The scaffold serves as 3D substrate for the cells adhesion and mechanical support for the newly grown tissue by maintaining the 3D structure for the regeneration of tissue and organ. In this paper, we proposed integrated scaffold fabrication system using multi-axis rapid prototyping (RP) technology. It can fabricate various types of scaffolds: arbitrary sculptured shape, primitive shape, and tube shape scaffolds by layered dispensing biocompatible/ biodegradable polymer strands in designated patterns. In order to fabricate the 3D scaffold, we need to generate the plotting path way for the scaffold fabrication system. We design a data processing program - scaffold plotting software, which can convert the 3D STL file, primitive and tube model images into the NC code for the system. Finally, we fabricated the customized 3D scaffolds with high accuracy using the plotting software and the fabrication system.
A novel approach to the manufacture of biocompatible ceramic scaffold for tissue engineering using micro-stereolithography system is introduced. Micro-stereolithography is a newly proposed technology that enables to make a 3D micro structure. The 3D micro structures made by this technology can have accurate and complex shape within a few micron error. Therefore, the application based on this technology can vary greatly in nano-bio fields. Recently, tissue-engineering techniques have been regarded as alternative candidate to treat patients with serious bone defects. So many techniques to design and fabricate 3D scaffolds have been developed. But the imperfection of scaffold such as random pore size and porosity causes a limitation in developing optimum scaffold. So scaffold development with controllable pore size and fully interconnected shape have been needed for a more progress in tissue engineering. In this paper, bone scaffold was developed by applying the micro-stereolithography to the mold technology. The scaffold material used was HA(Hydroxyapatite) nano powder. HA is a type of calcium phosphate ceramic with similar characteristic to human inorganic bone component. The bone scaffold made by HA is expected, in the near future, to be an efficient therapy for bone defect.

Silk protein and agarose are widely known as biocompatible materials in the human body. A three-dimensional (3D) scaffold composed of agarose and low-molecular- weight silk fibroin (LSF) was fabricated and examined in terms of structural characteristics and cellular responses in bone tissue engineering. This study showed that mouse pluripotent precursor cells attached to and proliferated uniformly on and within the LSF-containing 3D scaffold. Interestingly, cell proliferation and attachment was shown to be higher in a 3D scaffold containing 0.02% LSF, as compared to other LSF concentrations. The results of this study suggest that agarose-LSF scaffolds may be useful materials for tissue engineering.
This paper presents a novel structural composition for artificial bone scaffolds with an appropriate biocompatibility and biodegradability capability. To achieve this aim, carbon nanotubes, due to their prominent mechanical properties, high biocompatibility with the body and its structural similarities with the natural bone structure are selected in component of the artificial bone structure. Also, according to the piezoelectric properties of natural bone tissue, the barium titanate, which is one of the biocompatible material with body and has piezoelectric property, is used to create self-healing ability. Furthermore, due to the fact that, most of the bone tissue is consists of hydroxyapatite, this material is also added to the artificial bone structure. Finally, polycaprolactone is used in synthetic bone composition as a proper substrate for bone growth and repair. To demonstrate, performance of the presented composition, the mechanical behaviour of the bone scaffold is simulated using ANSYS Workbench software and three dimensional finite element modelling. The obtained results are compared with mechanical behaviour of the natural bone and the previous bone scaffold compositions. The results indicated that, the modulus of elasticity, strength and toughness of the proposed composition of bone scaffold is very close to the natural bone behaviour with respect to the previous bone scaffold compositions and this composition can be employed as an appropriate replacement for bone implants.
조직공학에서의 인공지지체는 세포의 부착과 증식 및 분화가 잘 되어야 하고, 우수한 생체친화성 및 생분해성을 지녀야 한다. 다양한 인공지지체 제작 방법이 시도되어지고 있으며, 최근들어 3D 프린팅 기술을 이용한 방식이 활발하게 연구되어지고 있다. 폴리카프로락톤(polycaprolactone, PCL)은 낮은 녹는점을 가지고 있어 3D 프린팅하기에 우수한 생체적합 고분자 합성재료이다. 본 연구에서는 3D 프린팅 기술을 이용하여 3차원 PCL 인공지지체를 제작하였고, 지지체의 표면개질을 위해 수산화나트륨(NaOH)을 이용하였다. 표면개질된 인공지지체의 표면특성을 SEM으로 확인한 결과, 수산화나트륨을 처리한 PCL 인공지지체가 처리하지 않은 PCL 인공지지체에 비해 거칠기가 증가함을 보였으며, 접촉각 측정을 통해 친수성이 증가함을 확인하였다. In vitro 실험결과, 수산화나트륨을 처리한 PCL 인공지지체가 처리하지 않은 PCL 인공지지체에 비해 세포의 증식과 분화가 증가함을 보였고, 세포의 부착 모습은 균일하고 밀집된 형태로 부착됨을 확인하였다. 따라서 조형가공기술을 이용하여 수산화나트륨을 처리한 표면개질된 PCL 인공지지체를 제작하고 분석함으로써, 세포적합성을 통해 체내 인공지지체 개발 적용 가능성을 제시하였다.
Microstereolithography technology has potential capability for fabrication of 3D microstructures. It evolved from conventional SLA which is one of the RP processes. In a microstereolithography process, 3D microstructures can be easily fabricated by continuously stacking 2D layer which is photopolymerized using a liquid prepolymer. Combination between biocompatible/biodegradable photocurable prepolymer and 3D complex fabrication in microstereolithography makes broad application areas such as medical, pharmaceutic, and bio devices. In particular, a 3D microneedle for transdermal drug delivery and a scaffold for tissue engineering are fabricated using this technology. In this paper, the authors address development of microstereolithography system adapted to large surface and fabrication of various microstructures. In addition, to apply human body we suggest a biodegradable 3D microneedle and a scaffold using biodegradable photocurable prepolymer.
3D 프린터를 이한 인공지지체 제작 기술은 손상된 골 조직 재생을 위해 개발되고 있다. 골 조직 재생에 적하기 위해 인공지지체는 생체적합성, 생분해성 그리고 적절한 기계적 특성을 지녀야 하며, 분한 양의 공극과 내부 연결성을 지닌 구조로 제작되어야 한다. 본 연구에서는 3 차원 이중 공극 인공지지체를 제작하기 위해서 열 해 적층법(fused deposition modeling, FDM) 기반의 폴리머 적층 시스템을 이하였다. 사된 재료는 폴리카프로락톤(polycaprolactone, PCL)과 알긴산 나트륨(sodium alginate, SA)이다. 제작된 3 차원 형상의 인공지지체에 이중 공극을 갖기 위해 염 침출법을 이하였다. 완성된 인공지지체는 주사 전자 현미경과 X 선 검출 분광기(scanning electron microscope-energy dispersive spectroscopy, SEM-EDS)를 통해 관찰하였으며, MG-63 세포를 이하여 in-vitro 평가를 하였다.
Conventional methods for fabricating three-dimensional (3-D) scaffolds have substantial limitations. In this paper, we present 3-D scaffolds that can be made repeatedly with the same dimensions using a microstereolithography system. This system allows the fabrication of a pre-designed internal structure, such as pore size and porosity, by stacking photopolymerized materials. The scaffolds must be manufactured in a material that is biocompatible and biodegradable. In this regard, we synthesized liquid photocurable biodegradable TMC/TMP, followed by acrylation at terminal ends. And also, solidification properties of TMC/TMP polymer are to be obtained through experiments. Cell adhesion to scaffolds significantly affects tissue regeneration. As a typical example, we seeded chondrocytes on two types of 3-D scaffold and compared the adhesion results. Based on these results, the scaffold geometry is one of the most important factors in chondrocyte adhesion. These 3-D scaffolds could be key factors for studying cell behavior in complex environments and eventually lead to the optimum design of scaffolds for the regeneration of various tissues, such as cartilage and bone.
생체 친화적이며 생분해성 고분자 소재인 poly($\varepsilon$-caprolactone)(PCL)을 rapid prototyping(RP) 공정인 바이오플로팅 시스템을 통해 세포 재생용 지지체(scaffold)를 제작하였다. 제작된 PCL 지지체는 DMA(dynamic mechanical analyzer)를 통해 동일한 재료로 제작된 기존 염침출법(salt-leaching)에 의한 지지체보다 월등히 향상된 기계적 강도를 갖고 있음을 확인하였고, 이는 기존 전통적인 세포지지체 제작에서 문제점중의 하나인 기계적인 강도적인 측면을 보완하여, 뼈조직 재생에 유용하게 활용될 수 있을 것으로 예상된다. 지지체 내부의 구조는 세포의 증식과 이동 및 영양분의 공급이 지속될 수 있도록 전체적으로 연결된 통로로 구성되어 있고, 다양한 세포의 증식이 가능하도록 지지체의 공극 크기와 strand의 굵기 등을 조절할 수 있으며, 이를 이용하여 대체하고자 하는 생체조직의 특성에 맞도록 기계적 강도를 조정할 수 있음을 확인하였다. 제조된 PCL지지체는 연골세포를 통하여 셀 컬쳐링 되었고, 3차원 세포 지지체로서의 충분한 가능성을 보여주었다.
쾌속 조형(Rapid Prototyping; RP) 기술은 다양한 분야에서 활용되고 있다. 본 연구에서는 RP 기술을 이용한 나노복합재 적층장치(Nano Composite Deposition System, NCDS)를 사용하여 이식 가능한 약물전달시스템을 제작하였다. 약물전달시스템 복합재는 약물 입자로 5-fluorouracil (5-FU)를 사용하였으며, 생분해 고분자 매트릭스로 PLGA85/15를 사용하였다. 제작된 약물전달시스템은 넓은 표면적을 가질 수 있도록 지지체(scaffold) 형상으로 제작되었으며, in vitro 환경에서의 약물방출실험이 수행되었다. 약물방출제어를 위하여 생체적합재료인 수산화아파타이트(Hydroxyapatite, HA)를 약물-고분자 복합재에 첨가하였다. 약 50일간의 방출실험을 통하여 약물방출의 가능성을 보임을 확인하였다.
