. based on routine biochemical tests and Biolog GN microplate results and as Enterobacter cloacae subsp. dissolvens by 16S rRNA gene sequence analysis (GenBank accession number EU429448). Influence of medium composition, medium initial pH, the influence of inoculum size, effect of various media and ferric ammonium citrate concentration were studied on metal removal in shake flask experiments. Under the optimized conditions studied, E. cloacae showed $94{\pm}2$, $95{\pm}2$ and $70{\pm}2%$ of cadmium, copper and mercury removal from a simulated waste in shake flask studies. In lab scale column reactor more than 85% of copper and mercury removal was achieved.
Biosorption process is an economic and potential process for metal sequestering from the water. The oxidized Undaria pinnatifida by nitric acid had high uptake capacity for heavy metals of 4 - 6 meq / g dry mass. For the application of oxidized Undaria pinnatifida, recovery of metal in plating wastewater was studied. The uptake capacity of the oxidized Undaria pinnatifida was high compared to the ion exchanger IR-120 plus. The treatment efficiency of chromium and copper in the wastewater was 85% In batch. Activated carbon was used to assist the recovery of water by removing organic matters of the wastewater.
BACKGROUND: Heavy-metal pollution represents an important environmental problem due to the toxic effects of metals, and their accumulation throughout the food chain leads to serious ecological and health problems. METHODS AND RESULTS: Optimum conditions in continuous-flow stirred tank reactor (CSTR) and packedbed column contactor (PBCC) using brown seaweed biosorbent were investigated. Under optimum conditions from both lab-scale biosorbent systems, removal efficiency of copper (Cu) in a large-scale PBCC system was investigated. Removal capacity of Cu using brown seaweed biosorbent in a lab-scale CSTR system was higher than that in a lab-scale PBCC system. On the other hand, over 48 L/day of flow rate in Cu solution, removal efficiency of Cu in a lab-scale PBCC system was higher than that in a lab-scale CSTR system. Optimum flow rate of Cu was 24 L/day, optimum Cu solution concentration was 100 mg/L. Removal capacity of Cu at different stages was higher in the order of double column biosorption system > single column biosorption system. Under different heavy metals, removal capacities of heavy metal were higher in the order of Pb > Cr > Ni > Mn ${\geq}$ Cu ${\geq}$ Cd ${\fallingdotseq}$ Zn ${\geq}$ Co. Removal capacity of Cu was 138 L in a large-scale PBCC system. Removal capacity of Cu a large-scale PBCC system was similar with in a lab-scale PBCC system. CONCLUSION(s): Therefore, PBCC system using brown seaweed biosorbent was suitable for treating heavy metal wastewater.
An ubiquitous bacterium, Pseudomonar cepacia KH410 was isolated from fresh water plant root and identified. Adsorption of heavy metals of lead, cadmium and copper by this strain was investigated. Optimal conditions foradsorption was 1.0 dry g-biomass, at pH 4.0 and temperature of $40^{\circ}C$. Adsorption equilibrium reached max-imum after 120 min in 1000 mg/l metal solutions. The adsorption capacity (K) of lead was 5.6 times higher thancadmium and 4.0 times higher than that of copper. Adsorption of lead was applicable for Langmuir modelwhereas Freundlich model for cadmium and copper, respectively. Adsorption strength (1/n) of heavy metal ionswere in the order of lead>copper>cadmium. Uptake capacity of lead, cadmium and copper by dried cell was83.2,42.0,65.2 mg/g-biomass, respectively. Effective desorption was induced 0.1 M HCI for lead and 0.1 $HNO_3$ for cadmium and copper. Pretreatment to increase ion strength was the most effective with 0.1 M KOH.Uptake by immobilized cell was 77.8,58.5,71.2 mg/g-biomass for lead, cadmium and copper, respectively. Theimmobilized cell was more effective than ion exchange resin on removal of heavy metals in solution containinglight metals.
Some rhizobacteria were isolated, that have copper resistance and can confer copper resistance to plants allowing growth under copper stress. Isolated strains Pseudomonas veronii MS1 and P. migulae MS2 produced 0.13 and 0.26 mmol/ml of siderophore, that is a metal-chelating agent, and also showed 64.6 and 77.9% of biosorption ability for Cu in 20 mg/L Cu solution, respectively. Copper can catalyze a formation of harmful free radicals, which may cause oxidative stress in organisms. Removal activity of 1,1-diphenyl-2-picryl hydrazyl radical and antioxidant capacity of strains MS1 and MS2 increased up to 82.6 and 78.1%, respectively compared to those of control at 24 h of incubation. They exhibited 7.10 and $6.42{\mu}mol$${\alpha}$-ketobutyrate mg/h of 1-aminocyclopropane-1-carboxylic acid deaminase activity, respectively, which reduced levels of stress hormone, ethylene in plants, and also produced indole-3-acetic acid and salicyclic acid that can help plant growth under abiotic stress. All these results indicated that these copper-resistant rhizobacteria could confer copper resistance and growth promotion to plants.
In the present study, we examined biosorption characteristics of heavy metals onto the extracellular polysaccharide (EPS) produced by the purple nonsulfur photosynthetic bacteria Rhodopseudomonas sp. KH4, which was isolated from a stream in Anyang, Kyonggi-Do. When Cd (100 mg/L) and Cu (100 mg/L) were added to EPS (1.0 g/L) in the optimal condition (Cd; pH 8, Cu; pH 5, $40^{\circ}C$), 84.2 mg/L of Cd and 70.0 mg/L of Cu were adsorbed within 30 min and 10 min, respectively. When 100 mg/L of Cd and Cu were present as mixture, 16.8 mg/L of Cd and 48.7 mg/L of Cu were adsorbed at $25^{\circ}C$, pH 5. The maximum adsorption capacity determined by fitting Langmuir isotherms model was suitable for describing the biosorption of Cd (76.9 mg/g) and Cu (67.1 mg/g) by EPS. The neutral monosaccharide in the EPS determined by GC consisted of arabinose (2.4%), glucose (7.1%) and mannose (90.5%).
A natural habit at bacterium, Enterobacter intermedious KH410 was isolated from freshwater plant root and identified. Adsorption of heavy metals such as lead, cadmium, and copper by this strain was examined. The minimal inhibitory concentrations(MIC) for each metal were 1.78 mM for lead, 0.17 mM for cadmium and 1.39 mM for lopper, respectively. Maximum production of dried cell was 2.56 g/$\ell$ in LB medium containing 0.5% NaCl, 1% yeast extract and 1% of lactose. Optimal conditions for adsorption were 0.6 dry g-biomass, at pH 4.0 and the temperature of $20^{\circ}C$. Adsorption equilibrium reached maximum after 30 min in 400 mg/$\ell$ metal solution. The adsorption capacity (K) of copper was 1.5 times higher than that of cadmium and lead was 1.1 times higher than that of cadmium. from the results obtained in this study, Freundlich adsorption model was applicable for all metals. Adsorption strength (1/n) of heavy metal ions were in the order of cadmium>copper>lead. The adsorption of dried cell for lead, cadmium, and copper was 56.2, 58.0, 55.8 mg/g-biomass, respectively. Pretreatment to increase ion strength was the most effective with 0.1 M NaOH whereas slight difference was found both KOH and $CaCl_2$ upon same concentration. Effective desorption was induced by 0.1 M EDTA for lead and 0.1 M $HNO_2$ for cadmium and copper.

A study on the removal of heavy metals using alginic acid, a kind of polysaccharides, was performed. Alginic acid adsorbed 480 mg Pb/g dry mass at pH 4, which was about twice as high as uptake capacity of other biosorbents. Isothermal adsorption curve for lead ions was described by the Langmuir model equation and the experimental data well fitted to model equation. The adsorption of lead ions was an endothermic process since binding strength increased with temperature. The effect of alkali metal ions ($Ca^{2+}$ and $Mg^{2+}$) on lead sorption capacity was negligible and most adsorption process was completed in 30min. The uptake capacity of other metals such as, copper, mercury, strontium, and cesium ions using alginic acid was also investigated.
The resistant effect of several heavy metal ions to Serratia marcescens strain P was studied by the method of minimal inhibitory concentration(MIC), and testing for their metal biosorption. S. marcescens strain P showed a good survival in the presence of high concentrations of some metal ions, namely cadmium, lead, iron, magnesium, and manganese. Copper had the most inhibitory effect among tested. The MIC value was ranged from 0.79 to 1.58 mM. Cells of S. marcescens strain P exhibit an abnormally long lag phase when incubated in high concentrations of zinc and cadmium. Pigment production was reduced by zinc and cadmium, but enhanced by lead and iron. S. marcescens strain P was resistant to ampicillin, tetracycline, cefamandole and chloramphenicol with minimal inhibitory concentration of 128 $\mu$g/ml, 32 $\mu$g/ml, 256 $\mu$g/ml, and 8 $\mu$g/ml, respectively. The kinetics study of biosorptive uptake by S. marcescens strain P revealed that 16.59% of cadmium and 35.38% of lead were eliminated from the media.
Bioremediation is a technique using microorganisms to clean up contaminated pollutants including heavy metals. It is well known that yeasts have a high capacity to remove a wide range of metals by biosorption. Therefore, this study was focused on to obtain yeast mutant that has strong tolerance to zinc (Zn), one of representative heavy metals. The Zn resistant yeast mutant (ZnR) was induced and isolated by growing yeast cells in media containing 1 mM $ZnCl_2$ and gradually increasing the concentration until 80 mM $ZnCl_2$, in which cells were adapted and survived. The induced ZnR cells showed strong tolerance to Zn stress compared with control cells. Moreover, the ZnR cells showed increased tolerance to cadmium and nickel stress but decreased tolerance to copper stress. The increased tolerance of ZnR cells to Zn stress was due to mutation of genes. This study can be useful in bioremediation of heavy metals as the metal tolerant microorganism was artificially induced in short time.
