Isolation and Properties of Cytoplasmic α-Glycerol 3-Phosphate Dehydrogenase from the Pectoral Muscle of the Fruit Bat, Eidolon helvum

  • Cytoplasmic $\alpha$-glycerol-3-phosphate dehydrogenase from fruit-bat-breast muscle was purified by ion-exchange and affinity chromatography. The specific activity of the purified enzyme was approximately 120 units/mg of protein. The apparent molecular weight of the native enzyme, as determined by gel filtration on Sephadex G-100 was $59,500{\pm}650$ daltons; its subunit size was estimated to be $35,700{\pm}140$ by SDS-polyacrylamide gel electrophoresis. The true Michaelis-Menten constants for all substrates at pH 7.5 were $3.9{\pm}0.7\;mM$, $0.65{\pm}0.05\;mM$, $0.26{\pm}0.06\;mM$, and $0.005{\pm}0.0004\;mM$ for L-glycerol-3-phosphate, $NAD^+$, DHAP, and NADH, respectively. The true Michaelis-Menten constants at pH 10.0 were $2.30{\pm}0.21\;mM$ and $0.20{\pm}0.01\;mM$ for L-glycerol-3-phosphate and $NAD^+$, respectively. The turnover number, $k_{cat}$, of the forward reaction was $1.9{\pm}0.2{\times}10^4\;s^{-1}$. The treatment of the enzyme with 5,5'-dithiobis-2-nitrobenzoic acid (DTNB) under denaturing conditions indicated that there were a total of eight cysteine residues, while only two of these residues were reactive towards DTNB in the native enzyme. The overall results of the in vitro experiments suggest that $\alpha$-glycerol-3-phosphate dehydrogenase of the fruit bat preferentially catalyses the reduction of dihydroxyacetone phosphate to glycerol-3-phosphate.

Echolocation Call Structure of Fourteen Bat Species in Korea

  • The echolocation calls of bats can provide useful information about species that are generally difficult to observe in the field. In many cases characteristics of call structure can be used to identify species and also to obtain information about aspects of the bat's ecology. We describe and compare the echolocation call structure of 14 of the 21 bat species found in Korea, for most of which the ecology and behavior are poorly understood. In total, 1,129 pulses were analyzed from 93 echolocation call sequences of 14 species. Analyzed pulses could be classified into three types according to the pulse shape: FM/CF/FM type, FM type and FM/QCF type. Pulse structures of all species were consistent with previous studies, although geographic variation may be indicated in some species. Overall classification rate provided by the canonical discriminant analysis was relatively low. Especially in the genera Myotis and Murina, there are large overlaps in spectral and temporal parameters between species. On the other hand, classification rates for the FM/QCF type species were relatively high. The results show that acoustic monitoring could be a powerful tool for assessing bat activity and distribution in Korea, at least for FM/QCF and FM/CF/FM species.

Bayesian estimation of the Korea professional baseball players' hitting ability based on the batting average (한국프로야구 선수들의 타율에 기반된 타격 능력의 베이지안 추정)

  • In baseball game, the hitting ability of batter is frequently assessed by a batting average, a run batted in, a home run, a run scored, an on-base percentage, etc. Recently, more comprehensive indicators such as OPS, ISO, SECA, TA, RC and XR are often used. But, these measures generally shows large deviations since they are calculated from the data for a certain period of time, and they are not an estimate of a population parameter, either. In this paper, we will presume the pure hitting ability of the korea professional baseball players as a parameter which is depend upon at bat. We will estimate the parameter by using the Bayesian method.

Presence of Rhodanese in the Cytosolic Fraction of the Fruit Bat (Eidolon helvum) Liver

  • Rhodanese was isolated and purified from the cytosolic fraction of liver tissue homogenate of the fruit bat, Eidolon helvum, by using ammonium sulphate precipitation and CM-Sephadex C-50 ion exchange chromatography. The specific activity was increased 130-fold with a 53% recovery. The $K_m$ values for KCN and $Na_2S_2O_3$ as substrates were $13.5{\pm}2.2\;mM$ and $19.5{\pm}0.7\;mM$, respectively. The apparent molecular weight was estimated by gel filtration on a Sephadex G-100 column to be 36,000 Da. The optimal activity was found at a high pH (pH 9.0) and the temperature optimum was $35^{\circ}C$. An Arrhenius plot of the heat stability data consisted of two linear segments with a break occurring at $35^{\circ}C$. The apparent activation energy values from these slopes were 11.5 kcal/mol and 76.6 kcal/mol. Inhibition studies on the enzyme with a number of cations showed that $Mg^{2+}$, $Mn^{2+}$, $Ca^{2+}$, and $Co^{2+}$ did not affect the activity of the enzyme, but $Hg^{2+}$ and $Ba^{2+}$ inhibited the enzyme.

  • The method for extracting the SPICE model parameter of Schottky diode under DC forward bias is proposed. A method for improving the accuracy of the SPICE model parameter at various temperatures is proposed. Three analysis steps according to the magnitude of the current is used in order to extract the parameters effectively. At each analysis step, initial parameters are calculated by using the current-voltage equations and the Levenberg-Marquardt analysis is proceeded. To verify the validity of the proposed method, the SPICE model parameters for the BAT45 and FSV1045 under DC forward bias is extracted. Schottky diode currents obtained from the proposed method shows the average relative error of 6.1% and 9% compared with the measured data for the BAT45 and FSV1045 sample at various temperatures.

A Comparative Analysis of Biomechanical Factors and Premotor Time of Body Muscles between Elite College and Amateur Baseball Players during the Baseball Batting Motion

  • Purpose: The aim of this study was to analyze biomechanical factors and PMT (premotor time) of body muscles between elite college and amateur baseball players during the baseball batting motion. Method: Kinematic and electromyographic data were obtained for 10 elite college baseball players and 10 amateur baseball players who participated in this study. All motion capture data were collected at 200 Hz using 8 VICON cameras and the PMT of muscles was recorded using a Delsys Trigno wireless system. The peak mean bat speed and the peak mean angular velocities of trunk, pelvis, and bat with PMT of 16 body muscles were computed. These kinematic and PMT data of both groups were compared by independent t-tests (p < .05). Results: The pelvis, trunk, and bat showed a sequence of angular velocity value during baseball batting. The PMTs of right tibialis anterior, left gastrocnemius, external oblique, and erector spinae were significantly different between the two groups. Conclusion: The PMT of body muscles was related to the shifting of body and rotation of the pelvis and the trunk segment, and this action can be considered the coordinated muscle activity of the lower and upper body.

Elemental analysis of the liver, kidney, and intestine tissues from a Hodgson's bat (Myotis formosus tsuensis)

  • Hodgson's bats are critically endangered in South Korea. This study analyzed the concentrations of elements in liver, kidney, and intestine tissues from a Hodgson's bat found dead in the wild. The concentrations of essential elements followed the order Fe > Zn > Cu > Mn > Se in the three tissues. Hg was detected at the highest concentrations among the non-essential elements analyzed in the liver and kidney tissues, while As was the most highly concentrated non-essential element in the intestine. To the best of our knowledge, this is the first study of tissue element concentrations in Hodgson's bats.

  • Mask cells are distributed in the animal tissue, The bat subject has not been studied with the electron microscope, The material was fixed in 2.5% phosphate buffered glutaraldehyde for 24 hours at $4^{\circ}C$ and then post-fixed in phosphate buffered 1% osmium tetroxide for 2 hours at $4^{\circ}C$ and then the cleared tissues were embedded in Epon. The mast cell has numerous cytoplasmic processes projecting into the surrounding connective tissue. In general, the cytoplasmic granules showed ovoid, round, lunal or irregular shape, most of them were fine particulate substructure in texture. Especially, the granules was enclosed by the lamella structure.

Preparation and electrical properties of thick PZT films deposited on alumina substrates with Ag-Pd electrodes and Pt plates by spin-on process (Ag-Pd/알루미나 및 Pt전극에 스핀온 방법으로 제조된 PZT후막의 전기적 특성)

  • The electrical properties of thick PZT films deposited on Ag-Pd/$Al_2O_3$ and Pt electrodes were carefully investigated according to the annealing methods and the sub-strates. For electrical properties measurements, silver was deposited on PZT films as top electrode. The crystallogaphic structure of the films was examined by standard X-ray diffraction method to determine which crystalline phase was present. Dielctric constant was measured at 1 kHz, 10 mV by using a HP4284A. The electrical properties of PZT films with 3 wt% PbO addition were not improved. It was also found that the Ag-Pd layer has a good possibility as electrode instead of Pt. It seems clear from the present experiments that the thick PZT films having the good electrical properties can certainly be obtained using spin on technique combined with rapid thermal annealing.

Spermiogenesis in the Korean Greater Horseshoe Bat, Rhinolophus ferrumequinum korai (한국산 관박쥐 (Rhinolophus ferrumequinum korai)에 있어서의 정자변태)

  • In order to study process of spermiogenesis of the Korean greater horseshoe bat, Rhinolophus ferrumequinum korai, the cycle of seminiferous epithelium was examined by the light and electron microscope and the following results were obtained based on the epithelial cell differentiation. 1. Spermiogenesis occurred from early July to mid-Octber, and spermatogenic activity was vigorous from mid-August to late September. Spermatocytes including spermatogonia were found to be degenerated in only July. It is deduced that the degeneration serves as the mechanism to regulate effective use of energy to prepare for mating and hibernating periods, and regulation of breeding cycle. 2. Spermiogenesis of the Korean greater horseshoe bat was divided according to differentiation of the cell structure, into Golgi, cap, acrosome, maturation and spermiation phases; Golgi, cap and spermiation phases were further divided into two steps of early and late phase respectively, and acrosome phase into three steps of early, mid and late phases, and maturation phase has only one step. Hence, the spermiogenesis consists of ten phases. The first research was done in this article on the changes of chromatin with nucleus, the time of appearance and disappearance of chromatin granules, in case of Korean greater horseshoe bat (Rhinolophus ferrumequinum korai). Chromatin granule began to be condensed in late Golgi and the condensation proceeded to form an irregular mass of a electron-dense chromatin in a form of circular cylinder in the center of nucleus at the phase of maturation. Finally, the chromatin condensation proceeded and perfect nucleus of sperm with homogeneous density was formed when the sperm was separated from Sertoli cell. Therefore, appearance and disappearance of chromatin granules occurred in the period of time between late Golgi and maturation phase, The tail of sperm began to develop in early cap phase, Numerous lipid droplets were obseved in the cytoplasm of spermatids during the maturation phase, which seemed to be used as energy source necessary to make mature sperm during spermiogenesis.
