Lactic acid bacteria were isolated from silage, which produce high level of hydrogen peroxide in cell culture supernatant. The 16S rDNA sequences of the isolate matched perfectly with that of Lactobacillus fermentum (99.9%), examined by a 16S rDNA gene sequence analysis and similarity search using the GenBank database, thus named L. fermentum CS12-1. L. fermentum CS12-1 showed resistance to low pH and bile acid. The production of hydrogen peroxide by L. fermentum CS12-1 was confirmed by catalase treatment and high-performance liquid chromatography. L. fermentum CS12-1 accumulated hydrogen peroxide in culture broth as cells grew, and the highest concentration of hydrogen peroxide reached 3.5 mM at the late stationary growth phase. The cell-free supernatant of L. fermentum CS12-1 both before and after neutralization inhibited the growth of enterotoxigenic Escherichia coli K88 that causes diarrhea in piglets.
The recent bloom of a very large jellyfish Nemopilema nomurai has caused a danger to sea fishery and sea bathers. Presently, Nemopilema nomurai is thrown away through a separator system in the sea. The objective of this work was to produce bio-gas from Nemopilema nomurai by using anaerobic digestion. The bio-gas includes the hydrogen or the methane gases. It relates that Nemopilema nomurai is effectually changed into the renewable energy. When the jellyfish biomass was used as an organic carbon source the bio-gases were evolved. The aim of this study was to determine the optimal conditions for hydrogen and methane gases production according to the substrate concentrations of Nemopilema nomurai, optimal culture condition and the sludge-pretreatment without pH control. The optimal culture condition was found to be $35^{\circ}C$ and the heat-treatments of jellyfish was done at $120^{\circ}C$ for 30 min. The production rate of hydrogen and methane gas were found to be 8.8 mL/L/h, 37.2 mL/L/h from 1.5 g of dry Nemopilema nomurai.
Anaerobic fermentation of food waste (FW) and waste activated sludge (WAS) for hydrogen production was performed in CSTR (Continuous Stirred tank reactor) under various HRTs and volumetric mixing ratio (V/V) of two substrates, FW and WAS. The specific hydrogen production potential of FW was higher than that of WAS. However, pH drop in the CSTR for hydrogen production from FW was higher than that from WAS. The maintenance of desired pH during fermentative hydrogen production is regarded as the most important operation parameter for the stable hydrogen production. Therefore, when the potential of hydrogen production from FW and better buffer capacity of WAS, the proper mixture of FW and WAS for fermentative hydrogen production were considered as a useful complementary substrate. The maximum yield of specific hydrogen production, 140 mL/g VSS, was found at HRT of 2 day and the volumetric mixing ratio of 20:80 (WAS : FW). The spatial distribution of hydrogen producing bacteria was observed in anaerobic fermentative reactor using fluorescent in situ hybridization (FISH) method.
본 연구에서는 혐기성 소화에서 aeration이 수소생성에 미치는 영향에 대해 살펴보았다. 혐기성 소화슬러지를 0, 1, 3, 6, 12, 24 시간 동안 aeration 실시 후 glucose(20 g/L)를 기질로 이용하여 batch test를 실시하였다. Aeration 시간이 길어질수록 메탄가스가 감소하고, 수소가스가 증가하였으며, 6시간 동안 aeration을 실시한 반응조에서 가장 높은 수소 생성율(570 ml/L)을 나타내었다. 연속운전의 경우 aerated reactor는 메탄가스의 생성 없이 수소가 지속적으로 발생하였으며, non-aerated reactor의 경우 낮은 pH와 짧은 HRT만으로는 메탄 생성균의 활성을 완전히 저해할 수 없었다. 그러나 미생물관점에서의 보다 명확한 규명을 위해 향후 연구가 추가적으로 진행되어야 하며, 현장 적용성을 고려한 aeration 처리의 최적조건 도출도 이루어져야 할 것이다.
다양한 수소에너지의 생산방법 중에서 진정으로 청정하고 지속가능한 유일한 기술이 물로부터 수소를 획득하는 태양-수소제조 시스템이다. 태양에너지를 활용한 물로부터 수소생산 연구는 1979년 일본 동경대학의 Honda와 Fujishima 교수의 광전기화학적 방법이 성공적으로 제시된 이래로 매우 많은 연구가 진행되어 오고 있다. 이러한 관심은 가시광 광촉매 제조, 광전기화학전지 등의 개발을 유발하였으며, 융합기술의 하나인 바이오-광촉매 복합시스템 구성 등의 연구를 도출시켰다. 본 고에서는 이들 태양의 광에너지를 직접 활용한 물분해 수소생산 기술을 소개하였으나 태양열을 이용한 수소 제조기술은 포함시키지 않았다.
This paper presents bio-ethanol production from fruit wastes as it possibly alternate fossil fuel in the future. To illustrate the component ratio in exocarps of fruit wastes such as pears, apples, and persimmons, the amount of moisture, lignin, $\alpha$, $\beta$, $\gamma$-cellulose, and ash content were respectively examined by the ingredient analysis. Also, the amount of the glucose obtained from the enzyme hydrolysis using the axocarps was investigated. It was found in our results that the energy efficient process requires different temperature conditions for the saccharification step($50^{\circ}C$ and the fermentation step($30^{\circ}C$ in ethanol synthesis.
CdS and $TiO_2$ nanoparticles were made by the precipitation method and sol-gel method, respectively, and they were mixed mechanically and then treated with the hydrothermal processing. CdS-$TiO_2$ composite particulate films were thus prepared by casting CdS-$TiO_2$ mixed sol onto $SnO_2$ conducting glass and a subsequent heat-treatment at $400^{\circ}C$. Again, the physico-chemical and photoelectrochemical properties of these films were controlled by the surface treatment with $TiCl_4$ aqueous solution. The photocurrents and the hydrogen production rates measured under the present experimental conditions varied in the range of $3.5{\sim}4.5mA/cm^2$ and $0.3{\sim}1.8cc/cm^2$-hr, respectively, and showed the maximum values at the $CdS/[CdS+TiO_2]$ mole ratio of 0.2. Also, the surface treatment with $TiCl_4$ aqueous solution caused a considerable improvement in the photocatalytic activity, Probably as a result of close contacts between the primary particles by the etching effect of $TiCl_4$ It was found that the photoelectrochemical performance of these particulate films could be effectively enhanced by this approach.
CdS-$TiO_2$ nano-composite sol was prepared by the sol-gel method in organic solvents at room temperature and further hydrothermal treatment at various temperatures to control the physical properties of the primary particles. Again, CdS-$TiO_2$ composite particulate films were made by casting CdS-$TiO_2$ sols onto $F:SnO_2$ conducting glass and then heat-treatment at $400^{\circ}C$. Physical properties of these 61ms were further controlled by the surface treatment with $TiCl_4$, aqueous solution. The photo currents and hydrogen production rates measured under the experimental conditions varied according to the $CdS/[CdS+TiO_2]$ mole ratio and the mixed-sol preparation method. For $CdS-TiO_2$ composite sols prepared in IPA, CdS particles were homogeneously surrounded by $TiO_2$ particles. Also, the surface treatment with $TiCl_4$ aqueous solution caused a considerable improvement in the photocatalytic activity, probably as a result of close contacts between the primary particles by the etching effect of $TiCl_4$. It was found that the photoelectrochemical performance of these particulate films could be effectively enhanced by this approach.
This study was conducted to investigate the effect of thermal pre-treatment on bio-hydrogen from food waste. Two continuous reactors operated and VFAs(volatile fatty acids) production and microbial communities were analyzed. The average hydrogen yield was 0.50 and 0.33mol $H_2/mol$$hexose_{added}$ in thermally treated food added reactor(R1) and control(R2), respectively. Butyrate concentration was similarly 7,500mg/L in both reactors, but two times higher lactate concentration was observed in R2(3,800mg/L). The results of FISH(fluorescence in situ hybridization) showed that the relative microorganism to hydrogen producing bacteria was 78 and 27% in R1 and R2, respectively.
