병원감염 예방을 위한 Film Cassette의 자외선 소독 효과 (Disinfection Efficacy of an Ultraviolet Light on Film Cassettes for Preventive of the Nosocomial Infection)

  • The bacteria infection on film cassette contact surface was examined at the diagnostic radiology department of the S. hospital. The objective of this study was to assess the contamination level on film cassette contact surface as a predictor of patient prevention from nosocomial infection and for improvement of the hospital environment. The laboratory result was identified non-pathologic bacterial in the five different cassette size of the contact surface. Film cassettes were exposed to ultraviolet light for 1, 2 and 3 minutes. Ultraviolet light disinfection is proven suitable for bacteria. The study concludes that presence of a bacterial infection will prevent a using antiseptic technique on film cassette contact surface. In addition education of nosocomial infection for radiographers will be required. In conclusion, ultraviolet is considered effective to irradate bacteria. Additionally, two minutes are required to sterilize film cassettes.

초유은행에서 수거한 초유의 병원체 오염과 면역수준의 평가 (Assessment of Immune Quality and Pathogen Contamination of Colostrums Collected from Colostrum Banks in Korea)

  • 송아지는 초유를 통해서만 모체항체를 받을 수 있기 때문에 신생우가 분만초기에 초유를 섭취하는 것은 질병에 대한 저항성을 갖추기 위해 필수적이다. 따라서 분만 후 잉여 초유를 저장하였다가 초유섭취가 부족한 신생우에 공급하여 충분한 항체를 갖도록 하는 것이 일반적인 사양법이다. 본 연구에서는 국내 3곳의 지역에서 운영되는 초유은행에서 저장된 35개의 초유를 무작위로 수거하여 일반세균오염과 주요 병원체들을 검사하였고 면역글로불린의 농도와 BVDV 특이 항체가를 측정하여 초유의 면역학적 품질을 평가하였다. 초유은행에서 수거된 대부분의 초유에서 중등도에서 고도의 세균오염이 관찰되었다. 또한 대부분의 초유가 좋은 품질의 초유라고 판단되는 50 g/L의 IgG 농도에 미치지 못하는 IgG를 포함하고 있었다. 따라서 초유은행 보관용 초유는 청결한 채취법에 따라 분만 후 2-3회 이하로 착유한 초유를 사용하여 안전성과 품질을 보장하여야 할 것이다. 또한 $63^{\circ}C$에서 30분간 실시하는 저온살균이 면역글로불린의 파괴를 최소화하는 동시에 초유에 의한 질병전파를 확연히 줄여주는 것으로 관찰되었으므로 냉동보관 전에 초유의 저온살균 과정을 반드시 실시하는 것이 필요할 것으로 판단된다.

소고기의 유통 단계별 병원성 미생물 오염도에 관한 연구 (A study on the contamination level of pathogenic microorganisms in beef distribution stages)

  • Contamination levels of pathogenic microorganisms in 145 cases of beef, which were distributed in Gwangju province, had been investigated in each distributed stage and also monitored by general bacterial count and E coli count index. General bacterial count of beef from the slaughterhouse was 10$^4$cfu/g less than the level of promotion(10 cfu/$\textrm{cm}^2$) and E coli count index was also under the level of 10$^2$cfu/$\textrm{cm}^2$ recommended level of the ministry of agriculture and forestry. Pathogenic microorganisms were detected from 23.2% of samples in the consumption stage, 12.5% in the slaughtering stage and 5.6% in the transporting and processing stage. Staphylococcus aureus was isolated in the largest number and its ratio was 9.0%, listeria monocytogenes 5.5% and salmonella spp 1.4%. There were no samples that bacteria had been detected dually. E coli O157:H7 and campylobacter jejuni were not isolated. In raw and chilled beef, isolation rate of pathogenic microorganisms were 13.3% and 16.5% each. Especially in raw beef, L monocytogenes was. isolated in 3 samples among 30 cases (10%) and S aureus in one sample (3.3%). According to a scale of meat store, isolation rates of pathogenic microorganisms were different. It was 28.6% in the small-scale meat store and 16.7% in the large-scale meat store each. Four cases (16.7%) of S aureus were isolated in the large-scale meat store and seven cases (20.0%) of L monocytogenes and 2 cases (5.7%) of salmonella spp were isolated in the small-scale meat store. S aureus was isolated in two places among 10 feeding facilities of the elementary school. This result shows that the sanitation of elementary school feeding facilities is so poor and more careful policy consideration is needed. Eleven strains of S aureus isolated showed ${\beta}$-hemolysis on blood agar, 1 strain ${\alpha}$-hemolysis, and 1 strain ${\gamma}$-hemolysis. Isolated strains of L monocytogenes were reconfirmed in 560 bp by PCR. Conclusively, these results show that the sanitary condition in the stages of slaughtering, transportation-processing and consumption influences the degree of pathogenic microorganisms contamination in beef severely It is necessary to apply thoroughly hazard analysis critical control point in a process of beef distribution and also to develop rapid test methods for microorganism diagnosis. This effort is very important for the supply of safe and clean meat from farm to table and helpful for the improvement of public health.

치과용 유니트 수관 물 빼기(Flushing)가 세균 오염도 감소에 미치는 영향 (Effects of Flushing in Dental Unit Waterlines on the Bacterial Contamination Level)

  • DCU의 물을 관리하는 다양한 방법들 중 치과용 유니트를 사용하기 전 수관 물 빼기 방법이 가장 사용이 쉽고 간단하기 때문에, 미국질병관리본부와 미국치과의사협회에서 환자 진료 전후에 20~30초 동안 물을 미리 배출시킬 것을 권장하고 있다. 하지만, 수관 물 빼기의 권장 시간인 20~30초와 관련한 과학적 근거가 명확하지 않고, 20~30초 이외의 시간 동안 수관 물 빼기 효과를 확인한 연구가 부족한 실정이다. 본 연구의 목적은 권장시간인 20~30초뿐만아니라 이보다 더 짧거나 긴 시간 동안 치과용 유니트 수관 물 빼기를 수행한 후 각 시간별 오염 세균 수의 감소 정도를 조사하여 20~30초 권장시간의 적절성을 확인하고 더 효과적인 수관 물 빼기 시간이 있는지를 조사하는 것이다. 물 시료 수집은 치과대학 학생 실습 시뮬레이션실 내 치과용 유니트를 대상으로 수행하였다. 7개 치과용 유니트의 초음파치석제거기에서 15분 동안 수관 물 빼기를 진행하면서 30초 간격으로 배출되는 물을 수집하였다. 추가적으로 5개의 치과용 유니트에서는 1분 동안 물을 배출시키면서 10초 간격으로 물을 수집하였다. 물 시료 내 오염 세균 수는 R2A agar 배지에 수집된 물을 도말한 후 배양하여 확인하였다. 7개의 치과용 유니트에서 최대 15분 동안 물을 배출시켰을 때 치과용 유니트의 오염 수준이 200 CFU/ml 이하로 감소되지 않았다. 추가적으로 5개의 치과용 유니트에서 1분 동안 10초 간격으로 배출되는 물을 수집하여 세균 수를 확인하였을 때 10초 동안 수관 물 빼기를 한 후 세균 수 감소율은 평균 63%였고, 20초 동안 수관 물 빼기를 한 후 세균 수 감소율은 평균 84%였다. 본 실험의 결론은 다음과 같다: 첫째, 수관 물빼기의 권장시간인 20~30초뿐만아니라 최대 15분 동안 수관 물 빼기를 시행하는 것은 권장수준(${\leq}200CFU/ml$)으로 세균 오염도를 감소시킬 수 없었다. 둘째, 수관 물 빼기의 권장 시간인 20~30초보다 짧게 10초 동안 수관 물 빼기를 하는 것도 세균 수 감소에 효과가 있었다. 셋째, 치과용 유니트 내 물의 세균 오염수준을 권장 수준으로 유지시키기 위해서는 수관 물 빼기와 함께 주기적으로 화학 소독제로 치과용 유니트 수관을 소독하는 것이 필요하다.

핵의학 검사실내 세균 분석 (Analysis of the Bacteria in Nuclear Medicine)

  • In this study, the bacterial contamination level of equipments and devices in the nuclear medicine department of a university hospital was investigated. CNS was detected from the sample collected from the door opening button of the nuclear medicine department. Bacillus sp. was detected from the table and CNS with Bacillus sp. were detected from the control button at the PET-CT room no.1. Also, CNS was detected from the table and the control button at the PET-CT room no.2. In the distribution room no.1, CNS and Bacillus sp. were detected while CNS being detected from the distribution room no.2 and CNS with Bacillus sp. being detected from the distribution room no.3. In the injection room, Enterrococcus faecium and Pontoea sp. were detected. On the table of the ecsomatics room, Pontoea sp. was detected. Bacillus sp. was detected from the inside of the syringe Pb shield and CNS was detected from the outside. Enterrococcus faecium was detected from the Gamma camera table and Bacillus sp. was detected from the door grip. On the chair at the patient waiting room, Pseudomonas aeruginosa abd Bacillus sp. were detected. Therefore, it was understood that infection should be prevented by securely sterilizing examination devices after each examination, maintaining cleanliness by regular sterilization of waiting chairs and such objects with a number of direct contacts with patients, and infection education for the features of nuclear department.

제주 동부 연안 넙치 양식장의 미생물학적 오염도 조사 (Investigation of Microbial Contamination the Level in Fish Farms of Jeju East Coast.)

  • 본 연구는 2007년 6윌부터 10월까지 제주도에서 양식장이 가장 많이 밀집되어 있는 제주도 동부 연안에 위치한 양식장 4개소를 대상으로 하여 사육 용수, 양어 사료, 양식 넙치의 미생물학적 오염도를 조사하였다. 식품병원성 미생물 중 일반세균과 총대장균군 그리고 Escherichia coli O157:H7, Salmonella spp., Vibrio parahaemolyticus, Staphylococcus aureus, Bacillus cereus를 조사 하였다. 이번 연구를 수행하는 동안 동부지역은 집중적인 호우로 인해 전년보다 400 mm 이상의 많은 강우량을 나타내었다. 사육 용수의 검사결과 V. parahaemolyticus, Salmonella spp., S. aureus, B. cereus가 빈번하게 검출되었고, 양어 사료의 경우 Salmonella spp, S. aureus, B. cereus는 각각 $0{\sim}4.3{\times}l0^3$ CFU/ml, $0{\sim}2.7{\times}l0^4$ CFU/ml, $0{\sim}7.6{\times}l0^3$ CFU/ml 검출 되었다. 또한 양식 넙치의 미생물 검사 결과 적은 양이지만 Salmonella spp., S. aureus, B. cereus가 검출되었고, 총대장균군과 E. coli Ol57:H7은 검출되지 않았다. 이번 조사 결과로 보아 미생물학적 위해요소는 양식장의 작업 환경이나 주변 환경 등의 환경적 요인이 크게 작용하는 것으로 사료되며, 양식장의 위생관리에 작업자들은 주의를 기울여야 할 것으로 생각된다. 앞으로 이와 관련하여 수산물 위생에 대한 좀 더 체계적인 시스템을 도입하여 적용한다면 수산물에서 야기되는 문제점들이 해결되어 제주도의 청정 브랜드의 이미지를 더욱 부각시킬 수 있으며, 안전하고 깨끗한 수산물의 생산이 가능할 것으로 생각된다.

양돈장 사료의 곰팡이독소 오염률 조사 (Prevalence of mycotoxin contamination in pig feedstuffs)

  • To analyze prevalence of mycotoxins, a total of 74 feedstuff samples were collected from silos (n=37) and hoppers (n=37) in nine pig farms. Six mycotoxins were tested with commercialized ELISA kits. All samples were contaminated with four or more mycotoxins. Zearalenone was detected in all of the tested samples. Ochratoxin, deoxynivalenol and H-2/HT-2 toxin were detected in more than 90% of the samples. And also, fumonisin was positive in 89.2% of the samples from the silos, 75.2% from the hoppers, respectively. On the other hand, aflatoxin was detected in about 40% of the samples. When the behavior of lactating sows was observed, possible mycotoxicosis was suspected. It was confirmed that their feedstuffs were contaminated with high levels of mycotoxins such as ochratoxin and T-2/HT-2 toxin. After cleaning the feedline, the clinical symptoms in sows suspected with mycotoxicosis were disappeared. Although mycotoxin concentration in most of the feedstuffs was below the acceptance level, these data indicate that what are required is more monitoring and continuous management for mycotoxins in pig feedstuffs.

칫솔모 사이에 형성한 구멍 수에 따른 세균 양 변화 (Comparison of the amount of bacteria according to the number of holes between bristles)

  • The purpose of this study was to determine the level of toothbrush bacterial growth, whether the dryness of the toothbrush head differs depending on the number of holes in the head, and to use these results as a reference for future toothbrush design. Two-millimeter holes were created on the head of the toothbrushes in groups of three, one, or zero holes. We made the solution with Streptococcus mutans, and the toothbrushes were placed in the solution and agitated. The toothbrushes were shaken to remove moisture and allowed to air-dry. The toothbrush heads were swabbed with saline and then placed in two inoculation groups. The first group was inoculated with a 102 dilution of the S. mutans culture and the second was inoculated with the original culture. After incubation, bacterial colony numbers were measured. The number of holes on the toothbrush head correlated with a decrease in number of cultured bacterial colonies. Our model of a toothbrush head with holes indicated that these holes in the toothbrush head were effective in reducing the level of microbial contamination and that a greater number of holes creates an improved toothbrush sanitation effect. The average number of colonies on the head of toothbrush by number of holes was high, followed by the number of holes 0, 1 and 3, and the average number of colony among toothbrush heads was same. The use of a toothbrush with holes between the toothbrush head indicates that it is effective in reducing the level of microbial contamination between the toothbrush head and toothbrush and the higher the number of holes, the better the effect.

학교 집단급식소 내 식기류 및 집기류의 미생물학적 분석 및 위해요인 평가 (A Microbiological Analysis and Hazard Factor Evaluation of Food Utensils and Fixtures of Food Service Operations in Schools)

  • Objectives: The aim of this study was to evaluate the microbial hazards posed by food utensils and fixtures in food service operations at selected middle and high schools located in Seoul, Korea. Methods: We collected 200 samples of utensils and fixtures including cups, spoons/chopsticks, food trays and tables from five different schools in Seoul. Target microorganisms of this study were divided into two groups: total bacterial count and total coliform as indicators of microbial contamination and Bacillus cereus and Staphylococcus aureus as pathogens of food poisoning. We used selective media to quantify microbial concentration and 16S rRNA PCR assay for qualitative analysis. In addition, intensive interviews with nutritionists were conducted and observations were made to identify factors that may affect microbial contamination. Logistic regression analysis was employed to examine the relationship between the microbial concentration and operation characteristics of each operation. Results: The level of microbial concentration in school B and C were significantly lower than in school A, D and E (p<0.05). Some samples from school A, D and E showed over 3.4 log CFU/100 $cm^2$ (total bacterial count) and 1.0 log CFU/100 $cm^2$ (total coliform), which requires immediate hygienic action. The number of customers per staff member, periodicity of hygiene education for staff and daily operation time of sterilizers were also found to be important factors related with the microbial contamination of food service operations. Conclusions: These results suggested that not only a HACCP (Hazard Analysis and Critical Control Point) approach, but also efforts to assess internal risk factors within operations be needed to reduce the microbial contamination of food utensils and fixtures. This study is expected to provide preliminary data for assessing microbial hazards in food service operations.

Degradation of Phenanthrene by Bacterial Strains Isolated from Soil in Oil Refinery Fields in Korea

  • The degradation of phenanthrene, a model PAH compound, by microorganisms either in the mixed culture or individual strain, isolated from oil-contaminated soil in oil refmery vicinity sites, was examined. The effects of pH, temperature, initial concentration of phenanthrene, and the addition of carbon sources on biodegradation potential were also investigated. Results showed that soil samples collected from four oil refinery sites in Korea had different degrees of PAH contamination and different indigenous phenanthrene-degrading microorganisms. The optimal conditions for phenanthrene biodegradation were determined to be 30$^{circ}C$ and pH 7.0. A significantly positive relationship was observed between the microbial growth and the rate of phenanthrene degradation. However, the phenanthrene biodegradation capability of the mixed culture was not related to the degree of PAH contamination in soil. In low phenanthrene concentration, the growth and biodegradation rates of the mixed cultures did not increase over those of the individual strain, especially IC10. High concentration of phenanthrene inhibited the growth of microbial strains and biodegradation of phenanthrene, but was less inhibitory on the mixed culture. Finally, when non-ionic surfactants such as Brij 30 and Brij 35 were present at the level above critical micelle concentrations (CMCs), phenanthrene degradation was completely inhibited and delayed by the addition of Triton X100 and Triton N101.
