Cellular Structural Change of Barley Seedling on Different Salt Concentration under Hydroponic Culture (보리 유묘의 염농도에 따른 세포의 형태반응)

  • The salt stress at seedling stage of winter barley was examined in different concentrations of NaCl containing 1/2 Hoagland solution. Fresh weight of seedling at 30 days after seeding was highest at 25mM of NaCl concentration containing 1/2 Hoagland solution but if the NaCl concentration was more than 50mM it began to decrease seriously. Water content in plant was decreased according to increase of NaCl concentration in 1/2 Hoagland solution, so physiological mechanism of NaCl in barley was different from saline plant. Stoma number per cm$^2$ of first leaf was higher than that of control in case of stressed by NaCl but in that case the leaf length was decreased so the number of stoma per first leaf was slightly decreased. Chloroplast shape was not changed by 75mM of high NaCl contained 1/2 Hoagland solution but cell division at root growing point was inhibited by 75mM of NaCl. As the result of salt stress mitochondria was ruined in structure and irregular solid was found to be transfered from the cytoplasm to the cell wall in root growing point.

Effects of Sprouting Degree and Threshing Methods on Germination and Seedling Vigor in Barley (보리의 수발아정도 및 탈곡방법이 종자활력 관련 형질에 미치는 영향)

  • In order for investigating the factors related to decrease of seedling vigor in the naked barley lines, two naked lines and a covered variety were sprouted with 20 mm rainfall in the natural condition, dried and the sprouted seeds were threshed by hand and small machine thresher, Chiyota. In the case of hand-threshing of the sprouted seeds, the germination rate index (GRI), germination rate, and TTC test value of the different varieties were not different due to the different sprouting degrees, but emergence rate index (ERI) became decreased with stronger sprouting damage. In comparison with hand-threshing, the embryos of sprouted seeds from machine threshing with 700 rpm were partially damaged, and the traits related to seedling vigor were decreased highly significantly. However, the diastatic activity was sharply increased by stronger sprouting and machine threshing. The sprouting degree in the hand-threshing plot was significantly correlated with the diastatic activity(r=0.931**, 0.951**) and GRI(r=0.461), respectively.

Barley Varietal Differences in Germination and Emergence Capacity under Different Water and Salt Stress Conditions (수분 및 Salt Stress조건하에서 대맥품종의 발아력과 출현력의 차이)

  • Germination and seedling emergence of 40 barley cultivars and lines having been bred in Korea were studied under different water and salt stress conditions containing different sucrose and KC1 concentration in laboratory at Suweon. This study revealed that the barley cultivars and lines differed markedly in their emergence rate index(ERI), total stand(TS), coleoptile length, seedling height, top and root weight. Those characters of the most cultivars were progressively reduced as water and salt stress increased. Jogangbori, Dongbori 1, SB 77460 and SB 77415 were less sensitive to both stress and will be useful for breeding sources. ERI was highly correlated with TS, coleoptile length and seedling height. ERI and TS between two different solutions were correlated each other, and this study indicated that for screening the varieties or lines tolerant to water and salt stress, both ways were available and should be screened genetic materials with a good germination and emergence rate in I stress conditions.

  • To establish a lawn Zoysia japonica Steud with seeds a win of experiments were conducted for the investigation of seedling root system. The results m summarized m follows; Zoysia japonica and maize elongated mesocotyle in germinating stage. but rye and barley did not. The mesocotyle of Z. japonica seed pushed the elongating coleoptile up throngh the soil, hence could emerge from more deeply planted. The crown roots of Z japonica originated from the coleoptile node. The crown roots of barley originated from the first foliage led node.

A Simple Method for Assessing Severity of Common Root Rot on Barley

  • Common root rot caused by Cochliobolus sativus is a serious disease of barley. A simple and reliable method for assessing this disease would enhance our capacity in identifying resistance sources and developing resistant barley cultivars. In searching for such a method, a conidial suspension of C. sativus was dropped onto sterilized elongated subcrown internodes and incubated in sandwich filter paper using polyethylene transparent envelopes. Initial disease symptoms were easily detected after 48h of inoculation. Highly significant correlation coefficients were found in each experiment (A, B and C) between sandwich filter paper and seedling assays, indicating that this testing procedure was reliable. The method presented facilitates a rapid pre-selection under uniform conditions which is of importance from a breeder's point of view.

Weed Control by Suppression Ability of Barley as Cover Crop in Dry Direct Seeded Rice Fields (보리를 이용한 벼 건답직파논에서의 잡초억제 효과)

  • This environment-friendly study was conducted to investigate the effects of winter barley as cover crop on the suppression of weeds and on the growth of dry direct seeded rice. The present study clarified the effects of winter barley as a cover crop on the weed vegetation, expressed as a multiplied ratio. In cultivation quality of rice, seedling stand was ranged from 104~112 $m^{-2}$. And, seeding stand of barley was the highest seedling stand 634 unit per $m^2$ in April 10 as compared to the other seeding dates. Furthermore, the dry weight of barley seeded in April 10 was heaviest at 146g $m^{-2}$. Rice yield was in 401kg $10a^{-1}$ in the April 10, and 517kg $10a^{-1}$ in the April 30, because of increased weeds caused by a low weed control. Effects of barley cover crop at same seeding with rice was ranged from 77~87% of control. The economic effect of barley as cover crop was analyzed to be increased by low herbicide using, despite of the slight decrease in rice yield during the period.

Physiological Response of Winter Barley to Salt Stress at Seedling Stage (보리 유묘기의 염처리가 생리적 반응에 미치는 영향)

  • Some physiological characteristics and cultivar differences of winter barley to salt stress were studied during seedling stage. Salt stress was caused by adding NaCl solution to the pot culture soil. Measurements of the responses to salt stress and of the responses after relief from stress were done in terms of leaf water potential, chlorophyll and free proline contents, seedling height and seedling dry weight, and survival rate of leaves. Under salt stress ($\Psi_{\pi}$ =-20bar) seedling height and seedling weight were decreased by 2~22% and by 25~39% respectively, showing some differences among cultivars. Chlorophyll contents was decreased by 33~49%, and free proline content was remarkably increased from control 0.2~0.3mg to salt stress 9.6~14.7mg. The leaf water potential of seedling grown under salt stress with NaCl solution($\Psi_{\pi}$ =-10 or -20bar) was decreased from control -3.3bar to salt stress -9.0bar or -16.2bar respectively but there were no large differences among cultivars with time after relief from salt stress. Leaf survival rate was high in order of Baegdong, Milyang12, Olbori, Durubori and Hyangmaeg, and decrease rate of seedling dry weight was low in the order of Baegdong, Olbori, Hyangmaeg, Milyang12, Durubori. The increase in free proline contents was high in the order of Milyang12, Hyangmaeg, Baegdong, Durubori and Olbori.

Effect of NaCl on Nitrogen Content of Barley Seedlings

  • This study was conducted to determine the effects of NaCl stress on nitrogen, ${NH_4}^+$, and ${NO_3}^-$ content of 4 barley cultivar seedlings that were cultured for 10 and 30 days with different NaCl levels (0, 50, 100, and 150 mM) contain-ing 1/4 Hoagland solutions. The sodium ion content in the shoot of barley seedlings sharply increased with an increase of NaCl concentration. After 30 days of NaCl treatment, the sodium content of the shoot at 150 mM NaCl was 27 times higher than in non-saline conditions. The sodium content in the root linearly increased with increasing NaCl concentration. Nitrogen content in the shoot linearly increased with increasing NaCl concentration, but nitrogen content in the root declined above the point where the $Na^+$ content was 3.0 mM/g $Na^+$ in the barley seedling. ${NO_3}^-$ content also decreased with increasing NaCl concentration. ${NH_4}^+$ content in the shoot decreased with NaCl condition, but its content in the root increased with NaCl condition. A positive correlation between ${NO_3}^-$ and ${NH_4}^+$ content was found in the shoot, but their relationship was negative in the root.
