Mesozooplankton Community Dynamics in Watan Stream, Yeonggwang, Korea (영광 와탄천의 중형동물플랑크톤 군집동태)

  • Mesozooplankton dynamics were studied in Watan stream which flows into the Yellow Sea in Yeonggwang, Korea. Samples were collected at eight stations bimonthly during April 2006~February 2007. Mesozooplankton community was consisted of 45 taxa (8 cladocerans and 29 cope-pods). The abundance varied between the highest one ($31{\sim}127,587\;indiv.m^{-3}$) in October, and the lowest ($12{\sim}233\;indiv.m^-3$) in December. Diversity index showed to be the highest value in July (0.671) and the lowest one in August (0.368). A euryhaline species, Acartia hongi was dominant in brackish water stations (the highest abundance at $13.4^{\circ}C$ and 14.1 psu). In freshwater stations of the upstream, two cladoceran species of Polyphemus pediculus and Moina weismani occurred in high abundance in August. Dominant taxa of copepod were clustered to two or three groups mainly due to the difference of salinity gradients. Spatial distribution of mesozooplankton revealed to be determined by salinity gradients which were affected by opening and closing of the artificial dam in Watan stream.

Intake, Digestibility, Nitrogen and Energy Utilization by Korean Native Goats Consuming Alfalfa and Birdsfoot Trefoil (Alfalfa와 Birdsfoot Trefoil의 급여가 산양의 섭취량, 소화율과 질소 및 에너지 이용성에 미치는 영향)

  • This study was conducted to compare the DM intake, the digestibility, and the utilization of nitrogen and energy of Korean native goats, when fed on orchardgrass supplemented with alfalfa or birdsfoot trefoil. Experimental diets included three treatments: orchardgrass(OG) 100%, orchardgrass 6%+alfalfa(AA) 40%(OG+AA), and orchardgrass 60%+birdsfoot trefoil(BT) 40%(OG+BT). The CP content was higher for OG+AA and OG+BT diets than for OG diets(P<0.05). The NDF content was high in OG diets(P<0.05) and the lignin content was high in OG+BT diets(P<0.05), but the ADF content did not differ among all diets. The DM intake and the digestibility of OM and cellular constituents were higher for OG+AA diets than for OG diets(P<0.05), but the NDF and ADF digestibility were not significant difference among all diets. The apparently digested N was higher for OG+AA diets than for other diets(P<0.05). However, the retained N % and biological value were higher for OG+BT diets than for OG diets(P<0.05), but those were not differ between OG+AA and OG+BT diets. On the other hand, the utilization of DE and ME did not show the difference among all diets. Based on the results, the utilization of nitrogen and energy was more improved for legume supplemented diets(AA, BT) than for OG diets(P<0.05). Furthermore, the utilization of nitrogen was slightly more improved for OG+BT diets than for OG+AA diets, but the significantly difference was not observed.

Comparison of Nerve Growth Factor Induction and Anti-aging Activity Using Dried Gastrodia and Fermented Gastrodia Extracts (건천마와 발효천마 추출물의 항노화 활성과 신경성장인자 유도 효과 비교)

  • This study was performed to examine the increasing biological activities of both nerve growth factor induction and anti-aging activity of gastrodia (Gastrodia elata) with fermentation process. Antioxidant activities, taken in the fermenting, were investigated to verify utility value of gastrodia for functional food and cosmetics. Fermented gastrodia extracts showed higher antioxidant activities than dried gastrodia extracts. During the routine check of all the practical use potential as functional food, inhibition effect of angiotensin converting enzyme (ACE) and xanthine oxidase (XOase) was tested among water extracted dried gastrodia (WEDG), water extracted fermented gastrodia (WEFG), 70% ethanol extracted dried gastrodia (EEDG) and 70% ethanol extracted fermented gastrodia (EEFG). DPPH was shown as WEDG = $64.14{\pm}0.89%$, WEFG = $66.21{\pm}1.03%$, EEDG = $82.25{\pm}0.52%$, and EEFG = $82.36{\pm}2.37%$. ABTS was shown as WEDG = $54.15{\pm}1.37%$, WEFG = $60.24{\pm}2.25%$, EEDG = $59.18{\pm}1.86%$, and EEFG = $77.17{\pm}4.23%$. Therefore, ACE activity was dramatically inhibited by EERG while there was no difference of XOase inhibition between EEFG and EERG. Nerve growth factor (NGF) activity was measured and indicated about 40% increased neurite growth effect. To conclude, biologically active compounds of gastrodia were increased by fermentation process. It seems to be that ferment gastrodia enhance the use ranges from functional food to fuctional cosmetics, and to all processing industry.

Effect of Dry Heating on the Pasting/retrogradation and Textural Properties of Starch-soy Protein Mixture (건조가열이 전분과 콩단백질 혼합물의 호화 및 노화특성과 조직감에 미치는 영향)

  • Effects of dry heating and pH on pasting, thermal, and textural properties of corn starch containing soy protein isolate (SPI) were investigated. Samples were prepared by drying corn starch dispersions containing 3% SPI at $40^{\circ}C$ (unheated sample) or additionally heating dried mixtures at $130^{\circ}C$ (heated sample). Pasting properties of starch samples and textural properties of starch gels were measured using Rapid Visco-Analyzer (RVA) and Texture Analyzer (TA), respectively. Effects of SPI addition and dry heating on retrogradation and microstructure of gel samples were investigated using DSC and SEM, respectively. Pasting viscosity of dry-heated samples was significantly higher than that of unheated samples, and at pH 8 compared to pH 6. Dry-heat treatment decreased fracture stress and strain of starch gels containing SPI. No significant effects of dry-heat treatment and pH were observed on retrogradation of starch gels. Addition of SPI retarded starch retrogradation and modified starch gel properties with increased nutritional value. Dry-heat treatment with SPI could be possible method for starch modification.

Mold Growth and Mycotoxin Contamination of Forages (조사료의 곰팡이 발생과 곰팡이독소 오염)

  • In order to ensure good animal health and performance, it is essential to produce forages with high feeding value and good hygienic quality. However, huge amounts of forages consumed by ruminants are contaminated with mold prior to harvest or during storage as hay, straw or silage. These mold can grow in forages only when nutrients are available, correct temperature exist, oxygen is present, and unbound water is available. Fungal 'species can be divided into two groups: field fungi and storage fungi. Field fungi invade the forages while the crop is still in the field, require high moisture conditions, and are such as species of Fusarium, Alternaria, Clodosporium, Diplodia, Gibberrella and Helminthosporium. Storage fungi invade forages during storage and need less moisture than field fungi. These such as species of Aspergillus and Penicillium usually do not occur any problem before harvest. Mold growth can spoil the nutritional aspects of the forages and also results in secondary metabolites that are highly toxic to animal, humans and plants. Moldy feeds are less palatable and may reduce dry matter intake. This, in turn, leads to a reduction of nutrition intake, reducing weight gains or milk production. Performance losses of 5 to 10 percent are typical with moldy feeds. Mycotoxins are toxic substances produced by fungi (molds) growing on crops in the field or storages. While greater than 400 mycotoxins have been chemically identified, the biological or veterinary medical impact of only several mycotoxins is known. Mycotoxins have attracted considerable attention as potential causes for poor performance and health disorders in domestic livestock. They can be carcinogenic, hepatotoxic, hematotoxic, immunosuppressive, estrogenic, or mutagenic. So, feeding moldy forages has adverse effects on animal health and milk consumers. Also, this author reported that rice straw hay was contaminated mycotoxigenic fungi such as Penicillium roqueforti and Fusarium culmorum in Korea. Therefore, it is an urgent need to develop an improved post harvest storage method to reduce nutrient loss and mycotoxin contamination of forages, which will have a positive impact on human health.

Chemical Composition and Biological Feed Value of Autoclaved Hatchery By-products for Poultry (가압열처리한 부화부산물의 화학적 조성과 닭에 대한 생물학적 사료가치)

  • This study was conducted to obtain the basic information for hatchery by-products as feed resources. Four kinds of by-products from a commercial hatchery, i. e., autoclaved day-old broiler or egg type chicks, dead embryos & infertile eggs of both chicks were used for chemical composition and bioavailability test. Chemical compositions were obtained by AOAC (1990) method, and amino acid and energy bioavailabilities were determined by the method of Sibbald (1976). The crude protein contents of day-old chicks (60.27∼62.59) were higher than those of dead embryos & infertile eggs (39.24∼40.09) , whereas the crude fat contents of all by-products were not different. The crude ash contents were higher in dead embryos & infertile eggs (29.18∼34.49%) than in day-old chicks (6.50∼6.71%). The Ca contents of dead embryos & infertile eggs (8.79∼10.82%) were higher than those of day-old chicks (1.29∼l.30%). The total amino acid contents, overall amino acid availabilities and ME contents were higher in day-old chicks than in dead embryos & infertile eggs. It seems that the day-old chicks of egg strain can be used as a high protein - high energy feed resources and the dead embryos & infertile eggs of broiler or egg type as a high protein-high calcium feed ingredients.

Chemical Composition and Biological Feed Value of Autoclaved Poultry By-products for Poultry (가압열처리한 도계부산물의 화학적 조성과 닭에 대한 생물학적 사료가치)

  • In order to obtain the basic information needed to utilize poultry by-products as feed resources, the yielding ratio, chemical composition and nutrient bioavailability of 5 offal components such as autoclaved head, feet, viscera, blood and feathers were investigated. Yielding ratios of head, feet, viscera, blood and feathers were 2.93% 4.78%, 10.98%, 3.91% and 4.83%, respectively. The crude protein contents of feathers (86.71%) and blood (82.99%) were higher than those of viscera (64.67%), feet (58.76%) and head (49.51%) , Inversely, the crude fat contents of blood (6.96%) and feathers (2.96%) were lower than those of head (26.19%), viscera (23.96%) and feet (13.73%). The crude ash contents of feet (21.69%) and head (20.38%) were higher than those of other by-products (0.96∼8.62%). The macro-mineral contents of head and feet were higher than those of other components, and the iron content of blood was higher than the other by-products. The total amino acid contents of poultry by-products showed the same trend as the crude protein contents. In addition, the Iysine content of feathers was very low compared to its high protein content. The essential amino acid contents of feathers were poorer than those of other offal components. Among the 5 offal components, the feathers showed the poorest amino acid availabilities. The ME contents were highest in viscera, and head, blood, feathers and feet. in decreasing order.

A New Histone Deacetylase Inhibitor, MHY4381, Induces Apoptosis via Generation of Reactive Oxygen Species in Human Prostate Cancer Cells

  • Histone deacetylase (HDAC) inhibitors represent a novel class of anticancer agents, which can be used to inhibit cell proliferation and induce apoptosis in several types of cancer cells. In this study, we investigated the anticancer activity of MHY4381, a newly synthesized HDAC inhibitor, against human prostate cancer cell lines and compared its efficacy with that of suberoylanilide hydroxamic acid (SAHA), a well-known HDAC inhibitor. We assessed cell viability, apoptosis, cell cycle regulation, and other biological effects in the prostate cancer cells. We also evaluated a possible mechanism of MHY4381 on the apoptotic cell death pathway. The IC50 value of MHY4381 was lower in DU145 cells (IC50=0.31 µM) than in LNCaP (IC50=0.85 µM) and PC-3 cells (IC50=5.23 µM). In addition, the IC50 values of MHY4381 measured in this assay were significantly lower than those of SAHA against prostate cancer cell lines. MHY4381 increased the levels of acetylated histones H3 and H4 and reduced the expression of HDAC proteins in the prostate cancer cell lines. MHY4381 increased G2/M phase arrest in DU145 cells, and G1 arrest in LNCaP cells. It also activated reactive oxygen species (ROS) generation, which induced apoptosis in the DU145 and LNCaP cells by increasing the ratio of Bax/Bcl-2 and releasing cytochrome c into the cytoplasm. Our results indicated that MHY4381 preferentially results in antitumor effects in DU145 and LNCaP cells via mitochondria-mediated apoptosis and ROS-facilitated cell death pathway, and therefore can be used as a promising prostate cancer therapeutic.

Evaluating Quality of Fertilizer Manufactured (livestock manure compost) with Different Sources in Korea (우리나라 퇴비의 축종별 성분함량 실태 평가)

  • The main objective of this research was to evaluate physicochemical properties of commercially available fertilizer manufactured with different sources (swine, cattle, poultry, and mixed manure) in Korea. Results of this research showed that average content of organics was ranged 39.11-40.78% depending on different sources with the highest value in swine manure. Average water contents in fertilizer was ordered swine > cattle > mixed > poultry, and ranged 37.00-42.16%. Total of 8 heavy metals (As, Cd, Hg, Pb, Cr, Cu, Ni, and Zn) were investigated and no significant difference was observed for average concentration of heavy metals depending on different sources except Zn and Cu. For both heavy metals, fertilizer manufactured with swine manure showed higher concentration (346.34 mg/kg for Zn and 117.00 mg $kg^{-1}$ for Cu) than other fertilizers mainly due to feed contents. More long term monitoring would be necessary to make best management practice for commercially available fertilizer in Korea.

A Study on Dissolve and Remove Analysis of Organic Chemicals Using a Mixed Method of Advanced Oxidation and Micro Filtering - Water Drinking Point - (고도산화와 정밀여과의 혼성공법을 이용한 유기화학물질의 분해 및 제거분석에 관한 연구 - 먹는 물을 중심으로 -)

  • It is impossible to remove toxic organic substances that are recognized as a cancer caused suspicious element in drinking water using the conventional water purification method. This study introduces groundwater into a reaction chamber as an effective amount of water to process this water using a mixed method of AOP oxidation and M/F membrane and purifies it as a desirable level by artificially mixing such toxic substances in order to effectively process the water. Based on this fact, this study configures an optimal operation condition. The VOCs existed in toxic substances was investigated as a term of phenol and toluene, and agricultural chemicals were also investigated as a term of parathion, diazinon and carbaryl. The experiment applied in this study was performed using a single and composite soolution. In the operation condition applied to fully dissolve and remove such substances, the amount of $H_2O_2$ injected in the process was 150 mL of a fixed quantity, the value of pH was configured as $5.5{\sim}6.0$, the temperature was controlled as a range of $12{\sim}16^{\circ}C$, the dissolved amount of ozone was applied more than 5.0 mg/L, the reaction time was determined as an optimal condition, such as $30{\sim}40$ minutes, and the segregation membrane in the same reactor was determined for acquire water drinking of large quantity using a pore size of $0.45{\mu}m$ M/F membrane.
