Role of tea catechins in prevention of aging and age-related disorders

  • Tea polyphenols especially catechins have long been studied for their antioxidant and radical scavenging properties. Scientists throughout the world have investigated the usefulness of the regular green tea consumption in several disease conditions. In-vitro and in-vivo experiments on catechins especially epigallocatechingallate have revealed a significant role in many ways. Reactive oxygen species have been increasingly implicated in the pathogenesis of many diseases and important biological processes. Toxic effects of these oxidants, commonly referred to as oxidative stress, can cause cellular damage by oxidizing nucleic acids, proteins, and membrane lipids. Oxidative stress has been related to aging and age related disorders. It is found that in a wide variety of pathological processes, including cancer, atherosclerosis, neurological degeneration, Alzheimer's disease, ageing and autoimmune disorders, oxidative stress has its implications. Catechins have been reported to be useful in combating aging and age related disorders like cancer, cardiovascular disorders and neurodegenerative diseases. In this mini review we will discuss such studies done across the globe.

Chestnut Extracts as new Anti-aging agent

  • Inner nutshell of Castanea crenata has been used as an anti-aging and anti-wrinkle agent from the ancient time in east Asia. In order to develop new anti-aging and anti-wrinkle, ethanolic extract of inner nutshell of Castanea crenata was prepared and various biological activities were evaluated. Cor-285 possessed potent tree radical scavenging activity in vitro compared to gallic acid. Cor-285 showed the preventive effect against UV-induced cytotoxicity of fibroblast at concentration of 25-250mg/ml. When Cor-285 was evaluated for its anti-allergic activity, it effectively inhibited histamine release from mast cells induced by compound 48/80. The inhibitory activity was stronger than that of glycyrrhizinate. Cor-285 also showed on vivo inhibition against delayed hypersensitivity as well as croton-oil induced ear edema in mice when topically applied. These results strongly suggest that Cor-285 may reduce immunoregulatory/inflammatory skin trouble. From the attempts to isolate the scavenger, were isolated. In a clinical trial of twenty healthy volunteers with aged skin, 6 weeks application of Cor-285 decreased wrinkle about 26% and increased moisturizing 20% on the skin. All of these results indicate that Cor-285 may be an effective anti-aging and anti-wrinkle agent.

A concise review of human brain methylome during aging and neurodegenerative diseases

  • DNA methylation at CpG sites is an essential epigenetic mark that regulates gene expression during mammalian development and diseases. Methylome refers to the entire set of methylation modifications present in the whole genome. Over the last several years, an increasing number of reports on brain DNA methylome reported the association between aberrant methylation and the abnormalities in the expression of critical genes known to have critical roles during aging and neurodegenerative diseases. Consequently, the role of methylation in understanding neurodegenerative diseases has been under focus. This review outlines the current knowledge of the human brain DNA methylomes during aging and neurodegenerative diseases. We describe the differentially methylated genes from fetal stage to old age and their biological functions. Additionally, we summarize the key aspects and methylated genes identified from brain methylome studies on neurodegenerative diseases. The brain methylome studies could provide a basis for studying the functional aspects of neurodegenerative diseases.

갈랑가 뿌리 추출물의 항산화, 항노화 효과 및 W/O형 에멀젼에서 항산화 효과의 보존성 (Antioxidant and anti-aging effects of Alpinia galanga L. rhizome extracts and preservation of antioxidant effects in W/O type emulsion)

  • 본 연구 결과 DPPH 라디칼 소거 활성은 갈랑가 뿌리 70% 에탄올 추출물(AG.E) 100 ㎍/mL 농도에서 81.8%이었으며 ABTS+ 라디칼 소거 활성은 AG.E 50 ㎍/mL의 낮은 농도에서 L-Ascorbic acid (AA)와 유사한 99.8%로 확인되었다. 항노화 활성을 측정하기 위하여 collagenase와 elastase 저해 활성을 측정한 결과 둘 모두에서 AG.E는 50 ㎍/mL의 낮은 농도부터 epigallocatechin (EGCG)보다 더 높은 저해 효과를 나타내었다. 특히 500 ㎍/mL 농도에서 EGCG 대비 3배 이상의 저해 효과를 보였다. CCD-986sk 세포내에서 AG.E의 항노화 효과를 검증하기 위해 UVB로 자극한 다양한 실험에서도 우수한 항노화 효과가 얻어졌다. RT-PCR을 통한 유전자 발현 분석 실험에서 COL1A mRNA 발현량은 AG.E 20 ㎍/mL 저농도에서 무첨가 대비 2.90배 증가시키는 결과가 얻어져 항노화관련 우수한 기능성 소재로 개발 가능성이 확인되었다. 제형에 소재의 적용 시 생리활성의 경시적 보전성에 대한 기초 연구로서 AG.E 및 AA등을 안정한 W/O type emulsion에 첨가하여 25 ℃ 항온조에 보관하면서 1일차, 30일차, 60일차에 DPPH와 ABTS+ 라디칼소거 활성을 측정한 결과 모두 경시적으로 항산화 효과가 높은수준 유지됨을 확인하였다.

IoT 디바이스 기반 노화진단을 위한 개념적 프레임워크 (A Conceptual Framework for Aging Diagnosis Using IoT Devices)

  • 사물인터넷 컴퓨팅의 등장으로 다양한 사물인터넷 디바이스를 통해 사용자에 대한 건강 컨텍스트의 수집과 수집된 건강 컨텍스트를 분석하여 노화진단이 가능해졌다. 하지만, 기존의 노화진단 기법들은 서로 다른 고정된 노화진단요소들을 사용하여 사용자에 따라 획득 가능한 건강 컨텍스트의 가변성을 고려하지 않아서 새로운 노화진단요소의 추가 및 삭제에 대해 동적 대응이 힘들다. 본 논문에서는 다양한 사물인터넷 디바이스를 기반으로 노화진단에 필요한 다양한 노화진단 요소를 수집하고, 사용자마다 가변적인 노화진단 요소의 구성에 따라 동적으로 적응 가능한 노화진단 프레임워크의 기법 및 설계를 제안한다. 제안된 노화진단 프레임워크를 이용하면 획득 가능한 건강 컨텍스트의 가변성과 관계없이 노화진단기법의 적용이 가능하며, 노화진단 요소의 동적 추가 및 삭제가 가능하다.

신품종 그린볼 사과의 광노화인자 조절효과 (Photo-aging regulation effects of newly bred Green ball apple)

  • 본 연구에서는 한국에서 육종한 신육성품종 그린볼 사과 껍질 추출물(GBE)이 피부 광노화 인자조절에 대한 억제효과를 확인하였다. 피부에서 광노화 인자 조절에 대한 억제 효과를 확인하기 위해 CCD986sk fibrobalst cell에 UVB로 광노화를 유도시킨 후 세포에 GBE를 처리하였다. 광노화 인자 조절 효과를 측정한 결과 GBE가 COL1A2, MMP-1, MMP-9, TIMP-1 단백질 발현량에서 UVB에 의해 자극된 MMP-1, MMP-9 단백질 합성을 억제하였으며, COL1A2, TIMP-1 단백질의 경우 발현량이 유의적으로 증가하는 것을 확인하였다. Photoaging-related factors인 COL1A2, MMP-1, MMP-9, HAS2, TGF-β, TIMP-1의 mRNA 발현량을 측정한 결과 GBE에 의해 MMP-1, MMP-9 단백질 발현을 효과적으로 억제하였으며, MMPs와 type procollagen 발현에 관여하는 조절인자인 TIMP-1과 TGF-β의 발현량이 증가함에 따라 COL1A2의 생성량이 증가하는 것을 확인할 수 있었다. 또한 피부를 구성하는 구조 단백질 중 하나인 hyaluronic acid 생성에 관여하는 HAS2의 발현량 증가도 확인하였다. 따라서 GBE는 광노화의 인자 억제 조절에 대한 우수한 효능을 가졌으며, 피부 노화를 예방하는 기능성 소재로서 활용이 가능할 것으로 판단되었다.

Estimation of Initial Concentrations of Phenanthrene and Atrazine from Soil Properties and Bioavailability During Aging

  • Contaminated sites are often abandoned for an extended period of time, thus organopollutants becoming sequestered. The information on the initial concentrations of the contaminants would be helpful for the economic bioremediation of the chemicals. The present study estimated the initial concentrations of atrazine and phenanthrene through multiple regression analyses using soil properties and the amount of chemicals available in situ. Percentage mineralized or extracted was best correlated with organic C or logarithm of organic C, and the $R^2$ values were 0.548 and 0.894 for atrazine and phenanthrene, respectively. Estimation of the initial concentration of the chemicals was then calculated from both the percentage mineralized or extracted and the amount of chemicals extracted or mineralized. Results showed that the estimation of the initial concentration of the chemical at the time of contamination is feasible.

MicroRNA controls of cellular senescence

  • Cellular senescence is a state of permanent cell-cycle arrest triggered by different internal and external stimuli. This phenomenon is considered to be both beneficial and detrimental depending on the cell types and biological contexts. During normal embryonic development and after tissue injury, cellular senescence is critical for tissue remodeling. In addition, this process is useful for arresting growth of tumor cells, particularly during early onset of tumorigenesis. However, accumulation of senescent cells decreases tissue regenerative capabilities and induces inflammation, which is responsible for cancer and organismal aging. Therefore cellular senescence has to be tightly regulated, and dysregulation might lead to the aging and human diseases. Among many regulators of cellular senescence, in this review, I will focus on microRNAs, small non-coding RNAs playing critical roles in diverse biological events including cellular senescence.

  • Defects in mitochondrial DNA (mtDNA) cause many human diseases and are critical factors that contribute to aging. The mechanisms of maternally-inherited mtDNA mutations are well studied. However, the role of acquired mutations during the aging process is still poorly understood. The most plausible mechanism is that increased reactive oxygen species (ROS) may affect the opening of mitochondrial voltage dependent anion channel (VDAC) and thus results in damage to mtDNA. This review focuses on recent trends in mtDNA research and the mutations that appear to be associated with increased ROS.

Anti-Aging Effect of Nigella Sativa Fixed Oil on D-Galactose-Induced Aging in Mice

  • Objectives: Aging is an unconscious and gradual process that can lead to changes in biological systems. Induction of oxidative stress and apoptosis, hepatotoxicity and neurotoxicity are involved in the aging process. Regarding the antioxidant property of black seed oil, the aim of this study was to evaluate the anti-aging effect of Nigella sativa (N. sativa) oil on d-galactose-induced aging in mice. Methods: For induction of aging, D-galactose (500 mg/kg, subcoutaneously SC) was administrated to male mice for 42 days. Animals were treated with D-galactose alone or with b lack seed oil (0.1, 0.2, 0.5 mL/kg, intraperitoneally (ip)). Additionally, vitamin E (200 mg/kg) was used as a positive control. At the end of treatment, the malondialdehyde (MDA) and the glutathione (GSH) contents in brain and liver tissues were measured. Also, enzymes in serum, including aspartate aminotransferase (AST) and alanine amino transferase (ALT), were determined. The levels of the proteins Bax, Bcl2, caspase-3 (pro and cleaved) in brain and liver tissues were evaluated. Results: Administration of D-galactose (500 mg/kg, SC) for 42 days increased serum levels of ALT and AST, as well as the MDA content, in brain and liver tissues, but decreased the GSH content. Additionally, the levels of apoptotic proteins, including Bax, procaspase-3 and caspase-3 cleaved, were markedly increased. N. sativa oil (0.1 and 0.2 mL/kg) diminished the levels of the biochemical markers ALT and AST. Administration of black seed oil (0.1, 0.2 and 0.5 mL/kg) reduced lipid peroxidation and at doses 0.1 and 0.2 mL/kg significantly recovered the GSH content. The oil decreased Bax/Bcl2 levels and at 0.1 mL/kg down-regulated the expressions of caspase-3 (pro and cleaved) proteins in brain and liver tissues. Conclusion: Through its antioxidant and anti-apoptosis properties, black seed oil exhibited an anti-aging effect in a model of aging induced with D-galactose.
