Transcriptional profiles of rock bream iridovirus (RBIV) using microarray approaches

  • Rock bream iridovirus (RBIV) causes high mortality and economic losses in the rock bream (Oplegnathus fasciatus) aquaculture industry in Korea. Viral open reading frames (ORFs) expression profiling at different RBIV infection stages was investigated using microarray approaches. Rock bream were exposed to the virus and held for 7 days at 23 ℃ before the water temperature was reduced to 17 ℃. Herein, 28% mortality was observed from 24 to 35 days post infection (dpi), after which no mortality was observed until 70 dpi (end of the experiment). A total of 27 ORFs were significantly up- or down-regulated after RBIV infection. In RBIV-infected rock bream, four viral genes were expressed after 2 dpi. Most RBIV ORFs (26 genes, 96.2%) were significantly elevated between 7 and 20 dpi. Among them, 12 ORF (44.4%) transcripts reached their peak expression intensity at 15 dpi, and 14 ORFs (51.8%) were at peak expression intensity at 20 dpi. Expression levels began to decrease after 25 dpi, and 92.6% of ORFs (25 genes) were expressed below 1-fold at 70 dpi. From the microarray data, in addition to the viral infection, viral gene expression profiles were categorized into three infection stages, namely, early (2 dpi), middle (7 to 20 dpi), and recovery (25 and 70 dpi). RBIV ORFs 009R, 023R, 032L, 049L, and 056L were remarkably expressed during RBIV infection. Furthermore, six ORFs (001L, 013R, 052L, 053L, 058L, and 061L) were significantly expressed only at 20 dpi. To verify the cDNA microarray data, we performed quantitative real-time PCR, and the results were similar to that of the microarray. Our results provide novel observations on broader RBIV gene expression at different stages of infection and the development of control strategies against RBIV infection.

  • Muscle fatigue models to predict maximum endurance time (MET) are broadly classified as either 'empirical' or 'theoretical'. Empirical models are based on fitting experimental data and theoretical models on mathematical representations of physiological process. This paper examines the effectiveness of dynamic muscle fatigue model as theoretical model to predict maximum endurance time during forearm isometric contraction. Forty volunteers (20 females, 20 males) are participated in this study. Empirical models (exponential model and power model) and theoretical model (dynamic muscle fatigue model) are used to compare. Mean absolute deviation (MAD), correlation coefficient (r) and intraclass correlation (ICC) are calculated between theoretical model and empirical models. MAD are below 3.5%p, r and ICC are above 0.93 and 0.87, respectively. This results demonstrate that dynamic muscle fatigue model as theoretical model is valid to predict MET.

  • The purpose of this review is to summarize studies that investigate blood and the main components of fetal bovine serum (FBS) in vertebrates, including major livestock, and review the current research on commercializing cultured meat. Detailed research on FBS is still lacking; however, some studies have shown that FBS consists of proteins, carbohydrates, growth factors, cytokines, fats, vitamins, minerals, hormones, non-protein nitrogen, and inorganic compounds. However, there are few studies on how the composition of FBS differs from blood or serum composition in adult animals, which is probably one of the main reasons for not successfully replacing FBS. Moreover, recent studies on the development of FBS replacers and serum-free media have shown that it is difficult to conclude whether FBS has been completely replaced or serum-free media have been developed successfully. Our review of the industrialization of cultured meat reveals that many basic studies on the development of cultured meat have been conducted, but it is assumed that the study to reduce or replace ingredients derived from fetuses such as FBS has not yet been actively developed. Therefore, developing inexpensive and edible media is necessary for the successful industrialization of cultured meat.

Perception and experience of dental communication education based on the role played by some dental hygiene students (치위생의사소통 교육에서의 역할극 수업에 대한 인식과 경험에 대한 연구)

  • Background: This study aimed to examine the effectiveness of role-play in dental hygiene education by analyzing learners' perceptions and experiences of role-play classes. Methods: Role-play was used as a teaching method to run dental medical communication and practice classes for 31 3rd graders in the Department of Dental Hygiene to investigate the learner's perception of role-play classes, effectiveness of the classes, and learner's perception of the experience. The Mann-Whitney U test was used to analyze the differences in learners' perceptions of the role-play classes according to the main characteristics related to dental health communication education. Results: The correlation coefficient between awareness of dental communication education (4.74 ± 0.35) and awareness of role-play classes (4.44 ± 0.48) was an Rs of 0.634, showing a significant positive correlation (p<0.001). Conclusion: Learners recognized that dental hygiene communication competency is important for dental hygienists. In addition, a strategic teaching design is necessary to reduce the learner's burden on role-play considering the learner's experience, which is perceived as a disadvantage of role-play

Effects of Magnetic Therapy to Improve on Pain Threshold, Blood Flow, and Balance in Patient with Knee Osteoarthritis (자기장 치료법이 무릎 뼈관절염 환자의 통증 역치, 혈류량 및 균형능력에 미치는 영향)

  • Purpose : This study was performed to evaluate the effects of magnetic therapy (MT) on pain threshold, blood flow, and balance in patients with knee osteoarthritis. Methods : A single-blind, randomized controlled trial (RCT) was conducted with 30 patient with knee osteoarthritis. They were randomly allocated 2 groups; magnetic therapy group (MTG; n=15) and placebo magnetic therapy group (PG; n=15). The MTG group received 30 minutes magnetic therapy and 20 minute conservative physical therapy (Hotpack, ICT), magnetic therapy was conducted in magnetic therapy device (OM-100, NUGA, Korea). In the placebo magnetic group received 30 minutes placebo magnetic therapy and 20 minute conservative physical therapy. Each group performed 50 minutes a day 3 times a week for 8 weeks. The primary outcome pressure pain threshold test, blood flow, balance ability were measured by a pressure threshold meter (Commander algometer, JTECH medical, USA), laser dofler image (Moor LDI2-IR, Moor instruments, USA), balance measurement system (BioRescue, Marseille, France). The measurement were performed before and after the 8 weeks intervention period. Results : Both groups demonstrated significant improvement of outcome in pain threshold, blood flow, and balance ability during intervention period. magnetic therapy group revealed significant differences in pain threshold, blood flow, and balance as compared to the placebo magnetic therapy group groups (p<.05). Our results showed that magnetic therapy was more effective than placebo therapy on pain threshold, blood flow, and balance in patients with knee osteoarthritis. Conclusion : Our findings indicate that magnetic therapy can improve pain threshold, blood flow, and Balance, highlight the benefits of magnetic therapy. This study will be able to be used as an intervention data for recovering pain threshold, blood flow, and balance in patients with knee osteoarthritis.

Cardiovascular-metabolic Diseases Affecting Hearing loss in Workers Exposed to Noise (소음 노출 근로자의 청력손실에 미치는 심혈관-대사성 질환의 영향)

  • Objectives: We examined the association of hearing with cardio-metabolic diseases, dyslipidemia, hypertension and diabetes mellitus according to the personal and occupational characteristics of workers exposed to noise. Methods: The subjects of the study were 237,028 workers who underwent 2, 3, and 4 kHz airway pure tone audiometry in 2015 and who underwent clinical tests to diagnose cardiovascular-metabolic diseases. Cardiovascular-metabolic diseases were defined using reference values for respective items including blood pressure (systolic/diastolic), fasting blood glucose, cholesterol, and triglycerides. The airway pure tone hearing threshold of 2, 3, and 4 kHz, the average threshold of 2-3-4 kHz, and the hearing loss by the average threshold of the primary examination were distinguished. Results: Workers with cardiovascular-metabolic disease had significantly higher average hearing thresholds and higher rates of hearing loss. Logistic regression analysis, which adjusted for demographic variables of gender and age and occupational variables such as workplace size, industry, and type of work, and cardiovascular-metabolic disease as independent variables, showed that the odds ratio of hypertension to hearing loss in the mid-frequency was 1.239 (95% confidence interval: 1.118-1.374). For hypertension was 1.159 (1.107-1.214) and for diabetes it was 1.166 (1.104-1.230) for hearing loss in the high-frequency. Hearing loss measured by mean hearing was 1.178 (1.105-1.256) for hypertension and 1.181 (1.097-1.271) for diabetes. Conclusions: Cardiovascular-metabolic diseases in noise-exposed workers are associated with an increased risk of hearing loss and should be accompanied by bio-monitoring of cardiovascular-metabolic diseases in addition to auditory surveillance.

The Education System for Cytotechnologists and Efforts to Improve Professionalism in the United States of America (미국 세포병리사 양성 교육제도와 전문성 향상을 위한 노력)

  • The path to becoming a cytotechnologist (also known as a cytologist) in the United States of America (USA) requires a minimum of a bachelor's degree and the completion of a training program certified by the Commission on Accreditation of Allied Health Education Programs. Aspirants are recognized as cytologists after passing a test administered by the American Society for Clinical Pathology Board of Certification on completion of a recognized cytotechnology training program. Cytotechnology training programs are currently provided by 18 universities and hospitals. Six of them provide master's level education, unifying existing programs to the master's level, which is necessitated due to a shortage of pathologists and the rapid development of the health care system. The improvement and unification of the curriculum, qualification renewal system, and the effort to change the title of qualified personnel from cytotechnologist to cytologist are seen as measures that would strengthen the capabilities of cytotechnologists as well as assist in keeping pace with the rapidly changing medical system. Knowledge about the educational courses available for cytotechnologists and the efforts to strengthen their expertise in the USA would be helpful to professors who guide students preparing for overseas employment and seeking professional improvement in Korea.

A Survey on the Utilization and Demand of Thermotherapy Devices in Korean Medical Institutions (한방의료기관의 온열 치료 기기 활용 현황 및 개발 수요에 대한 조사 연구)

  • Objectives : This study investigated the current utilization status of thermotherapy devices in Korean medicine (KM) institutions and identified areas for improvement and further development, as perceived by KM doctors (KMDs). Methods : An online survey was conducted, targeting KMDs primarily engaged in clinical patient care. The questionnaire included items about respondents' clinical practices, the extent of thermotherapy device usage, their opinions on these devices, and perceived improvement needs. The collected data underwent quantitative analysis. Results : From the 1,025 respondents, data from 862 respondents who provided complete responses were analyzed. On average, respondents utilized thermotherapy treatments for 80% of their patients. Infrared (IR) phototherapy unit, electrical moxibustion apparatus, and heater-based thermotherapy devices were predominantly owned by respondents, with IR being the most frequently used. The average satisfaction score for current thermotherapy devices was 79. A significant concern raised was the necessity for improved health insurance coverage and efficacy evaluation. Conclusions : This research has confirmed that the extensive use of thermotherapy devices by KMDs in treating primarily musculoskeletal and gastrointestinal ailments - common conditions among patients in KM institutions. The main areas identified for improvement encompass safety, cost-effectiveness, and device efficacy. Future enhancements in thermotherapy devices should address these crucial aspects.

The effects of dietary self-monitoring intervention on anthropometric and metabolic changes via a mobile application or paper-based diary: a randomized trial

  • BACKGROUND/OBJECTIVES: Weight loss via a mobile application (App) or a paper-based diary (Paper) may confer favorable metabolic and anthropometric changes. SUBJECTS/METHODS: A randomized parallel trial was conducted among 57 adults whose body mass indices (BMIs) were 25 kg/m2 or greater. Participants randomly assigned to either the App group (n = 30) or the Paper group (n = 27) were advised to record their foods and supplements through App or Paper during the 12-week intervention period. Relative changes of anthropometries and biomarker levels were compared between the 2 intervention groups. Untargeted metabolic profiling was identified to discriminate metabolic profiles. RESULTS: Out of the 57 participants, 54 participants completed the trial. Changes in body weight and BMI were not significantly different between the 2 groups (P = 0.11). However, body fat and low-density lipoprotein (LDL)-cholesterol levels increased in the App group but decreased in the Paper group, and the difference was statistically significant (P = 0.03 for body fat and 0.02 for LDL-cholesterol). In the metabolomics analysis, decreases in methylglyoxal and (S)-malate in pyruvate metabolism and phosphatidylcholine (lecithin) in linoleic acid metabolism from pre- to post-intervention were observed in the Paper group. CONCLUSIONS: In the 12-week randomized parallel trial of weight loss through a App or a Paper, we found no significant difference in change in BMI or weight between the App and Paper groups, but improvement in body fatness and LDL-cholesterol levels only in the Paper group under the circumstances with minimal contact by dietitians or health care providers.

Induction of antiviral mechanisms by interferon-related genes in rock bream (Oplegnathus fasciatus) infected with rock bream iridovirus (RBIV)

  • We evaluated the transcriptional response of interferon (IFN)-related genes in rock bream iridovirus (RBIV)-infected rock bream under high-, low-, or no-mortality conditions induced by different stocking water temperatures. Under the high susceptibility condition (group A, water temperature 26℃, 100% mortality), only the Mx gene was expressed early, with prolonged expression, and with heavy viral loads of approximately 106~107 major capsid protein gene copies/μL from 4 to 10 days post infection (dpi). However, IRF1, IRF3, IRF8, STAT1, ISG15, PKR, Viperin, GVIN1, IFI44, and ISG56 were activated at later time points (8 dpi) and then quickly decreased (10 dpi). For the low susceptibility condition, the water temperature was set at 23℃ for 7 days (group B) and then reduced to 17℃. Group B exhibited a 28% mortality rate, in which persistent and effective antiviral responses were observed for long periods of time. In particular, at 20 and 22 dpi, when virus replication was peaked at approximately 107/μL, the expressions of most of the IFN-related genes (IRF1, IRF3, IRF8, Mx, STAT1, ISG15, PKR, Viperin, GVIN1, IFI44, and ISG56) were significantly higher in group B than in the control group. Moreover, prolonged and higher levels of IRF3 (at least 30 dpi), IRF8 (at least 30 dpi), ISG15 (at least 30 dpi), PKR (at least 28 dpi), Viperin (at least 30 dpi), and IFI44 (at least 30 dpi) were also observed in the recovery stage of infection. Under the no-susceptibility condition at 17℃ (0% mortality), significantly elevated levels of IRF3, Mx, ISG15, and PKR were observed mostly until 20 dpi. The findings indicate that RBIV infection can induce an efficient IFN-mediated antiviral immune response in low- and no-susceptibility conditions. The findings could be valuable for effective control of viral pathogens in fish.
