Quercetin, A Bioflavonoid, Protects Against Oxidative Stress-related Gastric Mucosal Damage in Rats

  • Quercetin and its sugar conjugates are the most abundantly distributed bioflavonoids and represent the largest proportion of flavonols in the plant kingdom. The present study was undertaken to demonstrate the effect of quercetin on the role of reactive oxygen species (ROS) in the development of gastric ulcers in rats. Administration of quercetin in doses of 50, 100 and $200\;mg\;kg^{-1}$ twice daily for 5 days, showed dose dependent significant protection against ethanol (EtOH), aspirin (ASP), cold-restraint stress (CRS) and pylorus ligation (PL) -induced gastric ulcer models and the results were comparable with those elicited by sucralfate. The thiobarbituric acid reactive substances in the stomach mucosa, an index of lipid peroxidation and regulation of plasma corticosterone were significantly increased in CRS-induced gastric ulceration. The queroetin $(100\;mg\;kg^{-1})$ and reduced glutathione effectively inhibited gastric lesions induced by CRS with a significant decrease in the lipid peroxidation and plasma corticosterone. These results indicate that quercetin a bioflavonoid exerts its antiulcer effect in light of free radical scavenging and plasma corticosterone in cold restraint stress ulcers.

Extraction of Citrus Bioflavonoid with Vinegars and Effect on Blood Pressure (식초에 의한 감귤과피 Bioflavonoids의 추출과 혈압에 미치는 영향)

  • Effect of temperature(room temperature and boiling) and content of citrus peel(0.1∼6.0%) on the extractable hesperidin and naringin in the vinegars(apple, brown rice and persimmons) and also, dietary effect in hyperpiesia rats on blood pressure, biochemical changes were investigated. Extractable amounts of hesperidin and naringin in persimmons and brown rice vinegar were higher than those of apple vinegar in the both extraction of room temperature and boiling. Hesperidin and naringin contents showed highest in the one day extraction at room temperature when citrus peel was below 1% but, 3∼5 days extraction was optimum when the peel was above 2%. Hesperidin and naringin contents showed maximum at 2 hours extraction in case of boiling. Neutral lipids, total cholesterol and LDL cholesterol in blood, and blood pressure(reduction of 10.7∼33.2 mmHg) of the hyperpiesia rats took in 0.5% apple vinegar with 0.2, 0.4 and 0.6% citrus peel powder for 4 weeks were lower or reduced than those of control groups. But HDL cholesterol was higher than that of control groups.

  • Flavonoids are phenolic compounds widely distributed in wide variety of edible plants including leafy vegetables, fruits, beverages. Quercetin is one of bioflavonoid compounds and has anti-tumor effect by suppressing tumor growth in vitro and in vivo, including multiple biological effects by antioxidant and effective anti-inflammatory agent. The present study investigated whether quercetin can enhance antioxidant enzyme activities (glutathione peroxidase: GPX, superoxide dismutase : SOD, catalase: CAT) and intracellular reactive oxygen intermediate (ROI) levels in the presence of taurine on B16F10 murine melanoma cells. From this result, the antioxidant enzyme activities of quercetin in the presence of taurine was enhanced. In addition, the same treatments decreased intracellular reactive oxygen intermediate levels on B16F10 murine melanoma cells. Taken together, these results demonstrate that the antioxidant effect of quercetin can enhance in the presence of taurine and it might play an important role in anti-tumor effect.

Hesperidin Improves the IL-6-Mediated Hepatic Insulin Resistance in Hepa-1c1c7 Cells

  • Hesperidin (HES) is a bioflavonoid with antioxidant, anti-inflammatory and anti-diabetic properties. IL-6 is well known as a primary proinflammatory cytokine that contributes to impaired insulin signaling in liver. This study was to investigate whether HES improves IL-6-mediated impairment of insulin sensitivity in liver. Hepa-1c1c7 cells were pre-treated with 50 and $100{\mu}M$ HES in complete media for 1 h and then cultured in the presence or absence of IL-6 (20 ng/ml). These results demonstrated that HES restored IL-6-suppressed expression of IRS-1 protein, downregulated IL-6-increased expression of CRP and SOCS-3 mRNA, and inhibited LPS-induced production of IL-6 in Hepa-1c1c7 cells. These findings indicate that HES may ameliorate hepatic insulin resistance via improvement of IL-6-mediated impaired insulin signaling in hepatocytes.

  • Luteolin, 3', 4', 5,7-tetrahydroxyflavone, belongs to a group of naturally occurring compounds called flavonoids that are found widely in the plant kingdom. It possesses many beneficial properties including antioxidant, anti-inflammatory, anti-bacterial, anti-diabetic and anti-proliferative actions. Colorectal cancer (CRC) is a leading cause of cancer related deaths worldwide. Many signaling pathways are deregulated during the progression of colon cancer. In this review we aimed to analyze the protection offered by luteolin on colon cancer. During colon cancer genesis, luteolin known to reduce oxidative stress thereby protects the cell to undergo damage in vivo. Wnt/${\beta}$-catenin signaling, deregulated during neoplastic development, is modified by luteolin. Hence, luteolin can be considered as a potential drug to treat CRC.

Identfication of Phase I and Phase II Metabolites of Hesperetin in Rat Liver Microsomes by Liquid Chromatography-Electrospray Ionization-Tandem Mass Spectrometry

  • The purpose of this study is to investigate the in vitro metabolism of hesperetin, a bioflavonoid. Hesperetin was incubated with rat liver microsomes in the presence of NADPH and UDP-glucuronic acid for 30 min. The reaction mixture was analyzed by liquid chromatography-ion trap mass spectrometer and the chemical structures of hesperetin metabolites were characterzed based on their MS/MS spectra. As a result, a total of five metabolites were detected in rat liver microsomes. The metabolites were identified as a de-methylated metabolite (eriodictyol), two hesperetin glucuronides, and two eriodictyol glucuronides.

감귤과피 유색미의 취반 특성

  • 우리나라에서 생산되는 감귤은 지리적 기후 조건으로 내한성이 강한 만다린계 온주밀감이 주종을 이루나 최근 과잉생산으로 인한 가격하락과 수입자유화에 따른 대처의 일환으로 경쟁력이 높은 품종을 장려 보급하고 있다. 감귤의 국내 연간 생산량은 56만 톤으로 과일 전체 생산량의 약 30%를 차지하며 과실의 약 20%가 과피로서 그 일부가 한약재로 쓰이나 대부분이 버려지고 있다. 감귤의 과피에는 carotenoids, bioflavonoids, pectin 및 terpenes가 풍부하게 함유되어 있으며, 천연에서 발견되고 있는 약 300여종의 carotenoids계 색소 중 115종이 감귤에 존재한다. 감귤 과피의 주요 carotenoids로는 비타민 A의 역할을 하는 $\beta$-carotene과 cryptoxanthin을 비롯한 $\beta$-citraurin이며 천연 착색제로 활용되고 있다. 또, 주요 bioflavonoids로는 모세혈관의 수축을 촉진시켜 고혈압 예방과 이로 인한 각종 질환을 방지하는 작용을 가진 hesperidin과 혈액내 LDL 콜레스테롤의 양을 줄이는 작용이 알려진 naringin이 있다. 그 외의 감귤 flavonoids도 항산화작용, antimutagen 활성, 항암, 항알레르기 및 항바이러스 효과가 알려져 있다. 밀감 과피의 bioflavonoids는 약 60여종이 분리되어 그 구조가 밝혀져 있으나 90% 이상이 hesperidin이다. 또 과피유에는 $\delta$-limonene을 주성분으로 하는 휘발성 향미성분이 함유되어 있어 향미 개선제로서의 활용이 기대되고 있다. 본 연구는 최근 식생활의 서구화로 쌀 위주의 식생활에 많은 변화를 가져와 쌀 소비량은 해마다 줄어들고 있는 반면 기능성을 가미한 쌀의 소비가 늘고 있음을 감안하여, 우리나라 제주도에서 생산되고 있는 감귤 과피의 물 균질액을 쌀에 코팅하여 아름다운 색상과 기능성을 지닌 유색미를 제조함과 동시에 그 취반 특성을 조사하였다. 그 결과 유색미의 색상은 진노랑(L*; 63.6, a*; -7.87, b*; 46.35)으로 취반 후에도 유사한 색상을 나타내었다. Total carotenoids는 감귤과피에서는 10.74mg%, 과피의 물 균질액에는 0.46mg%이었으며 유색미에는 0.l2mg%, 유색미 취반에는 0.05mg%를 나타내었다. Hesperidin은 과피, 물 균질액, 유색미 및 유색미 취반에서 각각 2173.l2mg%, 108.65mg%, 21.73mg% 및 8.67mg%이었으며, naringin은 각각 1468.40mg%, 73.38mg%, 14.62mg% 및 5.87mg%를 나타내었다. 감귤과피의 유리아미노산은 asparagine이 94.22mg%로 가장 높았으며, methionine이 24.88mg%, alanine이 19.64mg%, ${\gamma}$ -aminoisobutylic acid가 15.37mg%로 이들 4종의 아미노산이 전체 유리아미노산함량의 70%를 나타내었다. 유색미 취반에는 백미 취반에 비하여 총 유리아미노산의 함량이 15%정도 증가되었으며, 그 중에서 glutamic acid는 2.5 배가 증가되었다. 감귤과피의 주요 무기질은 K(652.60mg%)와 Ca(222.50mg%)로 전체 무기질 함량의 86%를 차지하였으며 유색미 취반은 백미 취반에 비하여 K는 2.3배, Mn는 76%, Ca, P, Mg은 16~26%, Fe는 13%가 각각 증가되었다. 취반의 경도, 점착성 및 깨짐성은 유색미 취반과 백미 취반간에 차이가 없으나 응집성과 탄력성은 유색미 취반에서 높았다. 색상에 대한 기호도, 구수한 맛 및 종합적인 기호도는 유색미 취반에서 높았으며 단맛, 쓴맛은 백미 취반과 차이를 보이지 않았다.

Quantitative analysis of rutin content using silkworm genetic resources

  • Rutin is an important bioflavonoid that is consumed in the daily diet. This study compared the functional components of rutin from various silkworm species using a gene database with those of rutin produced by silkworms selectively bred through cross-combinations. We made comparisons between the geographical origin and species of silkworm using a gene database and discovered that rutin activity was ranked in the following order by species, Chinese (C5)> miscellaneous varieties (Jamsaeng 1 Ho) >Japanese (Jam 115) > European (E58) >Korean (Sun 3 ho). However, rutin levels with respect to various genetic traits (blood color, silk color, and egg color) were consistent. In order to study rutin changes that occurred during the cross breeding of the silkworm gene, we bred cross-combinations utilizing Jam 115 and the 4051 silkworms. In conclusion, in order to provide information about the constituents of functional materials contained in silkworm powder, it is imperative that silkworm cross breeding occurs so that the database of functional materials extracted from silkworms will expand.

Effects of Hesperidin and Naringin on Antioxidative Capacity in the Rat (Hesperidin 과 Naringin 이 흰쥐의 항산화능에 미치는 영향)

  • This study was performed to investigate effects of hesperidin and naringin on linpid peroxide formation and antioxidative enzyme activities in rats. Thiobarbituric acid reactive substance (TBARS) concentrations were measured in plasma and liver. Catalase, superoxide dismutase, and glutathione peroxidase activities were measured in erythrocyte and liver. Forty-nine male Sprague-Dauley rats weighing 275.3$\pm$3.3g were blocked into seven groups according to body weight and were raised fro four weeks on diets containing 0.25, 0.50 or 1.00%(w/w) hesperidin or naringin . Food intake, weight gain , food efficiency ratio, and weights of liver, kidney, spleen ,and epididymal fat pad were not significantly different among groups. In 0.50 and 1.00% naringin groups , plasma TBARS concentrations were significantly decreased with a dose response patter. In 0.25, 0.50 and 1.00% hesperidin groups, liver TBARS concentrations were significantly decreased without a dose dependent patter. Antiosidative enzyme activities in erythrocyte and liver were not significantly affected by type and amountof dietary bioflavonoid, but in the 1.00% hesperidin group, catalase, superoxide dismutase, and glutahione perosidase activities in linver showed a tendency to increase. In conclusion, naringin inhibited lipid peroxide formation with a dose response pattern in plasma without changing the activities of antioxidative enzymes. Hesperidin adminstration, regardless of the level in the diet, inhibited lipid peroxide formation in liver.
