Factors affecting the urease activity of native ureolytic bacteria isolated from coastal areas

  • Coastal erosion is becoming a significant problem in Greece, Bangladesh, and globally. For the prevention and minimization of damage from coastal erosion, combinations of various structures have been used conventionally. However, most of these methods are expensive. Therefore, creating artificial beachrock using local ureolytic bacteria and the MICP (Microbially Induced Carbonate Precipitation) method can be an alternative for coastal erosion protection, as it is a sustainable and eco-friendly biological ground improvement technique. Most research on MICP has been confined to land ureolytic bacteria and limited attention has been paid to coastal ureolytic bacteria for the measurement of urease activity. Subsequently, their various environmental effects have not been investigated. Therefore, for the successful application of MICP to coastal erosion protection, the type of bacteria, bacterial cell concentration, reaction temperature, cell culture duration, carbonate precipitation trend, pH of the media that controls the activity of the urease enzyme, etc., are evaluated. In this study, the effects of temperature, pH, and culture duration, as well as the trend in carbonate precipitation of coastal ureolytic bacteria isolated from two coastal regions in Greece and Bangladesh, were evaluated. The results showed that urease activity of coastal ureolytic bacteria species relies on some environmental parameters that are very important for successful sand solidification. In future, we aim to apply these findings towards the creation of artificial beachrock in combination with a geotextile tube for coastal erosion protection in Mediterranean countries, Bangladesh, and globally, for bio-mediated soil improvement.

A Study of Characteristics of Seeding Plants through Improvements of Dredge Vegetation-Base -Focus on Site 14 in Nakdong-gang- (준설토 파종식물의 생육경향 및 관리방안 연구 -낙동강 14공구 중심으로-)

  • This study was launched to verify the effective composition of plant species and its management program most suited for the dredged soiled area near Nakdong River Site 14. The improvement methods of planting base and the composition of plants such as silver grass, reed, and some colonies in aesthetic effect were studied. To search the management methods to decrease the confining pressure risen from the burried seeds which would consequently harm the previously seeded plants, experimental construction process was measured on the site. The purpose of this experiment was to figure out which part of the improvement on the plant base has the most significant effect for the revegetation of infertile, dredged soil, to verify the easily seeded, developing plants among seeded plants, and finally, to find the restoration model using plants near the dredged soil around riverside. 8 seeded plants and 23 invaded species were appeared which among the emerged plants, development of Aster yomena MAKINO, Lotus corniculatus var. japonica Regel, Trifolium repens L, and Dianthus longicalyx Miq were proved to be brought up well. Difference risen from the seed composition were not noticeable until 150day since the germination was proceeded mainly by Aster yomena MAKINO. The experimental plot with dredging sand+organic fertilizer method of construction and dredging sand+soil conditioner method showed most development while the effect of the plot with only the soil base of dredging sand stayed low. Another important method for the management of infertile, dredged soil base would be the removal of disturbing species which the experiment showed the tied relationship between the removal of disturbing species and development of seeding plants. Although this study was carried out focused on the Nakdong River Project, the study suggests the general management program that the removal of disturbing species such as Humulus japonicus Sieboid & Zucc. and Pueraria lobata (Willd.) Ohwi in times around rainy season(60days after seedling) would be effective for the easy growth of revegetation plants.

Improvement of a Rice Seed Pelleting Machine for Direct Seeding in Rice Cultivation(II) - Physical and cultural properties of the rice-seed pellets - (직파용 벼 펠렛종자 제조장치 개선 연구(II) - 펠렛종자의 물리적 특성과 재배특성 -)

  • Physical and cultural properties were investigated on the rice-seed pellets made by the pelleting machine(Yu, 2003) as the changes with mixing ratios of soil to rice seed of 6 : 1, 7 : 1, and 8 : I, and rotating speeds of forming rolls of 7, 10, and 13 rpm. Average weight, average diameter, and average sphericity of the pellets were 1.70 g, 12.0 ㎜, and 99.1 %, respectively. Average number of seeds per pellet was more than 3, and almost all pellets had more than 3 seeds in the cases of mixing ratios of 6 : 1, and 7 : 1 at the forming rolls' speed of 7 rpm. Gradual drying was needed because rapid drying caused cracks on surface of the pellets. Compression strength of the pellets dried in shady room was in the range of 132 ∼ 152 N, which was enough for handling. Comparing with the previous pellets(Park, 2002), average number of seeds per pellet, ratio of pellets including more than 3 seeds, and compression strength increased due to the effects of pressure feed of pellet materials, and improvements of the forming rolls. Emergence ratio of the pellets made at the mixing ratio of 6 : 1 and the forming rolls' speed of 7 rpm, was 100 % on dry paddy and was 97 % on flooded paddy surface. Good growth characteristics, and yield except number of seedling stand and ratio of missing plant were shown in planting of the pellets made at the mixing ratio of 7 : 1 and the forming rolls' speed of 7 rpm on flooded paddy surface field. Considering the cultural results, the mixing ratio of 6 : 1, and the forming rolls' speed of 7 rpm seems to be optimum operating condition for the improved pelleting machine.

Evaluation of SRI Water Management on the Reduction of Irrigation Supply and NPS Pollution in Paddies (SRI 물관리 방법이 논의 관개용수량과 비점오염원 저감에 미치는 영향)

  • Monitored data (rainfall runoff and water quality) from 4 different paddy sites over 3 years were compared to analyze the effect of irrigation water management on irrigation supply and rainfall runoff quality in Korea. The system of rice intensification water management was adopted at one site (SRI) while the conventional water management method was used for rice culture at the other three sites (CT, SD and HD). The soil texture at SRI, CT and SD was sandy loam while that at HD was silt loam. The average reduction of irrigation supply at SRI compared with CT, SD and HD during the 3 years studied was 49%, 51% and 55%, respectively. The average event mean concentration (EMC) at SRI compared with that at CT, SD and HD was decreased by 35% (BOD), 44% (COD), 47% (SS), 19% (TN) and 38% (TP). The correlation between rainfall runoff and the measured non-point source (NPS) pollutants was very good in general. The comparison revealed that SRI water management significantly reduced both irrigation supply and EMC in rainfall runoff. Paddy NPS pollution was closely related to factors that induce runoff such as rainfall and irrigation supply. It was concluded that SRI management could be an effective and practical option to cope with both water shortage due to climate change and water quality improvement in rural watersheds. However, further studies are recommended in large irrigation districts for use in the development and implementation of NPS pollution policies since the data was collected from field sized paddies.

Experimental Study on Reduction Effects of Non-Point Pollutants by Improvement of Infiltration Capacity of Soil Filter Strip (토양여과대의 침투능 향상을 통한 비점오염물질 저감 효과에 관한 실험적 연구)

  • Runoff of non-point pollutants has affected bad influence to water quality of river as reaching within short time. For this reason, reducing them prior to reaching aquatic systems or treating them after collection from discharge process of pollutants are desirable for efficient treatment of pollutants. This study was carried out to develop an ecotechnological method to prevent further aggravation of water quality by non-point source through vegetation filter strips. This study has placed a focus on improving infiltration capacity of soil for the optimum condition of vegetation filter strips. Therefore, we used titled soil filter strips instead of vegetation filter strips in this study. The three types of soil tilter strips were used in a bench scale experiment before applying to the field. The reduction efficiency of pollutants in soil filter strips (SS $84.5{\sim}92.5%$, BOD $67.9{\sim}80.6%$, T-N $43.4{\sim}76.6%$, T-P $40.6{\sim}87.4%$, Cu $28.3{\sim}48.1%$ Fe $92.1{\sim}97.7%$, Pb $81.4{\sim}97.3%$) was much higher than that of the controled group. And non-point pollutants reduction efficiency by soil filter strip's forms was estimated to be distinguishing in order of bio material, mixture of sand and gravel and lastly the whole gravel. In the event, the whole reduction efficiency of pollutants on the soil filter strips disclosed good results.

Analysis of the Effect of Bio-Retention Cells to Improve Water Cycle and Water Quality in Urban Streams (도시하천의 물순환 및 수질 개선을 위한 생태저류지의 효과분석)

  • Rapid urbanization poses three major problems in urban streams. The first problem is the reduction of soil wetting from rainfall as the impervious area increases. Decrease in soil wetting causes serious distortion in the water cycle of urban streams. The second problem is the increase of non-point sources pollutants by urban land use, and the third problem is the combined sewer overflows in the old city center. Increased non-point sources pollutants and combined sewer overflows are associated with water cycle distortion, which increases water pollution in urban streams. In this study, EPA SWMM was constructed for the Busan Oncheon-stream watershed in order to suggest solutions for these three problems, and the bio-retention cells installation project was planned by benchmarking the actual projects in New York City. Water cycle improvement and reduction of non-point sources pollutants and combined sewer overflows for each project scenario were analyzed together with required budgets.

Effect of Nurseries on Production of High Quality Seedlings in Panax ginseng C. A. Meyer (우량묘 생산을 위한 삼 묘대에 관한 연구)

  • This study was conducted to produce high quality ginseng seedlings by improvement of the physical properties of nursery soil. In order to select optimum nursery conditions, nursery types were treated with the yangiik nursery and semi-yangiik nursery. Soil conditioners were treated with two different materials of puffed rice husks and perlite. This experiments were tested at Gyunggido Agricultural Research and Extention Services, Hwaseng, from 2003 to 2004. Soil permeability was better at the plot of yangiik nursery than that of semi-yangjik nursery, and was better at plot of puffed rice husks than that of perlite. Soil porosity was improved 2.2% at plot of yangjiik nursery compare at that of semi-yangjik nursery, and was improved 2.0% at plot of puffed rice husks compared to control. There was no difference in the number of ginseng plants per kan among treatments. Root length was longer at plot of yangjik nursery than that of semi-yangjik nursery. More available ginseng plants per kan were obtained from the plot of yangjik nursery by more 252 seedlings than that of semi-yangjik nursery, but there was no significant difference between soil conditioners. The number of available seedlings per kan produced from yangjik nursery was 21.7% higher than those produced by semi-yangjik nursery. The number of available seedlings per kan produced from plot of puffed rice husks was 16.3% higher than that of control.

Improvement of Cyclosporin A Hydroxylation in Sebekia benihana by Conjugational Transfer of Streptomyces coelicolor SCO4967, a Secondary Metabolite Regulatory Gene (Sebekia benihana에서 Streptomyces coelicolor SCO4967 유전자 도입을 통한 하이드록실 사이클로스포린 A의 생전환)

  • Actinomycetes are Gram-positive soil bacteria and one of the most important industrial microorganisms due to superior biosynthetic capabilities of many valuable secondary metabolites as well as production of various valuable bioconversion enzymes. Among them are cytochrome P450 hydroxylase (CYP), which are hemoproteins encoded by a super family of genes, are universally distributed in most of the organisms from all biological kingdoms. Actinomycetes are a rich source of soluble CYP enzymes, which play critical roles in the bioactivation and detoxification of a wide variety of metabolite biosynthesis and xenobiotic transformation. Cyclosporin A (CyA), one of the most commonly-prescribed immunosuppressive drugs, was previously reported to be hydroxylated at the position of 4th N-methyl leucine by a rare actinomycetes called Sebekia benihana, leading to display different biological activity spectrum such as loss of immunosuppressive activities yet retaining hair growth-stimulating side effect. In order to improve this regio-selective CyA hydroxylation in S. benihana, previously-identified several secondary metabolite up-regulatory genes from Streptomyces coelicolor and S. avermitilis were heterologously overexpressed in S. benihana using an $ermE^*$ promoter-containing Streptomyces integrative expression vector. Among tested, SCO4967 encoding a conserved hypothetical protein significantly stimulated region-specific CyA hydroxylation in S. benihana, implying that some common regulatory systems functioning in both biosynthesis and bioconversion of secondary metabolite might be present in different actinomycetes species.

Debonding of microbially induced carbonate precipitation-stabilized sand by shearing and erosion

  • Microbially induced carbonate precipitation (MICP) is an innovative soil improvement approach utilizing metabolic activity of microbes to hydrolyze urea. In this paper, the shear response and the erodibility of MICP-treated sand under axial compression and submerged impinging jet were evaluated at a low confining stress range. Loose, poorly graded silica sand was used in testing. Specimens were cemented at low confining stresses until target shear wave velocities were achieved. Results indicated that the erodibility parameters of cemented specimens showed an increase in the critical shear stress by up to three orders of magnitude, while the erodibility coefficient decreased by up to four orders of magnitude. Such a trend was observed to be dependent on the level of cementation. The treated sand showed dilative behavior while the untreated sands showed contractive behavior. The shear modulus as a function of strain level, based on monitored shear wave velocity, indicated mineral debonding may commence at 0.05% axial strain. The peak strength was enhanced in terms of emerging cohesion parameter based on utilizing the Mohr-Coulomb failure criteria.

Water/nutrient use efficiency and effect of fertigation: a review

  • Fertigation, which has been introduced in agricultural fields since 1990, has been widely practiced in upland fields as well as in plastic film houses as part of the crop production system. In accordance with demands in the agricultural sector, a huge number of scientific studies on fertigation have been conducted worldwide. Moreover, with a combination of advanced technologies such as big-data, machine learning, etc., fertigation is positioned as an indispensable tool to achieve sustainable crop production and to enhance nutrient and water use efficiency. In this review, we focused on providing valuable information in terms of crop production and nutrient/water use efficiency. A variety of fertigation studies have described that enhancement of crop production did not differ relative to conventional method or slightly increased. In contrast, fertigation significantly improved nutrient/water use efficiency, with a reduction in use ranging from 20 to 50%. Water-soluble organic resources such as livestock manure and agricultural byproducts also have been identified as useful resources like chemical fertilizers. Furthermore, the initial irrigation point was generally recommended in a range of -10 - -40 kPa, although the point differed according to the crop and crop growth stage. From this review, we suggest that fertigation, which is closely integrated with advanced technology, could be a leading technology to attain not only food security but also carbon neutrality via improvement of nutrient/water use efficiency.
