위장관내 조건에서 이중코팅 처리 된 프로바이오틱 비피도박테리움의 생존력 향상 (Dual Coating Improves the Survival of Probiotic Bifidobacterium Strains during Exposure to Simulated Gastro-Intestinal Conditions)

  • 프로바이오틱 박테리아는 면역력 활성 조절, 콜레스테롤 수치 억제, 유당내성 강화, 항종양 활성 등의 다양한 생리활성 기능으로 건강 증진 효과가 있는 것으로 보고되고 있다. 프로바이오틱 박테리아는 일단 섭취하게 되면 위장관을 통과하는 동안 산도가 낮거나 단백질분해 효소가 많은 열악한 환경에서 생존해야 하며 프로바이오틱 효과를 발휘하기 위해 증식해야 한다. 이중 코팅 기술은 펩타이드와 다당류의 이중코팅으로 섭취된 프로바이오틱 박테리아를 열악한 조건으로부터 보호하기 위해 개발되었다. 본 연구에서는 이중코팅 된 4종의 비피도박테리움 혼합물의 생존 안정성을 평가하기 위해 코팅이 되지 않은 비피도박테리움 혼합물과 담즙, 산 저항성 및 열 안정성을 비교 평가하였다. 이중 코팅 된 균주와 코팅이 되지 않은 균주를 산과 담즙 조건 및 $40^{\circ}C$에 노출 시킨 후 한천배지에 배양하여 생존생육 세포수를 측정하였으며, BacLigtht 키트를 이용하여 염색 한 후 유세포 분석기를 이용하여 생균과 사균의 세포수를 평가하였다. 이중코팅 된 균주 혼합물의 경우 코팅이 되지 않은 균주 혼합물 보다 산, 담즙 내성이 더 높았으며, 열 안전성 또한 코팅 되지 않은 균주 혼합물보다 높은 것으로 나타났다. 이 같은 결과들로 이중코팅 기술은 프로바이오틱 박테리아의 안정성 및 섭취 후 위장관 트랙을 통과하는 동안 균주의 생존률을 향상시킬 수 있음을 확인하였다.

프로바이오틱스의 동결보호 및 장관안정성 개선을 위한 첨가제 효과 분석 (Analysis of Ingredient Mixtures for Cryoprotection and Gastrointestinal Stability of Probiotics)

  • Current drying and encapsulation methods for probiotics manufacturing are complicate and cost-burdened processes. The aim of this study was to develop a simple ingredient mixture to make probiotic granules via one-step process, providing not only a cryoprotective effect during freezing and drying but also high survival ratio in gastrointestinal tract. As cryoprotectans, commercially available ingredients including skim milk, monosaccharide (trehalose or glycerin), maltodextrins (with low or high degree of equivalents) were used. Their cryoprotective effect during lyophilization and survival ratios in artificial gastric juice and bile salt were measured against 3 strains of lactic acid bacteria (LAB) (Lactobacillus plantarum, Lb. brevis, and Lactococcus lactis). As results, 3 mixtures with different compositions showed a cryprotective effect on LAB tested and the best compostion was dependant upon LAB; skim milk 10%, trehalose 15%, glycerin 0.5%, and NaCl 1% was for Lb. plantarum and Lc. lactis, and maltodextrin 10% instead of skim milk was for Lb. brevis. In addition, those mixtures showed similar survival effect on LAB tested. These results demonstrate that skim milk or maltodextrins with trehalose, glycerin, and NACl can be effectively used for onestep lyophilization of LAB as an alternative method of encapsulation.

인체분변으로부터 분리한 유산균 Lactobacillus pentosus Miny-148의 생균제 특성 연구 (Probiotic Property of Lactobacillus pentosus Miny-148 Isolated from Human Feces)

  • 우수한 생균제를 개발하기 위하여 안전성이 알려진 유산균을 대상으로 인체의 분변으로부터 300여 균주를 분리하고 내산성, 내담즙성, 내열성, 항균력, 항암 및 항바이러스 효과를 가지는 균주들을 선발하여 생균제 특성을 나타내는지를 알아보기 위해 본 실험을 수행하였다. 인체에서 분리한 여러 균주 중 Miny-148 균주는 낮은 pH 및 높은 담즙산에 대한 내성, 열처리에 대한 열안정성을 지녀 기초적인 probiotic 특성을 가진 균주로 선발되어, Lactobacillus pentosus (99.9% 상동성)로 동정되었다. 항균력 실험에서 Escherichia coli O157:H7을 비롯한 Shigella flexneri, Bacillus anthracis, Staphylococcus aureus, E. coli, Vibrio cholerae, V. vulnificus, Salmonella typhimurium, 그리고 및 methicillin-resistant S. aureus (MRSA) 균주 8종 등 총 16종의 병원성세균을 억제하였다. 또한 Miny-148은 결장암 세포인 HT-29 cell을 억제하였을 뿐 아니라, transmissible gastroenteritis virus의 생육을 저해하여 세포변성 억제효과를 가진 우수한 probiotic 특성을 지닌 균주로 분석되었다.

전통발효식품 유래 유산균의 프로바이오틱스 특성 연구 (Probiotic Properties of Lactic Acid Bacteria Isolated Traditional Fermented Foods)

  • 본 연구에서는 전통발효식품에서 분리한 유산균 200여종에 대하여 프로바이오틱 특성을 확인하였다. 내담즙성 및 내산성이 높게 측정된 유산균 4종을 선발하였다. 선발된 유산균은 Lb. plantarum SRCM102224, Lb. plantarum SRCM102227, Lb. paracasei SRCM102329, Lb. paracasei SRCM102343이다. 항균활성, 내산성, 내담즙성, 용혈성, 세포표면 소수성, 장내 세포 부착성, 항생제 내성을 조사하였다. 선발된 유산균 4종 중 SRCM102343은 세포표면 소수성이 95.9%로 대조구 Lb. rhamnosus GG는 13.4%보다 높게 측정되었다. 4종의 유산균의 장내상피세포 caco-2에 대한 부착능을 조사하였다. 그 결과, SRCM102343와 SRCM102329의 Lb. rhamnosus GG보다 부착율이 높았다. 선발된 유산균 4종에 대하여 항균활성 조사 결과, 그 중 3종이 Escherichia coli ATCC 10798, Staphylococcus aureus KCCM 11593, Listeria invanovii KCTC3444, Bacillus cereus ATCC11778 및 S. enterica serovar. Typhi KCTC1926에 대하여 항균활성이 뛰어났다. 이들 결과를 바탕으로 선발된 균주는 프로바이오틱의 가능성으로 기능성식품에 활용이 기대된다.

Antioxidant and Anti-Inflammatory Effect of Probiotic Lactobacillus plantarum KU15149 Derived from Korean Homemade Diced-Radish Kimchi

  • Lactobacillus plantarum KU15149 was demonstrated to have probiotic behavior and functions, including antioxidant and anti-inflammatory activity. L. plantarum KU15149 obtained from homemade diced-radish kimchi has a high survival rate under artificial gastric acid (pH 2.5, 0.3% pepsin) and bile salt (0.3% oxgall) conditions. However, L. plantarum KU15149 did not produce β-glucuronidase, which is known to be a carcinogenic enzyme with resistance to several antibiotics, such as gentamycin, kanamycin, streptomycin, tetracycline, and ciprofloxacin. L. plantarum KU15149 strongly adhered to HT-29 cells and had high antioxidant activity in terms of 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical-scavenging and β-carotene bleaching assays. L. plantarum KU15149 also exhibited a pronounced inhibition of nitric oxide (NO) production, along with expression of nitric oxide synthase (iNOS) and cyclooxygenase -2 (COX-2) as well as pro-inflammatory cytokines, such as TNF-α, IL-1β, and IL-6, when RAW 264.7 cells were stimulated with LPS. Therefore, L. plantarum KU15149 exhibited pharmaceutical functionality as a potential probiotic.

Lactobacillus plantarumCLP-1이 돼지바이러스에 미치는 효과 (The Effect of Lactobacillus plantarumCLP-1 on the Swine Viruses)

  • To isolate Lactic acid bacteria for animals, we have screened from Kim-chi, swine intestine, swine feces, and dairy products by random selection and anti-viral, antipathogenic bacteria test. Among them, CLP-1 shown that inhibitory effect against rotavirus, porcine epidemic diarrhea (PED) virus, Salmonella sp, and E.coli. By examining biological property, API-ZYM and identified Lactobacillus plantarum by 16S rDNAgene sequence. CLP-1 determined resistance to low pH and bile salt. Futhermore, the cell body of CLP-1 adhered to the intestinal epithelium tissue of swine and Caco-2 cell. CLP-1 was examined on cell immune system modulating activity in vitro. The whole cell and cell culture supernatant was increasing of interferon-${\beta}$ activity. And then, CLP-1 increased prevention effect by Salmonella enteritidis infection in SPF chickens. And we determined similar result in pigs.

질 내 유산균의 Candida albicans에 대한 항진균 효과 연구 (Characterization and Antifungal Activity against Candida albicans of Vaginal Lactobacillus spp. Isolated from Korean Women)

  • Vulvovaginal candidiasis (VVC) is one of the urogenital infections occurring in women worldwide. Candida albicans is generally observed among various types of microorganisms causing VVC. Antibiotic therapy is typical, and the use of Lactobacilli probiotics is to be recognized as a promising alternative. The aim of this study was to select vaginal lactobacilli with probiotic properties against C. albicans. In a previous study, we isolated 38 lactobacilli from vagina of Korean women and 20 isolates were shown to inhibit C. albicans. We further selected 10 isolates which were able to inhibit C. albicans less than $10^5CFU/mL$. Among these selected strains, Lactobacillus salivarius MG242 (identified by 16s rRNA sequencing) was finally selected based on its strong anti-candidal activity, acid/bile salt resistance and adhesion property. Indirect adhesion activity of MG242 measured by auto-aggregation assay showed more than 60% auto-aggregation after 5 h standing. Taken these results together, the selected strain MG242 may have potential for application in vagina health related products.

An updated review on probiotics as an alternative of antibiotics in poultry - A review

  • Antibiotics used to be supplemented to animal feeds as growth promoter and as an effective strategy to reduce the burden of pathogenic bacteria present in the gastro-intestinal tract. However, in-feed antibiotics also kill bacteria that may be beneficial to the animal. Secondly, unrestricted use of antibiotics enhanced the antibiotic resistance in pathogenic bacteria. To overcome above problems, scientists are taking a great deal of measures to develop alternatives of antibiotics. There is convincing evidence that probiotics could replace in-feed antibiotics in poultry production. Because they have beneficial effects on growth performance, meat quality, bone health and eggshell quality in poultry. Better immune responses, healthier intestinal microflora and morphology which help the birds to resist against disease attack were also identified with the supplementation of probiotics. Probiotics establish cross-feeding between different bacterial strains of gut ecosystem and reduce the blood cholesterol level via bile salt hydrolase activity. The action mode of probiotics was also updated according to recently published literatures, i.e antimicrobial substances generation or toxin reduction. This comprehensive review of probiotics is aimed to highlight the beneficial effects of probiotics as a potential alternative strategy to replace the antibiotics in poultry.

김치로부터 분리한 Lactobacillus plantarum JK-01의 동정 및 생리적 특성 (The Identification and Physiological Properties of Lactobacillus plantarum JK-01 Isolated from Kimchi)

  • 김치의 L. plantarum의 생균제적 특성을 조사하기 위하여 김치에서 산생산과 성장능력 등이 우수한 25종의 Lactobacillus sp.를 분리하였고, API kit 분석에 의하여 L. plantarum을 동정하고 재차 16S rDNA 염기서열(99.9% 상동성)을 비교한 후 L. plantarum JK-01로 표기하였다. L. plantarum JK-01은 MRS broth에서 배양시 18시간 후 $2.9{\times}10^{10}CFU/ml$로 최대로 증식하는 빠른 성장특성을 나타냈으며 pH도 4.5로 조속히 하강하였다. 효소활성은 xylanase, amylase, protease, phytase의 순으로 높은 활성을 포함하고 있는 것으로 예측되었다. L. plantarum JK-01은 pH 2에서도 $1.36{\times}10^5CFU/mL$가 생존하였고, 1%의 담즙산에서도 약 $10^6CFU/mL$이상이 생존하여 내산성과 내담즙산성이 강한 것으로 나타났다. 또, $60^{\circ}C$에서도 $3.3{\times}10^3$ CFU/mL 정도로 생존하는 내열성이 있었으며, 대장균과 함께 배양시 18시간 후 대장균을 사멸시키는 것으로 나타났다. 이상의 결과로부터 분리한 L. plantarum JK-01은 생균제로서 충분히 이용가치가 있는 것으로 사료되었다.

Screening of Probiotic Activities of Lactobacilli Strains Isolated from Traditional Tibetan Qula, A Raw Yak Milk Cheese

  • In this study, 69 lactobacilli isolated from Tibetan Qula, a raw yak milk cheese, were screened for their potential use as probiotics. The isolates were tested in terms of: Their ability to survive at pH 2.0, pH 3.0, and in the presence of 0.3% bile salts; tolerance of simulated gastric and intestinal juices; antimicrobial activity; sensitivity against 11 specific antibiotics; and their cell surface hydrophobicity. The results show that out of the 69 strains, 29 strains (42%) had survival rates above 90% after 2 h of incubation at pH values of 2.0 or 3.0. Of these 29 strains, 21 strains showed a tolerance for 0.3% bile salt. Incubation of these 21 isolates in simulated gastrointestinal fluid for 3 h revealed survival rates above 90%; the survival rate for 20 of these isolates remained above 90% after 4 h of incubation in simulated intestinal fluid. The viable counts of bacteria after incubation in simulated gastric fluid for 3 h and simulated intestinal fluid for 4 h were both significantly different compared with the counts at 0 h (p<0.001). Further screening performed on the above 20 isolates indicated that all 20 lactobacilli strains exhibited inhibitory activity against Micrococcus luteus ATCC 4698, Bacillus subtilis ATCC 6633, Listeria monocytogenes ATCC 19115, and Salmonella enterica ATCC 43971. Moreover, all of the strains were resistant to vancomycin and streptomycin. Of the 20 strains, three were resistant to all 11 elected antibiotics (ciprofloxacin, erythromycin, tetracycline, penicillin G, ampicillin, streptomycin, polymyxin B, vancomycin, chloramphenicol, rifampicin, and gentamicin) in this study, and five were sensitive to more than half of the antibiotics. Additionally, the cell surface hydrophobicity of seven of the 20 lactobacilli strains was above 70%, including strains Lactobacillus casei 1,133 (92%), Lactobacillus plantarum 1086-1 (82%), Lactobacillus casei 1089 (81%), Lactobacillus casei 1138 (79%), Lactobacillus buchneri 1059 (78%), Lactobacillus plantarum 1141 (75%), and Lactobacillus plantarum 1197 (71%). Together, these results suggest that these seven strains are good probiotic candidates, and that tolerance against bile acid, simulated gastric and intestinal juices, antimicrobial activity, antibiotic resistance, and cell surface hydrophobicity could be adopted for preliminary screening of potentially probiotic lactobacilli.
