살충제(Fenitrothion) 살포가 소나무림의 토양생물에 미치는 영향 (Effects of Pesticide (Fenitrothion) Application on Soil Organisms in Pine Stand)

  • 피해가 급속히 확산중인 소나무재선충병 방제를 위해 항공방제를 실시하는 소나무림이 증가하면서, 항공방제시 농약침투로 인한 토양생태계 교란의 가능성이 제기되고 있다. 본 연구는 이러한 가능성을 평가하기 위해, 현지 외 야외실험을 BACI 실험설계하에 서울 홍릉시험림의 소나무림에서 면적 $25m^2$인 3개의 방제구와 3개의 대조구를 설치하여 실시하였다. 항공방제 약종인 fenitrothion을 항공방제직후 낙엽에서 검출된 잔류량인 0.0335 kg a.i/ha를 지면에 살포하였다. 농약살포는 2002년 6월 4일, 6월 11일, 6월 29일 3회에 걸쳐 실시하였는데, 살포일은 전년도 부산지역 항공방제일과 동일한 날짜로 선정하였다. 절족동물, 곰팡이, 세균, 방선균 등의 토양생물의 개체수 조사를 살포 전에 2회, 살포기간중에 2회, 마지막 살포후 3회 실시하였다. 살충제의 살포는 토양에 서식하는 절족동물, 곰팡이, 세균, 방선균의 개체수 변화와 이들 토양생물군집의 구조에 유의한 영향을 주지 않아. 항공방제가 소나무림의 토양생물에 큰 영향을 줄 가능성이 낮음을 나타내었다.

Quality and Lactic Acid Bacteria Diversity of Pork Salami Containing Kimchi Powder

  • We investigated the quality properties and bacteria diversity of pork salami containing homemade kimchi powder (KP). Pork salamis were manufactured with commercial starter culture (control), and 1% KP (KP1), 3% KP (KP3), and 5% KP (KP5). The salami was fermented and aged for 2 days and 56 days, respectively. The pH and $A_W$ values of salamis with KP were significantly lower than that of the control (p<0.05). The 2-thiobarbituric acid reactive substance values of all salamis with KP increased but were below 0.2 mg MDA/kg. Salamis with KP, decreased the $L^*$ value, but increased the $a^*$ and $b^*$ values (p<0.05). The Lactobacillus count was significantly higher in the salamis with KP than in the control by day 14 (p<0.05). The number of Leuconostoc was higher in salamis with KP than in the control and was the highest in salamis in KP1. The KP1 protected lipid oxidation and showed low TBARS value of pork salami compared to the control. This study demonstrates that KP can improve pork salami properties and can serve as a potential natural compounds for fermented meat prodcuts like Metzgerei.

Bacterial Population in Intestines of Litopenaeus vannamei Fed Different Probiotics or Probiotic SupernatantS

  • The interactions of microbiota in the gut play an important role in promoting or maintaining the health of hosts. In this study, in order to investigate and compare the effects of dietary supplementation with Lactobacillus pentosus HC-2 (HC-2), Enterococcus faecium NRW-2, or the bacteria-free supernatant of a HC-2 culture on the bacterial composition of Litopenaeus vannamei, Illumina sequencing of the V1-V2 region of the 16S rRNA gene was used. The results showed that unique species exclusively existed in specific dietary groups, and the abundance of Actinobacteria was significantly increased in the intestinal bacterial community of shrimp fed with the bacteria-free supernatant of an HC-2 culture compared with the control. In addition, the histology of intestines of the shrimp from the four dietary groups was also described, but no obvious improvements in the intestinal histology were observed. The findings in this work will help to promote the understanding of the roles of intestinal bacteria in shrimps when fed with probiotics or probiotic supernatant.

The impact of diet on the composition and relative abundance of rumen microbes in goat

  • Objective: This experiment was conducted to explore the impact of diet on the ruminal microbial community in goats. Methods: Twelve goats were divided into two groups and fed complete feed (CF) or all forage (AF) diet. The total microbial DNAs in the rumen liquid were extracted. The V4 region of microbial 16S rRNA genes was amplified and sequenced using high-throughput. Information of sequences was mainly analyzed by QIIME 1.8.0. Results: The results showed that Bacteroidetes and Firmicutes were the most predominant microbial phyla in the rumen of all goats. At genus level, the abundance of fiber-digesting bacteria such as Ruminococcus and Lachnospiracea incertae sedis was significantly higher in AF than that in CF, while the levels of fat-degrading bacterium Anaerovibrio and protein-degrading bacterium Pseudomonas were opposite. The core shared genera, Prevotella and Butyrivibrio were widespread in the rumen of goats and no significant difference was observed in relative abundance between groups. Conclusion: We concluded that the richness of fiber-, protein-, and fat-digesting bacteria was affected by diet and tended to increase with the rise of their corresponding substrate contents in the ration; some bacteria shared by all goats maintained stable despite the difference in the ration, and they might be essential in maintaining the normal function of rumen.

  • Because of high population diversity in soil microbial communities, it is difficult to accurately assess the capability of biodegradation of toxicant by microbes in soil and sediment. Identifying biodegradative microorganisms is an important step in designing and analyzing soil bioremediation. To remove non-important noise information, it is necessary to selectively enrich genomes of biodegradative microorganisms fromnon-biodegradative populations. For this purpose, a stable isotope probing (SIP) technique was applied in selectively harvesting the genomes of biphenyl-utilizing bacteria from soil microbial communities. Since many biphenyl-using microorganisms are responsible for aerobic PCB degradation In soil and sediments, biphenyl-utilizing bacteria were chosen as the target organisms. In soil microcosms, 13C-biphenyl was added as a selective carbon source for biphenyl users, According to $13C-CO_2$ analysis by GC-MS, 13C-biphenyl mineralization was detected after a 7-day of incubation. The heavy portion of DNA(13C-DNA) was separated from the light portion of DNA (12C-DNA) using equilibrium density gradient ultracentrifuge. Bacterial community structure in the 13C-DNAsample was analyzed by t-RFLP (terminal restriction fragment length polymorphism) method. The t-RFLP result demonstates that the use of SIP efficiently and selectively enriched the genomes of biphenyl degrading bacteria from non-degradative microbes. Furthermore, the bacterial diversity of biphenyl degrading populations was small enough for environmental genomes tools (metagenomics and DNA microarrays) to be used to detect functional (biphenyl degradation) genes from soil microbial communities, which may provide a significant progress in assessing microbial capability of PCB bioremediation in soil and groundwater.

백두산의 식생에 따른 토양 미생물의 분포 및 특성 (Distribution and Properties of Soil Microorganisms Isolated from Representative Plant Communities of Mt. Paektu)

  • Physicochemical factors, microbial population size and the properties of the bacterial isolates were assessed to find out the nature of soil ecosystem of Mt. Paektu. Samples were obtained from the surface layer of soils on which specific plant community is developed. Average content of moisture, organic matter and avaiable phosphate of the soils were 21.6%, 17.3% and 2.48mg/100g, respectively. These values were similar to those of developing forest soils, but were slightly lower than those of climax ecosystem such as Piagol in Mt. Chiri. The population size of soil bacteria ranged from 2.7 to $202.5{\times}10^5$ CFU/g.dry soil, and the size is somewhat dependent on the content of moisture and oranic matter of the forest soil. A large number of bacteria was able to decompose macromolecules such as starch, elastin and gelatin. While the distribution rate of resistant bacteria to antibiotics was high, that to toxic chemicals was low. This means that the competition between microorgani는 predominate over the interference with artificial behaviour such as spread of pesticides in the surveyed region. Bacterial species composition of each soil was comparatively simple. Pseudomonas, Agrobacterium, Flavobacterium and Xanthomonas which are Gram-negative short rods were widely distributed in the forest soils. The endospore forming Bacillus species were also main constituents of the soil microflroa. any one of the strains was not identified as Azospirillum or Micrococcus which are known to be one of major constituents of the forest soil. for the correct identification of isolates chemotaxonomic studies will be proceeded, and the strains are to be stored in the Type collection Center.

Removal of Organic Load from Olive Washing Water by an Aerated Submerged Biofilter and Profiling of the Bacterial Community Involved in the Process

  • The present work aims to use a biofilter technology(aerated submerged filters) for the aerobic transformation at laboratory-scale of olive washing water(OWW) generated in the first steps of olive oil processing, as well as the genetic profiling and identification to the species level of the bacteria involved in the formation of the biofilm, by means of TGGE. Chemical parameters, such as biological oxygen demand at five days($BOD_5$) and chemical oxygen demand(COD), decreased markedly(up to 90 and 85%, respectively) by the biological treatment, and the efficiency of the process was significantly affected by aeration and inlet flow rates. The total polyphenol content of inlet OWW was only moderately reduced(around 50% decrease of the inlet content) after the biofilter treatment, under the conditions tested. Partial 16S rRNA genes were amplified using total DNA extracted from the biofilm and separated by TGGE. Sequences of isolated bands were mostly affiliated to the $\alpha-subclass$ of Proteobacteria, and often branched in the periphery of bacteria] genera commonly present in soil(Rhizobium, Reichenowia, Agrobacterium, and Sphingomonas). The data obtained by the experimentation at laboratory scale provided results that support the suitability of the submerged filter technology for the treatment of olive washing waters with the purpose of its reutilization.

Changes in Gut Microbial Community of Pig Feces in Response to Different Dietary Animal Protein Media

  • Beef, pork, chicken and milk are considered representative protein sources in the human diet. Since the digestion of protein is important, the role of intestinal microflora is also important. Despite this, the pure effects of meat and milk intake on the microbiome are yet to be fully elucidated. To evaluate the effect of beef, pork, chicken and milk on intestinal microflora, we observed changes in the microbiome in response to different types of dietary animal proteins in vitro. Feces were collected from five 6-week-old pigs. The suspensions were pooled and inoculated into four different media containing beef, pork, chicken, or skim milk powder in distilled water. Changes in microbial communities were analyzed using 16S rRNA sequencing. The feces alone had the highest microbial alpha diversity. Among the treatment groups, beef showed the highest microbial diversity, followed by pork, chicken, and milk. The three dominant phyla were Proteobacteria, Firmicutes, and Bacteroidetes in all the groups. The most abundant genera in beef, pork, and chicken were Rummeliibacillus, Clostridium, and Phascolarctobacterium, whereas milk was enriched with Streptococcus, Lactobacillus, and Enterococcus. Aerobic bacteria decreased while anaerobic and facultative anaerobic bacteria increased in protein-rich nutrients. Functional gene groups were found to be over-represented in protein-rich nutrients. Our results provide baseline information for understanding the roles of dietary animal proteins in reshaping the gut microbiome. Furthermore, growth-promotion by specific species/genus may be used as a cultivation tool for uncultured gut microorganisms.

Antifungal Activity of Lactic Acid Bacteria Strains Isolated from Natural Honey against Pathogenic Candida Species

  • The role of lactic acid bacteria (LAB) in honey as antifungal activity has received little attention and their mechanism of inhibitory of fungi is not fully understood. In this study, LAB were isolated from honey samples from Malaysia, Libya, Saudi Arabia, and Yemen. Twenty-five isolates were confirmed LAB by catalase test and Gram staining, and were screened for antifungal activity. Four LAB showed inhibitory activity against Candida spp. using the dual agar overlay method. And they were identified as Lactobacillus plantarum HS isolated from Al-Seder honey, Lactobacillus curvatus HH isolated from Al-Hanon honey, Pediococcus acidilactici HC isolated from Tualang honey and Pediococcus pentosaceus HM isolated from Al-Maray honey by the 16S rDNA sequence. The growth of Candida glabrata ATCC 2001 was strongly inhibited (>15.0 mm) and (10~15 mm) by the isolates of L. curvatus HH and P. pentosaceus HM, respectively. The antifungal activity of the crude supernatant (cell free supernatant, CFS) was evaluated using well diffusion method. The CFS showed high antifungal activity against Candida spp. especially The CFS of L. curvatus HH was significantly (p < 0.05) inhibited growth of C. glabrata ATCC 2001, C. parapsilosis ATCC 2201, and C. tropicalis ATCC 750 with inhibitory zone 22.0, 15.6, and 14.7 mm, respectively. While CFS of P. pentosaceus HM was significantly (p < 0.05) effective against C. krusei, C. glabrata, and C. albicans with inhibition zone 17.2, 16.0, and 13.3 mm, respectively. The results indicated that LAB isolated from honey produced compounds which can be used to inhibit the growth of the pathogenic Candida species.

체류시간이 서로 다른 부영양 수계에서 플랑크톤군집의 생태학적 특성 (Community Structure of Plankton in Eutrophic Water Systems with Different Residence Time)

  • 체류시간이 서로 다른 두 부영양 수계에서 미소먹이망의 생태학적 정보를 파악하고자, 기초적 환경요인과 bacterioplankton 및 nanoplankton의 현존량 및 탄소생체량을 각각 조사하였다. 조사기간동안 두 수계 공히 강우는 주로 7월에서 9월에 집중하였으며, 수온 역시 큰 차이는 없었다. 두 수계의 플랑크톤 군집은 공히 저온기 동안에는 비교적 세포크기가 큰 nanoplankton이 우점한 반면, 고온기 동안에는 소형 bacterioplankton이 높은 현존량을 나타냈다. 그러나 비교적 수계가 안정된 석촌호수에서는 강우에 큰 영향을 받지 않은 반면, 경안천에서 nanoplankton은 현존량은 물론 탄소생체량의 다양한 변화를 보였다. 두 수계의 미세먹이망은 석촌호수에서는 박테리아와 phytoplankton 또는 cyanobacteria간에 밀접한 분포 관계를 보인 반면, 경안천에서는 박테리아와 protist 사이에 뚜렷한 먹이관계를 보였다. 또한 경안천에서 개체수와 탄소생체량간의 비대칭적인 현상이 뚜렷하였다. 따라서 경안천의 미세먹이망은 수온이나 영양물질보다 강우에 의한 빠른 체류시간에 의해 강하게 영향을 받고 있지만, 안정된 수계에 비해 bacterioplankton보다 nanoplankton들의 높은 성장과 종 다양성을 유도하였다.
