Expression profile of defense-related genes in response to gamma radiation stress (방사선 스트레스 반응 방어 유전자의 탐색 및 발현 분석)

  • Tradescantia is a perennial plant in the family of Commelinaceae. It is known to be sensitive to radiation. In this study, Tradescantia BNL 4430 was irradiated with gamma radiation at doses of 50 to 1,000 mGy in a phytotron equipped with a $^{60}Co$ radiation source at Korea Atomic Energy Research Institute, Korea. At 13 days after irradiation, we extracted RNA from irradiated floral tissues for RNA-seq. Transcriptome assembly produced a total of 77, 326 unique transcripts. In plantlets exposed to 50, 250, 500, and 1000 mGy, the numbers of up-regulated genes with more than 2-fold of expression compared that in the control were 116, 222, 246, and 308, respectively. Most of the up-regulated genes induced by 50 mGy were heat shock proteins (HSPs) such as HSP 70, indicating that protein misfolding, aggregation, and translocation might have occurred during radiation stress. Similarly, highly up-regulated transcripts of the IQ-domain 6 were induced by 250 mGy, KAR-UP oxidoreductase 1 was induced by 500 mGy, and zinc transporter 1 precursor was induced by 1000 mGy. Reverse transcriptase (RT) PCR and quantitative real time PCR (qRT-PCR) further validated the increased mRNA expression levels of selected genes, consistent with DEG analysis results. However, 2.3 to 97- fold higher expression activities were induced by different doses of radiation based on qRT-PCR results. Results on the transcriptome of Tradescantia in response to radiation might provide unique identifiers to develop in situ monitoring kit for measuring radiation exposure around radiation facilities.

A study on the concentration of trace metal elements in whole blood of the women in reproductive ages in urban and rural area (도시 및 농촌지역 가임연령 여성들의 혈중 미량금속원소의 함량에 관한 조사)

  • To acquire the essential basic data to the establishment of control measure for the hazardous health effect that could be caused by harmful metals, the author measured the concentrations of trace metals in whole blood of women of $20{\sim}39$ years old living in urban and rural area using atomic absorption spectrophotometer. The summarized results were as follows; 1. The mean concentration of zinc in whole blood was $10.69{\pm}8.07{\mu}g/ml$ in rural area. The frequency distribution by zinc concentration level was nearly L-type and the cumulative frequency distribution was showed bimodal type in both area. 2. The mean iron concentration in whole blood was $323.09{\pm}87.15{\mu}g/ml$ and $322.07{\pm}104.74{\mu}g/ml$ in urban and rural area, respectively. The frequency distribution was similar to normal distribution type in both area, but the cumulative distribution was unimodal type in urban area and bimodal type in rural area. 3. The mean magnesium concentration was $41.08{\pm}19.58{\mu}g/ml$ and $40.28{\pm}16.82{\mu}g/ml$ in the area, respectively. The frequency distribution type had skewness to the right and the cumulative frequency distribution was unimodal type in both area. 4. The mean copper concentration was $1.417{\pm}0.761{\mu}g/ml$ and $1.375{\pm}0.743{\mu}g/ml$ in the area, respectively. The frequency distribution type had skewness to the right and the cumulative frequency distribution was bimodal type in both area. 5. The mean manganese concentration was $0.079{\pm}0.039{\mu}g/ml$ and $0.07{\pm}0.058{\mu}g/ml$ in the area, respectively. The frequency distribution type had skewness to the right in both area but slight irregular in rural area and the cumulative distribution was unimodal and bimodal type in urban and rural area, respectively. 6. The mean cadmium concentration in whole blood was $0.031{\pm}0.026{\mu}g/ml$ in urban and $0.028{\pm}0.023{\mu}g/ml$ in rural area. The frequency distribution type had skewness to the right and cumulative frequency distribution was bimodal type in both area.

Blood Lead, Manganese, Aluminium and Silicon Concentrations in Korean Adults (한국인의 연, 망간, 알루미늄 및 실리콘의 혈중 농도)

  • Objectives : This study was peformed to determine the reference values of blood lead, manganese, aluminium, and silicon in healthy adults. Methods : The subjects were 132 (67 male and 65 female), and classified to three age groups $(\leq39,\;40\sim49,\;and\;50\leq)$. Blood lead, manganese and aluminium were analyzed by atomic absorption spectrophotometer, and blood silicon was analyzed by direct current plasma optical emission spectrometer. Results : Blood lead levels(geometric mean, S.D) were (3.49, 1.70) ${\mu}g/dL$ in male and (3.04, 1.65) ${\mu}g/dL$ in female, but the difference is not significant, and there was no significant difference between age groups. Mean blood manganese level was $0.99{\pm}0.41{\mu}g/dL$, and there was no significant difference between sex or age groups. Mean blood aluminium level was $0.59{\pm}0.35{\mu}g/dL$, and there was no significant difference between sex or age groups. Mean blood silicon level was $54.41{\pm}27.64{\mu}g/dL$ in male and $43.34{\pm}23.51{\mu}g/dL$ in female, and the level in male was significantly higher than that in female (p<0.05). There was significant difference between age groups, and the oldest showed the highest level in male (p<0.05), but no significant difference between age groups in female. Conclusions : Authors hope that this study would provide basic data for determining reference values and evaluating health effects.

  • 양자점은 전자와 양공을 3차원으로 속박 시키므로 기존의 bulk나 양자우물보다 양자점을 이용한 레이저 다이오드의 경우 낮은 문턱 전류, 높은 미분이득 및 온도 안전성의 장점이 있을 거라 기대되고 있다. 그러나, 양자점은 낮은 areal coverage 때문에 높은 속박효율을 얻지 못하고 있다. 이러한 양자점의 문제점을 해결하기 위해 양자점을 양자우물 안에 성장시켜 운반자들의 포획을 향상시키는 방법들이 연구되고 있다. 양자우물 안에 양자점을 넣으면 양자우물이 운반자들의 포획을 증가 시키고, 열적 방출도 억제하여 온도 안정성이 향상 되는 것으로 알려져 있다. 광통신 대역의 1.3 ${\mu}m$ 경우, GaAs계를 이용하여 InAs 양자점을 strained InGaAs 박막을 우물층으로 한 dot-in-a-well 구조의 연구는 몇몇 보고된 바 있다. 그러나 InP계를 사용하는 1.55 ${\mu}m$ 대역에서 dot-in-a-well구조의 연구는 아직 미미하다. 본 연구에서는 유기 금속 화학 증착법(metal organic chemical vapor deposition)을 이용하여 InP 기판 위에 InAs 양자점을 자발성장법으로 성장하였으며 dot-in-a-well 구조에서 우물층으로 1.35 ${\mu}m$ 파장의 $In_{0.69}Ga_{0.31}As_{0.67}P_{0.33}$ (1.35Q)를, 장벽층으로는 1.1 ${\mu}m$ 파장의 $In_{0.85}Ga_{0.15}As_{0.32}P_{0.68}$(1.1Q)를 사용하였다. 양자우물층과 장벽층은 모두 InP 기판과 격자가 일치하는 조건으로 성장하였다. III족 원료로는 trimethylindium (TMI)와 trimethylgalium (TMGa)을 사용하였으며 V족 원료 가스로는 $PH_3$ 100%, $AsH_3$ 100%를, carrier gas로는 $H_2$를 사용하였다. InP buffer층의 성장 온도는 640$^{\circ}C$이며 양자점 성장 온도는 520$^{\circ}C$이다. 양자점 형성은 원자력간 현미경(Atomic force microscopy)를 이용하여 확인하였으며, 박막의 결정성은 쌍결정 회절분석(Double crystal x-ray deffractometry)를 이용하여 확인하였다. 확인된 성장 조건을 이용하여 양자점 시료를 성장하였으며 광여기분광법(Photoluminescence)을 이용하여 광특성을 분석하였다. Fig. 1은 dot in a barrier 와 dot-in-a-well 시료의 성장구조이다. Fig. 1(a)는 일반적인 dot-in-a-barrier 구조로 InP buffer층을 성장하고 1.1Q를 100 nm 성장한 후 양자점을 성장하였다. 그 후 1.1Q 100 nm와 InP 100 nm로 capping하였다. Fig. 1(b)는 dot-in-a-well 구조로 InP buffer층을 성장하고 1.1Q를 100 nm 성장 후 1.35Q 우물층을 4 nm 성장하였다. 그 위에 InAs 양자점을 성장하였다. 그 후에 1.35Q 우물층을 4 nm 성장하고 1.1Q 100 nm와 InP 100 nm로 capping하였다. Fig. 2는 dot-in-a-barrier 시료와 dot-in-a-well 시료의 상온 PL data이다. Dot-in-a-barrier 시료의 PL 파장은 1544 nm이며 반치폭은 79.70 meV이다. Dot-in-a-well 시료의 파장은 1546 nm이며 반치폭은 70.80 meV이다. 두 시료의 PL 파장 변화는 없으며, 반치폭은 dot-in-a-well 시료가 8.9 meV 감소하였다. Dot-in-a-well 시료의 PL peak 강도는 57% 증가하였으며 적분강도(integration intensity)는 45%가 증가하였다. PL 데이터에서 높은 에너지의 반치폭 변화는 없으며 낮은 에너지의 반치폭은 8 meV 감소하였다. 적분강도 증가에서 dot-in-a-well 구조가 dot-in-a-barrier 구조보다 전자-양공의 재결합이 증가한다는 것을 알 수 있으며, 반치폭 변화로부터 특히 높은 에너지를 갖는 작은 양자점에서의 재결합이 증가 된 것을 알 수 있다. 이는 양자우물이 장벽보다 전자-양공의 구속력을 증가시키기 때문에 양자점에 전자와 양공의 공급을 증가시키기 때문이다. 따라서 낮은 에너지를 가지는 양자점을 모두 채우고 높은 에너지를 가지는 양자점까지 채우게 되므로, 높은 에너지를 가지는 양자점에서의 전자-양공 재결합이 증가되었기 때문이다. 뿐만 아니라 파장 변화 없이 PL peak 강도와 적분강도가 증가하고 낮은 에너지 쪽의 반치폭이 감소한 것으로부터 에너지가 낮은 양자점보다는 에너지가 높은 양자점에서의 전자-양공 재결합율이 급증하였음을 알 수 있다. 우리는 이와 같은 연구에서 InP계를 이용해 1.55 ${\mu}m$에서도 dot in a well구조를 성장 하여 더 좋은 특성을 낼 수 있으며 앞으로 많은 연구가 필요할 것이라 생각한다.

Seasonal Changes of Copper and Manganese Content in the Raw Bovine Milk in Korea (국내산 원유 중의 구리 및 망간 함량의 계절적 변화)

  • This study was conducted by using the ICP-AES (Inductively Coupled Plasma - Atomic Emission Spectrometry) to investigate the seasonal changes of copper and manganese content in the raw bovine milk. From July 2003 to June 2004, the milk samples were collected from the two geographical locations, Chungcheong-do and Jeolla-do, in Korean peninsula. Copper (Cu) content was determined in the range of min. 10.10 to max. 21.00 ${\mu}g$/100 mL (13.79 ${\mu}g$/100 mL) in spring, 5.06 to 15.41 (10.17) in summer, 5.04 to 19.70 (10.70) in autumn and 6.96 to 17.90 (12.11) in winter. For manganese (Mn), 3.00 to 8.30 (4.87) in spring, 2.30 to 6.44 (3.75) in summer, 2.81 to 6.04 (3.82) in autumn and 2.25 to 9.02 (4.48) in winter. Those data have shown that levels of copper and manganese was relatively constant but higher in Spring than other seasons, suggesting that the levels were not affected by seasons and different locations in Korea.

Spectroscopic Characterization of Soil Humic Acid Fractions by Molecular Weight (토양에서 추출한 흄산의 분자량별 분류에 따른 분광학적 성질 비교)

  • The soil humic acid was subdivided into four subfractions by molecular weight (F1: >100.000 dalton; F2: >100.000 dalton; F3: >10.000 dalton; F4: >2.000 dalton) using MP-dual hollow fiber ultrafiltration system. The characterization using IR, 1H and 13C NMR spectroscopy, showed similar spectroscopic features of HA, demonstrating that the bulk properties of HA subfractions are very similar to one another. IR spectral data showed a decrease in polysaccharide contents and increase in carboxylate functionality as molecular weight become smaller.functions. The structure of (NO) can be described by two interactions (N${\cdot}{\cdot}{\cdot}$N, N${\cdot}{\cdot}{\cdot}$O). One is the ONNO structure with an (N${\cdot}{\cdot}{\cdot}$N) interaction. In this structure, acyclic cis-ONNO with $C_{2v}$-symmetry, acyclic trans-ONNO with $C_{2h}$, and cyclic ONNO with trapezoidal structure ($C_{2v}$) are optimized at the MP2 level. The other structure is the ONON structure with an (N${\cdot}{\cdot}{\cdot}$O) interaction. In the structure, acyclic cis-ONON with Cs$^{-symmetry}$ and cyclic ONON of the rectangular ($C_{2h}$), square $(D_{2h})$, rhombic $(D_{2h})$, and parallelogramic $(D_{2h})$ geometries are also optimized. It is found that acyclic cis-ONNO $(^1A_1$) is the most stable structure and cyclic ONNO ($^3A_1$) is the least stable. Acyclic trans-ONNO ($^3A_1$) with an (N${\cdot}{\cdot}{\cdot}$N) interaction, acyclic trans-ONON and bicyclic ONON $(C_{2v})$ with (N${\cdot}{\cdot}{\cdot}$O) interaction, and acyclic cis- and trans-NOON with an (O${\cdot}{\cdot}{\cdot}$O) interaction can not be optimized at the MP2 level. Particularly, acyclic trans-ONNO with $C_{2h}$-symmetry can not be optimized at the CCSD(T) level. Meanwhile, acyclic NNOO ($^1A_1$, $C_s)$ and trianglic NNOO ($^1A_1$,$C_{2v})$ formed by the (O${\cdot}{\cdot}{\cdot}$N) interaction between $O_2$and $N_2$are optimized at the MP2 level. The binding energies and the relative energy gaps among the isomers are found to be relatively small./sec. Spiral CT scans during the arterial phase were obtained 35 seconds after the injection of contrast medium. CT findings of 78 lesions less than 4cm in diameter were correlated with angiographic findings. Results : The attenuation of lesions was high(n = 69), iso(n = 5), and low(n = 4) compared with liver parenchyma during the arterial phase of spiral CT. In lesions with high-, iso-, and low-attenuation during the arterial phase of spiral CT, hypervascularity on angiograms was found in 63 of 69(91.3%), three of five(60%), and three of four lesions(75%), respectively. Six lesions with high-attenuation on the arterial phase of spiral CT were not seen on angiography. Two iso-attenuated and one low-attenuated lesion were hypovascular on angiograms. Conclusion : The results of this study suggest that with some exceptions there was good correlation between the arterial phase of spiral CT and angiography.

  • To elucidate the effect of Amo-1618(4-hydroxyl-5-isopropyl-2-methlphenyl trimethyl ammonium chloride, 1-piperidine carboxylate) known as a kind of growth retardant, on the growth, grain yield, increasing the efficiency of nitrogen fertilizer, behavior of mineral nutritions and the rate of phosphorus utilization, this experiment was conducted pot culture method in a vinyl house. Two nitrogen level, namely, practical nitrogen level(1 N) and three times nitrogen level(3 N) was made and labelled double-calcium-superphosphate $Ca(H_2P^{32}O_4)_2.\;2H_2O)$ as a source of radioactive phosphorus(P-32) was employed $80\;{\mu}c/pot$, respectively. Rice seedlings, variety 'Suwon No. 82', was transplanted to a 1/50,000 a china pot on June 13 in 1968. For treatment, at early stage of tillering, 10,000 ppm solution of Amo-1618 was foliar sprayed only one time. The Duncan's new mutiple-range test was adopted for statistical analysis evaluating experimental data at 5% level significance. The results obtained may be summarized as follows; 1) No significant difference was found among the treatments in plant height, but in plot of Amo-1618 treatment and 3 N level, number of tillers was significantly increased than that of others. 2) Weight of 10,000 kernels and seed-setting rate was also remarkably increased in same treatment above. 3) Grain yield per pot was significantly increased in Amo-1618 and 3 N level application. This results seemed to be due to the increased the factors on the yield. 4) Contents of nitrogen and phosphorus per cent in the grain was likewise increased in Amo-1618 and 3 N application. There is, however, no difference among treatments in the content of nitrogen and phosphorus in the leaves and culms of rice plants. 5) On the other hand, the contents of potassium and magnesium, no distinctly tendeny showed among treatments. 6) The rate of phosphorus utilization was significantly increased in the plot of Amo-1618 and 3 N application.

Coupled Hydro-Mechanical Modelling of Fault Reactivation Induced by Water Injection: DECOVALEX-2019 TASK B (Benchmark Model Test) (유체 주입에 의한 단층 재활성 해석기법 개발: 국제공동연구 DECOVALEX-2019 Task B(Benchmark Model Test))

  • This study presents the research results of the BMT(Benchmark Model Test) simulations of the DECOVALEX-2019 project Task B. Task B named 'Fault slip modelling' is aiming at developing a numerical method to predict fault reactivation and the coupled hydro-mechanical behavior of fault. BMT scenario simulations of Task B were conducted to improve each numerical model of participating group by demonstrating the feasibility of reproducing the fault behavior induced by water injection. The BMT simulations consist of seven different conditions depending on injection pressure, fault properties and the hydro-mechanical coupling relations. TOUGH-FLAC simulator was used to reproduce the coupled hydro-mechanical process of fault slip. A coupling module to update the changes in hydrological properties and geometric features of the numerical mesh in the present study. We made modifications to the numerical model developed in Task B Step 1 to consider the changes in compressibility, Permeability and geometric features with hydraulic aperture of fault due to mechanical deformation. The effects of the storativity and transmissivity of the fault on the hydro-mechanical behavior such as the pressure distribution, injection rate, displacement and stress of the fault were examined, and the results of the previous step 1 simulation were updated using the modified numerical model. The simulation results indicate that the developed model can provide a reasonable prediction of the hydro-mechanical behavior related to fault reactivation. The numerical model will be enhanced by continuing interaction and collaboration with other research teams of DECOVALEX-2019 Task B and validated using the field experiment data in a further study.

Positron Annihilation Spectroscopy of Active Galactic Nuclei

  • This paper focuses on the interpretation of radiation fluxes from active galactic nuclei. The advantage of positron annihilation spectroscopy over other methods of spectral diagnostics of active galactic nuclei (therefore AGN) is demonstrated. A relationship between regular and random components in both bolometric and spectral composition of fluxes of quanta and particles generated in AGN is found. We consider their diffuse component separately and also detect radiative feedback after the passage of high-velocity cosmic rays and hard quanta through gas-and-dust aggregates surrounding massive black holes in AGN. The motion of relativistic positrons and electrons in such complex systems produces secondary radiation throughout the whole investigated region of active galactic nuclei in form of cylinder with radius R= 400-1000 pc and height H=200-400 pc, thus causing their visible luminescence across all spectral bands. We obtain radiation and electron energy distribution functions depending on the spatial distribution of the investigated bulk of matter in AGN. Radiation luminescence of the non-central part of AGN is a response to the effects of particles and quanta falling from its center created by atoms, molecules and dust of its diffuse component. The cross-sections for the single-photon annihilation of positrons of different energies with atoms in these active galactic nuclei are determined. For the first time we use the data on the change in chemical composition due to spallation reactions induced by high-energy particles. We establish or define more accurately how the energies of the incident positron, emitted ${\gamma}-quantum$ and recoiling nucleus correlate with the atomic number and weight of the target nucleus. For light elements, we provide detailed tables of all indicated parameters. A new criterion is proposed, based on the use of the ratio of the fluxes of ${\gamma}-quanta$ formed in one- and two-photon annihilation of positrons in a diffuse medium. It is concluded that, as is the case in young supernova remnants, the two-photon annihilation tends to occur in solid-state grains as a result of active loss of kinetic energy of positrons due to ionisation down to thermal energy of free electrons. The single-photon annihilation of positrons manifests itself in the gas component of active galactic nuclei. Such annihilation occurs as interaction between positrons and K-shell electrons; hence, it is suitable for identification of the chemical state of substances comprising the gas component of the investigated media. Specific physical media producing high fluxes of positrons are discussed; it allowed a significant reduction in the number of reaction channels generating positrons. We estimate the brightness distribution in the ${\gamma}-ray$ spectra of the gas-and-dust media through which positron fluxes travel with the energy range similar to that recorded by the Payload for Antimatter Matter Exploration and Light-nuclei Astrophysics (PAMELA) research module. Based on the results of our calculations, we analyse the reasons for such a high power of positrons to penetrate through gas-and-dust aggregates. The energy loss of positrons by ionisation is compared to the production of secondary positrons by high-energy cosmic rays in order to determine the depth of their penetration into gas-and-dust aggregations clustered in active galactic nuclei. The relationship between the energy of ${\gamma}-quanta$ emitted upon the single-photon annihilation and the energy of incident electrons is established. The obtained cross sections for positron interactions with bound electrons of the diffuse component of the non-central, peripheral AGN regions allowed us to obtain new spectroscopic characteristics of the atoms involved in single-photon annihilation.

  • This study organized data from 2000 to 2014 for 20 grid-connected countries in Europe and analyzed the determinants of carbon emissions through the panel GLS method considering the problem of heteroscedasticity and autocorrelation. At the same time, the effect of introducing ETS was considered by dividing the sample period as of 2005 when the European emission trading system was introduced. Carbon emissions from individual countries were used as dependent variables, and proportion of generation by each source, power self-sufficiency ratio of neighboring countries, power production from resource-holding countries, concentration of power sources, total energy consumption per capita in the industrial sector, tax of electricity, net electricity export per capita, and size of national territory per capita. According to the estimation results, the proportion of nuclear power and renewable energy generation, concentration of power sources, and size of the national territory area per capita had a negative (-) effect on carbon emissions both before and after 2005. On the other hand, the proportion of coal power generation, the power supply and demand rate of neighboring countries, the power production of resource-holding countries, and the total energy consumption per capita in the industrial sector were found to have a positive (+) effect on carbon emissions. In addition, the proportion of gas generation had a negative (-) effect on carbon emissions, and tax of electricity were found to have a positive (+) effect. However, all of these were only significant before 2005. It was found that net electricity export per capita had a negative (-) effect on carbon emissions only after 2005. The results of this study suggest macroscopic strategies to reduce carbon emissions to green growth, suggesting mid- to long-term power mix optimization measures considering the electricity trade market and their role.
