6탄당과 5탄당을 이용할 수 있는 효모 Picha stipitis를 이용하여 해조류인 구멍갈파래 가수분해 추출물의 단당류로부터 바이오 에탄올을 생산하는 반복 회분식 공정에 대하여 연구하였다. 이러한 공정이 180시간 까지 반복적으로 이루어질 수 있었으며, 약 30 g/L의 총환원당으로 부터 최고 평균 11.9 g/L의 바이오 에탄올이 생산됨을 확인하였다. 이 때 바이오 에탄올 수율은 0.40 (DNS 방법 기준)과 0.37 (TLC 방법 기준)이었으며, 이는 이론치의 78.4%와 72.5%에 해당하는 바이오 에탄올 수율에 해당한다. 이 결과를 다른 측면에서 분석하면, 본 연구 결과로 얻어진 반복 회분식공정에서 건조 구멍갈파래 1 kg에서 39.67 g의 바이오 에탄올을 생산 할 수 있다는 결론을 얻게 되었다. 본 연구를 통하여 구멍갈파래의 가수분해 추출물로부터 바이오 에탄올을 생산할 수 있다는 것을 실험적으로 증명하였고, 상업적인 대량생산이 가능한 공정기술로서 반복 회분식 방법이 적합하다는 것을 확인할 수 있었다.
가스 산업계에서 분산 제어 시스템 (DCS)이나 공정 정보 시스템 (PIS)이 널리 도입됨에 따라 데이터를 기반으로 하는 공정 이상 감시 기술이 많은 관심을 끌고 있다. 하지만 회분식 공정의 경우는 공정의 강한 비선형성으로 인해 이러한 기술들이 효과적으로 이용되지 못했다. Multiway principal component analysis (MPCA)가 개발됨에 따라 이러한 문제점을 해결하면서 산업계에 널리 이용되고 있는데 이 또한 다양한 정보를 해석하기 위해서는 상당한 통계적 지식이 요구되고 이 결과 운전원들이 이용하기가 어렵다는 단점이 있다. 이를 해결하기 위해서 본 논문에서는 운전원들이 쉽게 이해하고 활용할 수 있는 도구들을 제-공하는 회분식 가스 제조공정용 실시간 감시 시스템을 소개한다. 본 시스템은 데이터의 수집부터 이상의 원인을 파악하는 진단에 이르기까지 감시와 진단에 필요한 기능들을 모두 제공하는 총괄적인 시스템으로 개발된 시스템은 회분식 가스제조, 정밀화학제품, 의약품 등의 회분식 고부가가치 제품생산에 널리 이용될 것으로 보인다. 개발된 시스템은 산업체의 전형적인 회분식 반응기 감시를 위한 감시 시스템을 구축하여 봄으로써 검증하였으며 검증 결과 감시와 진단에 매우 효과적이라는 것을 알 수 있었다.
파이로프로세싱의 전해환원공정에서 생산된 금속전환체의 조성은 전해정련공정 운전의 중요한 운전변수인 용융염 중 $UCl_3$의 농도변화에 영향을 미친다. 따라서, 본 연구에서는 금속전환체에 함유된 TRU와 RE 원소의 함량 및 금속전환체에 동반되어 전해정련 전해조에 유입될 수 있는 $Li_2O$ 농도가 전해정련 전해조의 $UCl_3$ 농도 변화에 미치는 영향을 검토하였다. 금속전환체의 TRU 원소와 RE 원소의 농도만을 고려하였을 때 전해정련 운전 batch 수 증가에 따라 $UCl_3$ 농도가 감소하였다. 전해정련 1 campaign(20 batch)를 운전하기 위해서는 $UCl_3$를 3회 이상 추가 보충해야 함을 알 수 있었다. 한편, 금속전환체에 동반되어 전해정련 전해조에 유입되는 $Li_2O$의 유입량 증가에 따라 $UCl_3$ 농도 감소의 영향이 크게 나타났으며, 이에 따라 운전 가능 batch 수가 급격히 감소하게 되어 전해정련 운전에 중요한 운전 변수임을 보여주었다. 이러한 결과는 전해정련 운전 중 $UCl_3$ 농도 유지를 위해 금속전환체에 포함된 TRU 및 RE 원소뿐만 아니라 금속전환체에 동반되어 유입될 가능성이 있는 $Li_2O$의 영향도 고려하여 전해정련 운전모드를 설정하여야 함을 보여주었다.
화장품용 방부제인 CPN-Gal은 사람 피부세포에 대하여 보다 안전하다고 알려져있다. 장기간에 걸쳐서 CPN-Gal을 생산하는 공정을 확립하기 위하여, β-gal을 생산하는 재조합 대장균을 alginate bead에 고정화 시켜서, 반복 회분식 반응으로 CPN으로부터 CPN-Gal로의 전환반응(transgalactosylation)을 조사하여 보았다. 이 전환반응은 300 ml flask에서 수행하였으며, 반응액에는 고정화된 대장균, 33.8 mM CPN, 400 g/l lactose가 포함되어있으며, pH와 온도는 7.0과 40℃ 였다. 이 때, galactose 한 분자가 lactose로부터 CPN으로 전달된다. 반복 회분식 전환반응은 4회 연속적으로 192시간 동안 성공적으로 수행이 가능하였고, 이 때, 64%의 전환수율을 보였으며, 이 결과는 선행연구 결과에 비하여 보다 우수한 결과이다. 그런데, 반복 회분식 반응이 진행되면서, 192시간 이후부터는 점진적으로 전환수율의 감소가 관찰되었다. western blotting으로 β-gal의 양을 관찰하여본 결과, β-gal의 양의 감소가 관찰되었으며, alginate beads에서 깨어진 금이 또한 발견되었다. 이러한 bead의 깨어짐 현상과 아울러서 β-gal의 불활성화, 그리고 galactose에 의한 product inhibition 등이 192시간 이후의 전환수율 감소의 원인으로 생각된다. 본 연구의 전환공정은 β-gal의 정제가 필요 없이 진행할 수 있기 때문에, CPN-Gal 합성이 보다 실용적이고, 가격 경쟁력 있게 수행될 수 있을 것으로 생각된다. 그래서, 이러한 전환공정이 CPN-Gal을 상업화하기 위한 공정으로 발전하길 기대하고 있다.
Sequencing Batch Reactor (SBR) is well adopted for community wastewater treatment for its simplicity, performance and various advantageous treatment options. SBR is now drawing attention for its process modification such as coupled with membrane bioreactor, reverse osmosis or applying different media to achieve high removal efficiency. This study focused on the improved efficiency of carbon, nitrogen and phosphorous removal by applying zeolite materials called bioceramics to the SBR. Two laboratory-scale SBR units were operated in the same operating conditions - one with bioceramics called Bioceramic SBR (BCSBR) and the other without bioceramics used as control. Routine monitoring of COD, TP, $NH_3-N$, $NO_3-N$ was performed throughout this study. COD removal was about 80% to 100% and phosphorous removal was about 60% in the process whereas $NH_3-N$ removal efficiency was found to be 99.9% in the BCSBR unit. Addition of bioceramics also improved sludge characteristics such as sludge dewaterability, specific gravity and particle size. BCSBR can withstand high ammonia shock loading leading to the better treatment capacity of high ammonia containing wastewater. The cause of improved removal efficiencies within the biological reactor could be attributed to the biochemosorption mechanisms of bioceramics. Absorption/adsorption or desorption capacity of bioceramics was tested through laboratory experiments.
It has been proved that co-cultivation of human neroblastoma cells and human fibroblast cells can enhance nerve cell growth and the production of BDNF in perfusion cultivation. In batch co-cultivation, maximum cell density was increased up to 1.76${\times}$106 viable cells/mL from 9${\times}$105 viable cells/mL of only neuroblastoma cell culture. The growth of neuroblastoma cells was greatly improved by culturing both nerve and fibroblast cells in a perfusion process, maintaining 1.5${\times}$106 viable cells/mL, which was much higher than that form fed-batch cultivation. The nerve cell growth was greatly enhance in both fed-batch and perfusion cultivations while the growth of fibroblast cells was not. It strongly implies that the factors secreted from human fibrobast cells and/or the environments of co-culture system can enhance both cell growth and BDNF secretion. Specific BDNF production rate was not enhanced in co-cultures; however, the production period was increased as the cell growth was lengthened in the co-culture case. Competitive growth between nerve cells and fibroblast cells was not observed in all cases, showing no changes of fibroblast cell growth and only enhancement of the neuroblastoma cell growth and overall BDNF production. It was also found that the perfusion cultivation was the most appropriate process for cultivating two cell lines simultaneously in a bioreactor.
Kojic acid production by Aspergillus flavus strain S44-1 using sucrose as a carbon source was carried out in a 250-mL shake flask and a 2-L stirred tank fermenter. For comparison, production of kojic acid using glucose, fructose and its mixture was also carried out. Kojic acid production in shake flask fermentation was 25.8 g/L using glucose as the sole carbon source, 23.6 g/L with sucrose, and 6.4 g/L from fructose. Reduced kojic acid production (13.5 g/L) was observed when a combination of glucose and fructose was used as a carbon source. The highest production of kojic acid (40.2 g/L) was obtained from 150 g/L sucrose in a 2 L fermenter, while the lowest kojic acid production (10.3 g/L) was seen in fermentation using fructose as the sole carbon source. The experimental data from batch fermentation and resuspended cell system was analysed in order to form the basis for a kinetic model of the process. An unstructured model based on logistic and Luedeking-Piret equations was found suitable to describe the growth, substrate consumption, and efficiency of kojic acid production by A. flavus in batch fermentation using sucrose. From this model, it was found that kojic acid production by A. flavus was not a growth-associated process. Fermentation without pH control (from an initial culture pH of 3.0) showed higher kojic acid production than single-phase pH-controlled fermentation (pH 2.5, 2.75, and 3.0).
본 연구는 크루즈선에 적합한 생물학적 오 폐수처리장치 개발을 위하여 SBR, MBR, MSBR 공정을 Lab scale 실험을 수행하여 오염물 처리 효율과 크루즈선이라는 특수 환경과의 접목성을 검토한 결과 MSBR공정이 처리효율과 장치 운영 면에서 가장 적합한 공정으로 평가되었다. MSBR 공정은 처리 대상 물질이 특정 성분에 국한 되지 않고 유기물, 영양염류, 병원성 미생물 처리에 있어 모두 안정적인 효율을 나타내었으며, 소요용적 및 장치의 운영 면에서도 우수한 결과를 나타내어 선박이라는 특수한 현장 적용에 매우 유리한 공정으로 확인되었다. MSBR 공정의 BOD, COD 및 SS 제거 효율은 99%, 98%, 99%로 나타나 IMO의 규제 기준을 모두 만족하였으며, 총질소와 인의 제거 효율도 76%, 59%로 강화되어 가는 해양오염기준을 충만족시킬 수 있는 공정으로 판단되었다.

It has been known that continuous cultivation of hairy root is difficult to maintain for a long period of time compared to the microbial and callus cultures. Chemostat cultivation was successfully carried out in order to economically produce a plant-based colorant, betacyanin, from red beet hairy root for more than 85 days in a 14-1 fermentor. The result from the chemostat cultivation was compared to those of the batch and fed-batch cultivations of red beet hairy roots. It was shown that hairy root reached its steady state within 50 days of the cultivation, and then maintained for about 25-30 days in a wide range of dilution rates. Total betacyanin production from the continuous process was also calculated to be 2.65g at 0.28(l/d) of dilution rate, compared to 0.196g from fed-batch cultivation. It was found that betacyanin production was a partially growth related process, yielding 0.376 mg/g-fresh wt. cell and $1.89{\times}10^{-5}$ mg/g-fresh wt. cell/d, with 0.92 of correlation factor in a partial growth-product model. It was also shown that the cell growth required was relatively large for maintenance amount of energy at a low dilution rate. The growth of hairy root was inhibited by high light intensity in following a photo-inhibition model. The growth parameters were estimated to be 0.3(l/d), $10.56kcal/\textrm{m}^2/h$,{\;}and{\;}35.81kcal/\textrm{m}^2/h$ for the maximum specific growth rate, half saturation light intensity, and inhibition light intensity, respectively.
