Effect of Natural Product Extracts on Inhibition of Macrophage and Basophil (천연물 추출물이 대식세포 및 호염구 활성 억제에 미치는 영향)

  • Asthma is a chronic inflammatory disease characterized by recurring symptoms, airflow obstruction, and bronchial hyper-responsiveness. The onset of asthma for most patients begins early in life, and current asthma treatment with anti-inflammatory agents can have adverse effects, eventually leading to impaired quality of life. In the pathogenesis of asthma, macrophages and basophils play a vital role during progression. Macrophages not only induce inflammation by secreting inflammatory cytokines but also promote DNA damage and mucus production through nitric oxide (NO) production. Basophils enhance eosinophil recruitment and aggravate asthma through the FcεRIα receptor with high affinity for histamine and IgE. Therefore, in this study, we investigated whether the activation of macrophages and basophils is suppressed by the individual extracts of 28 natural products. RAW 264.7 cells (mouse macrophages) were treated with the natural products in LPS, and 4 natural product extracts resulted in decreased NO production. In β-hexosaminidase assay using RBL-2H3 cells (rat basophils), 19 natural product extracts decreased β-hexosaminidase production. In NO production and β-hexosaminidase assay using macrophages and basophils, 3 natural product extracts (Plantago asiatica, Centella asiatica, and Perilla frutescens var. japonica) significantly inhibited NO production and β-hexosaminidase release. Overall, we examined the inhibitory effects of 28 natural product extracts on macrophage and basophil activity, and the findings demonstrated the potential of natural product extracts for treating asthma and macrophage- and basophil-related diseases.

  • 세균 바이러스 원충 등의 병원체가 생체내에 침입하면 그 부위에 inflammation이 생긴다. 동시에 혈관속에서 흐르고 있는 백혈구가 inflammation site에 집결하는 생체방어반응이 나타난다. 백혈구에는 lymphocyte, monocyte, neutrophil, basophil, eosinophil 그리고 macrophage 등의 세포들이 있는데 염증부위에 이들 모든 세포들이 집결하는 것이 아니다. 또 염증의 종류에 따라 모이는 백혈구의 종류도 다르고 또 염증부위의 시간경과에 따라 침윤되는 백혈구 종류도 다르다. 이런 차이점이 어떠한 mechanism으로 나타나는 것일까? 그리고 혈관내를 맹렬한 속도로 흐르고 있는 백혈구가 어떻게 inflammation site를 인지하고 또 급속하게 정지하고 혈관벽에 부착되어 내피세포를 통과한 후 inflammation site에 모이게 되는가? 이런 의문은 최근의 molecular pathology 연구발전으로 거의 해명하게 되었다. 여기 Kyoa Hakko Institute의 Nishi가 J. Bioscience and Biotechnology, 33, 1995에 발표한 Leukocyte Move into Inflammed Tissues 논문을 간추려서 알기쉽게 해설하고자 한다.

Hematological and blood chemical changes of rabbits infected with viral hemorrhagic disease virus (토끼 출혈성 바이러스에 감염된 토끼의 혈액상과 혈액화학치의 변화)

  • Hematological and blood chemical changes of rabbits infected with rabbit hemorrhagic disease virus were studied and the results were summarized as follows: 1. Total leukocyte count ($2,410{\pm}1,076/{\mu}l$), lymphocyte count ($1,582{\pm}632.5{\mu}l$) and heterophil count ($705{\pm}411.1{\mu}l$) were significantly decreased after 24 hours of the infection (p<0.01). However, no significant changes were observed in monocyte, eosinophil and basophil numbers. 2. A significant increase of aspartate aminotransferase (96IU/L), alanine aminotransferase (96IU/L) and alkaline phosphatase ($401.1{\pm}131.8IU/L$) was observed (p<0.01). 3. A moderate increase of BUN ($26.9{\pm}3.6mg/100ml$) and creatinine ($3.2{\pm}1.9mg/100ml$) was observed (p<0.05). 4. No significant changes of r-GTP, thymol turbidity, glucose, cholesterol, albumin, total serum protein, fibrinogen were observed.

Historical Control Data from 4-week Repeated Toxicity Studies in Crj:CD (SD) Rats

  • Reference ranges of standard experimental parameters are useful for comparisons in toxicology. The aim of this study was to collect data from 4-week repeated toxicity studies in Crl:CD (SD) rats, a strain widely used for toxicity and efficacy research, for establishing domestic reference values. Data on body weight, food consumption; urinalysis, hematological, and blood biochemical parameters; and organ weights were collected from 16 toxicity studies in 220 Crl:CD (SD) rats (110 males and 110 females). The studies had been performed at a single testing facility over the last 3 years and involved animals sourced from a single breeder. The findings were collated as means, standard deviations, percentages, and ranges. Urine volume, uterus weight, eosinophil, and basophil counts, and triglyceride, total bilirubin, and gammaglutamyl transpeptidase levels showed standard deviations of 30% or more. These historical control data would help to interpret the effects of test substances in routine toxicity and efficacy studies.

Biodegradation and Preparation of Biodegradable Film by using Natural Gums (천연 Gum류를 이용한 생분해성 필름의 제조와 생체분해)

  • Gum is known as natural polymer. Biodegradable films were prepared by solution blend method in the weight ratio of natural gums(Xanthan, Locust bean, Guar) for the purpose of useful bioimplants. The possibility of bioimplants, which prepared from natural gums as a skin substitute was evaluated by measuring biodegradability. This biodegradable films were inserted in the back of rats and their of biodegradability were investigated by hematological change evaluation as a function of time to the biotransformation. Rats implantation test results showed that Guar induced increments of monocyte and basophil after 48 hours of implantation. And Locust bean showed increase of monocyte and neutrophile counts after 48 hours of implantation. And Xanthan induced decrease of monocyte and neutrophile at 24 hours after implantation. Guar and Locust showed high hemoglobin contents and hematocrit after 48 hours of implantation. Guar and Locust induced some suspects of incompatibility in the tissue after 48 hours, but three were little effects to the skin inflammation at 24 hours. These values of biodegradable films, which prepared from prepared from natural gums measured in this were some satisfiable results at short period with those of ideal skin bioimplants.

The Effect of Norepinephrine on Immune function in elderly people (노르에피네프린이 노인의 면역기능에 미치는 영향)

  • Purpose: The purpose of this study was to examine how norepinephrine affects immunity in patients over age 65. Methods: We enrolled 25 male and female subjects age 65 or older. A low frequency electroacupuncture (EA) device was used to stimulate acupoint Hogu (L14). The 2 Hz frequency EA was applied to the acupoint for 20 minutes. Leukocyte subtypes-including neutrophils, lymphocytes, monocytes, eosinophils, and basophil-were then measured. The immunoglobulins IgG and IgM were also quantified. The data were finally analyzed using Wilcoxon singed-rank test and regression test as part of the SPSS WIN v. 10.1 program. Results: As norepinephrine levels decreased after EA stimulation, neutrophil, lymphocyte, and monocyte levels increased, and eosinophil and neutrophils levels decreased. Neutrophil and monocyte levels did not change to a statistically significant degree, but eosinophil levels showed a statistically significant decrease (p<0.05). Immunoglobulin IgG showed a statistically significant increase (p<0.05). Conclusion: This study showed that norepinephrine does affect immunity in persons over the age of 65. This indicates that there is an interaction between the nervous system and the immune system, and interaction that plays a crucial roles in the body's immune resistance and homeostasis.

  • Biodegradable films were prepared by solution blending method in the weight ratio of Chitosan and Algin for the purpose of useful bioimplants. The possibility of bioimplants, which prepared from natural polymers as a skin substitute and food wrapping materials were evaluated by measuring the biodegradability. these biodegradable films were inserted in the back of rats and their biodegradability was investigated by hematological change as a function of time. Rats study showed that low-Chitosan induced increments of monocyte and basophil after 48 hours of implantation. And medium-Chitosan showed increase of lymphocyte and decreased neutrophil counts after 48 hours of implantation. Low, medium Chitosan showed high hemoglobin contents, medium and high Chitosan showed high hematocrit value after 48 hours of implantation. As a result, medium, high-Chitosan induced potential incompatibility in the tissue after 48 hours, but there was little effects to the skin inflammation. The values of biodegradable films, which prepared from natural polymers measured in this study were some satisfiable results at short period with those of ideal skin bioimplants and artificial skin.

Excavation of Lead Compounds that Inhibit Mast Cell Degranulation by Combinatorial Chemistry and Activity-Guided

  • An allergic reaction ensues after antigen binds to mast cell or basophil high affinity IgE receptor, Fc$\varepsilon$RI, resulting in degranulation of various inflammatory mediators that produce various allergic symptoms. In this study, i) we isolated the active component for the inhibition of mast cell degranulation from the extract of leaves of Castanea crenata and identified it as quercetin; ii) we established the total synthesis procedure of quercetin; iii) using quercetin as positive control, we excavated some lead compounds that possess inhibitory activities for mast cell degranulation by screening the chemical libraries of 1,3-oxazolidine derivatives prepared by solid phase combinatorial chemistry. Some of 1,3-oxazolidine compounds possessing acetyl and 3',4'-dichlorophenyl group displayed strong inhibitory activities on Fc$\varepsilon$RI-mediated mast cell degranulation, suggesting that they can be used as lead compounds for the development of anti-allergic agents.

  • Seven cases of canine babesiasis were observed by clinically, hematologically and immunologically in the Kyungbook and Pusan area from May 1961 to October 1963. And a survey was conducted on the rising pups for their babesia, infection in the broken out districts of the disease. The results obtained were as follows: 1. Severe cases of canine babesiasis due to the same agent have been encountered in various breed, of dogs in widely separated location in the Kyungpook and Pusan area. 2. The principal symptoms oserved in infected dogs were severe anemia, weakness, inappetence, moderate fever or subnormal body temperature, increased respiration and pulse, palpitation, icterus, hemogrobinuria and redish yellow defecation. 3. Of hematological findings, decrease in erythrocytes count, hemoglobin content was evident and severe anemic changes occured which were associated with macrocytic anisocytosis. The differential leucocytes counts showed a tendency to increased monocytes and decreased monocytes and decreased basophil. 4. The principal anatomic chabges observed in two cases of infected dogs were severe anemic and icteric changes, markedly enlarged spleen, and enlarged liver with distended gall bladder. 5. Haemaphysalis bispinosa was suspected as being the principal vector of the infection. 6. It was suspected that chronic canine babesiasis would prevailed widely in Kyungpook and Pusan area, and that dogs are raised in such infected environment usually suffer only from the mild, chronic form, which may be practically symptomless, while imported dogs usually suffer from the acute form of the disease.

Taenia saginata Infection Misdiagnosed as Acute Cholecystitis in a Tibetan Patient, in China

  • The present study reports a rare case of Taenia saginata infection, which was initially diagnosed as acute cholecystitis in a Tibetan patient at the Qinghai-Tibetan Plateau pastoral area, China. A 45-year-old female was initially diagnosed with acute cholecystitis at a hospital in China. She had a slight fever, weight loss and constipation and complained of pain in the upper abdomen and left back areas. Increase of monocyte, eosinophil and basophil levels were shown. Taenia sp. eggs were detected in a fecal examination. An adult tapeworm approximately 146 cm in length, whitish-yellow color, was collected from the patient after treatment with traditional Chinese medicine. The adult tapeworm had a scolex and proglottids with genital pores. The scolex was rectangular shape with 4 suckers and rostellum without hooklet. The cox1 gene sequence shared 99.5-99.8% homology with that of T. saginata from other regions in China. The patient was diagnosed finally infected with T. saginata by morphological and molecular charateristics.
