Practical Synthesis of $\alpha$-Galactosyl Ceramide, KRN 7000.

  • Galactosyl ceramides paly important roles in biological system as immunomodulator and essential constituents of membranes and cell walls. An efficient synthesis of $\alpha$-galactosyl ceramide, KRN 7000, derived from marine sponge Agelas mauritianus as accomplished via a short reaction involving the coupling ceramide moiety and trichloroacetimidate as glycosylation donor. We could synthesize $\alpha$-galactosyl ceramide stereoselectivity without $\beta$-anomer formation on a multigram scale.

Identification of the active components inhibiting the expression of matrix metallopeptidase-9 by TNFα in ethyl acetate extract of Euphorbia humifusa Willd

  • Euphorbia humifusa Willd (EuH), called Ttang-Bin-Dae in Korea, is a traditional medicinal plant widely used for its anti-inflammatory and antiviral activity. Ethyl acetate (EA) extracts of EuH (EA/EuH) inhibit invasion and metastasis by inhibiting tumor necrosis factor TNFá-induced matrix metalloproteinases (MMP)-9 expression in human breast cancer cells. However, the bioactive components of EA/EuH mediating the inhibition of MMP-9 expression have not been identified. In the present study, three bioactive constituents of EA/EuH were isolated using high-performance liquid chromatography. Nuclear magnetic resonance spectroscopy revealed isoquercetin, avicularin, and astragalin as the bioactive compounds responsible for preventing TNFα-induced MMP-9 mRNA expression in breast cancer cells. These findings suggest that isoquercetin, avicularin, and astragalin could be used as valuable anti-metastatic agents against metastatic cancers.

  • Mushrooms are ubiquitous in nature and have high nutritional attributes. They have demonstrated diverse biological effects and therefore have been used in treatments of various diseases, including cancer, diabetes, bacterial and viral infections, and ulcer. In particular, polysaccharides, including ${\beta}$-glucan, are considered as the major constituents responsible for the biological activity of mushrooms. Although an overwhelming number of reports have been published on the importance of polysaccharides as immunomodulating agents, not all of the healing properties found in these mushrooms could be fully accounted for. Recently, many research groups have begun investigations on biologically active small-molecular weight compounds in wild mushrooms. In this mini-review, both structural diversity and biological activities of novel bioactive substances from Korean native mushrooms are described.

The Cytotoxic Constituents of Betula platyphylla and their Effects on Human Lung A549 Cancer Cells

  • During the screening for cytotoxic compounds from plants grown in Korea, Betula platyphylla (BP) showed potent activity against the adenocarcinomic human alveolar basal epithelial A549 cell line. To identify the cytotoxic components from BP, the $CH_2Cl_2$ fraction with the most significant cytotoxic effect was applied to the column chromatographies. Seven compounds were isolated: lupeol (1), betulinic acid (2), (-)-rhododendrol (3), platyphyllenone (4), platyphyllone (5), (-)-centrolobol (6), and oleanolic acid (7). Among them, three diarylheptanoids (4 - 6) exhibited cytotoxicity toward A549 cells. Especially, $50{\mu}M$ of 4 reduced A549 cell viability to $18.93{\pm}0.82%$ compared to control ($100.00{\pm}21.48%$). Lactate dehydrogenase (LDH) leakage and intracellular reactive oxygen species (ROS) production were also induced by $50{\mu}M$ 4. This is the first report on the cytotoxic effect of BP-derived diarylheptanoids 4-6 against A549 cells. The compound 4 may be useful for the development of early hit compounds for non-small cell lung carcinoma, but the consideration about selectivity of 4 is required since 4 also showed the cytotoxicity in the human normal lung epithelial BEAS-2B cell line.

Isolation and Determination of Phenolic Compounds in Peony (Paeonia lactiflora Pall.)Root

  • The five phenolic compounds of peony root were isolated by Sephadex LH-20 column chromatography. Their chemical structures were identified by spectroscopic methods (UV, FT-IR, FAB-MS and $^1H$ .&^13C$-NMR). The complete structures of these compounds were elucidated to be (+)-taxifolin-3-0-$\beta$-D-glucopyranoside, benzoic acid, gallic acid, (-)-epicatechin and (+)-catechin. The concentrations of five phenolic compounds in the peony root of three Korean cultivars (Youngchonjakyak, Euisungjakyak and Jomjakyak) were determined by reverse-phase HPLC. The constituents concentration in Youngchonjakyak were generally higher than in Euisungjakyak and Jomjakyak. The concentrations of (+)-taxifolin-3-O-$\beta$-D-glucopyranoside, benzoic acid, gallic acid, (-)-epicatechin and (+)-catechin in three different cultivars were ranged 0.23-0.52%, 0.20-0.30%, 0.26-0.28%, 0.09-0.12% and 0.34-0.63%, respectively.

Chemical Constituents from the Aerial Parts of Bupleurum falcatum L. and Biological Evidences

  • In this study, phytochemical investigation on the aerial parts of Bupleurum falcatum resulted in the isolation of fourteen compounds including three quinic acid derivatives (1 - 3), five flavonoids (4 - 8), three monoterpene glycosides (9 - 11), and three saikosaponins (12 - 14). Compound 1 was first isolated from nature and unambiguously determined to be 3-O-feruloyl 5-O-caffeoylquinic acid on the basis of the extensive spectroscopic evidence. Biological testing revealed that saikosaponin A (12) and saikosaponin D (13) showed moderate antiproliferative effects on HL-60 and HepG2 cancer cell lines.

Inhibitory effect of Plant Essential Oils on Malassezia pachydermatis

  • Effect of the plant essential oils on the growth of Malassezia pachydermatis was evaluated and the essential oils of Ocimum basilicum L., Melaleuca alternifolia (Maid. & Bet.) Cheel, and Rosa damascene Mill. were the most active against M. pachydermatis and their activity were high than that of itraconazole at 2 mg/mL. The major constituents of the three oils by GC-MS analysis were linalool (21.83%) and estragole (74.29%) for O. basilicum, a-terpinolene (17.96%) and terpinen-4-01 (45.54%) for M. alternifolia, and a-citronellol (59.98%) and geraniol (27.58%) for R. damascene. Results showed that these selected three oils could be effective toward controlling M. pachydermatis opportunistic infections.

Three Phenolic Glycosides from Gastrodia elata

  • Three phenolic glycosides, gastrodin (1), parishin (2), and parishin B (3) were isolated from the n-BuOH fraction of Gastrodia elata (Orchidaseae) by medium pressure liquid chromatography. Their chemical structures were identified by the interpretation of their spectral data and direct comparison with literature values. Gastrodin and parishin were isolated as the major constituents in G. elata. Gastrodin may be a suitable index component for quality control of G. elata.

  • Three limonoids and two phenolics were isolated from stem bark of Pellodendron amurense Rupr. (Rutaceae) and were defined clearly as calodendrolide (1), obacunone (2), limonin (3), methyl 4-hydroxybenzoate (4), and syringin (5) based on NMR spectroscopy results obtained with the aid of X-ray crystallographic analysis. This is the first report on the isolation of calodendrolide (1) from this plant.

  • Metal thin films, which are indispensable constituents of ULSI (Ultra Large Scale Integration) circuits, have been fabricated by physical or chemical methods. However, these methods have a drawback of using expensive high vacuum instruments. In this work, the fabrication of tungsten metal film by spin coating was investigated. First of all, inorganic peroxopolytungstic acid (W-IPA) powder, which is soluble in water, was prepared by dissolving metal tungsten in hydrogen peroxide and by evaporating residual solvent. Then, the solution of W-IPA was mixed with organic solvent, which was spin-coated on wafers. And then, tungsten metal films, were obtained after reduction procedure. By selecting an appropriate organic solvent and irradiating UV, the sheet resistance of the tungsten metal film could be remarkably reduced.
