The objective of this study is to estimate the effect of the bed expansion and the characteristics of attached biomass in the start-up in the anaerobic fluidized bed reactor(AFBR). The fluidized bed reactor was operated with bacteria supported on the bed of granular activated carbon(GAC). The reactor was operated at 35$\circ$C, 5 kg $COD/m^3\cdot day$ at bed expansion varying from 0 to 100% with soluble glucose wastewater(5,000 mg/l). When the effluent reached a steady state at 100% of bed expansion, maximum COD removal efficiency of 87.3% and 0.031 $m^3CH_4/kg COD_{removed}$ were obtained. At higher bed expansion, COD removal efficiency, methane production rate and biogas production rate increased. Especially, at 50% of bed expansion, the efficiency of the treatment increasedg rapidly in the AFBR. The biomass colonized in the pits and crevices of the GAC particle and no complete biofilm was established in the bioreactor during the experiment.
Biofuel based on available biomass in rural area could contribute to increase the vitality and income of rural residents. This study estimates the potential production and environmental benefits of biofuel production from surplus rice, rice straw, cow manure, and food waste in South Korea. We could produce about 2.4 billion liters of ethanol using surplus rice and rice straw and 841 million $m^3$ of methane from cow manure and food waste. 1.7 billion liters of gasoline and 800 million liters of diesel could be replaced with these potential biofuel productions, which can annually reduce 4.9 million ton $CO_2$ eq. GHGs.
One osmotolerant strain from among 44 yeast isolates was selected based on its growth abilities in media containing high concentrations of sucrose. This selected strain, named SK-ENNY, was identified as Meyerozyma guilliermondii by sequencing the internal transcribed spacer regions and partial D1/D2 large-subunit domains of the 26S ribosomal RNA. SK-ENNY was utilized to produce high-fructose glucose syrup (HFGS) from sucrose-containing biomass. Conversion rates to HFGS from 310-610 g/l of pure sucrose and from 75-310 g/l of sugar beet molasses were 73.5-94.1% and 76.2-91.1%, respectively. In the syrups produced, fructose yields were 89.4-100% and 96.5-100% and glucose yields were 57.6-82.5% and 55.3-79.5% of the theoretical values for pure sucrose and molasses sugars, respectively. This is the first report of employing M. guilliermondii for production of HFGS from sucrose-containing biomass.
Callus and cell suspension cultures of Eurycoma longifolia Jack could be an alternative supply of 9-hydroxycanthin-6-one and 9-methoxycanthin-6-one. The callus tissues were initiated from leaves of different trees. The friable calli were used for the preparation of the cell suspension cultures of E. longifolia. The leaf explant of tree Eu-9 produced the most callus and also induced high cell biomass in the cell suspension culture, but it produced low quantity of 9-methoxycanthin- 6-one and 9-hydroxycanthin-6-one. The leaf explant from tree Eu-8 produced low quantity of callus and cell biomass, but produced the highest quantity of 9-methoxycanthin- 6-one and 9-hydroxycanthin-6-one. Optimum production of cell biomass was obtained on cell culture medium with pH 5.75 prior to autoclaving, but high alkaloids content could be induced in culture medium in acidic condition with pH 4.75 and 5.25 prior to autoclaving.
Production of yeast diet has been carried out in batch fermenters under optimum culture conditions. The fermentation of Candida utilis on a $2\%$ complex medium resulted in 1.22 g/L/h productivity and $65\times10^8$ viable cells/ml, and the addition of $15{\mu}M$ zinc to the medium increased both the productivity and the number of viable cells just a little more. In the case of the fermentation of Kluyveromyces fragilis, the highest value of the biomass productivity, 1.94 g/L/h, was obtained on a $2.5\%$ fructose medium with $70\times10^8$ viable cells/ml, and $1\%$ peptone was found to be a growth factor in this fermentation. When $3.5\%$ NaCl was added to the given medium, both the biomass productivity and the number of viable cells decreased significantly in each fermentation, but this may be considered to preserve yeast diet long without osmotic lysis.
Production of acetic acid from cellulosic biomass by Simultaneous Saccharification and Extractive Fermentation (SSEF) was investigated. The homoacetate organism used in this study was a strain of Clostridium thermoaceticum, ATCC # 49707. A batch operation of Simultaneous Saccharification and Fermentation(SSF) using ${\alpha}$-cellulose at pH 5.5 and 55$^{\circ}C$ yielded 40% conversion of cellulose to acetic acid, while a fed-batch SSF operation produced a maximum acetic acid concentration of 25 g/L, with 50% overall yield. In-situ extractive fermentation to reduce the end-product inhibition on both bacteria and enzyme was carried out. in a batch SSEF using 200 g/L IRA-400 resin, acetic acid concentration reached to 23.9 g/L and acetic acid yield and productivity were observed to be 48% and 0.20 g/L-hr, respectively.
Enzymatic hydrolysate from non pre-treated biomass of yellow poplar (Liriodendron tulipifera) was prepared and used as resource for bioethanol production. Fresh branch (1 year old) of yellow poplar biomass was found to be a good resource for achieving high saccharification yields and bioethanol production. Chemical composition of yellow poplar varied significantly depending upon age of tree. Cellulose content in fresh branch and log (12 years old) of yellow poplar was 44.7 and 46.7% respectively. Enzymatic hydrolysis of raw biomass was carried out with commercial enzymes. Fresh branch of yellow poplar hydrolyzed more easily than log of yellow poplar tree. After 72 h of enzyme treatment the glucose concentration from Fresh branch of yellow poplar was 1.46 g/L and for the same treatment period log of yellow poplar produced 1.23 g/L of glucose. Saccharomyces cerevisiae KCTC 7296 fermented the enzyme hydrolysate to ethanol, however ethanol production was similar (~1.4 g/L) from both fresh branch and log yellow poplar hydrolysates after 96 h.
원유 가격의 상승과 지구온난화로 인하여 재생 가능한 바이오매스를 이용하여 산업적으로 중요한 화합물을 생산하는 연구가 주목받고 있다. 특히, 3세대 바이오매스인 해조류는 비식량 자원, 높은 생산 수율, 온실가스 저감 등 장점을 가지고 있기에 연구 되어야 할 필요가 있다. 본 연구에서는 고체분석 방법을 이용하여 4종의 해조류 중에 미역이 가장 많은 당류를 함유하는 것을 확인하였다. 미역의 효과적인 전처리를 위해 반응표면분석법을 이용하였으며 이를 통해 고체의 부하 및 촉매의 농도 증가가 총 당의 추출률과 관계 있음을 확인하였다. 4종의 락토바실러스 균주에서 미역의 전처리물을 이용하여 젖산 생산 수행하였으며 L. alimentarius와 L. brevis가 해조류 이용한 젖산생산에 적합한 균주임을 확인하였다.
Cleaning up the landfill soil by phytoremediation in association with biomass production and utilization of biosolid as a soil amendment will be an attractive green technology. In order to examine this integrated green technology, in the current study of pot trial, heavy metal removal rate and biomass production were determined following cultivation of three different plant species in the landfill soil incorporated with biosolid at two different levels (25 ton $ha^{-1}$ and 50 ton $ha^{-1}$). Among the three plant species including Indian mustard (Brassica juncea), giant sunflower (Helianthus giganteus. L), and giant cane (Arundo donax. L), sunflower appeared to produce the largest biomass yield (19.2 ton $ha^{-1}$) and the produced amounts were magnificently increased with biosolid treatment compared to the control (no biosoild treatment). The increased production associated with biosolid treatment was common for other plant species and this was attributed to the biosolid originated nutrients as well as the improved soil physical properties due to the organic matter from biosolid. The elevated heavy metals in soil which was originated from the incorporated biosolid were Cu and Zn. Based on the phytoavailable amount of heavy metals from biosolid, the removed amount by plant shoots were 95% and 165% for Cu and Zn, respectively, when sunflower was grown. This indicated that mitigation of heavy metal accumulation in soils achieved by the removal of metal through sunflower cultivation enables the successive treatment of biosolid to soils. Moreover, sunflower showed heavy metal stabilization ability in the rhizosphere resulting in alleviation of metal release to ground water.

Itaconic acid (IA) is a dicarboxylic acid included in the US Department of Energy's (DOE) 2004 list of the most promising chemical platforms derived from sugars. IA is produced industrially using liquid-state fermentation (LSF) by Aspergillus terreus with glucose as the carbon source. To utilize IA production in renewable resource-based biorefinery, the present study investigated the use of lignocellulosic biomass as a carbon source for LSF. We also investigated the production of fumaric acid (FA), which is also on the DOE's list. FA is a primary metabolite, whereas IA is a secondary metabolite and requires the enzyme cis-aconitate decarboxylase for its production. Two lignocellulosic biomasses (wheat bran and corn cobs) were tested for fungal fermentation. Liquid hydrolysates obtained after acid or enzymatic treatment were used in LSF. We show that each treatment resulted in different concentrations of sugars, metals, or inhibitors. Furthermore, different acid yields (IA and FA) were obtained depending on which of the four Aspergillus strains tested were employed. The maximum FA yield was obtained when A. terreus was used for LSF of corn cob hydrolysate (1.9% total glucose); whereas an IA yield of 0.14% was obtained by LSF of corn cob hydrolysates by A. oryzae.
