돼지도축장의 생물학적 위해요소에 대한 중요관리점 설정 (A study on establishment of bio-hazard analysis and critical control point for pork slaughter house)

  • To ensure the safety of domestic livestock products, the government made it obligatory to enforce the hazard analysis critical control point(HACCP) in all domestic slaughterhouses. Under the HACCP, most of the hazards generated in slaughterhouses are bio-hazards, especially pathogenic bacteria. In order to reduce to the pathogenic bacteria, critical control point (CCP) is established and controled in the process of slaughter. A study was carried out to measure the level of bacterial contamination of swine carcass in 6 slaughterhouses selected. As a result, the aerobic plate counts(cfu/$\textrm{cm}^2$) of all samples was $10^2$-10 in average, except slaughterhouse C. The level of the aerobic plate counts on the surface of lower loin in slaughterhouse C was $10^4$ and it was considered that slaughterhouse C should set the process of manual transport of carcass as the CCP. Escherichia coli level was the highest in middle line cut surface. Especially, E coli level of slaughterhouses C and D were about 6.5- and 3.0-fold higher than that of other 4 slaughterhouses. Thus, it was considered that the slaughterhouses C and D should set the process of the entrails treatment as the CCP. The air contamination was measured at two point in a slaughterhouse. The air contamination level was 4-13 times higher than that of the standard Japanese slaughterhouses.

Characteristics of low-nitrite pork emulsified-sausages with paprika oleoresin solution during refrigerated storage

  • The objective of this study was to evaluate quality characteristics of low-nitrite emulsified-sausages (ESs, < 75 ppm) containing paprika oleoresin solution (POS) for replacing sodium nitrite (NaNO2). Pork ESs were prepared with four treatments (reference (REF), 150 ppm NaNO2; TRT1, 0 ppm NaNO2 + 0.1% POS; TRT2, 37.5 ppm NaNO2 + 0.1% POS; and TRT3, 75 ppm NaNO2 + 0.1% POS). The physicochemical and texture properties, microbial counts, residual nitrite and thiobarbituric acid reactant substances (TBARS) were measured during refrigerated storage of 35 days. Although TRT2 and TRT3 had lower levels of NaNO2, they had higher redness and yellowness than REF (p < 0.05). Microbial counts of total bacterial counts and Enterobacteriaceae of TRT2 and TRT3 were similar to those of REF (p > 0.05). Expressible moisture percentages (EM, %) of TRT2 and TRT3 were lower than those of REF (p < 0.05). TBARS values of TRT2 and TRT3 were not different from those of REF (p > 0.05). Among treatments, TRT1 had the highest TBARS values (p < 0.05). In conclusion, 0.1% POS in combination with 37.5 ppm NaNO2 would have quality characteristics similar to those of REF. Therefore, approximately 3/4 of the initial nitrite level could be replaced with 0.1% POS, and eventually developed healthier pork products.

낙동강 상·중·하 수역에서의 질화세균군의 변화 (Changes of Nitrifying Bacteria in the Different Zone (Upper·Mid·Lower Part) of the Nak-Dong River)

  • Nitrifying bacteria were detected by fluorescent in situ hybridization (FISH) method at 6 sampling sites with different eutrophication degree in the Nak-Dong River and their tributaries. And conventional physico-chemical parameters including $NH_4-N$, $NO_3-N$, and TN were determined concurrently. In rainy period (July), there was no noticeable difference between the number of ammonia/nitrite-oxidizing bacteria detected at each site except Sang-Ju and the ratio of nitrifying bacteria to total counts stained by DAPI varied in 6~33%. By contrast, in the dry period (October), both of bacterial population was increased differently and the ratio of nitrifying bacteria to total counts ranged more widely from 6% in heavily polluted water zone, Hwa-Won to 60% in upper tributary with high agricultural land use. Byung-Sung-Chun. In January, the numbers of ammonia-oxidizing bacteria was reduced up to one tenth, while those of nitrite-oxidizing bacteria was apparently increased maybe due to high DO and low DOC.

Microscopic Detection of Urinary Tract Infection in Nepalese Patients

  • Urinary tract infection (UTI) is one of the most common domiciliary and nosocomial bacterial infections prevalent in both males and females. UTI is diagnosed on the basis of clinical symptoms, microscopy and culture of urine. In order to evaluate the efficacy of microscopic detection for presumptive diagnosis of UTI we analyzed urine samples of Nepalese patients. We have conducted Gram staining and counting of pus cells, red blood cells (RBC) and epithelial cells. We observed that RBC and epithelial cell counts were not sensitive enough to be used for presumptive diagnosis of UTI. However, pus cell counts as well as Gram stain are sensitive and significant enough to presume UTI. When the Gram stain result was compared with the culture result, it was statistically significant. From this, we suggest that Gram stain of centrifuged urine is a very sensitive screening method to detect bacteriuria. In addition, we found that E. coli was the most predominant microorganism causing UTI and nitrofurantoin was the most effective antibiotic against the isolated urinary pathogens.

알지네이트 인상체에 대한 상업성 소독제의 효과 (The effect of disinfectants for Alginate impression)

  • To evaluate the effect of disinfectants for Alginate impression, several commercially available disinfectants were tested to Alginate model contaminated by Methicillin resistant Staphylococcus aureus. Alginate specimens were artificially contaminated by Staphylococcus aureus and soaking in four commercially available disinfectants for 1, 5, 10 minutes and colonies were counted for each case. Significant reduction of bacterial colony counts was noted after soaking in 0.5% and 0.05% sodium hypochlorite and after soaking in 2% glutaraldehyde, but not after soaking in Hypochlorus add. Commercially available sodium hypochlorite and glutaraldehyde were found to be effective disinfectants for Alginate impression. But hypochlorus add was not adequate disinfectant.

Efficacy of apitoxin for the treatment of otitis externa in dogs

  • This study examined the efficacy of apitoxin for the treatment of otitis externa in dogs. Ten dogs with otitis externa were allocated randomly to two groups. The control group was treated with the susceptible antibiotics and the experimental group was injected with apitoxin into the tragus subcutaneously. There were no significant differences in the clinical scores, blood WBC counts and neutrophil/lymphocyte ratios between the control and experimental groups. By 2 weeks, the bacterial cell counts were significantly lower in the experimental group than the control (p < 0.05). No adverse reactions were observed in any of the dogs during the study. This suggests that a topical injection of apitoxin is an effective treatment for otitis externa in dogs.

부산시 일부 지하상가 공기중 부유 균주 (Airborne Microorganisms of Some Underground Shops in Pusan)

  • This study was investigated to evaluate the distribution of airborne microorganisms at underground shops in Seomyun and Nampodong, Pusan. The number of bacterial colonies on the nutrient agar plate plates were calculated by the open petri dish method for 30 minutes in indoor air of underground shops at every seasons in a year. There was no statistically significant difference between Seomyun and Nampodong in mean colony counts. Isolation rates of Gram positive rods was highest, and Gram positive cocci and Gram negative rods were followed. In Nampodong underground shops, Enterobacteriaceae strains was isolated. Mean colony counts according to seasons was higher at summer and autumn in Seomyun, and spring and winter in Nampodong. In near future, a study on the distribution of bacteria causing respiratory infection should be followed.

레토르트 및 감마선 조사에 의한 화닭 덮밥 소스의 미생물 제어 효과 비교 (Antimicrobial Effects of Retort and Gamma Irradiation on Bacterial Populations in Spicy Chicken Sauce)

  • 극한 환경에서 취식이 가능하도록 안전성 및 기호성이 확보된 덮밥 소스류 개발을 위한 기초연구로 많이 소비되고 있는 화닭 덮밥소스의 위생화를 위해 감마선 조사 기술을 적용하고 레토르트 살균과의 비교연구를 진행하였다. 화닭 덮밥 소스의 미생물 오염도 평가 결과 레토르트 비처리군은 총세균수가 4.5 log CFU/g 검출되었고 레토르트 처리군은 2.1 log CFU/g의 총 세균이 검출되었다. 반면 3 kGy의 감마선 처리군 에서는 검출한계 이하의 세균이 검출되었다. 검출된 미생물을 동정한 결과 Bacillus spp.로 확인되었으며, B. cereus 및 B. subtilis에 대한 방사선 감수성 측정 결과 각각 0.39 및 0.28 kGy로 확인되었다. SOS chromotest를 이용한 유전독성학적 안전성 평가 결과 감마선 조사에 의한 돌연변이원성은 관찰되지 않았다. 이상의 결과를 종합하여 볼 때, 화닭 덮밥 소스의 효과적인 위생화를 위하여 3 kGy의 감마선 조사처리가 레토르트 처리보다 효과적으로 미생물을 사멸하는 것으로 확인되었다.

돼지 도축폐수 처리를 위한 RABC 공정의 생물막 세균군집 구조 (The Bacterial Community Structure in Biofilms of the RABC Process for Swine Butchery Wastewater Treatment)

  • 돼지 도축폐수를 처리하는 Rotating Activated Bacillus Contactors (RABC) 공정의 세균군집 특성을 파악하기 위하여 그람양성세균수와 총세균수를 계수하여 기존 고도폐수처리공정인 A2O (Anaerobic-Anoxic-Oxic) 공정과 비교하였다. RABC 공정의 생물막 세균군집구조는 비배양기법인 16S rDNA 염기서열결정법을 이용하여 분석하였다. RABC 공정의 총세균수에 대한 그람양성세균수 비율은 생물막(32%)에 비해 최종포기조(1282%) 및 반송슬러지(958%)에서 현저히 증가한 반면, A2O 공정의 그람양성세균수 비율은 호기조(40%)와 반송슬러지(49%) 모두에서 상대적으로 훨씬 낮았다. 총 9개 문에 해당하는 92개의 클론이 검출되었으며, 이 가운데 최우점 집단은 Proteobacteria (64.1%)와 Actinobacteria (18.4)%로서 이들 2개 문이 전체의 82.5%를 차지하였다. 3번째로 많이 검출된 것은 내생포자형성세균집단이 속하는 Firmicutes (5.4%) 문이었다. 소량 검출된 나머지 6개 문은 Bacteroidetes (3.3%), Chlorobi (2.2%), Nitrospirae (1.1%), Chlorofleix (1.1%), Acidobacteria (1.1%), Fusobacteria (1.1%)의 순이었다. Proteobacteria 문 중에서는 Betaproteobacteria 강 34.8%, Alphaproteobacteria 강 26.1%로서 대부분을 차지하였고, Gammaproteobacteria 강은 3.2%이었다. 내생포자형성세균집단의 비율은 모두 19.4%로서, Firmicutes 문 5.4%와 Actinobacteria 문의 Intrasporangiaceae과 14.0%이었다. 질화세균 및 탈질세균과 관련된 클론 비율 6.5%, 인축적세균과 관련된 클론 비율 5.4%를 기록함으로써 무기영양소 및 악취 제거능력을 가진 세균집단이 RABC 생물막에 많이 서식하고 있음이 확인되었다.

명태 (Gadus chalcogrammus)의 성장 단계별 질병 모니터링 (Disease monitoring of Alaska pollock (Gadus chalcogrammus) based on growth stages)

  • 본 연구에서는 명태 양식 기술 개발 과정 중 수산생물 병원체의 영향을 알아보고자 사육수 내 총 세균수 변화, 분포와 명태에 대한 영향을 분석하였으며, 성장 단계별 명태의 병원체(세균, 기생충 및 바이러스) 조사 및 비정상 개체 발생에 대한 원인 구명을 수행하였다. 자어 단계에서 사육수의 평균 세균수는 $2.11{\times}10^3cfu/mL$로 나타났으며, 비브리오균이 52%로 가장 높게 분포하였다. 치어 및 미성어 단계에서 사육수의 세균수는 $3.43{\times}10^3cfu/mL$(슈도모나스균 90% 분포) 그리고 성어 단계에서는 $3.2{\times}10^2cfu/mL$(슈도모나스 52%)로 나타났다. 명태 사육수 세균총의 변화는 먹이생물의 종류, 공급 및 잔류량에 의해 영향을 받는 것으로 판단된다. 외관상 건강한 개체에서는 대량 폐사를 일으킬 수 있는 세균, 바이러스가 검출되지 않았으나, 일부 안구돌출, 턱 주변 조직 출혈을 보이는 비정상 개체에서 아가미 새변의 기포 형성, 각막 조직 및 맥락막 주변 조직 괴사가 관찰되었다. 특히, 병변 부위에서 검출된 세균(P. fluorescens, Vibrio sp., V. pelagius, Planococcus maritumus 그리고 Pseudoalteromonas sp.)이 사육수에도 동일하게 분포하였으며, 사육 과정 중 외상 발생 이후 2차 세균 감염이 발생할 수 있음을 시사한다. 명태의 안정적인 양식 산업화를 위한 요소로 기초 질병 데이터 구축과 더불어 지속적인 질병관리가 필요하다.
