Comparison of Bio-ethanol Productivity Using Food Wastes by Various Culture Modes (에탄올 발효방법에 따른 음식물류 폐기물의 바이오에탄올 생산성 비교)

  • In order to improve bio-ethanol productivity by various cultivation methods in this paper, the culture modes using food wastes, such as batch culture, high-cell-density fermentation, SSF (simultaneous saccharification and fermentation) by fill & draw, continuous culture by fill & draw were performed and their productivities were compared. SSFs by fill & draw were performed by continuous decompression using 1 L evaporator system, and by 10 L bioreactor without decompression. In addition, the continuous cultures by fill & draw mode using SFW (saccharafied food wastes) medium were performed by changes of 40% culture broth with intervals of 12 h (0.03 $h^{-1}$), 6 h (0.07 $h^{-1}$), 3 h (0.13 $h^{-1}$). Consequently, productivities of bio-ethanol were 2.52 g/L-h and 1.30 g/L-h in batch culture and high- cell-density fermentation, respectively. The productivities of SSF by fill & draw showed 2.24 g/L-h and 2.03 g/L-h in continuous decompression with 1 L evaporator and 10 L bioreactor without decompression, respectively. Also, the productivities in continuous culture by fill & draw modes showed 2.02 g/L-h, 4.07 g/L-h and 6.25 g/L-h by medium change with intervals of 12 h, 6 h, and 3 h, respectively. In conclusion, the highest ethanol productivity was obtained in the continuous culture mode by fill & draw with dilution rate of 0.13 $h^{-1}$.

Quality Properties of Pear Vinegars with High-Acidity under Different Fermentation Conditions (고산도 배식초 제조 시 발효조건에 따른 품질특성)

  • High-acidity vinegar was manufactured using pear concentrate by fed-batch fermentation without additional nutrients, and the physicochemical properties and volatile components were investigated at different fermentation stages (Stages 1-4) and at various initial alcohol concentrations (IAC; 6-9%). The levels of reducing sugar, free amino acids, total phenolic content, total flavonoid content, and radical scavenging ability increased slightly during Stage 4 (high-acidity vinegar), which was affected by alcohol feeding. The contents of approximately 20 types of volatile compounds differed between the moderate- and high-acidity vinegar samples, as determined by solid-phase microextraction/gas chromatography-mass spectroscopy. The level of acetic acid in high-acidity vinegar increased according to the initial alcoholic content applied. The high-acidity vinegar produced by fed-batch culture at an IAC of 6-7% showed improved physicochemical and volatile properties as compared to the moderate-acidity vinegar.

  • This study was conducted to evaluate the performance of methane fermentation from effluent of hydrogen fermentation reactor in anaerobic baffled reactor (ABR) and anaerobic sequencing batch reactor (ASBR). Two reactors were operated at organic loading rate of $1.0kg\;COD/m^3{\cdot}d$ and hydraulic retention time (HRT) of 20 day. Methane production rates of ABR and ASBR for start-up periods were 0.04 L/L/d and 0.19 L/L/d, respectively, whereas maximum methane production rates of ABR and ASBR were 0.25 L/L/d and 0.31 L/L/d, respectively. Removal rates of chemical oxygen demand (COD) in ABR and ASBR for start-up periods were 89% and 92%, respectively. After startup periods, removal rates of COD and volatile solids (VS) in ABR and ASBR were maintained over 90%. The specific methanogenic activity (SMA) increased as microorganism acclimated to the substrate.

The Accelerative Effect on Fermentation of Salted and Fermented Anchovy by Homogenates of Sea Tangle, Laminaria japonica Aresschoug (다시마 마쇄물을 이용한 멸치젓갈의 숙성 촉진)

  • The present study was directed to investigate the accelerative effects of fermentation of salted anchovy on sea tangle homogenates. With the addition of $8\%$ (w/w) sea tangle homogenates(T), there was an increase of amino-N content in both the muscle and juice of salted anchovy during all fermentation periods. It was only in VBN value that there was exhibited the same characteristics as the control batch. Viable cell count in muscle was increased rapidly after 60 days of fermentation, but in juice the content was maximal after 60 days of fermentation. When $8\%$ (w/w) sea tangle homogenates was added, the pH value in muscle and juice were maintained same degree of control until 80 days of fermentation, but showed increase in pH value of muscle and juice more rapidly than the control system after 80 days of fermentation. Then a large percentage of muscle turned to juice after 100 days of fermentation. The degree of fermentation In salted anchovy, when sea tangle homogenates were added, accelerated more than the control batch. Concerning the factors related to the accelerative effects on fermentation of salted anchovy, there was a continuous increase in amino-N content, and it was a sudden change of viable cell count and pH value at a certain point in the fermentation periods.

  • Improvement of productivity in ethanol fermentation was attempted using a hollow fiber bioreactor (HFR) where Saccharomyces cerevisiac var. ellipsoideus cells were recycled to achieve a high yeast concentration. Industrial wort was used as the fermentation media without supplying any additional nutrients. The performances in hollow fiber recycle reactor (HFR) were compared with those of batch and continuous cultures. In a continuous culture with 11$^{\circ}$P and 15$^{\circ}$P wort media final ethanol concentrations were 4.71% and 5.82% (v/v) and yields 86.2% and 78.6% respectively when the dilution rate (D) was 0.1 h$^{-1}$, in contrast, the ethanol concentration and productivity in HFR were 7.64%(v/v) and 6.1g/l/h at D=0.1h$^{-1}$ with 15$^{\circ}$P media. When the dilution rate was increased to 0.2 h$^{-1}$, the concentration and the Productivity were 7.62% (v/v) and 12.2g/l/h. At D=0.3h$^{-1}$ the sugar was completely consumed and the productivity was 18.1g/l/h. This correponds to 4 times that in continuous system and 16.3 times that in the batch system performed in comparable conditions.

Invertase Production by Fed-batch Fermentations of Recombinant Saccharomyces cerevisiae

  • Fed-batch fermentations with different feeding media were carried out in order to increase the productivity of invertase expression using a recombinant Saccharomyces cerevisiae containing plasmid pRB58. Two batch cultures showed the expression of the SUC2 gene at a low concentration of glucose, suggesting that glucose concentration could be used as a control variable in a fed-batch operation mode. In the fed-batch culture by feeding the basal medium, cell mass and specific invertase activity did not increase much as compared with the simple batch culture. A series of fed-batch cultures revealed that the sucrose-supplemented medium increased cell mass whereas the enriched medium did specific invertase activity. To capitalize on the synergism of the sucrose-supplemented medium and the enriched medium, the sucrose-supplemented enriched medium was used as a feeding medium. The fed-batch culture using this medium resulted in a 2.4-fold increase in cell mass and a 1.9-fold enhancement in specific invertase activity compared with those of the batch culture. The increase in cell mass and specific invertase activity led to a marked increase in total invertase activity, 250U/ml, which was 6.3 times higher than that of the batch culture.

Process Kinetics of Nisin Production in Batch and Continuous Culture (회분식 및 연속식 배양시 Nisin의 생산특성)

  • Fermentation condition of Streptococcus lactis IFO 12007 for nisin production was examined. The optimal glucose concentration was 60g/ι. The pH and temperature optimum were 6.5 and 31$^{\circ}C$, respectively. The maximum nisin activity in batch culture was 2000IU/$m\ell$. The fermentation quotients after 7 hours of fermentation in batch culture were; specific glucose uptake rate:0.59g/g/h , specific nisin productivity: 34924IU/g/h, product yield: 5944IU/g, growth yield:0.24, biomass:4.81g/ι. The specific growth rate was affected by pH and temperature and the activation energy for growth was 1.35kcal/mole. pH control was essential for nisin production. Fed-batch culture using 20g/$\ell$ glucose medium produced 1420IU/$m\ell$ after 14 hours. The continuous culture could be operated at below 0.38h$^{-1}$ for nisin production. The steady state nisin concentration and specific nisin productivity were 740IU/$m\ell$ and 45000IU/g/h. The growth yield and maintenance energy were 0.144 and 207mg glucose/g-cell/h.

  • Pichia pastoris and Hansenula polymorpha, the methylotrophic yeasts have been widely used as a host for the production of e eudaryotic proteins due to the advantages related to their inherited characters. This paper describes the method to enhance t the productivity of recombinant proteins by P. pastoris and H. po$\psi$morpha. In the production of recombinant proteins using a f fed batch fermentation system, the effects of specific growth rate on the specific expression rate of re$\infty$mbinant proteins w were studied. In both species, the expression system of recombinant proteins using the fed batch fermentation was optimezed.

  • Effect of glucose feeding strategy and initial concentration of glucose on Serratia marcescens ATCC 27117 in high cell density fed-batch fermentation was investigated. The final biomasses in batch, constant feeding, constant and exponentially feeding strategy at glucose starvation condition in fed-batch were 1.40, 5,07, 6,93 and 7.60 g/L at 40, 41, 24 and 40 hrs, respectively. Productivities of biomass were 0.035, 0.124, 0.289 and 0.190 g/L$.$h, respectively. As a result, constant feeding strategy at starvation condition was 1.5∼8.6 times higher than other strategies. The relationship between dissolved oxygen and glucose feeding times was good identified in exponential feeding strategy and constant feeding strategy at starvation condition. And high cell density cultivation was obtained when minimal media was used.

  • A novel control method with automatic tuning of PID controller parameters has been developed for efficient regulation of dissolved oxygen concentration in fed-batch fermentations of Escherichia coli. Agitation speed and oxygen partial pressure in the inlet gas stream were chosen to be the manipulated variables. A heuristic reasoning allowed improved tuning decisions from the supervision of control performance indices and it coule obviate the needs for process assumptions or disturbance patterns. The control input consisted of feedback and feedforword parts. The feedback part was determined by PID control and the feedforward part is determined from the feed rate. The proportional gain was updated on-line by a set of heuristics rules based on the supervision of three performance indices. These indices were output error covariance, the average value of output error, and input covariance, which were calculated on-line using a moving window. The integral and derivative time constants were determined from the period of output response. The specific growth rate was maintained at a low level to avoid acetic acid accumulation and thus to achieve a high cell density. The specific growthe rate was estimated from the carbon dioxide evolution rate. In fed-batch fermentation, the simutaneous control of dissolved oxygen concentration (at 0.2; fraction of saturated value) and specific growth rate (at 0.25$hr^{-1}$) was satisfactory for the entire culture period in spite of the changes in the feed rate and the switching of control input.
