Recognition of Microorganisms Using SPR Biosensor Immobilized with Thiolated Antibody (티올화 항체고정형 SPR 바이오센서를 이용한 미생물 인식)

  • This study was performed to fabricate a batch-type SPR biosensing system using a thiolated E. coli antibody coupling, and to explore the feasibility of real-time detection of E. coii in a stagnant sample solution. In advance. “O” and “K” antigenic serotype E. coli antibodies were thiolated with sulfo-LC-SPDP and dithiothreitol, and immobilized by chemisorption in the gold surface of compact SPR sensors. When the SPR biosensor immobilized with E. coli antibody monitored a E. coli solution, it took 3 to 5 min to stabilize. The SPR biosensing system developed in this study was able to detect E. coli in the range above 10$^4$ CFU/mL at the 0.05 significant level. Also, the SPR biosensor had possibility to significantly detect E. coli in the range of 10$^2$ to 10$^4$ CFU/mL in E. coli solutions. Meanwhile, when the SPR biosensor immobilized with 5. coli antibody was cleaned with NaOH solutions, its ability to detect E. coli largely decreased due to wash-out of the immobilized antibody. In order to reuse the SPR sensor, it should be antibody-immobilized newly.

A Real-Time Surveillance System for Vaccine Cold Chain Based o n Internet of Things Technology

  • In this study, a real-time surveillance system using Internet of Things technology is proposed for vaccine cold chains. This system fully visualizes vaccine transport and storage. It comprises a 4G gateway module, lowpower and low-cost wireless temperature and humidity collection module (WTHCM), cloud service software platform, and phone app. The WTHCM is installed in freezers or truck-mounted cold chain cabinets to collect the temperature and humidity information of the vaccine storage environment. It then transmits the collected data to a gateway module in the radiofrequency_physical layer (RF_PHY). The RF_PHY is an interface for calling the bottom 2.4-GHz transceiver, which can realize a more flexible communication mode. The gateway module can simultaneously receive data from multiple acquisition terminals, process the received data depending on the protocol, and transmit the collated data to the cloud server platform via 4G or Wi-Fi. The cloud server platform primarily provides data storage, chart views, short-message warnings, and other functions. The phone app is designed to help users view and print temperature and humidity data concerning the transportation and storage of vaccines anytime and anywhere. Thus, this system provides a new vaccine management model for ensuring the safety and reliability of vaccines to a greater extent.

Nano-biomarker-Based Surface-Enhanced Raman Spectroscopy for Selective Diagnosis of Gallbladder and Liver Injury

  • During living donor liver transplantation, a number of blood vessels and bile ducts are anastomosed while the liver and gallbladder are resected in the donor and recipient. Early detection and treatment of complications after surgery by evaluating the function of blood vessels and the biliary tract is crucial. A biosensing chip that can monitor patient health status from the bile excreted during the recovery process has been developed using a surface-enhanced Raman sensing chip. Surface-enhanced Raman spectroscopy signals of bile obtained from normal bile duct ligation and gallbladder damage mouse models using a cautery device were identified and analyzed. The surface-enhanced Raman chip with a nanometer-level porous structure can selectively separate the nanometer biomarkers and measure the Raman signal. Through the detection of nanometer biomarkers in the bile and comparative analysis of histopathology, the Raman signal in the damaged gallbladder was compared with that caused by liver damage due to bile duct ligation, showing that it becomes a biosensing chip for monitoring recovery.

  • Liposomes and proteoliposomes, artificial membranes, can interact with many solutes, such as drugs, peptides and proteins. Immobilization of (proteo)liposomes as supramolecular aggregates on gold surfaces have potential applications in nanotechnology and biosensors. We demonstrate a quartz crystal analyzer (QCA) method to monitor the construction of multi layers of unilamellar liposomes based on avidin-biotin binding on gold surface using quartz crystal microbalance(QCM). Thus, QCA provides an on line and efficient method to detect the protein membranes construction and have applications to biosensing system.

  • Among the many studies of carbon-based nanomaterials, carbon-based dots (CDs) have attracted considerable interest owing to their large surface area, intrinsic low-toxicity, excellent biocompatibility, high solubility, and low-cost with environmentally friendly routes, as well as their ability for modification with other nanomaterials. CDs have several applications in biosensing, photocatalysis, bioimaging, and nanomedicine. In addition, the fascinating electrochemical properties of CDs, including high active surface area, excellent electrical conductivity, electrocatalytic activity, high porosity, and adsorption capability, make them potential candidates for electrochemical sensing materials. This paper reviews the recent developments and synthesis of CDs and their composites for the proposed electrochemical sensing platforms. The electrochemical principles and future perspective and challenges of electrochemical biosensors are also discussed based on CDs-nanocomposites.

  • Inorganic III-V light emitting diodes (LEDs) have superior characteristics, such as long-term stability, high efficiency, and strong brightness compared to conventional incandescent lamps and OLED. However, due to the brittle property of bulk inorganic semiconductor materials, III-V LED limits its applications in the field of high performance flexible electronics. This seminar introduces the first flexible and implantable GaN LED on plastic substrates that is transferred from bulk GaN on Si substrates. The superb properties of the flexible GaN thin film in terms of its wide band gap and high efficiency enable the dramatic extension of not only consumer electronic applications but also the biosensing scale. The flexible white LEDs are demonstrated for the feasibility of using a white light source for future flexible BLU devices. Finally a water-resist and a biocompatible PTFE-coated flexible LED biosensor can detect PSA at a detection limit of 1 ng/mL. These results show that the nitride-based flexible LED can be used as a type of implantable LED biosensor and as a therapy tool.

  • 바이오센서 산업은 국민 삶의 질을 향상시키고 미래 국가 경쟁력을 증진시킬 수 있다는 기대 속에 고부가가치 산업 육성을 제고할 수 있는 분야로 주목 받고 있는 가운데, 실시간 진단, 고감도화, 소형화를 위한 집적화, 저가격화, 다기능화 등에 대한 이슈가 지속적으로 제기되고 있다. 이러한 이슈들을 해결할 수 있는 대안 기술로써, 높은 Q 지수를 갖는 광 마이크로 공진기에 대한 관심이 높아지고 있으며, 최근의 성공적인 연구 결과들은 차세대 미세 바이오 센서 시스템 구현과 실용화의 가능성을 높여주고 있다. 특히, 단일 분자 및 나노 입자까지도 측정 가능한 고감도의 무표지 센서 기술은 특성은, 각종 감염성 질환 및 암표지자의 실시간 조기 진단과 개인별 맞춤 치료, 환경 유해물질 검출, 신약물질 발굴 등 새로운 시장창출에 대한 기대감을 증폭시키고 있다.

Single Nanoparticle Photoluminescence Studies of Visible Light-Sensitive TiO2 and ZnO Nanostructures

  • Visible light-sensitive $TiO_2$ and ZnO nanostructure materials have attracted great attention as the promising material for solar energy conversion systems such as photocatalysts for water splitting and environmental purification as well as nano-biosensors. Success of their applications relies on how to control their surface state behaviors related to the exciton dynamics and optoelectronic properties. In this paper, we briefly review some recent works on single nanoparticle photoluminescence (PL) technique and its application to observation of their surface state behaviors which are raveled by the conventional ensemble-averaged spectroscopic techniques. This review provides an opportunity to understand the temporal and spatial heterogeneities within an individual nanostructure, allowing for the potential use of single-nanoparticle approaches in studies of their photoenergy conversion and nano-scale optical biosensing.
