  • Effects of dimethipin on ${\alpha}$-amylase activity of barley seeds were investigated. In the treatments of $1{\mu}M\;and\;10{\mu}M$ dimethipin, the indexes of germination were reduced to 17% and 24 % respectively. After seed germination, dimethipin was added to germinated seedlings and then the seedlings were kept to measure seedling length under illumination for 7 days. In control, the length of seedling was 5.7 cm, but in the treatments of $1{\mu}M$ dimethipin and $10{\mu}M$ dimethipin, seedling lengths were 5.5 cm and 1.2 cm respectively. In the relationship between dimethipin concentrations and ${\alpha}$-amylase activities, there was a linear curve. The more dimethipin was added to the seeds, the more ${\alpha}$-amylase activities were inhibited. In the treatments of $1{\mu}M$ dimethipin and $10{\mu}M$ dimethipin, ${\alpha}$-amylase activities were reduced to 33% and 71% respectively. Dimethipin also inhibited ${\alpha}$-amylase activities increased by gibberellin and the content of soluble protein. Therefore, it could be suggested that dimethipin might inhibit directly the activities of hydrolysis enzymes including ${\alpha}$-amylase or the expression of ${\alpha}$-amylase genes as germination and seedling growth were severely disturbed.

  • This study was conducted to develop a easy seedling method to test large amounts of barley and wheat cultivars for resistance of powdery mildew. In addition, we also examined the resistance of genetic resources that have been used in barley and wheat breeding programs in Korea. This seedling test used paper bag with seeds can be completed within three weeks. to-day seedlings were available to inoculation and 8 days was needed for result reading. This method can test at least 180 collections at one time. we can identify the two resistant types by leave symptoms showed non infection and necrotic spots. Among the 79 Korean barley cultivars, only two cultivar, 'Sangrokbori' and 'Dajinbori' were resistant and 'Jejubori' showed moderate resistant. There was no resistant in hulless barley and wheat cultivars. It was same results in comparison of earlier resistant reports in field test. We confirm that this method could using in test of powdery mildew resistance in barley and wheat. Among the 1,401 genetic resources using in Korean breeding program, malting barley has more resistant collections comparing to 796 hulled and hulless barley and 273 wheat germplasms.

  • To investigate the effect of seeding date, seeding rate, and sprouted seeds on seedling establishment, yield and its components of rice in barley /rice reley cropping as a minimum tillage, Gancheokbyeo rice seeds sowed by hand broadcast in various overlapped days befer barley harvest. The highest rice seedling stand was at sowed on the day just before barley harvest and l00kg per hectare of seeding rate with sprouted seeds. Also number of tiller and panicle showed the similar tendency. Delayed rice heading dates were found 2 days at l0-day overlap, 2 days at 5-day overlap, and 4 days at 0-day overlap before barley harvest compared with machine transplanting rice at just after barley harvest using l0-days old seedling. The culm length of broadcast rice shortened 2∼4cm compared with the transplanting rice. The rice plants lodged slightly at seeding rate l00kg per hectare and at sowed on the day of barley harvest. The rice panicle number increased with later date sowing and higher seeding rate. Number of spikelets per panicle and ripened grains decreased at higher seeding rates. Sprouted seeds and higher seeding rates showed higher rice yield at later sowing dates.

  • This study was conducted to investigate the variation of chemical components in stagnant water under different mulching amount of barley straw after combine harvesting and the effect of stagnent water on the germinatidn and seedling elongation at early growth stage in rice. pH was lower in barley straw mulching than non-mulching and decreased with increasing of mulched barley straw but was not different among treatments in stagnant water at 15 days after flooding. Dissolved oxygen decreased with the increasing of mulched barley straw and increased with the passing of time, while saline content increased with the increasing of mulched barley straw, showing significant difference among treatments, was peaked at 15 days after flooding and thereafter, decreased. NH4-N was higher 5 days after flooding with thicker barley straw mulching but was not different at 10 days. N $O_3$-N increased up to 10 to 15 days after flooding and increased with thicker barley straw mulching. Water soluble phosphorus and cation content ( $K^{+}$, $Ca^{2+}$, $Mg^{2+}$)were higher in thicker barley straw mulching and peaked at 10 and 15 days after flooding. Germination rate was decreased by thicker straw mulching and was high in order of medium late, medium and early maturing rice variety. Keumobyeo, Gancheokbyeo, Ilpumbyeo, Dongjinbyeo and Gyehwabyeo showed higher germination rate. Seedling elongation at early growth stage was poorer with increasing of barley. straw mulching. Ilpumbyeo had good seedling elongation in one time amount of barley straw mulching and Shinunbongbyeo, Unjangbyeo, Nonganbyeo and Dongjinbyeo had poorer, in two times.mes.

  • This study was carried out to investigate the effects of induced wet-injury conditions on barley seedling growth. Barley seedlings at the three leaf stage were grown in culture solutions with various combinations of dissolved oxygen (DO), nutrients and citric acid (CA) concentrations. Seedling growth was reduced by hypoxia. Root length and root fresh weight were most severely reduced under 1 ppm DO. Shoot growth was slightly reduced but not root growth under low nutrient condition. Seedling growth was reduced by CA in a concentration-dependant manner, Response of seedling growth was efficiently differentiated in a solution containing 1 ppm DO, one tenth of the normal P, K and Mg concentration and 3 mM CA. Under this induced wet-injury condition, overall seedling growth of 6 cultivars was reduced by 11 to 19% compared to the control condition. The results suggest that the induced wet-injury condition can be used for the screening of resistant genotypes at the seedling stage.

  • Barley, a monocotyledonous plant, is relatively recalcitrant to the process of Agrobacterium-mediated genetic transformation. In this study, seedlings of six barley cultivars (Keunal-1-Ho, Saessal, Ol, Saechalssal, Seodunchal and Pungsanchalssal) were injured using alkali, oxidizing or reducing agents. They were then transformed using Agrobacterium via vacuum infiltration for the analysis of comparative GUS gene expression. It was determined that chemical injuries causing a slight growth retardation could overall enhance the GUS transformation rate. Hydrogen peroxide was determined to be the most effective.

  • The production of 8-ketotrichothecenes, deoxynivalenol (DON), nivalenol (NIV), and their monoacetyl derivatives was studied in rice and corn cultures using 8 isolates of Fusarium graminearum which were obtained from corn and barley samples. Higher concentrations of trichothecenes were produced on rice than corn, and production of the toxins on rice was enhanced by growing the fungi at $25^{\circ}C$. The isolates were used for evaluation of toxin production and pathogenicity after artificial inoculation to 5 corn and 3 barley cultivars. The kinds and the relative amounts of trichothecenes produced in cultures were consistent with those in infected kernels of corn and barley with some exceptions. As for DON chemotypes, the ratios of 15-acetyl-DON to 3-acetyl-DON were varied among the pathogen-cultivar interactions. The corn and barley cultivars showed the significant differences of resistance to the Fusarium isolates in disease severity and seedling blight, and resistance ranking to the different isolates was varied. However, significant correlations were observed between the total concentrations of trichothecenes in infected kernels of corn and barley and pathogenicities of the Fusarium isolates to the hosts.

  • The seedling of barley and weed would be generally inhibited by rice straw residues in the field. This experiment was carried out to investigate the allelopathic effects of rice straw and environmental conditions of germination such as osmotic potential, pH on the seedling of barley and water foxtail. The pH did not affect the germination of barley and water foxtail. Osmotic potential to -2.9bar, -0.6bar did not affect the germination of barley and water foxtail, respectively. Rice straw mixed with soil at the concentration of 4%(w/w) reduced the dry weight of shoot and root, and volume of barley root, as compared to the untreated control. Dry weight of water foxtail by rice straw mulching was decreased by 87.1%, 86.2%, respectively.

  • Effects of seed disinfectants on loose smut (Ustilago nuda) and leaf stripe (Pyrenophora graminea) of barley were investigated in field tests. For these experiments, seed samples carrying natural infection of Ustilago nuda and Pyrenophora graminea were used and the following fungicides were used: Baytan, Baytan U, Benlate T, Busan 30, KAC-7703, P 242, Panoctine, Sisthane Ec, Sisthane Wp, Sisthane Ds, Sodium Omadine, Terracoat Zn, Vitathiram and Zinc Omadine, respectively. Results have shown that Sisthane and Benlate T have equal effect to Vitathiram against Ustilago nuda and Pyrenophora graminea. Baytan U was effective against loose smut but inferior to leaf stripe of barley. P 242 was effective against leaf stripe but inferior to loose smut of barley. Busan 30 have shown moderately inferior effect to Vitathiram against loose smut and leaf stripe of barley. A mild inhibition of seed germination and seedling growth of barley, naked barley and wheat seeds was observed when high concentration of Sisthane and Baytan U were treated in seedling box placed in green house conditions, but no symptom observed in field conditions.
