Clinical Applicability of Multi-Tumor Marker Protein Chips for Diagnosing Ovarian Cancer

  • Purpose: To assess the value of multi-tumor marker protein chips in the diagnosis and treatment of ovarian cancer. Materials and Methods: Twelve tumor markers (CA19-9, NSE, CEA, CA242, CK19, ${\beta}$-HCG, AFP, SCC, c-PSA, CA125, CA724 and CA15-3) were detected by protein biochip in 220 patients with ovarian carcinomas, 205 with benign ovarian tumors and 200 healthy subjects. Results: The positivity rate was obviously higher in ovarian cancer (77.7%), than that in the benign cases (26.3%, p<0.01) and healthy subjects (4.5%, p<0.01). Serum levels of tumor markers were furthermore significantly higher in cases with lymph node metastasis (86.8%) than those without metastasis (44.7%), p<0.01. Conclusions: Multi-tumor marker protein chips provide important assistance in the diagnosis and treatment evaluation in ovarian cancers.

MEMS Technology for Biophotonic Applications (바이오포토닉스응용을 위한 MEMS 미세광학소자의 개발)

  • Biophotonics is an emerging area in a fusion of biology and photonics, especially in advanced bioimaging, optical biosensors, photomodulation, and biochip optical read-out, and optical manipulation. This emerging area also creates many opportunities for interdisciplinary study of biology and photonics. Micro-Electro-Mechanical-System(MEMS) is an attractive technology in miniaturizing sensors and actuactors. For last decade, it has contributed to the development for active and passive small and integrated optical components in optical communication. Recently, this technology is also merging into biology for high sensitive biosensing and high resolution and fast bioimaging in small form factor. In this talk, some key advantages of small optical components and recent biophotonic MEMS achievement will be discussed for miniaturized advanced biophotonic systems.

Electrochemical Immunosensing of GOx-labeled CRP Antigen on Capture Antibody Monolayer Immobilized on Calixcrown-5 SAMs

  • Insulating effects on Au electrode according to the thickness and density of coated materials are well-known. To do electrochemical immunoassay reproducibly the glod electrode would be coated with self-assembled monolayers and antobodies. To get reproducibility, the antobody monolayer should be packed at highest density so that the amount of immobilized antibody at defined area should be the same. The calix[4]crown-5 SAMs could provide the basis for the antibodies to be immobilized reproducibly and at highest density. But the insulating effect would be highest too. We proved that the compactly packed protein monolayers on SAMs inhibited the electron transfer by block the free shuttling of redox molecules. The inhibition was minimized by inserting redox molecules in between the proteins during immobilization process. In this paper, we demonstrated that the calix[4]crown-5 SAMs would provide the protein monolayers with highest density and new method to minimize the insulating effect by inserted redox molecules in between the compactly packed protein monolayers.

Development of 3D Micro-Nano Hybrid Patterns Using Anodized Aluminum and Micro-Indentation (양극산화된 알루미늄과 마이크로 인덴데이션을 이용한 3차원 마이크로-나노 하이브리드 패턴 제작)

  • A simple method for the fabrication of 3D micro-nano hybrid patterns was presented. In conventional fabrication methods of the micro-nano hybrid patterns, micro-patterns were firstly fabricated and then nano-patterns were formatted on the micro-patterns. Moreover, these micro-nano hybrid patterns could be fabricated on the flat substrate. In this paper, we suggested the fabrication method of 3D micro-nano hybrid patterns using micro-indentation on the anodized aluminum substrate. Since diameter of the hemispherical nano-pattern can be controlled by electrolyte and applied voltage in the anodizing process, we can easily fabricated nano-patterns of diameter of loom to 300nm. Nano-patterns were firstly formatted on the aluminum substrate, and then micro-patterns were fabricated by deforming the nano-patterned aluminum substrate. Hemispherical nano-patterns of diameter of 150nm were fabricated by anodizing process, and then micro-pyramid patterns of the side-length of $50{\mu}m$ were formatted on the nano-patterns using micro-indentation. Finally we successfully replicated 3D micro-nano hybrid patterns by hot-embossing process. 3D micro-nano hybrid patterns can be applied to nano-photonic device and nano-biochip application.

Facile Cell Patterning Based on Selectively Patterned Polydimethylsiloxane (PDMS) and Polyelectrolyte Surface (PDMS와 고분자 전해질 표면을 이용한 간편한 세포 패터닝 방법)

  • This study presented facile method of cell patterning using fabricated PDMS patterns on polyelectrolyte coated surface. This basic principle is the fabrication of functional surface presenting two orthogonal surfaces such as cell adhesive and repellent properties. Cell adhesive surface was firstly fabricated with simple coating of polyelectrolyte multilayer. And then, the desired patterns of PDMS for the prevention of nonspecific binding of cells were transferred onto the previously formed thin film of polyelectrolyte multilayer. Thus, we could prepare novel functional surface simultaneously containing PDMS and polyelectrolyte region. As expected, the PDMS regions showed effective prevention of nonspecific binding of cell and the other region, exposed polyelectrolyte area, provided cell adhesive environment. The height of formed PDMS structure was about 100 nm. Based on this method, cell patterning can be successfully obtained with various pattern shapes and sizes. Therefore, we expect that this simple method will be useful platform technology for the development of cell chip, cell based assay system, and biochip.

  • 많은 압전 후막은 여러 감지소자, 통신 및 사무자동화 기기, 전기 및 전자부품, 의료장비 및 국방산업에 까지 널리 응용되어 왔다. 그 중에서도 압전특성이 뛰어난 PZT 후막은 마이크로 펌프, 밸브, 헤드, 모터, 트랜스듀서 뿐 아니라 최근 바이오칩용 센서와 액추에이터로서 널리 연구되고 있다. 또한 마이크로 센서와 액추에이터 의 제작 및 구동을 위한 MEMS 기술의 도입으로 실리콘 베이스의 소자 개발이 집중되고 있다. 스크린 프린팅 방법은수 마이크론에서 수십 마이크론 후막의 실현이 용이하고 비교적 경제적이며 소자신뢰도가 높고 대량생산에 유리하여 활발한 연구가 진행 중이다. 그러나 후막은 벌크에 비해 기공률이 높고, 또 소자응용에 있어서 고온소결 시 MEMS공정을 위한 실리콘 베이스 기판과의 확산 및 반응에 의 한 계면 및 활물질 성능의 저하가 문제가 되고 있다. 따라서 본 연구에서는 스크린 프린팅과 더불어 졸 코팅 방법의 도입으로 후막의 성형 및 소결 밀도를 높임과 동시에 여러 확산 방지 막의 증착으로 capacitor 형 PZT 후막의 물성 및 전기 적 특성을 향상시키고자 하였다.

Microfilter Chip Fabrication for Bead-Based Immunoassay (비드를 이용한 면역분석용 마이크로필터 칩의 제작)

  • Immunoassay is one of the important analytical methods for clinical diagnoses and biochemical studies, but needs a long time, troublesome procedures and expensive reagents. In this study, therefore, we propose the micro filter chip with microbeads for immunoassay, which has pillar structures. The advantage of the proposed micro filter chip is to use simple fabrication process and cheap materials. The mold was made by the photolithography technique with Si wafer and negative photoresist SU-8. The replica was made of PDMS, bonded on the pyrex glass. The micro filter chip consists of inlet channel, filter chamber and outlet channel. HBV (Hepatitius B virus) monoclonal antibody (Ag1) labeled with biotin were immobilized onto streptavidin coated beads of 30∼50 $\mu$m size. Fluorescein isothiocyanate (FITC)-labeled HBV monoclonal antibody (Ag8) was used to detect HBsAg (Hebatitis B virus surface Antigen), and fluorescence intensity was monitored by epi-fluorescence microscope. In this study, the immune response of less than 30 min was obtained with with the use of 100 $m\ell$ of sample.

A Study About PDMS-Glass Based Thermopneumatic Micropump Integrated with Check Valve (체크밸브가 달린 열공압 방식의 PDMS-유리마이크로 펌프에 관한 연구)

  • Microfluidic single chip integrating thermopneumatic micropump and micro check valve are developed. The micropump and micorvalve are made of biocompatible materials, glass and PDMS, so as to be applicable to the biochip. By using the passive-type check valve, backward flow and fluid leakage are blocked and flow control is stable and precise. The chip is composed of three PDMS layers and a glass substrate. In the chip, flow channel and pump chamber were made on the PDMS layers by the replica molding technique and pump heater was made on the glass substrate by Cr/Au deposition. Diameter of the pump chamber is 7 mm and the width and depth of the channel are 200 and $180{\mu}m$, respectively. The PDMS layers chip and the heater deposited glass chip are combined by a jig and a clamp for pumping operation, and they are separable so that PDMS chip is used as a disposable but the heater chip is able to be used repeatedly. Pumping performance was simulated by CFD software and investigated experimentally. The performance was the best when the duty ratio of the applied voltage to the heater was 33%.

The protective effect of anti-oxidant in cadmium-induced hair cell death

  • Cadmium ($Cd^{2+}$) is a heavy metal and a potent carcinogen implicated in tumor development through occupational and environmental exposure. However, the auditory effect of $Cd^{2+}$ is not poorly understood. The purpose of the present study was to investigate whether prevent the ototoxic effects of $Cd^{2+}$ by antioxidnat in auditory cell line, HEI-OC1. Rosmarinic acid is a naturally occurring phenolic compound and also known to possess marked antioxidant properties. We showed that antioxidnat inhibited the the cytochrome c release induced by $Cd^{2+}$. In addition, we showed that the regulatory effect of antioxidnat on apoptosis is through the caspase-9 activation, and extracellular signal-regulated kinase activation in auditory cells. These results suggested antioxidnat its therapeutic usefulness, against $Cd^{2+}$ induced activation of caspase-9 and ERK.

Characterization of Microfluidic Channels using DVD Pick-up Fluorescent Scanner (광 픽업 방식 형광스캐너를 이용한 미소유체 특성 분석)

  • Microfluidics deals with the behavior, precise control and manipulation of fluids at a micro scale. It has become increasingly prevalent in various applications such as biomedical applications (diagnostics, therapeutics, and cell/tissue engineering), inkjet head, and fuel cells etc. The issue of inspection and characterization of microfluidics has emerged as a major consideration in design, fabrication, and detection of microfluidic devices. In this paper, we characterize a diffusion based mixing in Y-microchannel using a fluorescent optical scanner based on a DVD pick-up module, which is widely used in optical storages. Using fluorescent dye, we measure the fluorescent intensity that represents the mixing patterns in Y-microchannel. We also compare these experimental results with computational fluid dynamics (CFD) simulation ones. It is shown that the proposed optical scanner can be used as an alternative measurement system with high performance and cost-effectiveness, compared to conventional optical tools such as epifluorescent microscopes using high resolution CCD camera and confocal microscopes with photomultiplier (PMT) detectors.
