Mapping QTLs for Tissue Culture Response of Mature Wheat Embryos

  • The mature wheat embryo is arguably one of the best explants for genetic transformation because of its unlimited availability and lack of growth season restriction. However, an efficient regeneration system using mature wheat embryos (Triticum aestivum L.) is still not available. To identify genes related to the tissue culture response (TCR) of wheat, QTLs for callus induction from mature embryos and callus regeneration were mapped using an RIL population derived from the cross of 'Wangshuibai' with 'Nanda2419', which has a good TCR. By whole genome scanning we identified five, four and four chromosome regions conditioning, respectively, percent embryos forming a callus (PEFC), percent calli regenerating plantlets (PCRP), and number of plantlets per regenerating callus (NPRC). The major QTLs QPefc.nau-2A and QPcrp.nau-2A were mapped to the long arm of chromosome 2A, explaining up to 22.8% and 17.6% of the respective phenotypic variance. Moreover, two major QTLs for NPRC were detected on chromosomes 2D and 5D; these together explained 51.6% of the phenotypic variance. We found that chromosomes 2A, 2D, 5A, 5B and 5D were associated via different intervals with at least two of the three TCR indexes used. Based on this study and other reports, the TCRs of different explant types of wheat may be under the control of shared or tightly linked genes, while different genes or gene combinations may govern the stages from callus induction to plantlet regeneration. The importance of group 2 and 5 chromosomes in controlling the TCRs of Triticeae crops and the likely conservation of the corresponding genes in cereals are discussed.

Pathogen Associated Molecular Pattern (PAMP)-Triggered Immunity Is Compromised under C-Limited Growth

  • In the interaction between plants and pathogens, carbon (C) resources provide energy and C skeletons to maintain, among many functions, the plant immune system. However, variations in C availability on pathogen associated molecular pattern (PAMP) triggered immunity (PTI) have not been systematically examined. Here, three types of starch mutants with enhanced susceptibility to Pseudomonas syringae pv. tomato DC3000 hrcC were examined for PTI. In a dark period-dependent manner, the mutants showed compromised induction of a PTI marker, and callose accumulation in response to the bacterial PAMP flagellin, flg22. In combination with weakened PTI responses in wild type by inhibition of the TCA cycle, the experiments determined the necessity of C-derived energy in establishing PTI. Global gene expression analyses identified flg22 responsive genes displaying C supply-dependent patterns. Nutrient recycling-related genes were regulated similarly by C-limitation and flg22, indicating re-arrangements of expression programs to redirect resources that establish or strengthen PTI. Ethylene and NAC transcription factors appear to play roles in these processes. Under C-limitation, PTI appears compromised based on suppression of genes required for continued biosynthetic capacity and defenses through flg22. Our results provide a foundation for the intuitive perception of the interplay between plant nutrition status and pathogen defense.

Climate Change in Corn Fields of the Coastal Region of Ecuador

  • The Ecuadorian coast has two different climate regions. One is humid region where the annual rainfall is above 2000 mm and rain falls in almost all months of the year, and the other is dry region where the annual rainfall can fall below 50 mm and rainfall can be very seasonal. The agriculture is frequently limited by the seasons during the year and the availability of rainfall amounts. The corn fields in Ecuador are cultivated during the rainy season, due to this reason. The weather conditions for optimum development of corn growth require a monthly average rainfall of 120 mm to 140 mm and a temperature range of $22^{\circ}C{\sim}32^{\circ}C$ for the dry region, and a monthly average rainfall of 200 mm to 400 mm and a temperature range of $25^{\circ}C{\sim}30^{\circ}C$ for the humid area. The objective of this study is to predict how the weather conditions are going to change in corn fields of the coastal region of Ecuador in the future decades. For this purpose, this study selected six General Circulation Models (GCM) including BCC-CSM1-1, IPSL-CM5A-MR, MIROC5, MIROC-ESM, MIROC-ESM-CHEM, MRIC-CGC3 with different climate scenarios of the RCP 4.5, RCP 6.0, and RCP 8.5, and applied for the period from 2011 to 2100. The climate variables information was obtained from the INAMHI (National Institute of Meteorology and Hydrology) in Ecuador for the a base line period from 1986 to 2012. The results indicates that two regions would experience significant changes in rainfall and temperature compared to the historical data. In the case of temperature, an increment of $1^{\circ}C{\sim}1.2^{\circ}C$ in 2025s, $1.6^{\circ}C{\sim}2.2^{\circ}C$ in 2055s, $2.1^{\circ}C{\sim}3.5^{\circ}C$ in 2085s were obtained from the dry region while less increment were shown from the humid region with having an increment of $1^{\circ}C$ in 2025s, $1.4^{\circ}C{\sim}1.8^{\circ}C$ in 2055s, $1.9^{\circ}C{\sim}3.2^{\circ}C$ in 2085s. Significant changes in rainfall are also projected. The rainfall projections showed an increment of 8%~11% in 2025s, 21%~33% in 2055s, and 34%~70% in 2085s for the dry region, and an increment of 2%~10%, 14%~30% and 23%~57% in 2025s, 2055s and 2085s decade respectively for humid region.

Effect of Slurry Composting and Bio-filtration (SCB) by Fertigation on Soil Chemical Properties and Growth of Red Pepper (Capsicum annuum L.)

  • The slurry composting and bio-filtration (SCB) liquid manure has some obvious advantages including a good source of N, P and K, local availability, effective microorganism and the ability to improve soil properties. This study was conducted to evaluate the influence on the changes of soil chemical properties and yield of red pepper by fertigation cultivation with SCB application for 2 years. Red pepper was transplanted in early May in 2013 and 2014. The treatment with three replication was composed of 4 types as control (N 1.0), SCB 0.5N, SCB 1.0N, and SCB 2.0N standards of recommended nitrogen fertilizer ($19kg\;N\;10a^{-1}$). The fertigation cultivation which was installed the surface drip irrigation system was splitted 10 times as $2.5Mg\;10a^{-1}$ nutritional solution included with chemical fertilizer and SCB every 10 days during the cultivation. The height and width of pepper plant were 7.0% and 5.8% higher in SCB 2.0N treatment than that in control. The yield of red pepper increased with the increasing of SCB application rates from SCB 0.5N to 2.0N. The yield of SCB 1.0N was much better 10% in average than that of control, and there was significant differences among all treatments. pH of control soil after final harvest decreased to 6.1, however pH of SCB treated soils increased from 6.7 to 7.1 depending on SCB application rates. The Exch.-K contents of SCB treated soils were increased 13.7 to 56.9% after final harvest compared with control by $0.51cmol_c\;kg^{-1}$. Accordingly, these results showed that SCB 1.0N application rate as a recommended nitrogen level based on soil testing can be used as an alternative nitrogen management as well as plant nutrition for red pepper cultivation.

Daily Variation of Size-Fractionated Chlorophyll a Concentrations and Water Conditions Associated with Freshwater Discharge during Summer in the Yeongsan River Estuary (영산강 하구의 하계 담수 방류와 연관된 크기별 Chlorophyll a와 수환경의 일간 변동)

  • A sea dike in the Yeongsan River estuary was constructed in 1981 to supply water and reclaim tidal flats for agriculture, separating the estuary into the freshwater and seawater zones. However, the sluice gates are frequently opened and freshwater is discharged in summer when more rainfall is recorded than other seasons, then converting the estuary to brackish water system. In this study, the direct effect of freshwater discharge was investigated by monitoring daily variation in water properties and phytoplankton size structure before and after the freshwater discharge events from 2013 to 2015. Freshwater discharge resulted in a sharp decrease in salinity and dissolved oxygen (DO) at surface water whereas it increased the turbidity of water column. However, salinity did not decrease sharply in 2014 when freshwater was discharged one day before the monitoring and salinity remained low prior to the monitoring. Levels of nutrients especially dissolved inorganic nitrogen (DIN) increased after the discharge and this contributed to potential limitation of nutrients such as P or Si rather than N in the estuary. Freshwater discharge also caused the changes in phytoplankton biomass (chlorophyll a) and size structure although their responses were different between years. The changes may affect growth of grazers and thus structure of marine food web by alternating food availability in the Yeongsan River estuary.

Status Quo Bias in Ocean Marine Insurance and Implications for Korean Trade

  • Purpose - This research uses ocean marine insurance (OMI) statistics, international emails, focus-group interviews, and surveys to fill the gap between the theory of behavioral insurance, particularly status quo bias (SQB), and the practice of OMI in Korea. The contractual forms of OMI, the oldest and most globalized form of commercial insurance, were developed in the UK as the Institute Cargo Clauses in 1906 and revised in 1963, 1982, and 2009. SQB has been academically explored, mostly in health insurance and the financial services sector, but never in OMI. Thanks to the availability of OMI statistics in Korea, we can conduct SQB research here for the first time in this field. Design/methodology - We show the existence of SQB in the OMI of Korea through Korean statistics between 2009 and 2018, email correspondence with experts in the UK, Germany, and Japan, focus-group interviews with Korean OMI underwriters, an in-depth interview with one underwriter, and a survey of 15 OMI insureds (company representatives). Findings - We find that Korean foreign traders rely on the old-type OMI contracts developed in 1963, whereas other industrialized countries use the newest type of OMI contract developed in 2009. With a simple loss ratio analysis during 2009-2018, we show that the behavior of insurers has little to do with rational profit maximization and is instead driven by irrational bias, as they forgo the more profitable contracts provided by the new clauses by keeping the old clauses. The consistent addiction to old types of contracts in the OMI market suggests strong SQB among Korean exporters, importers, bankers, or insurers, which we confirmed in our interviews and survey. Originality/value - This research has significant originality and academic value because it reports new findings with crucial implications for the development of efficient trade practices and policy. First, this research is based on actual statistics that have not been used in previous Korean research on OMI. Second, this research shows that all-risk OMI policies provide more value to insureds, in terms of coverage given premium, than partial coverage policies, which differs from arguments previously made in Korea. Third, this research reveals strong SQB in Korea, where foreign trade plays a pivotal role in economic growth. That bias could be attributable to uninformed traders, informed but idle insurers, or conservative bankers. Fourth, to further develop foreign trade, policy initiatives are needed to review the current practices of OMI contracts and move forward with the new contract forms. All of these findings and arguments are both new and important.

A Study on the Relationships among ICT Capability, Global Orientation and Export Marketing in Korean SMEs (중소기업의 ICT 역량, 글로벌지향성 및 수출마케팅의 관계에 대한 연구)

  • The utilization of Digital and Social Network has become be a key success factor in business operations as information technologies evolve rapidly. This study is to identity the relationship between ICT & digital utilization, global orientation and export marketing capability in Korean SMEs. Survey data of Korean SMEs in south regional areas was collected and analyzed for empirical hypothesis test by PLS structural equation method. The key findings are as follows. First, ICT utilization such as SNS, homepage and online portal does not impact directly on export marketing capability. It means that SMEs would not still take advantage of utilization of social network availability. Second, ICT utilization has positive influences on global market orientation and global network orientation. Finally, both global market orientation and network have significant impacts on export marketing capability. In conclusion, global orientation has full-mediation effects on relationship between ICT capability and global marketing. This study provides evidences and some practical insights that digital utilization have indirect influences on global marketing through global markets and network orientation in Korean SMEs. In information technology era, ICT utilization will play an important role in the process of SMEs' internationalization. Therefore, for sustainable growth and survival SMEs should manage and intensify ICT capability more strategically as one of core competence. The value of ICT capability in SMEs would be depended on their strategic perspectives toward digitalization. Additional empirical research about digital capability based on larger sample will be needed.

  • Due to high cost of domestic production structure, steep growth of China's manufacturing business, and increase in oversea's production, building integrated system to support user-centered product design system based on cognitive and affective information is required to restore development of domestic manufacturing business. This paper put purpose on analysing requirements of end users, especially on information equipment business which works as a major industry in manufacturing businesses, and planning system design direction, prior to constructing user-centered product design support system based on cognitive and affective information. Research was conducted to identify current manufacturing process, application data on manufacturing, availability of cognitive and affective information data and its method of use, and necessity of user-centered product design support system based on cognitive and affective information, by carrying out in-depth interview with 6 related manufacturing companies. Need for user's character information was deducted from the interview, especially cognitive and affective information which is demanding for small to medium manufacturing business to research on its own.

Effects of Nitrogen Fertilizers on Growth and Nutrient Uptake of Rice Plants and Chemical Properties of Paddy Soil (질소질(窒素質) 비종(肥種)이 수도(水稻)의 생육(生育)과 양분흡수(養分吸收) 및 토양(土壤)의 화학적(化學的) 성질(性質)에 미치는 영향(影響))

  • For the purpose of investigating the effect of chemical composition of nitrogenous fertilizers such as urea, ammonium sulfate, ammonium phosphate(dibasic) and aqua-ammonia on growth, yield, efficiency and chemical properties of paddy soil this study was carried out at the same nitrogen concentration through field experiment. The results obtained was summarized as follows. 1. Nitrogenous fertilizers had an effect on plant height at heading stage, plant weight at harvest and No. of spikelet per panicle of rice plants in decreasing order: Ammonium sulfate > Diammonium phosphate > Urea > Ammonia water. But they didn't effect on culm length, spike length, dry matter weight, No. of grain per spike, ripening ratio and weight of 1000 grains of rice plants. 2. The absorbed amount of nitrogen and that of sulfur and nitrogen availability were highest in ammonium sulfate application and lowest in ammonia water application and these were positively correlated with yield. 3. The influence of nitrogenous fertilizers on nitrogen and sulfur efficiency and translocation efficiency was insignificant but that of ammonium sulfate on nitrogen efficiency was higher than that of the others. 4. The effect of nitrogenous fertilizers on a basic chemical properties of paddy soil experimented such as pH, C.E.C, exchangeable cation and organic matter was insignificant. But the sulfur content of ammonium sulfate application and phosphorous content of diammonium phosphate application were higher than that of the others.

Growth and Survival of Rhizobium meliloti M14 on Korean Peat Carrier (Rhizobium meliloti M14의 니탄배양(泥炭培養)에 관(關)한 연구)

  • This experiment was carried out to study the availability of Korean peat as a main carrier material of rhizobial inoculant, using the alfalfa strain Rhizobium meliloti M 14 which was isolated in the previous report. Modification of powdered peat with calcium carbonate and other materials was studied; inoculation of the peat with culture broth, maturation of the mixture under different conditions, and survival of the strain in the peat culture was examined. The results obtained were as follows. 1. Peat produced in Pyongtak was highly acidic, pH 3.8, and addition of calcium carbonate by 14% was required for pH adjustment to 6.4. However the amount of calcium carbonate could be reduced by 4 to 8% when carbon or charcoal was mixed with the peat. 2. Viable number of the strain reached to $7-9{\times}10^9cells/g$ after 3 days, when inoculated with the culture broth of early stationary growth phase and matured in unsteriled peat of open trays; and the number in steriled peat was $1.1-6.2{\times}10^{10}cells/g$ after 5 days, when matured in closed bottles. 3. Survival of the strain was affected markedly by storage temperature, and positive effect of D-sorbitol on the viability was recognized at elevated temperatures, when added as an additional carbon source and moistening agent. Glycerol, sorbitol, or sodium lactate was utilized by the strain as a sole source of carbon, and the decimal reduction time of viable number in the peat culture was was found to be 8 to 9 weeks at $25^{\circ}C$ when these agents were added by 0.5%.
