Red pepper, one of the major economic crops in Korea, is being affected by anthracnose disease caused by Colletotrichum acutatum. To control this disease, an antagonistic bacterial strain, Bacillus subtilis YGB36 identified by 16S rDNA sequencing, physiological and biochemical analyses is used as a biological control agent. In vitro screening revealed that the strain YGB36 possess strong antifungal activity against the pathogen Cylindrocarpon destructans. The strain exhibited cellulase, protease, amylase, siderophore production and phosphate solubility. In vitro conidial germination of C. acutatum was most drastically inhibited by YGB36 cell suspensions (106 cfu/ml) or culture filtrate. Development of anthracnose symptoms was reduced on detached immature green pepper fruits by treatment with cell suspensions, and its control value was recorded as 65.7%. The YGB36 bacterial suspension treatment enhanced the germination rate of red pepper seeds and promoted root development and growth under greenhouse conditions. The in vitro screening of fungicide and insecticide sensitivity test against YGB36 revealed that the bacterial growth was not affected by any of the insecticides, and 11 fungicides out of 21 used. Collectively, our results clearly suggest that the strain YGB36 is considered as one of the potential biocontrol agents against anthracnose disease in red pepper.

This study aimed to examine the harmful effects of Mn and Fe, which may be generated as dust or fume in the industrial sites, on the body and genital organs by their inhalation. It is intended to find the characteristics and differences of the hazardousness by inhaling a single and the mixed materials of Mn and Fe. Male F344 rats were divided into the control group and 3 exposed groups on the basis of the test material compound (Mn $1.5mg/m^3$, Mn 1.5 and Fe $3.0mg/m^3$, Fe $3.0mg/m^3$). The 4 groups were divided into 4 subgroups again on the basis of the exposure period (4 and 13 weeks) and the recovery period (4 and 13 weeks). The exposure condition was 6 hours a day, 5 days a week for the whole body. Clinical tests including changes in weight and feed rate, blood biochemical test, motility change, changes in the number and the amount of spermatozoon (sperm count), daily sperm production (DSP), deformity test of spermatozoon and changes in the accumulation of Mn and Fe in blood and internal organs were performed. Motility was reduced by Mn exposure. Especially, the effect of Mn was exposure period responsible. By mixing with Fe, no significant change in motility Mn and Fe accumulation in organs was observed. Sperm count and daily sperm production (DSP) were decreased by Mn. Additional effect like the reduction of sperm count and DSP, and delayed restoration of sperm count and DSP during the recovery period were observed in the mixed exposure group. These results indicate that Mn and Fe may affect the motility reduced and has male reproductive toxicity. Mixed exposure of Mn and Fe lead to synergic effects on the male reproductive toxicity.

This study was conducted to investigate the physiological responses of sevenband grouper, Epinephelus septemfasciatus at low temperature condition in winter season. Water temperature was dropped from $12^{\circ}C$ to $5^{\circ}C$ by two different interval (interval I: $-1^{\circ}C/24h$ and interval II: $-1^{\circ}C/8h$). The experimental fishes were exposed for 9 days at each water temperature condition, and were observed survival rate. The blood sample was collected from fishes for analysis of cortisol, glucose, total protein (TP), and electrolytes ($Na^+$, $Cl^-$, $K^+$). Mortality of fish was not observed until $5^{\circ}C$. The mortality (12 individuals) began after 170 h and all fish were death after 174 h in interval I case, whereas the mortality (9 individuals) began after 72 h and all fish were death after 76 h in interval II case. Little different was observed in the physiological responses, but trend was similar between two intervals. The physiological metabolism of plasma cortisol and glucose were increased rapidly, and swimming ability and respiration response was faster at $7^{\circ}C$. This result means that the critical temperature of sevenband grouper was $7^{\circ}C$ at low water temperature condition.
The nutritive value of lentil straw (Lens culinaris) relative to 4% urea treated wheat straw (w/w) was assessed in adult bucks and milking buffaloes on-station and on-farm, respectively. A digestion-cum-nitrogen balance experiment was conducted with three bucks (24.35 kg) fed ad libitum lentil straw (LS), urea treated wheat straw (UTS) and a 1:1 (w/w DM basis) mixture of LS and UTS (LS:UTS) in a 3$\times$3 Latin square design. Though daily dry matter intake (g kg $W^{0.75}$ or % LW) of bucks fed LS and UTS did not differ significantly, that of LS:UTS was significantly (p<0.05) low. Similarly, digestibility of nutrients (DM, OM and CP) and nitrogen balance (g/day) did not differ significantly for bucks given LS or UTS although a significant positive improvement was evident in these parameters for LS:UTS. The digestibility of fibre fractions (NDF and ADF) was significantly (p<0.05) lower in LS as compared to UTS or LS:UTS. Except for the significantly (p<0.05) lower intake of DCP (g kg$W^{0.75}$), the bucks fed LS had statistically similar nutrient intake as compared to UTS. Feeding of LS:UTS resulted in significantly higher nutrient (DCP, TDN) intake by bucks. The important blood-biochemical parameters (haemoglobin,packed cell volume, glucose, urea nitrogen, total protein) and activity of serum enzymes (alanine aminotransferase and aspartate aminotransferase) did not differ significantly in goats irrespective of dietary treatment. In the onfarm study, eighteen multiparous milking buffaloes were randomly assigned into three equal groups to assess the relative effect of feeding untreated wheat straw (WS), UTS and LS:UTS ad libitum as basal feed with a supplement. The study continued for 3 months and revealed that daily dry matter intake and average milk production (liters/day) by buffaloes offered LS:UTS and UTS was statistically similar but it was significantly higher compare to WS. However, milk composition did not differ significantly among the dietary treatments. It may be concluded that the nutritive value of lentil straw appeared to be no different to UTS, however, a positive synergistic effect was evident by feeding a mixture of LS and UTS on performance of goats and lactating buffaloes.

The present experiment was conducted to evaluate the effect of simulated heat stress on digestibility and methane ($CH_4$) emission. Four non-lactating crossbred cattle were exposed to $25^{\circ}C$, $30^{\circ}C$, $35^{\circ}C$, and $40^{\circ}C$ temperature with a relative humidity of 40% to 50% in a climatic chamber from 10:00 hours to 15:00 hours every day for 27 days. The physiological responses were recorded at 15:00 hours every day. The blood samples were collected at 15:00 hours on 1st, 6th, 11th, 16th, and 21st days and serum was collected for biochemical analysis. After 21 days, fecal and feed samples were collected continuously for six days for the estimation of digestibility. In the last 48 hours gas samples were collected continuously to estimate $CH_4$ emission. Heat stress in experimental animals at $35^{\circ}C$ and $40^{\circ}C$ was evident from an alteration (p<0.05) in rectal temperature, respiratory rate, pulse rate, water intake and serum thyroxin levels. The serum lactate dehydrogenase, aspartate aminotransferase, alanine aminotransferase, alkaline phosphatase activity and protein, urea, creatinine and triglyceride concentration changed (p<0.05), and body weight of the animals decreased (p<0.05) after temperature exposure at $40^{\circ}C$. The dry matter intake (DMI) was lower (p<0.05) at $40^{\circ}C$ exposure. The dry matter and neutral detergent fibre digestibilities were higher (p<0.05) at $35^{\circ}C$ compared to $25^{\circ}C$ and $30^{\circ}C$ exposure whereas, organic matter (OM) and acid detergent fibre digestibilities were higher (p<0.05) at $35^{\circ}C$ than $40^{\circ}C$ thermal exposure. The $CH_4$ emission/kg DMI and organic matter intake (OMI) declined (p<0.05) with increase in exposure temperature and reached its lowest levels at $40^{\circ}C$. It can be concluded from the present study that the digestibility and $CH_4$ emission were affected by intensity of heat stress. Further studies are necessary with respect to ruminal microbial changes to justify the variation in the digestibility and $CH_4$ emission during differential heat stress.
A cDNA of bovine brain glutamate dehydrogenase (GDH) was isolated from a cDNA library by recombinant PCR. The isolated cDNA has an open-reading frame of 1677 nucleotides, which codes for 559 amino acids. The expression of the recombinant bovine brain GDH enzyme was achieved in E. coli. BL21 (DE3) by using the pET-15b expression vector containing a T7 promoter. The recombinant GDH protein was also purified and characterized. The amino acid sequence was found 90% homologous to the human GDH. The molecular mass of the expressed GDH enzyme was estimated as 50 kDa by SDS-PAGE and Western blot using monoclonal antibodies against bovine brain GDH. The kinetic parameters of the expressed recombinant GDH enzymes were quite similar to those of the purified bovine brain GDH. The $K_m$ and $V_{max}$ values for $NAD^+$ were 0.1 mM and $1.08\;{\mu}mol/min/mg$, respectively. The catalytic activities of the recombinant GDH enzymes were inhibited by ATP in a concentration-dependent manner over the range of 10 - $100\;{\mu}M$, whereas, ADP increased the enzyme activity up to 2.3-fold. These results indicate that the recombinant-expressed bovine brain GDH that is produced has biochemical properties that are very similar to those of the purified GDH enzyme.
Purpose: This study was conducted in order to develop morning bread using powdered citrus peels and to examine the effect of its several biological activities for 12 weeks in overweight adults ($$BMI{\geq_-}23$$) living in dormitories of Chonbuk National University. Methods: Control bread was prepared using the same formula except for replacing the wheat flour with 3% citrus peel powder. Subjects were classified according to two groups: Intake group of 3% citrus peel powder fortified morning bread (n = 30) and general morning bread (n = 30). Results: The results of our study showed no significant effects of citrus peel powdered morning bread on anthropometric indices and biochemical characteristics among overweight adults. Despite the slight reduction in total-cholesterol, LDL-cholesterol, GOT, and GPT (p < 0.05), no significant differences in changes of these values were observed between the two groups. Conclusion: The short duration of our intervention might have resulted in the lack of any significant effect. Therefore, conduct of further studies with longer duration is warranted in order that the results from this study can be utilized as a basis for the development of food products with citrus peel powder.

A bacterium producing the halotolerant extracellular protease was isolated from squid jeotgal, and was identified as Bacillus sp. SJ-10 based on morphological, physiological and biochemical characteristics, as well as phylogenetic analysis using 16S rRNA gene sequence. The strain grew at $20^{\circ}C\sim55^{\circ}C$, pH 5~8, and 0%~14% NaCl and optimal growth conditions were $35{\pm}5^{\circ}C$, pH 7, and 5% NaCl. The major cellular fatty acids were anteiso-$C_{15:0}$, anteiso-$C_{17:0}$, and $C_{16:0}$ DNA G+C content was 50.58 mol% and menaquinone consisted of MK-7 Phylogenic analysis based on the 16S rRNA gene sequence indicated that SJ-10T belongs to the genus Bacillus. About 40 kDa of the salt-tolerant protease was purified by 40% ammonium sulfate saturation and Mono Q column chromatography. The optimal activity of the protease was pH 8 and stable at pH 5~10. The optimum temperature and NaCl concentration were $35{\pm}5^{\circ}C$ and $5{\pm}1%$, respectively.
We have isolated a bacterial strain that tends to kill P. micans from the mixed culture of p. minns plus seawater filtrate (poresize, 0.8 $\mu$m) collected at Masan bay in July 1996, in which the mixed culture grown in the f/2 medium. According to the experimental results of the isolated bacterium such as fatty acids analysis, morphological and biochemical characteristic tests, the strain was supposed to be a Pseudomonas and then it was named as Pseudomonas sp. LG-2. The killing effect of Pseudomonas sp. LG-2 against P. micans was proportionally increased with the concentrations of culture filtrate (pore size, 0.8 $\mu$m) is well as with the number of bacterium inoculated. In the mixed culture inoculated with $1.3\times10^6$ cells/ml of Pseudomonas sp. LG-2, the number of P. micans (2,000 cells/ml) was gradually decreased and then killed below 100 cells/ml within 7 days. In addition, the culture filtrate with $30\%$ of final concentration revealed a significant killing effect against P. micans around 3 days after culture. In the relationship between killing effects and growth stage of Pseudomonas sp. LG-2, the culture filtrate at lag phase has little effects on P. micans. In constant, the culture filtrate at mid-log phase showed the killing effect by decreasing P. micans to 112 in number within 5 days. In particular, the culture filtrate at stationary phase showed a significant killing effect against P. micans in which the majority of it was killed after 3 day culture. The species specificity of killing effects of Pseudomonas sp. LG-2 against 5 species of dinoflagellate was only found in P. micans and Scrippsiella trochoidea.
Osteoporosis, a disease characterized by low bone mass and microarchitectural deterioration of bone tissue leading to enhanced bone fragility and fracture risk, is a major public health problem. The diagnostic methods for osteoporosis include simple radiography, bone scan, DXA (Dual energy X-ray Absortiometry) and biochemical markers of bone turnover. Optimal treatment and prevention of osteoporosis require modification of risk factors, particularly smoking cessation, adequate physical activity, and attention to diet, in addition to pharmacologic intervention. The estrogens and raloxifene both prevent bone loss in postmenopausal women, and the estrogens probably also decrease the risk of first fracture. There is good evidence that raloxifene prevents further fractures in postmenopausal women who already have had fractures and some evidence that estrogen does as well. Bisphosphonate prevents bone loss and reduces fractures in healthy and osteoporotic postmenopausal women and in osteoporotic men as well. Risedronate is more potent and has fewer side effects than alendronate and reduces the incidence of fractures in osteoporotic women. Calcitonin increases bone mineral density in early postmenopausal women and men with idiopathic osteoporosis, and also reduces the risk of new fractures in osteoporotic women. All of the agents discussed above prevent bone resorption, whereas teriparatide and strontium increase bone formation and are effective in the treatment of osteoporotic women and men. New avenues for targeting osteoporosis will emerge as our knowledge of the regulatory mechanisms of bone remodeling increases, although issues of tissue specificity may remain to be addressed.
