Effect of Glutaraldehyde Treatment on Stability of Permeabilized Ochrobactrum anthropi SY509 in Nitrate Removal

  • For practical application, the stability of permeabilized Ochrobactrum anthropi SY509 needs to be increased, as its half-life of enzymatic denitrification is only 90 days. As the cells become viable after permeabilization treatment, this can cause decreased activity in a long-term operation and induce breakage of the immobilization matrix. However, the organic solvent concentration causing zero cell viability was 50%, which is too high for industrial application. Thus, whole-cell immobilization using glutaraldehyde was performed, and 0.1% (v/v) glutaraldehyde was determined as the optimum concentration to maintain activity and increase the half-life. It was also found that 0.1% (v/v) glutaraldehyde reacted with 41.9% of the total amine residues on the surface of the cells during the treatment. As a result, the half-life of the permeabilized cells was increased from 90 to 210 days by glutaraldehyde treatment after permeabilization, and no cell viability was detected.

  • Herein, we have fabricated an ultrathin aluminum oxide ($Al_2O_3$) coated PP separator by using a RF sputter deposition process. Approximately 20 nm thickness coating layer on the bare PP separator was formed at the power of 55 W for 2 minutes without thermal damage. Whereas only permeability of the coated separator was degraded slightly, other properties such as thermal stability, uptake amount of liquid electrolyte, and ionic conductivity were improved comparing to the bare PP separator. As a result, an only 20-nm-thick $Al_2O_3$ coating layer could improve the rate capability compared with a bare PP separator under a high current density.

The Corrosion Study of Al Current Collector in Phosphonium Ionic Liquid as Solvent for Lithium Ion Battery

  • A room temperature ionic liquid (RTIL) based on trihexyl (tetradecyl)phosphonium bis(trifluoromethanesulfonyl) imide ([$(C_6H_{13})_3P(C_{14}H_{29)}$] [TFSI];P66614TFSI) was synthesized and analyzed to determine their characteristics and properties. The bis(trifluoromethanesulfonyl)imide (TFSI) anion is widely studied as an ionic liquid (IL) forming anion which imparts many useful properties, notably electrochemical stability. Especially its electrochemical and physical characteristics for solvent of lithium ion battery were investigated in detail. $P_{66614}$ TFSI exhibits fairly low conductivity (0.89 mS $cm^{-1}$) and higher viscosity (298 K: 277 cP; 343 K: 39 cP) than other ionic liquids, but it exhibits a high thermal stability (over $400^{\circ}C$). Especially corrosion behavior on Al current collector was tested at room temperature and further it was confirmed that thermal resistivity for Al corrosion was highly increased in 1.0M LiTFSI/$P_{66614}$-TFSI electrolyte comparing with other RTILs by linear sweep thermometry.

  • A study was condoled to investigate the effects of phosphate salts $(Na_2HPO_4\;and\;K_2HPO_4)$ on the emulsion stability of soy protein isolate (SPI) in terms of the salts concentration and addition order. When phosphates were added before emulsification, emulsion stability (ES) of SPI was improved at the concentration of 10 mM, while ES was decreased by addition of phosphates after emulsification. At high phosphate concentrations, ES of SPI was decreased by the addition of phosphates, regardless of the addition order. ES of SPI at the isoelectric point (pH 4.5) or in the presence of $CaCl_2$ was greatly enhanced by the phosphates. In both cases, the overall ES profile was found to be nearly similar to the solubility profile of SPI, indicating the positive relationship between solutibility and emulsion stability of SPI.

  • Cholesterol plays an important role in various physiological responses and membrane stability. To investigate the effect of cholesterol in liposome on the stability of encapsulated ascorbic acid, the physico-chemical experiments using various amounts of cholesterol-containing liposomes were performed. The encapsulation efficiency of ascorbic acid was decreased with increasing cholesterol content in liposome, whereas size of liposome was increased. Furthermore, the stability of encapsulated ascorbic acid was increased with increasing the content of cholesterol. The stability was not affected by pH. Encapsulated ascorbic acid in liposome stored at $37^{\circ}C$ was rapidly oxidized compared to those stored at $4^{\circ}C$. These results suggest that cholesterol in liposome affects largely to the stability of encapsulated ascorbic acid.

  • The reduction behavior of silver ions to silver nanoparticles is an important topic in polymer/silver salt complex membranes to facilitate olefin transport, as this has a significant effect on the long-term performance stability of the membrane. In this study, the effects ofthe solvent type on the formation of silver nanoparticles, as well as the long-term membrane performance of a solid polymer/silver salt complex membrane were investigated. These effects were assessed for solid complexes of poly(N-vinyl pyrrolidone) $(PVP)/AgBF_4$, using either an ionic liquid (IL), acetonitrile (ACN) or water as the solvent for the membrane preparation. The membrane performance test showed that long-term stability was strongly dependent on the solvent type, which increased in the following order: IL > ACN >> water. The formation of silver nanoparticles was more favorable with the solvent type in the reverse order, as supported by UV-visible spectroscopy. The poor stability of the $(PVP)/AgBF_4$ membrane when water was used as the solvent might have been due to the small amount of water present in the silver-polymer complex membranes actively participating in the reduction reaction of the silver ions into silver nanoparticles. Conversely, the higher stability of the $(PVP)/AgBF_4$, membrane when an IL was used as the solvent was attributable to the cooperative coordination of silver ions with the IL, as well as with the polymer matrix, as confirmed by FTIR spectroscopy.

  • This was investigated on the major issues and research trends regarding the heat-treatment of woods through literature reviews. The principal heat-treatment technologies utilized for industrial purposes include the Plato-process (Netherlands), the Retification process (France), the OHT-process (Germany), and the Thermowood Process (Finland). Factors that mainly influence the heat-treatment process are the wood species, process temperature, processing time, and the heating medium (air, steam, vacuum, N2, oil, etc.). Researches on investigating the optimal conditions with these process conditions being the variables stand as the mainstream. Heat-treated woods present dimensional stability improvement, but mass loss and strength reduction, a wide variations for decaying inhibition, and insufficient resistance against mold, wood borer, and termites. For further improvement in respects of durability or resistance to biological degradation, necessity to search for more suitable heat treatment process and processing conditions fit for each wood species has been suggested. Exploiting new ways to utilize heat-treated wood and extending its range of use have been considered to be important matters that need more effort put into for the sustainable and sound environment as well as saving the wood resources.

  • In this study, as a tool of evaluating biological stability of landfilled waste, influencing factors of $AT_4$ method was studied for standardizing the method. As influencing factors, initial lag time, exchanging period of $CO_2$ absorbing agent, and interval of pressure measurement were discussed, and also the relationship between content of dried food waste and $AT_4$ value were compared. Considering heterogeneity of landfilled waste and statistical error range of measurement, authors suggest that the criteria of stabilized landfill waste is $10mg\;O_2/g\;DM$ by $AT_4$ method.

  • A mature mutant ribose binding protein (RBP) of Escherichia coli was obtained by site-directed mutagenesis, replacing Thr-3 in the N-domain of wild-type mature RBP (WT -mRBP) with a Trp residue (N- Trp-mRBP). The equilibrium unfolding properties and the refolding kinetics of this protein were monitored by fluorescence and circular dichroism (CD). (omitted)

  • Posturography stands or quantitative assessment of body postural stability analysis. The present study developed a balance plate system to monitor patient's center of pressure (COP) movement and to analyze its stability. An equilateral triangular shaped plate was made of duralumin and forces were measured on the three vertices of the plate using industrial load cells. Specially designed electronic circuit picked up force signals ed into data acquisition system to calculate the cartesian coordinates of COP. COP calculation error was less than 2%. The force signals enabled to compute stability measures, which consisted of a variety of clinical parameters related to postural stability. Clinical experiments were carefully designed and performed on 40 normal subjects. The results were that 1) postural stability decreased with age and 2) the best parameters were those of posture deviation measures. A customized PC-based software package was developed to apply the present technique with a great convenience to monitoring and analyzing postural stability in an accurate and quantitative way.
